Aim: To review in vitro, in vivo, and in silico studies examining the antibacterial and immunomodulatory properties of piperine (PPN). Methods: This systematic review followed PRISMA guidelines, and five databases were searched. Results: A total of 40 articles were included in this study. Six aspects of PPN activity were identified, including antibacterial spectrum, association with antibiotics, efflux pump inhibition, biofilm effects, protein target binding, and modulation of immune functions/virulence factors. Most studies focused on Mycobacterium spp. and Staphylococcus aureus. Cell lineages and in vivo models were employed to study PPN antibacterial effects. Conclusion: We highlight PPN as a potential adjuvant in the treatment of bacterial infections. PPN possesses several antibacterial properties that need further exploration to determine the mechanisms behind its pharmacological activity.

Resumen Introducción: la enfermedad diarreica es una de las principales causas de muerte en niños menores de cinco años. Uno de los agentes etiológicos bacterianos más reconocido es Escherichia coli, que puede causar infecciones a través de diversos mecanismos asociados a la adquisición de genes de virulencia. Metodología: se recolectaron muestras de aguas residuales en los afluentes y los efluentes de cinco plantas de tratamiento de la Gran Área Metropolitana, durante tres días consecutivos, en los meses de marzo, mayo, octubre y diciembre del año 2013, a fin de cuantificar la concentración de E. coli, mediante la técnica de fermentación en tubos múltiples, y evaluar la presencia de los genes de virulencia aatA, bfpA, eaeA, ipaH, lt, st, stx1 y stx2, vinculados con patotipos de E. coli diarrogénica, con la técnica de PCR punto final. Resultados: el estudio reveló niveles de E. coli similares a los encontrados en afluentes de otras regiones del mundo, en cambio, los efluentes presentaron concentraciones superiores a los 4 Log10 NMP/100 mL. La circulación anual de genes de virulencia fue relacionada con los patotipos enterotoxigénica, productores de toxina Shiga/enterohemorrágica, enteroagregativa y enteropatógena. No se detectaron genes referentes a la variedad enteroinvasora. Además, se determinó que no todas las plantas de tratamiento remueven significativamente la carga de E. coli. Conclusiones: el flujo de E. coli diarrogénica es común en nuestro país y su diseminación en el ambiente es posible cuando los efluentes se descargan en cuerpos de aguas superficiales


Abstract Introduction. Diarrheic disease is one of the most important diseases causes in children under five years. One of the most recognized etiological agents is Escherichia coli. This bacterium has the capacity of causing infections by multiple mechanisms associated with the acquisition of different virulence genes. Methodology: water samples were collected from the affluents and effluents of five wastewater treatment plants located in the Costa Rican Great Metropolitan Area, during three sequential days, in March, May, October, and December from 2013. The aim was to quantify E. colis concentration by multiple tubes fermentation method and to evaluate the presence of aatA, bfpA, eaeA, ipaH, lt, st, stx1, and stx2 virulence genes, related with diarrheagenic E. coli, by PCR end-point method. Results: The study revealed that the levels of E. coli into the affluents are similar to those reported in other world regions. The effluents concentration presented higher than 4 Log10 NMP/100 mL level of E. coli. A year-round circulation of virulence genes was demonstrated, and it was associated with enterotoxigenic, Shiga-toxin producers/ enterohemorrhagic, enteroaggregative, and enteropathogenic pathotypes. Genes related to enteroinvasive variety did not were detected. Not all wastewater treatment plants removed E. coli efficiently. Conclusions: Diarrheagenic E. coli circulation is common in our county with the possibility of dissemination to the environment by effluents wastewater discharge in surface water bodies.

