RESUMO

Anaerobic technologies with downstream autotrophic nitrogen removal have been proposed to enhance bioenergy recovery and transform a wastewater treatment plant from an energy consumer to an energy exporter. However, approximately 20-50 % of the produced methane is dissolved in the anaerobically treated effluent and is easily stripped into the atmosphere in the downstream aerobic process, contributing to the release of greenhouse gas emissions. This study aims to develop a solution to beneficially utilize dissolved methane to support high-level nitrogen removal from anaerobically treated mainstream wastewater. A novel technology, integrating Partial Nitritation, Anammox and Methane-dependent nitrite/nitrate reduction (i.e. PNAM) was demonstrated in a membrane-aerated biofilm reactor (MABR). With the feeding of ∼50 mg NH4+-N/L and ∼20 mg/L dissolved methane at a hydraulic retention time of 15 h, around 90 % of nitrogen and ∼100 % of dissolved methane can be removed together in the MABR. Microbial community characterization revealed that ammonia-oxidizing bacteria (AOB), nitrite-oxidizing bacteria (NOB), anammox bacteria, nitrite/nitrate-dependent anaerobic methane oxidation microorganisms (n-DAMO bacteria and archaea) and aerobic methanotrophs co-existed in the established biofilm. Batch tests confirmed the active microbial pathways and showed that AOB, anammox bacteria and n-DAMO microbes were jointly responsible for the nitrogen removal, and dissolved methane was mainly removed by the n-DAMO process, with aerobic methane oxidation making a minor contribution. In addition, the established system was robust against dynamic changes in influent composition. The study provides a promising technology for the simultaneous removal of dissolved methane and nitrogen from domestic wastewater, which can support the transformation of wastewater treatment from an energy- and carbon-intensive process, to one that is energy- and carbon-neutral.

Anaerobic treatment of domestic wastewater has the advantages of lower biomass yield, lower energy demand and higher energy recover over the conventional aerobic treatment process. However, the anaerobic process has the inherent issues of excessive phosphate and sulfide in effluent and superfluous H2S and CO2 in biogas. An electrochemical method allowing for in-situ generation of Fe2+ in the anode and hydroxide ion (OH-) and H2 in the cathode was proposed to overcome the challenges simultaneously. The effect of electrochemically generated iron (e­iron) on the performance of anaerobic wastewater treatment process was explored with four different dosages in this work. The results showed that compared to control, the experimental system displayed an increase of 13.4-28.4 % in COD removal efficiency, 12.0-21.3 % in CH4 production rate, 79.8-98.5 % in dissolved sulfide reduction, 26.0-96.0 % in phosphate removal efficiency, depending on the e­iron dosage between 40 and 200 mg Fe/L. Dosing of the e­iron significantly upgraded the quality of produced biogas, showing a much lower CO2 and H2S contents in biogas in experimental reactor than that in control reactor. The results thus demonstrated that e­iron can significantly improve the performance of anaerobic wastewater treatment process, bringing multiple benefits with the increase of its dosage regarding effluent and biogas quality.

Tetramethylammonium hydroxide (TMAH) is a known toxic chemical used in the photolithography process of semiconductor photoelectronic processes. Significant amounts of wastewater containing TMAH are discharged from electronic industries. It is therefore attractive to apply anaerobic treatment to industrial wastewater containing TMAH. In this study, a novel TMAH-degrading methanogenic archaeon was isolated from the granular sludge of a psychrophilic upflow anaerobic sludge blanket (UASB) reactor treating synthetic wastewater containing TMAH. Although the isolate (strain NY-STAYD) was phylogenetically related to Methanomethylovorans uponensis, it was the only isolated Methanomethylovorans strain capable of TMAH degradation. Strain NY-STAYD was capable of degrading methylamine compounds, similar to the previously isolated Methanomethylovorans spp. While the strain was able to grow at temperatures ranging from 15 to 37°C, the cell yield was higher at lower temperatures. The distribution of archaeal cells affiliated with the genus Methanomethylovorans in the original granular sludge was investigated by fluorescence in situ hybridization (FISH) using specific oligonucleotide probe targeting 16S rRNA. The results demonstrated that the TMAH-degrading cells associated with the genus Methanomethylovorans were not intermingled with other microorganisms but rather isolated on the granule's surface as a lone dominant archaeon. KEY POINTS: • A TMAH-degrading methanogenic Methanomethylovorans strain was isolated • This strain was the only known Methanomethylovorans isolate that can degrade TMAH • The highest cell yield of the isolate was obtained at psychrophilic conditions.

