Wild waterfowl serve as a reservoir of some astroviruses. Fecal samples from wild waterfowl collected at Hong Kong's Marshes were tested using pan-astrovirus reverse transcription-PCR. Positive samples underwent subsequent host identification using DNA barcoding. Based on deduced partial sequences, noteworthy samples from three astrovirus groups (mammalian, avian and unclassified astroviruses) were further analyzed by next-generation sequencing. One sample of Avastrovirus 4 clade, MP22-196, had a nearly complete genome identified. The results of ORF2 phylogenetic analysis and genetic distance analysis indicate that Avastrovirus 4 is classified as a distinct subclade within Avastrovirus. MP22-196 has typical astrovirus genome characteristics. The unique characteristics and potential differences of this genome, compared to other avian astrovirus sequences, involve the identification of a modified sgRNA sequence situated near the ORF2 start codon, which precedes the ORF1b stop codon. Additionally, the 3' UTR of MP22-196 is shorter than other avian astroviruses. This study expands our understanding of the Avastrovirus 4 clade.

Aquatic bird bornavirus 1 (ABBV-1) is a neurotropic virus that infects waterfowls, resulting in persistent infection. Experimental infection showed that both Muscovy ducks and chickens support persistent ABBV-1 infection in the central nervous system (CNS), up to 12 weeks post-infection (wpi), without the development of clinical disease. The aim of the present study was to describe the transcriptomic profiles in the brains of experimentally infected Muscovy ducks and chickens infected with ABBV-1 at 4 and 12 wpi. Transcribed RNA was sequenced by next-generation sequencing and analyzed by principal component analysis (PCA) and differential gene expression. The functional annotation of differentially expressed genes was evaluated by gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis. The PCA showed that the infected ducks sampled at both 4 and 12 wpi clustered separately from the controls, while only the samples from the chickens at 12 wpi, but not at 4 wpi, formed a separate cluster. In the ducks, more genes were differentially expressed at 4 wpi than 12 wpi, and the majority of the highly differentially expressed genes (DEG) were upregulated. On the other hand, the infected chickens had fewer DEGs at 4 wpi than at 12 wpi, and the majority of those with high numbers of DEGs were downregulated at 4 wpi and upregulated at 12 wpi. The functional annotation showed that the most enriched GO terms were immune-associated in both species; however, the terms associated with the innate immune response were predominantly enriched in the ducks, whereas the chickens had enrichment of both the innate and adaptive immune response. Immune-associated pathways were also enriched according to the KEGG pathway analysis in both species. Overall, the transcriptomic analysis of the duck and chicken brains showed that the main biological responses to ABBV-1 infection were immune-associated and corresponded with the levels of inflammation in the CNS.

Vibrio parahaemolyticus is a Gram-negative, foodborne pathogenic bacterium that causes human gastroenteritis. This organism is ubiquitously present in the marine environment. Detection of V. parahaemolyticus in aquatic birds has been previously reported; however, the characterization of isolates of this bacterium recovered from these birds remains limited. The present study isolated and characterized V. parahaemolyticus from aquatic bird feces at the Bangpu Recreation Center (Samut Prakan province, Thailand) from 2016 to 2017, using multilocus sequence typing (MLST) and genome analysis. The results showed that V. parahaemolyticus was present in 34.9% (76/218) of the collected bird fecal samples. Among the ldh-positive V. parahaemolyticus isolates (n = 308), 1% (3/308) were positive for tdh, 1.3% (4/308) were positive for trh, and 0.3% (1/308) were positive for both tdh and trh. In turn, the MLST analysis revealed that 49 selected V. parahaemolyticus isolates resolved to 36 STs, 26 of which were novel (72.2%). Moreover, a total of 10 identified STs were identical to globally reported pathogenic strains (ST1309, ST1919, ST491, ST799, and ST2516) and environmental strains (ST1879, ST985, ST288, ST1925, and ST260). The genome analysis of isolates possessing tdh and/or trh (ST985, ST1923, ST1924, ST1929 and ST2516) demonstrated that the organization of the T3SS2α and T3SS2ß genes in bird fecal isolates were almost identical to those of human clinical strains posing public health concerns of pathogen dissemination in the recreational area. The results of this study suggest that aquatic birds are natural reservoirs of new strains with high genetic diversity and are alternative sources of potentially pathogenic V. parahaemolyticus in the marine environment. IMPORTANCE To our knowledge, infection of foodborne bacterium V. parahamolyticus occurs via the consumption of undercooked seafood contaminated with pathogenic strains. Aquatic bird is a neglectable source that can transmit V. parahaemolyticus along coastal areas. This study reported the detection of potentially pathogenic V. parahamolyticus harboring virulence genes from aquatic bird feces at the recreational center situated near the Gulf of Thailand. These strains shared identical genetic profile to the clinical isolates that previously reported in many countries. Furthermore, the strains from aquatic birds showed extremely high genetic diversity. Our research pointed out that the aquatic bird is possibly involved in the evolution of novel strains of V. parahaemolyticus and play a role in dissimilation of the potentially pathogenic strains across geographical distance.