BACKGROUND: Tick populations have expanded in many parts of the globe, bringing with them an enhanced appreciation and discovery of novel tickborne pathogens, as well an increased in reported human cases of tickborne disease. Targeted and unbiased (shotgun) clinical metagenomic sequencing tests are increasingly used for detection of known and emerging infectious agents and have recently been employed for detection of tickborne pathogens. CONTENT: This review describes the types of metagenomic sequencing assays used for detection of emerging tickborne pathogens and reviews the recent literature on this topic. Important diagnostic and interpretative challenges are also covered. SUMMARY: Metagenomic analysis has emerged as a powerful tool for detection, discovery, characterization, and classification of tickborne pathogens. Shotgun metagenomics is especially promising because it allows for detection of all tickborne bacteria, viruses, and parasites in a single specimen. Despite the potential advantages, there are several important challenges, including high cost, complexity of testing and interpretation, and slow turnaround time. No doubt, these challenges will diminish with increased use and advances in the field.

Bacterial diseases represent the main impediment to the development of fish aquaculture. Granulomatous diseases caused by bacteria lead to fish culture losses by high mortality rates and slow growth. Bacteria belonging to genera Streptococcus spp., Mycobacterium sp., Nocardia sp., Francisella sp., and Staphylococcus sp. have been implicated in the development of granulomatous processes. The granuloma formation and the fish's immune response continue to be the subject of scientific research. In fish, the first defense line is constituted by non-specific humoral factors through growth-inhibiting substances such as transferrin and antiproteases, or lytic effectors as lysozyme and antimicrobial peptides, and linking with non-specific phagocyte responses. If the first line is breached, fish produce antibody constituents for a specific humoral defense inhibiting bacterial adherence, as well as the mobilization of non-phagocytic host cells and counteracting toxins from bacteria. However, bacteria causing granulomatous diseases can be persistent microorganisms, difficult to eliminate that can cause chronic diseases, even using some immune system components to survive. Understanding the infectious process leading to granulomatosis and how the host's immune system responds against granulomatous diseases is crucial to know more about fish immunology and develop strategies to overcome granulomatous diseases.

Huanglongbing (HLB, associated with Candidatus Liberibacter asiaticus and transmitted by the Asian citrus psyllid Diaphorina citri) and citrus variegated chlorosis (CVC, caused by Xylella fastidiosa subsp. pauca and transmitted by sharpshooter species) have been managed by vector control and removal of symptomatic trees. Although vectors and new symptomatic trees can be detected year round, peaks of vector populations are higher in spring and summer, and the most symptomatic trees are found in autumn and winter. This work aimed to compare the management of both diseases during these favorable periods. The experiment was conducted during 5 years in a commercial orchard and had a three-by-two factorial design. The factor "vector control" had three levels: monthly vector control year round (VCYR), monthly vector control in spring and summer (VCSS), and vector control when a threshold level of 10% occupancy was detected (VCOT). The factor "inoculum removal" had two levels: monthly eradication year round (TEYR) and monthly eradication in autumn and winter (TEAW). Host flush, both HLB and CVC vector populations, and the number of symptomatic citrus plants were visually assessed. The level of vectors over the seasons, as measured using the average area under the curve (AUC), was similar for all treatments with the exception of psyllid abundance, which was approximately 4.5 times higher for VCSS than for other treatments. For both diseases, no difference in the average AUC of disease progress and disease final incidence was observed. VCOT or adjusted VCSS associated with TEAW could be integrated for sustainable citrus production.

BACKGROUND: With the increase in unprecedented and unpredictable disease outbreaks due to human-driven environmental changes in recent years, we need new analytical tools to map and predict the spatial distribution of emerging infectious diseases and identify the biogeographic drivers underpinning their emergence. The aim of the study was to identify and compare the local and global biogeographic predictors such as landscape and climate that determine the spatial structure of leptospirosis and Buruli Ulcer (BU). METHODS: We obtained 232 hospital-confirmed leptospirosis (2007-2017) cases and 236 BU cases (1969-2017) in French Guiana. We performed non-spatial and spatial Bayesian regression modeling with landscape and climate predictor variables to characterize the spatial structure and the environmental drivers influencing the distribution of the two diseases. RESULTS: Our results show that the distribution of both diseases is spatially dependent on environmental predictors such as elevation, topological wetness index, proximity to cropland and increasing minimum temperature at the month of potential infection. However, the spatial structure of the two diseases caused by bacterial pathogens occupying similar aquatic niche was different. Leptospirosis was widely distributed across the territory while BU was restricted to the coastal riverbeds. CONCLUSIONS: Our study shows that a biogeographic approach is an effective tool to identify, compare and predict the geographic distribution of emerging diseases at an ecological scale which are spatially dependent to environmental factors such as topography, land cover and climate.