Purple bacteria (PPB), anoxygenic photoorganoheterotrophic organisms with a hyper-versatile metabolism and high biomass yields over substrate, are promising candidates for the recovery of nutrient resources from wastewater. Infrared light is a pivotal parameter to control and design PPB-based resource recovery. However, the effects of light intensities on the physiology and selection of PPB in mixed cultures have not been studied to date. Here, we examined the effect of infrared irradiance on PPB physiology, enrichment, and growth over a large range of irradiance (0 to 350 W m-2) in an anaerobic mixed-culture sequencing batch photobioreactor. We developed an empirical mathematical model that suggests higher PPB growth rates as response to higher irradiance. Moreover, PPB adapted to light intensity by modulating the abundances of their phototrophic complexes. The obtained results provide an in-depth phylogenetic and metabolic insight the impact of irradiance on PPB. Our findings deliver the fundamental information for guiding the design of light-driven, anaerobic mixed-culture PPB processes for wastewater treatment and bioproduct valorization.

Although iron salts such as iron(III) chloride (FeCl3) have widespread application in wastewater treatment, safety concerns limit their use, due to the corrosive nature of concentrated solutions. This study demonstrates that local, electrochemical generation of iron is a viable alternative to the use of iron salts. Three laboratory systems with anaerobic membrane processes were set up to treat real wastewater; two systems used the production of either in-situ or ex-situ electrochemical iron (as Fe2+ and Fe2+(Fe3+)2O4, respectively), while the other system served as a control. These systems were operated for over one year to assess the impact of electrochemically produced iron on system performance. The results showed that dosing of electrochemical iron significantly reduced sulfide concentration in effluent and hydrogen sulfide content in biogas, and mitigated organics-based membrane fouling, all of which are critical issues inherently related to sustainability of anaerobic wastewater treatment. The electrochemical iron strategy can generate multiple benefits for wastewater management including increased removal efficiencies for total and volatile suspended solids, chemical oxygen demand and phosphorus. The rate of methane production also increased with electrochemically produced iron. Economic analysis revealed the viability of electrochemical iron with total cost reduced by one quarter to a third compared with using FeCl3. These benefits indicate that electrochemical iron dosing can greatly enhance the overall operation and performance of anaerobic membrane processes, and this particularly facilitates wastewater management in a decentralized scenario.

A process for the valorization of seagrass residues has been developed, aiming to investigate its potential as a phosphate adsorbent and the production of added-value products, which can be assessed in agricultural applications. Posidonia oceanica residues were thermally treated (500 °C, 1 h) and were tested as an adsorbent of phosphate from real wastewater. Chemical leaching experiments of phosphate from dewatered anaerobic sludge (DWAS) were conducted, evaluating sonication and inorganic acids (sulfuric acid (SA), thermal-sulfuric acid (TSA), and nitric acid (NA)) as extraction methods. Τhe extraction efficiency of each method tested demonstrated that the most suitable acid medium to leach out phosphate were SA and TSA processes with 84.9 and 93.2% removal efficiency, respectively. Moreover, the saturation capacity of thermally treated P. oceanica was assessed, and the results confirmed the high phosphate adsorption capacity (179.1 mg g-1). Adsorption batch experiments with real wastes (anaerobic effluent wastewater and leached solution from DWAS), demonstrated that thermally treated seagrass could have a high adsorption efficiency and selectivity towards phosphate. After phosphate adsorption, the solid residues were mixed with compost in different ratio and then tested as fertilizer substitutes on plant growth. The solid residue after adsorption produced from anaerobic effluent or synthetic solutions imposed a positive effect on plant growth with germination index (GI) values 96.7-111.14%, for all types of seeds tested (Solanum Lycopersicum, Lepidium sativum, and Sinapis alba), while the solid residue after adsorption produced from DWAS leached solution negatively affected the germination of plants, probably due to potentially refractory compounds contained in DWAS.