RESUMO

Radiocesium concentrations were measured in the pectoralis muscles of great cormorant (n = 36) collected from Gunma Prefecture between November 2011 and July 2012. The 137Cs contamination levels in great cormorant were consistent with the 137Cs deposition levels on ground soil and 137Cs concentrations in freshwater fishes. The Tag of great cormorants were comparable with those of other birds except for copper pheasant.

Bornaviruses are a diverse family of viruses infecting various hosts, including birds. Aquatic bird bornavirus 1 (ABBV-1) and aquatic bird bornavirus 2 (ABBV-2) have been found in wild waterfowl but data on their prevalence are scarce. To gain knowledge on the occurrence of ABBVs in Poland, samples originating from dead birds of the Anseriformes order collected in 2016-2021 were tested with a real time RT-PCR method targeting the ABBVs genome. A total of 514 birds were examined, including 401 swans, 96 ducks and 17 geese. The presence of ABBV-1 RNA was detected in 52 swans (10.1% of all tested birds) from 40 different locations. No positive results were obtained for ducks and geese. Sequences of about 2300 bases were generated for 18 viruses and phylogenetic analysis was performed. A relatively low genetic diversity of the examined ABBV-1 strains was observed as all were gathered in a single cluster in the phylogenetic tree and the minimum nucleotide identity was 99.14%. The Polish strains were closely related to ABBV-1 identified previously in Denmark and Germany, but a limited number of sequences from Europe hinders the drawing of conclusions about interconnections between Polish and other European ABBVs. The results of the present study provide new insights into the distribution and genetic characteristics of ABBVs in wild birds in Europe.

Due to their unique breeding pattern, aquatic bird farms are increasingly considered as hotspots in the development and spread of antimicrobial resistance. However, comprehensive studies addressing the whole-genomic features of colistin-resistant bacteria in aquatic bird farms are scarce. Over a 2-year period, we conducted surveillance to determine the whole-genome epidemiology and characterisation of mcr-1-positive Escherichia coli in aquatic bird farms in southeastern coastal China. A total of 100 mcr-1-producing isolates among 654 E. coli strains were recovered from 781 samples collected in 11 aquatic bird farms and 1 veterinary clinic in the Pearl River Delta area. Higher resistance phenotypes to 17 antibiotics were found in mcr-1-positive isolates compared with other isolates. Subsequently, 20 mcr-1-carrying isolates were sequenced to analyse the whole-genomic features. Molecular typing as well as antimicrobial resistance gene and virulence factor profiles of the isolates showed considerable diversity. Three types of genetic backbones of mcr-1 in the isolates were assembled and were identified in diverse broad-host-range plasmids and bacterial species. Pangenome analyses revealed a large genetic pool composed of the isolates. Furthermore, phylogenetic trees both of the isolates in this study and a global data set were built, indicating the spread of the three mcr-1 backbones and the mcr-1-positive isolates among different habitats, farms and even countries. This study highlights that aquatic bird farms may act as an important reservoir for mcr-1-producing E. coli, from which colistin resistance may be spread to diverse habitats, different geographical locations and even across bacterial species.

String-pulling is among the most widespread cognitive tasks used to test problem-solving skills in mammals and birds. The task requires animals to comprehend that pulling on a non-valuable string moves an otherwise inaccessible food reward to within their reach. Although at least 90 avian species have been administered the string-pull test, all but five of them were perching birds (passeriformes) or parrots (psittaciformes). Waterbirds (Aequorlitornithes) are poorly represented in the cognitive literature, yet are known to engage in complex foraging behaviours. In this study, we tested whether free-living ring-billed gulls (Larus delawarensis), a species known for their behavioural flexibility and foraging innovativeness, could solve a horizontal string-pull test. Here, we show that 25% (26/104) of the ring-billed gulls that attempted to solve the test at least once over a maximum of three trials were successful, and that 21% of them (22/104) succeeded during their first attempt. Ring-billed gulls are thus the first waterbird known to solve a horizontal single-string-rewarded string-pull test. Since innovation rate and problem-solving are associated with species' ability to endure environmental alterations, we suggest that testing the problem-solving skills of other species facing environmental challenges will inform us of their vulnerability in a rapidly changing world.