Bovine mastitis is the most frequent disease worldwide in dairy herds, causing high economic losses to producers and industry, as well as having implications for public health due to the zoonotic potential of some agents involved in its etiology and the increased risk of antimicrobial residues in milk and its derivatives. Considering the multifactorial aspect of this disease, knowledge of the agents involved in its etiology and their antimicrobial susceptibility profiles is very important. This study was conducted with 306 dairy herds from the Campo das Vertentes region, located in the south of Minas Gerais state, whose owners were milk suppliers to a dairy in the same region. The study involved approximately 34,000 dairy cows and covered an area of approximately 12,564 km2. In these herds, prevalence rates of Staphylococcus aureus and Streptococcus agalactiae and their relationship with bulk milk somatic cell counts (BMSCC), total bacterial counts (TBC), and daily production were evaluated. In addition, analyses of resistance of these pathogens to the antimicrobials most commonly used in the treatment of mastitis in dairy herds were performed. Microbiological analyses of milk samples from collect from bulk milk tanks were performed aiming to evaluate the prevalence of S. aureus and S. agalactiae. For these proposes, the modified Baird-Parker Agar medium was used for detection of S. aureus and the modified Edwards Agar medium, enriched with 5% defibrinated sheep blood, was used for detection of S. agalactiae. The disc diffusion technique was applied to evaluate antimicrobial resistance. Results show high prevalence rates of S. aureus (70.3%) and S. agalactiae (67.0%) in the dairy farms studied, with 47.71% of the herds showing both pathogens. Associations between BMSCC and the presence of pathogens S. aureus and S. agalactiae and between TBC and the presence of S. agalactiae were observed, demonstrating the influence of these pathogens in milk quality. No variation was observed in the distribution of S. aureus and S. agalactiae in the different strata of daily production. High levels of resistance and multi-resistance were observed among the pathogens S. aureus and S. agalactiae. The results indicate the need for more effective control measures for mastitis caused by S. aureus and S. agalactiae in the dairy herds of the region studied and more judicious use of antimicrobials in order to reduce the problem of resistance to them.(AU)


A mastite bovina é a doença de maior frequência em rebanhos leiteiros em nível mundial, acarretando grandes prejuízos econômicos aos produtores e à indústria. Além disso, esta enfermidade tem implicações na saúde pública, devido ao potencial zoonótico de alguns agentes envolvidos em sua etiologia e por aumentar os riscos de resíduos de antimicrobianos no leite e derivados. Considerando o aspecto multifatorial da mastite bovina, o conhecimento dos agentes envolvidos em sua etiologia e os perfis de suscetibilidade aos antibióticos é de suma importância. O estudo envolveu 306 fazendas de leite da região de Campo das Vertentes, localizada no sul de Minas Gerais, cujos proprietários eram fornecedores de leite para um laticínio da região, totalizando aproximadamente 34.000 animais e abrangendo uma área aproximada 12.564 km2. Nestes rebanhos, avaliaram-se a prevalência de Staphylococcus aureus e Streptococcus agalactiae e a relação destes agentes com os índices de contagem de células somáticas do leite do tanque de expansão (CCSt), contagem bacteriana total (CBT) e produção diária. Analisou-se também a resistência destes patógenos aos antimicrobianos mais comumente utilizados no tratamento da mastite em rebanhos leiteiros. Análises microbiológicas de amostras de leite dos tanques de expansão foram realizadas para se determinar as prevalências dos patógenos S. aureus e S. agalactiae. Para a detecção de S. aureus, utilizou-se o meio seletivo Ágar Baird-Parker modificado e para a detecção de S. agalactiae, o meio seletivo Ágar Edwards modificado, enriquecido com 5% de sangue ovino desfibrinado. Foi utilizada a técnica de difusão em discos para a avaliação de resistência aos antimicrobianos. Os resultados apontaram altas prevalências de S. aureus (70,3%) e de S. agalactiae (67,0%), com 47,71% dos rebanhos examinados apresentando ambos os agentes. Verificaram-se associações entre a CCSt e a presença dos patógenos S. aureus e S. agalactiae, e também entre a CBT e a presença de S. agalactiae, demonstrando a interferência negativa destes patógenos nestes quesitos de qualidade. Não se observaram variações nas distribuições dos patógenos S. aureus e nem S. agalactiae em função da produção diária das propriedades estudadas. Níveis elevados de resistência e de multirresistência foram observados para ambos os agentes. Os resultados apontam a necessidade de medidas mais efetivas de controle para S. aureus e S. agalactiae nos rebanhos da região estudada e do uso mais criterioso dos antimicrobianos, visando minimizar o problema da resistência aos mesmos.(AU)