Maximum specific growth rate (µmax) and substrate saturation constant (Ks) are widely used in determining the growth of microorganisms. The ratio (µmax/Ks), also referred to as specific affinity, aA0, is a better parameter to assess the advantage in competition for substrates by bridging microbial growth and the kinetics of enzymatic substrate uptake, but is not well studied. This study investigated the effect of hydraulic retention time (HRT) on the aA0 of anaerobic sludge from an anaerobic membrane bioreactor (AnMBR), associated microbial communities and the overall wastewater treatment performance. The AnMBR was fed with acetate wastewater (~500 mg COD/L) and operated at fixed solids retention time (45 d) while HRT continued to decrease. There was no significant difference in Ks (ranging from 170 to 243 mg COD/L) at different HRTs. However, aA0 increased from (4.0 ± 0.2) × 10-4 to (4.9 ± 0.2) × 10-4 and to (6.5 ± 0.1) × 10-4 L/mg COD/d as HRT decreased from 24 h to 12 h and further to 6 h, respectively. This was accompanied by the increase in acetoclastic methanogens (mainly Methanosaeta) from 3.85 × 1010, 8.82 × 1010 to 1.05 × 1011 cells/L, respectively. The fraction of Methanosaeta in the anaerobic biomass increased from 33.67% to 61.08% as HRT decreased from 24 h to 6 h. Correspondingly, effluent quality was improved, as evidenced from the COD concentrations of 32 ± 6, 21 ± 4, and 13 ± 5 mg/L at the HRTs of 24 h, 12 h, and 6 h, respectively. The results confirm that microorganisms are able to adapt to growth conditions by adjusting their kinetic properties and suggest that short HRTs in the AnMBR favor the growth and accumulation of Methanosaeta with high specific affinity likely because they can compete for acetate at low concentrations by increasing substrate uptake rate and thus specific microbial growth rate.

Interspecies electron transfer plays an important role in syntrophic methanogenesis. Direct interspecies electron transfer (DIET) between syntrophic oxidizers and methanogens via conductive pili has been only confirmed in some specific co-cultures. This study examined potential syntrophic cooperation via type IV pili and quorum sensing between widespread syntrophic bacteria and methanogens through a metagenomic analysis of 12 anaerobic sludge samples. We found that Methanosaeta and Methanosarcina, which are reported to have DIET ability, were dominant in most methanogenic samples. Putative conductive pili genes were found in some typical syntrophic bacteria, which has rarely been reported previously. The existence of diverse quorum-sensing genes suggested that various quorum-sensing systems might participate in the communication of anaerobic microorganisms. Specifically, the diffusible signal factor and 3'-5' cyclic diguanosine monophosphate related genes were mainly assigned to syntrophic bacteria. These results suggest that the combined regulation of these signals might be responsible for the biosynthesis of type IV pili and affect syntrophic interaction during methanogenesis. These novel results provide fresh evidence to support the widespread existence of DIET in anaerobic methanogenic systems; therefore, regulating the quorum-sensing system may promote syntrophic interaction.

The H2-assisted biogas upgrading approach has recently attracted much interest as a low-cost and environmentally friendly alternative to commonly used ex-situ/ physiochemical biogas upgrading techniques. However, most studies conducted to date have been limited to anaerobic solid-waste treatment characterized by flocculant sludge and low organic loading rates (OLR). In an attempt to expand its application to high-rate anaerobic wastewater treatment, an innovative two-stage up-flow anaerobic sludge blanket reactor system was employed using anaerobic granular sludge. We found that the CH4 content of product gas was consistently >90% and that H2 and CO2 concentrations stayed below 5%, even when OLR was increased from 1 to 5 g L-1 d-1 and H2 feeding rates were increased from 0.13 to 0.63 g L-1 d-1. We were also able to show that CO (5-10%) in H2-rich syngas didn't inhibit methanogenesis or had significant impact on microbial community structure, suggesting that H2-assisted biogas upgrading with H2-rich syngas is feasible.