The increasing involvement of wild waterfowl in H5 Highly Pathogenic Avian Influenza Virus (HPAIV) circulation continues to pose a threat to animal and public health worldwide. In winter 2020-2021, two field surveillance activities were carried out on a weekly basis, through virological and serological analyses, in 823 hunted and 521 trapped migratory aquatic birds in northeast Italy. Sixty Eurasian teals were recaptured several times, which allowed us to follow the progression of the HPAI H5 infection in naturally infected wild waterfowl. Oropharyngeal, cloacal, and feather swabs (OS, CS and FS) were collected from each duck and tested by real time rRT-PCR Type A influenza. The identified viruses were characterized and pathotyped by sequencing. Several viruses belonging to three different HPAI H5 subtypes were detected: H5N8, H5N5, and H5N1. High prevalence of infection with HPAI H5 clade 2.3.4.4b during November-December 2020 (up to 27.1%) was observed in captured Eurasian teals, while infection rates in hunted dabbling ducks, mainly Eurasian wigeons, showed the highest prevalence of infection in November 2020 (8.9%) and January 2021 (10.2%). All HPAI positive birds were also clinically healthy when recaptured weeks apart. The OS and FS showed the highest detection efficiency of HPAIV. Our results highlight that HPAI passive surveillance should be complemented by a targeted active surveillance to more efficiently detect novel HPAI viruses.

BACKGROUND: Aquatic bird bornavirus 1 (ABBV-1) has been associated with neurological diseases in wild waterfowls. In Canada, presence of ABBV-1 was demonstrated by RT-qPCR and immunohistochemistry in tissues of waterfowls with history of neurological disease and inflammation of the central and peripheral nervous tissue, although causation has not been proven by pathogenesis experiments, yet. To date, in vitro characterization of ABBV-1 is limited to isolation in primary duck embryo fibroblasts. The objectives of this study were to describe isolation of ABBV-1 in primary duck embryonic fibroblasts (DEF), and characterize replication in DEF and three immortalized avian fibroblast cell lines (duck CCL-141, quail QT-35, chicken DF-1) in order to evaluate cellular permissivity and identify suitable cell lines for routine virus propagation. METHODS: The virus was sequenced, and phylogenetic analysis performed on a segment of the N gene coding region. Virus spread in cell cultures, viral RNA and protein production, and titres were evaluated at different passages using immunofluorescence, RT-qPCR, western blotting, and tissue culture dose 50% (TCID50) assay, respectively. RESULTS: The isolated ABBV-1 showed 97 and 99% identity to European ABBV-1 isolate AF-168 and North American ABBV-1 isolates 062-CQ and CG-N1489, and could infect and replicate in DEF, CCL-141, QT-35 and DF-1 cultures. Viral RNA was detected in all four cultures with highest levels observed in DEF and CCL-141, moderate in QT-35, and lowest in DF-1. N protein was detected in western blots from infected DEF, CCL-141 and QT-35 at moderate to high levels, but minimally in infected DF-1. Infectious titre was highest in DEF (between approximately 105 to 106 FFU / 106 cells). Regarding immortalized cell lines, CCL-141 showed the highest titre between approximately 104 to 105 FFU / 106 cells. DF-1 produced minimal infectious titre. CONCLUSIONS: This study confirms the presence of ABBV-1 among waterfowl in Canada and reported additional in vitro characterization of this virus in different avian cell lines. ABBV-1 replicated to highest titre in DEF, followed by CCL-141 and QT-35, and poorly in DF-1. Our results showed that CCL-141 can be used instead of DEF for routine ABBV-1 production, if a lower titre is an acceptable trade-off for the simplicity of using immortalized cell line over primary culture.