BACKGROUND: In the last decade, diseases of bacterial origin in onions have increased and this has led to significant losses in production. These diseases are currently observed in both the Old and New Worlds. The present study aimed to evaluate whether the irrigation method influences the incidence of diseases of bacterial origin. RESULTS: In cases where the inoculum was natural, the initial incidence of soft bacterial rot did not manifest in any treatment in the first year, whereas, at the end of the conservation period, all treatments had increased incidences of infection. Sprinkler irrigation (8%) was statistically differentiated from the other treatments, for which the final incidence was similar (4.5%). For all irrigation treatments, the final incidence of bacterial soft rot decreased or remained stable towards the end of the cycle, with the exception of sprinkler irrigation in 2015, which increased. CONCLUSION: From the results of the present study, it can be inferred that the irrigation method does have an influence on the incidence of diseases of bacterial origin in the post-harvest stage for onions. © 2018 Society of Chemical Industry.

BACKGROUND: Hypervirulent strain of Klebsiella pneumoniae genotype K1 isolates have recently emerged, causing severe pyogenic liver abscess complicated by devastating metastatic infections in humans. METHODS: We describe a short outbreak of the non-human primate (NHP) research center, associated with a hypervirulent K. pneumoniae. The genetic similarity of the strains was evaluated by pulsed-field gel electrophoresis (PFGE) and multilocus sequence typing (MLST) techniques, and virulence encoding genes were detected by polymerase chain reaction (PCR). RESULTS: The isolates were phenotypically like strains causing community-acquired invasive liver abscess syndrome in humans. All strains exhibited identical PFGE patterns and were found to belong to ST23 and presented a hypermucovisity phenotype and possessed magA and rmpA gene. CONCLUSION: This is the first case report of NHPs caused by K. pneumoniae displaying a hypermucoviscosity phenotype and belonging to capsular serotypes K1 and ST23.

Las enfermedades bacterianas y parasitarias pueden llegar a comprometer los niveles de sobrevivencia de un cultivo de camarón marino. La frecuencia de aparición de camarones muertos en las orillas de los estanques o que aparecen en las atarrayas, son solo un signo o señal de que algo marcha mal en el estanque. Cuando los camarones están sometidos a factores estresantes se reducen su actividad limpiadora, no mudan y por tanto, son altamente susceptibles a la invasión de ectoparásitos. En grados de severidad 3 y 4 de la infestación por ectoparásitos en la superficie corporal del camarón, trae como consecuencia dificultades en la locomoción, la alimentación, la respiración, y finalmente sobreviene la muerte. Por otra parte, las enfermedades de origen bacteriano reportados en los sistemas de cultivo causadas por el género Vibrio, ha llegado a ser la enfermedad económicamente más importante en el cultivo de organismos marinos, afectando un gran número de especies; es catalogada como la causa de serias pérdidas económicas en la producción de camarón de cultivo en diversos países.