Kinetic parameters affecting effluent water quality including half saturation constant (Ks), maximum specific growth rate (µmax), and specific affinity ([Formula: see text], defined as µmax/Ks) were investigated using three types of anaerobic sludge (raw anaerobic digestion sludge referred to as unacclimated sludge, unacclimated sludge after endogenous decay, and sludge acclimated to low-strength wastewater in an anaerobic membrane bioreactor (AnMBR) for 360 days). Long-term acclimation to low-strength wastewater resulted in sludge with high specific affinity (1.6 × 10-3 L/mg COD/day for acclimated sludge compared to 4.1 × 10-4 L/mg COD/day for unacclimated sludge). The µmax values for unacclimated sludge and acclimated sludge were 0.08 and 0.07 day-1, respectively. The Ks values for unacclimated sludge and acclimated sludge were 194 ± 81 mg COD/L and 45 ± 13 mg COD/L, respectively. Although the Ks of unacclimated sludge after endogenous decay increased to 772 ± 74 mg COD/L, µmax increased to 0.35 day-1 as well, resulting in no statistically significant difference of [Formula: see text] between the two types of unacclimated sludge. Overall, [Formula: see text] is a better indicator than µmax or Ks alone for determining effluent water quality, as effluent substrate concentration is approximately inversely proportional to the specific affinity. 16S rRNA sequencing data analysis indicated a high abundance (85.8% of total archaea) of Methanosaeta in the microbial community after long-term acclimation. High [Formula: see text] associated with the enrichment of Methanosaeta appears to ensure successful anaerobic treatment of low-strength wastewater.

In this study, the suitability of an anaerobic biofilter (AnBF) as an efficient and low-cost wastewater treatment for safer irrigation water production for Sub-Saharan Africa was investigated. To determine the influence of different ubiquitous available materials on the treatment efficiency of the AnBF, rice husks and their pyrolysed equivalent, rice husk biochar, were used as filtration media and compared with sand as a common reference material. Raw sewage from a municipal full-scale wastewater treatment plant pretreated with an anaerobic filter (AF) was used in this experiment. The filters were operated at 22 °C room temperature with a hydraulic loading rate of 0.05 m·h-1 for 400 days. The mean organic loading rate (OLR) of the AF was 194 ±â€¯74 and 63 ±â€¯16 gCOD·m-3·d-1 for the AnBF. Fecal indicator bacteria (FIB) (up to 3.9 log10-units), bacteriophages (up to 2.7 log10-units), chemical oxygen demand (COD) (up to 94%) and turbidity (up to 97%) could be significantly reduced. Additionally, the essential plant nutrients nitrogen and phosphorous were not significantly affected by the water treatment. Overall, the performance of the biochar filters was significantly better than or equal to the sand and rice husk filters. By using the treated wastewater for irrigating lettuce plants in a pot experiment, the contamination with FIB was >2.5 log-units lower (for most of the plants below the detection limit of 5.6 MPN per gram fresh weight) than for plants irrigated with raw wastewater. Respective soil samples were minimally contaminated and nearly in the same range as that of tap water.

Conventional completely mixed anaerobic treatment systems limit the chances of the different species of bacteria to spatially group together according to their mutual cooperation and as a result, show a lower efficiency and vulnerability towards shock situations. It is interesting to know about the stratification of the different bacterial species participating in the degradation process and the intermediates that they produce. In this study, we established and optimized a two-phase anaerobic packed bed biofilm reactor system (AnPBR) with porous PVA gel beads used as bio-carriers and ran the reactor system in a steady state to observe the VFAs produced along with the microbial diversity of the predominant species at different stages of the reactor system. We observed that acetate and butyrate were the predominant intermediate VFAs while concentrations of other VFAs such that propionic acid were low. Acetobacterium and Clostridium were found to be the most abundant bacterial species in acidogenic reactor while methanogenic reactor was highly enriched with Methanobacterium and Methanosarcina. Apart from the above, syntrophic populations such as Syntrophobactor wolinii were also observed to be dominant in both the reactors - especially towards the end of acidogenic reactor and the initial part of the methanogenic reactor.