Babesia species are tickborne hemoprotozoans. Although experiments have shown that primaquine is highly effective in the treatment of Babesia species infections in mammals, this drug has not been widely used for the treatment of avian babesiosis. Consequently, the treatment of this disease for avian patients has traditionally relied on an empirically established imidocarb treatment. In this study, the authors examined the efficacy of primaquine as a treatment alternative for avian babesiosis (Babesia peircei and Babesia ugwidiensis) in seabirds. Retrospective analysis was performed on the medical records and blood smears of 446 B peircei-positive African penguins (Spheniscus demersus) and 41 B ugwidiensis-positive Cape cormorants (Phalacrocorax capensis) admitted for rehabilitation at the Southern African Foundation for the Conservation of Coastal Birds (SANCCOB, Cape Town, South Africa). Treatment with primaquine (1 mg/kg PO q24h for 10 days) was effective in rapidly and markedly decreasing the proportion of Babesia-positive blood smears in African penguins and Cape cormorants. No regurgitation, loss of appetite, or any other signs after administration of primaquine were observed during the study period. The use of primaquine can be a particularly advantageous treatment alternative for avian babesiosis in circumstances in which it is not possible to determine confidently whether the intraerythrocytic inclusions seen in blood smears correspond to Babesia or Plasmodium or in cases in which there is a coinfection by Babesia and Plasmodium.

The water quality of Koumoundourou Lake, a heavily modified, peri-urban, brackish and shallow lake near Athens, Greece, is under multiple stressors due to the industrial activities in the catchment area and natural pollution pressures. Although the broader area has undergone significant land use changes since the beginning of the 20th century converting it from rural to heavily industrialized, Koumoundourou Lake remained as one of the few habitats in Attica Region, for large numbers of aquatic birds. The water quality of the lake has been recently improved, which contributed to the increase of aquatic birds' population in the area and to the higher, in relation to the past, nitrogen inputs from natural sources. Therefore, a monitoring program has been implemented to assess the pollution pressure factors in the lake. A water quality model has been also used to simulate the lake's processes and estimate the nutrient mass budget focusing on the various nitrogen loading mechanisms (natural and anthropogenic). Based on the model output, the main polluting factor of the lake is the groundwater inflows. Aquatic birds affect slightly the lake's water quality, while the dry and wet atmospheric deposition contributes insignificantly to the total nitrogen loading.

During the winter of 2013-14, 22 Canada geese ( Branta canadensis ) were admitted to the Wildlife Clinic at the Cummings School of Veterinary Medicine at Tufts University with nonspecific neurologic abnormalities and emaciation. Five of these geese, along with three geese that were submitted dead, were evaluated via histopathology, immunohistochemistry, and reverse transcription PCR (RT-PCR) for bornaviruses. Histopathologically, six of the eight birds had lymphoplasmacytic encephalitis. One bird, which also had encephalitis, had a dilated esophagus. Lead poisoning, West Nile virus, avian influenza, and avian paramyxovirus infection were excluded from the diagnosis. Brain tissue from all eight geese was positive for bornaviral N-antigen on immunohistochemistry. Frozen brain tissue from five birds was available for bornavirus RT-PCR. Three of the five birds were positive for the bornavirus M gene. Formalin-fixed paraffin-embedded brain tissue was evaluated on the remaining three geese via RT-PCR, with one of these geese testing positive. A bornavirus was subsequently cultured in duck embryo fibroblasts from the brain of one Canada Goose. This virus genome was sequenced, and the virus was identified as aquatic bird bornavirus 1. We were unable to identify any unusual features of this genome that would account for its apparent pathogenicity, given that subclinical infection with bornavirus in waterfowl is common in North America.

Abstract Species of Hystrichis are parasite nematodes of the digestive tract of aquatic birds in South America, Europe and Asia. In Brazil, Hystrichis acanthocephalicus has been reported in Phimosus infuscatus. There are few data on the morphometry of this species and there are no reports on pathological conditions that it causes. Therefore, the purpose of this study was to report morphometric data from H. acanthocephalicus and describe the pathological effects of this parasite on the Phimosus infuscatus proventriculus. Thirty gastrointestinal tracts of P. infuscatus were examined to search for nematodes and H. acanthocephalicus occurred in 83% of hosts. Were measured the total length and body width of males and females, and of their respective cuticular spines, esophagus, spicules and eggs, and the internal and external diameter of copulatory bursa. Histopathological examination revealed parasitic structures in the proventriculus from the lumen (anterior end) to the outer layers of the organ (intermediate and posterior parts), in which we observed inflammatory reaction with infiltration of heterophils, hemorrhage and hemosiderin. The results of this study of histopathology, morphometry and parasitological indices are the first ones reported to H. acanthocephalicus and should contribute to the identification and recognition in cases of outbreaks in the Neotropical region.