The frequency of the appearance of dead shrimp on the banks of the ponds or that appear on the cast nets, are just a sign that something is wrong in the pond. When shrimp are subjected to stress, their cleaning activity is reduced, they do not molt and therefore are highly susceptible to invasion by ectoparasites. In degrees of severity 3 and 4, the infestation by ectoparasites on the body surface of the shrimp, results in difficulties in locomotion, feeding, breathing, and finally, death ensues. On the other hand, the diseases of bacterial origin reported in culture systems caused by the Vibrio genus have become the most economically important disease in the cultivation of marine organisms, affecting a large number of species; It is classified as the cause of serious economic losses in the production of farmed shrimp in various countries.

Background: Dermatophilosis is an infectious-contagious disease of acute, subacute or chronic evolution caused by the etiologic agent Dermatophilus congolensis, and presents as hyperplastic or exudative dermatitis with crusty and scaly skin eruptions. Although it is a disease with important economic impact on Brazilian beef cattle, the reports of outbreaks in zebu cattle are restricted to the states of São Paulo and Mato Grosso do Sul. The present paper aimed to report the epidemiological, clinical, laboratorial and pathological findings of a dermatophilosis outbreak in Nelore calves raised extensively in Goiás, Midwestern Brazil. Case: An outbreak of skin lesions in Nelore calves occurred on a farm located in Cocalzinho de Goiás, Goiás, Midwestern Brazil. Fifty one (25.5%) of the 200 calves from 3-11-months-old presented cutaneous lesions. Forty nine of the calves were still suckling and two were weaned recently, and the most affected calves were offspring’s of cows with the lowest body condition score. Three severely affected calves were clinically examined presenting regular body condition score, pale pink ocular mucosa, fever (39.2-40.3°C), tachycardia (80-100 beats per minute), tachypnea (48-56 breaths per minute) and moderate presence of ticks. Skin inspection revealed crusty, elevated, non-pruritic lesions of grayish color that easily detached from the skin exposing a humid and hemorrhagic superficial lesion with pus. Lesions were widespread on the body with predominance on the face, ears, neck and dorsal region. Hematological findings of these calves presented leukocytosis (16,083 ± 1,910/µL) by netrophilia (11,121 ± 2,349/µL) and hyperfibrinogenemia (966.6 ± 208.16 mg/dL). Biochemical alterations consisted of hypoproteinemia (6.5 ± 0.5 g/dL) and a slight increase in GGT activity in two calves (28 and 19 U/L, respectively)...

Leptospirosis is been considered an important infectious disease that affects humans and animals worldwide. This review summarizes our current knowledge of bacterial attachment to extracellular matrix (ECM) components and discusses the possible role of these interactions for leptospiral pathogenesis. Leptospiral proteins show different binding specificity for ECM molecules: some are exclusive laminin-binding proteins (Lsa24/LfhA/LenA, Lsa27), while others have broader spectrum binding profiles (LigB, Lsa21, LipL53). These proteins may play a primary role in the colonization of host tissues. Moreover, there are multifunctional proteins that exhibit binding activities toward a number of target proteins including plasminogen/plasmin and regulators of the complement system, and as such, might also act in bacterial dissemination and immune evasion processes. Many ECM-interacting proteins are recognized by human leptospirosis serum samples indicating their expression during infection. This compilation of data should enhance our understanding of the molecular mechanisms of leptospiral pathogenesis.

This study aimed to report the epidemiological, clinical-pathological and laboratory findings in a 2-month-old kid with meningoencephalitis by Escherichia coli. Clinical examination revealed lateral recumbency, vocalization, opisthotonos, neck stiffness, bilateral corneal opacity, fever (41.3C), tachycardia, dehydration, reddish urine and paddling. Hematology revealed light leukocytosis with absolute monocitosis. Serum biochemistry was within physiological standards for the specie. Cerebrospinal fluid (CSF) analysis showed turbid aspect, light yellow coloration, neutrophilic pleocytosis (598 leukocytes/L) and bacteria. The animal died about two hours after the clinical exam. Necropsy findings consisted of hyperemia, opacity and presence of fibrinopurulent exudate in the meninges, cerebral cortex sulci and brain stem; while the kidneys showed severe congestion. Microscopically, there was a mixed inflammatory infiltrate involving the meninges of the regions of the parietal, temporal cortex and brainstem associated to bacterial colonies. Edema and neuronal degeneration was also noted. Microbiological culture of a CSF aliquot allowed the isolation of E. coli.