Since anaerobic wastewater treatment is a nonlinear and complex biochemical process, reasonable monitoring and control are needed to keep it operating stably and efficiently. In this paper, a least-square support-vector machine (LS-SVM) was employed to construct models for the prediction of effluent chemical oxygen demand (COD) in an anaerobic wastewater treatment system. The result revealed that the performance of the steady-state model based on LS-SVM for predicting effluent COD was acceptable, with the maximum relative error (RE) of 11.45%, the mean average percentage error (MAPE) of 0.79% and the root mean square error (RMSE) of 3.08 when training, and the performance fell slightly when testing. Even though, the correlation coefficient value (R) between the predicted value and the actual value of 0.9752 could be achieved, which means this model can predict the variation of effluent COD in general. The dynamic-state models under three kinds of shock loads, which were concentration, hydraulic, and bicarbonate buffer absent, showed good forecasting performance, the correlation coefficient values (R) all excelled 0.99. Among these three shocks, the dynamic LS-SVM model under bicarbonate buffer absent shock achieved the optimal performance and followed by the dynamic-state model under hydraulic shock. This paper provides a meaningful reference to improve the monitoring level of the anaerobic wastewater treatment process.

Anaerobic wastewater potentially was an ideal medium for cultivating microalgae. The coupled effect of ammonium, temperature and pH on lipids accumulation was a core issue during algal culture using anaerobic wastewater. Therefore, their combined effects on Chlorella pyrenoidosa culture and lipids accumulation in anaerobic effluent were investigated. Free ammonia induced from the rising pH and temperature inhibited algal growth, but significantly promoted lipid accumulation. The highest lipids content reached 30.2% when pH rose to 8.3-8.5 (25 °C, ammonium 280 mg/L), which was 1.6-fold higher than that under neutral condition. Moreover, the percentage of unsaturated fatty acids (un-SFAs) increased to 74.8-77.9% at pH 8.3-8.5, whereas it was only 56.1-58.9% under neutral condition. The C18:2 and C18:3 dominated the un-SFAs increase at high pH, typically the percentage of C18:3 increased by 74.5-153.1%. This study provides a potential way for lipid accumulation in algal culture using anaerobic wastewater.

Effects of Fe(III)/sulfate (Fe/S) ratio on organic carbon oxidation kinetics and microbial ecology of a novel Fe(III)-dosed anaerobic wastewater treatment system were investigated in this study. Fixed-film batch bioreactors under three Fe/S molar ratios (1, 2, and 3) yielded COD oxidation rates that increased with the Fe/S ratio, and estimated Michaelis-Menten model parameters Vmax ranging in 0.47-1.09 mg/L⋅min and Km in 2503-3267 mg/L. Both iron and sulfate reducing bacteria contributed to the organics oxidation, and the produced sludge materials contained both biomass (32-45 wt.%) and inorganic precipitates from biogenic ferrous iron and sulfide (68-55 wt.%). Spectroscopic and chemical elemental analyses indicated that the inorganic fraction of the sludge materials contained both FeS and FeS2, and had Fe/S stoichiometric ratios close to 1. Microbiological analyses of the biofilm samples revealed that the major putative iron- and sulfate reducers were Geobacter sp. and Desulfovibrio sp. along with noticeable N-fixing and fermentative bacteria. The COD oxidation rate had a positive correlation with the relative abundance of iron reducers, and both increased with the Fe/S ratio. This study evaluated the methanogens responsible for methanogenic degradation of tetramethylammonium hydroxide (TMAH) in a continuous flow bioreactor. The enriched methanogens attained an estimated maximum specific TMAH degradation rate and half-saturation constant of 39.5 mg TMAH/gVSS/h and 820 mg/L, following the Monod-type kinetic expression for methanogenic TMAH degradation. Presence of sulfide more than 20 mg/L significantly extended lag period and slowed down specific TMAH degradation rates. The results of terminal restriction fragment length polymorphism (T-RFLP), cloning/sequencing, and quantitative real-time PCR analyses targeting on the methyl coenzyme M reductase alpha subunit (mcrA) genes retrieved from the bioreactor and batch experiments indicated that Methanomethylovorans species were the dominant methanogens responsible for methanogenic degradation of TMAH. The isolated TMAH-degrading methanogen from the bioreactor, however, was identified closely related to Methanosarcina mazei. It is likely that a very low TMAH environment in the bioreactor favored the growth of Methanomethylovorans hollandica, while the much higher TMAH in the isolation growth medium proliferated Methanosarcina mazei.