Resumo Espécie de Hystrichis são nematoides parasitos do trato digestório de aves aquáticas, na América do Sul, Europa e Ásia. No Brasil, foi registrado Hystrichis acanthocephalicus, em Phimosus infuscatus. Existem poucos dados sobre a morfometria e não há registros relativos à patologia desta espécie. Portanto, o objetivo deste estudo foi relatar dados morfométricos de H. acanthocephalicus e descrever os efeitos patológicos deste parasito no proventrículo de Phimosus infuscatus. Foram examinados trinta tratos gastrintestinais de P. infuscatus para a pesquisa de nematoides e H. acanthocephalicus ocorreu em 83% dos hospedeiros. Mediu-se o comprimento total e largura do corpo de machos e fêmeas, dos espinhos cuticulares, do esôfago, dos espículos, dos ovos, respectivamente, e a largura interna e externa da bolsa copuladora. Nos exames histopatológicos foram encontradas estruturas parasitárias no proventrículo, desde o lúmen (porção anterior) até as camadas mais externas do órgão (porções intermediária e posterior), nas quais se observou reação inflamatória com infiltrado de heterófilos, presença de hemorragia e deposição de hemossiderina. Os resultados desse estudo sobre histopatologia, morfometria e índices parasitológicos são os primeiros relatados para H. acanthocephalicus e deverão contribuir para a identificação e reconhecimento em casos de surtos na região Neotropical.

To investigate aquatic bird bornavirus 1 in Europe, we examined 333 brains from hunter-killed geese in Denmark in 2014. Seven samples were positive by reverse transcription PCR and were 98.2%-99.8% identical; they were also 97.4%-98.1% identical to reference strains of aquatic bird bornavirus 1 from geese in North America.

Avian bornaviruses, recently described members of the family Bornaviridae, have been isolated from captive parrots and passerines as well as wild waterfowl in which they may cause lethal neurologic disease. We report detection of avian bornavirus RNA in the brains of apparently healthy gulls. We tested 439 gull brain samples from 18 states, primarily in the northeastern US, using a reverse-transcriptase PCR assay with primers designed to detect a conserved region of the bornavirus M gene. Nine birds yielded a PCR product of appropriate size. Sequencing of PCR products indicated that the virus was closely related to aquatic bird bornavirus 1 (ABBV-1). Viral RNA was detected in Herring Gulls (Larus argentatus), Ring-billed Gulls (Larus delawarensis), and Laughing Gulls (Leucophaeus atricilla). Eight of the nine positive birds came from the New York/New Jersey area. One positive Herring Gull came from New Hampshire. Histopathologic examination of one well-preserved brain from a Herring Gull from Union County New Jersey, showed a lymphocytic encephalitis similar to that observed in bornavirus-infected parrots and geese. Bornavirus N protein was confirmed in two Herring Gull brains by immunohistochemistry. Thus ABBV-1 can infect gulls and cause encephalitic brain lesions similar to those observed in other birds.

OBJECTIVE: To determine the helminth parasites of Anas crecca (A. crecca) in one of proper refuges of Iran, Fereydunkenar. METHODS: A total number of one hundred thirty-six gastrointestinal tracts of green-winged teal (A. crecca) were collected from Fereydunkenar, Mazandaran province during September and October 2011. The gastrointestinal tracts were examined for helminth infection. RESULTS: The total infection rate was 70.50% (96) that 68.96% (40) of males and 71.79% (56) of females shown helminthes infection. The examined A. crecca harbored one species of Nematoda, Cestoda and two species of Digenea which were as following: Contracaecum larvae (from stomach wall), Diorchis stefanskii (D. stefanskii) (from small intestine), Hypoderaeum conoideum (from small intestine) and Notocotylus attenuatus (N. attenuatus) (from caecum), respectively. There was no significant difference in the prevalence of infection between examined males and females ducks in Hypoderaeum conoideum, D. stefanskii and N. attenuatus (P>0.05) whereas a significant relationship was observed between males and females in Contracaecum larvae (P<0.05). CONCLUSIONS: Based on the results of the present study, we conclude that A. crecca plays a prominent role in transmission of mentioned parasites. In addition, this is the first report of Contracaecum larvae, D. stefanskii and N. attenuatus from A. crecca in Iran.