O presente trabalho objetiva relatar os achados epidemiológicos, clínico-patológicos e laboratoriais de um caso de meningoencefalite por Escherichia coli em um cabrito de dois meses de idade. Ao exame clínico observou-se decúbito lateral, vocalização, opistótono, rigidez muscular cervical, opacidade bilateral de córnea, febre (41,3ºC), taquicardia, desidratação, urina avermelhada e movimentos de pedalagem. A hematologia mostrou discreta leucocitose com monocitose absoluta. A bioquímica sérica apresentou-se dentro dos padrões fisiológicos para a espécie. A análise do líquido cefalorraquiano (LCR) revelou aspecto turvo, coloração amarelo claro, pleocitose (598 leucócitos/µL) neutrofílica e presença de bactérias. O animal veio a óbito aproximadamente duas horas após o exame clínico. A necropsia revelou hiperemia, opacidade e presença de exsudato fibrinopurulento nas meninges, sulcos do córtex cerebral e tronco encefálico; e os rins com severa congestão. Microscopicamente, observou-se infiltrado inflamatório misto envolvendo as meninges das regiões do córtex parietal, temporal e tronco encefálico; além de edema e degeneração neuronal. A cultura microbiológica de alíquota do LCR permitiu o isolamento de E. coli.

The aim of this work was to relate the occurrence of bacterial illnesses in two wild birds raised under captive conditions in Rio Grande do Norte State, Brazil, focusing on their clinical and microbiological findings. Clinical exams were performed in both birds in order to collect data and to determine the clinical and pathological outcome. In bird [A1] was diagnosed an infection in the respiratory system, and in bird [A2] the diagnosis was pododermatitis. Samples of shedding were collected using a sterile swab to perform a bacterial culture in blood agar and McConkey agar. An antibiogram was performed as well. The bacterial culture revealed the growing of Proteus sp. and Proteus vulgares, respectively. The antibiogram performed with the samples of bird [A1] showed the resistance to the follow antibiotics: ampicillin, amoxicillin/clavulanic acid, aztreonam, cefepime, ceftazidime, ciprofloxacin, tetracycline, sulfamethoxazole/trimethoprim and gentamicin. In bird [A2], strain was resistant to ampicillin, amoxicillin/clavulanic acid, aztreonam, cefotoxin, chloramphenicol, tetraciclin, sulfamethoxazole/trimethoprim and gentamicina. Proteus spp. appears to be a potencial multiresistant pathogen and causes severe lesions and diseases for wild birds in captivity.


O objetivo deste trabalho é relatar a ocorrência de enfermidades bacterianas em duas aves exóticas criadas em cativeiro no Rio Grande do Norte, com enfoque nos achados clínicos e microbiológicos. Realizou-se o exame clínico nas aves para a coleta de dados e caracterização do quadro clínico-patológico, sendo diagnosticada na ave [A1] infecção no trato respiratório superior, e na ave [A2] pododermatite profunda. O material biológico das secreções foi colhido com o auxílio de swabs estéreis para a determinação do agente etiológico através de cultura bacteriana em ágar sangue e ágar MacConkey. Realizou-se também o teste de susceptibilidade a antimicrobianos. A cultura bacteriológica das aves [A1] e [A2] resultou em crescimento de Proteus sp. e Proteus vulgares, respectivamente. O antibiograma da ave [A1] demonstrou resistência aos antibióticos: ampicilina, amoxicilina/ácido clavulâmico, aztreonam, cefepime, ceftazidima, ciprofloxacina, tetraciclina, sulfametoxazol/trimetroprim e gentamicina. Na ave [A2] a cepa isolada apresentou resistência a ampicilina, amoxicilina/ácido clavulâmico, aztreonam, cefoxitina, cloranfenicol, tetraciclina, sulfametoxazol/trimetroprim e gentamicina. Proteus spp. pode ser considerado um patógeno multirresistente, causador de lesões graves e doenças em aves selvagens criadas em cativeiro.