The hydrogen sulphide (H2S) levels from wastewater treatment plants (WWTPs) in Curitiba, Brazil have been quantified for the first time. H2S generated by anaerobic decomposition of organic matter in WWTPs is a cause for concern because it is an air pollutant, which can cause eye and respiratory irritation, headaches, and nausea. Considering the requirement for WWTPs in all communities, it is necessary to assess the concentrations and effects of gases such as H2S on populations living and/or working near WWTPs. The primary objective of this study was to evaluate the indoor and outdoor concentration of H2S in the neighbourhood of two WWTPs located in Curitiba, as well as its human health impacts. Between August 2013 and March 2014 eight sampling campaigns were performed using passive samplers and the analyses were carried out by spectrophotometry, presenting mean concentrations ranging from 0.14 to 32µgm-3. Eleven points at WWTP-A reported H2S average concentrations above the WHO recommendation of 10µgm-3, and 15 points above the US EPA guideline of 2µgm-3. At WWTP-B the H2S concentration was above US EPA guideline at all the sampling points. The I/O ratio on the different sampling sites showed accumulation of indoor H2S in some instances and result in exacerbating the exposure of the residents. The highest H2S concentrations were recorded during the summer in houses located closest to the sewage treatment stations, and towards the main wind direction, showing the importance of these factors when planning a WWTP. Lifetime risk assessments of hydrogen sulphide exposure showed a significant non-carcinogenic adverse health risk for local residents and workers, especially those close to anaerobic WWTPs. The data indicated that WWTPs operated under these conditions should be recognized as a significant air pollution source, putting local populations at risk.

The aim of this study was to evaluate the feasibility of treating the kitchen food waste (FW) jointly with urban wastewater (WW) in a wastewater treatment plant (WWTP) by anaerobic membrane technology (AnMBR). The experience was carried out in six different periods in an AnMBR pilot-plant for a total of 536days, varying the SRT, HRT and the food waste penetration factor (PF) of food waste disposers. The results showed increased methane production of up to 190% at 70days SRT, 24h HRT and 80% PF, compared with WW treatment only. FW COD and biodegradability were higher than in WW, so that the incorporation of FW into the treatment increases the organic load and the methane production and reduces sludge production (0.142 vs 0.614kgVSSkgremovedCOD-1, at 70days SRT, 24h HRT and 80% PF, as compared to WW treatment only).

This paper describes a new option for integrated recovery and esterification of carboxylates produced by anaerobic digestion at a pH above the pKa. The carboxylates (acetate, propionate, butyrate, valerate and lactate) are recovered using a strong anion exchange resin in the bicarbonate form, and the resin is regenerated using a CO2-expanded alcohol technique, which allows for low chemicals consumption and direct esterification. Paper mill wastewater was used to study the effect of pH and the presence of other inorganic anions and cations on the adsorption and desorption with CO2-expanded methanol. Calcium, which is present in paper mill wastewater, can cause precipitation problems, especially at high pH. Esters yields ranged from 1.08±0.04mol methyl acetate/mol of acetatein to 0.57±0.02mol methyl valerate/mol of valeratein.

This study investigated the recovery of H2 and CH4 from bagasse bioethanol fermentation residues (bagasse BEFR) using a two-stage bioprocess. In the hydrogen fermentation bioreactor (HFB), carbohydrate removal efficiency was maintained at 82-93% and the highest hydrogen yield was 8.24mL/gCOD at volumetric loading rate (VLR) of 80kgCOD/m(3)/day. The results indicated a positive correlation between hydrogen yield and butyrate-to-acetate ratio, which might be due to the mechanisms of lactate/acetate utilization for hydrogen production and acetogenesis occurring in the HFB. Remaining volatile fatty acids and alcohols in the HFB effluent were further utilized for methane production in methane fermentation bioreactor (MFB), in which the highest methane yield of 345.2mL/gCOD was attained at VLR of 2.5kgCOD/m(3)/day. Overall, the two-stage bioprocess achieved a maximum COD removal of 81% from bagasse BEFR, and converted 0.3% and 72.8% of COD in the forms of H2 and CH4, respectively.
