Microorganisms are important indicators of soil quality due to their sensitivity to changes, reflecting the impacts caused by different land uses. The objective of this study was to evaluate the microbiological and physical-chemical attributes of the soil in areas cultivated with coffee under three different management systems (shaded coffee and full sun coffee with two spacings), as well as in adjacent areas under pasture and native forest, in Bahia, Brazil. The microbiological and physicochemical indicators evaluated were basal soil respiration (MBR), soil total organic carbon (TOC), microbial biomass carbon (MBC), metabolic quotient (qCO2), microbial quotient (qMic), enzyme activities (urease, acid phosphatase and fluorescein diacetate hydrolysis (FDA)). Physical and chemical indicators (particle size, texture, pH, P, K+, Ca2+, Mg2+, Al3+, and sum of bases) were also evaluated. Biological and chemical attributes were much more discriminative of study areas in the dry season. Microbial quotient (qMic) and metabolic quotient (qCO2) in the dry season showed that pasture is the most degraded land use. Conversely, nature forest and coffee with Grevillea were similar and were the best ones. In general, soil quality indicators were more sensitive to discriminate pasture and native forest from coffee systems, which, in turn, were not well discriminated among themselves.

The cryopreservation of plant germplasm at ultralow temperatures is an alternative technique for the long-term storage of seeds of the genus Coffea sp. However, for this technique to be successful, cell integrity must be maintained at all stages of the process on the basis of scientific research. The present study investigated validated cryopreservation protocols for Coffea arabica L. seeds and evaluate the effects on the physiological and biochemical characteristics of the seeds at each stage of the process. Seeds were dried on silica gel or with saturated saline solution, precooled or not in a biofreezer, immersed in nitrogen, and reheated in a water bath. After each of these steps, the physiological and biochemical quality of the seeds was determined. Pre-cooling is a step that can be dispensed with in the cryopreservation of Coffea arabica seeds, direct immersion in liquid nitrogen being more indicated. Coffea arabica L. seeds tolerate cryopreservation after rapid drying in silica gel up to water contents of 17 or 20% (wb), with greater survival at 17%. The enzyme activities of catalase, polyphenol oxidase and peroxidase are indicators of the quality of C. arabica L. seeds subjected to cryopreservation.


A criopreservação de germoplasma vegetal em temperaturas ultrabaixas é uma alternativa para o armazenamento em longo prazo de sementes do gênero Coffea sp. Entretanto, para que essa técnica apresente sucesso, a realização de pesquisas que garantam a manutenção da integridade celular em todas as etapas do processo é de fundamental importância. O objetivo com o presente trabalho foi investigar protocolos de criopreservação validados para sementes de Coffea arabica L., avaliando separadamente, os efeitos sobre as características fisiológicas e bioquímicas das sementes, em cada etapa do processo. As sementes foram secadas em sílica gel ou em solução salina saturada, submetidas ou não ao pré-resfriamento em biocongelador, em seguida imersas no nitrogênio e reaquecidas em banho-maria. Após cada uma dessas etapas, a qualidade fisiológica e bioquímica das sementes foi determinada. O pré-resfriamento é uma etapa que pode ser dispensada na criopreservação de sementes de Coffea arabica, sendo mais indicada a imersão direta em nitrogênio líquido. Sementes de Coffea arabica L. toleram a criopreservação após secagem rápida em sílica gel até teores de água de 17 ou 20% (wb), com maior sobrevivência a 17%. A atividade das enzimas catalase, polifenoloxidase e peroxidase são indicadoras da qualidade de sementes de Coffea arabica L. submetidas à criopreservação.

The adaptive potential of plants is commonly used as an indicator of genotypes with higher breeding program potential. However, the complexity and interaction of plant metabolic parameters pose a challenge to selection strategies. In this context, this study aimed to explore phenotypic plasticity within the germplasm of Hybrid Timor coffee. Additionally, we assessed the utility of the multivariate phenotypic plasticity index (MVPi) as a promising tool to predict genotype performance across diverse climatic conditions. To achieve this, we evaluated the performance of seven accessions from the Hybrid Timor germplasm in comparison to the Rubi and IPR 100 cultivars, known for their susceptibility and resistance to drought, respectively. The experiment took place in a greenhouse under two conditions: one with normal soil moisture levels near maximum capacity, and the other with a water deficit scenario involving a period of no irrigation followed by rehydration. Data on physiological and biochemical factors were collected at three stages: before applying the water deficit, during its imposition, and after rehydration. Growth data were obtained by the difference between the beginning and end of the experimental period Furthermore, field evaluations of the productivity of the same genotypes were carried out over two consecutive seasons. Based on physiological and biochemical assessments, the MVPi was computed, employing Euclidean distance between principal component multivariate analysis scores. Subsequently, this index was correlated with growth and productivity data through linear regressions. Our findings reveal that the plastic genotypes that are capable of significantly altering physiological and biochemical parameters in response to environmental stimuli exhibited reduced biomass loss in both aerial and root parts. As a result, this positively influenced their productivity. Enhanced plasticity was particularly prominent in accessions from the MG Germplasm Collection: MG 311-Hybrid Timor UFV 428-02, MG 270-Hybrid Timor UFV 377-21, and MG 279-Hybrid Timor UFV 376-31, alongside the Rubi MG 1192 cultivar. The MVPi emerged as a valuable instrument to assess genotype adaptability and predict their performance under varying climatic scenarios.

Weed management in areas adjacent to coffee plantations makes herbicide drift a constant concern, especially with the use of nonselective products such as dicamba. The objective of this study was to evaluate the phytotoxic effects of the herbicide dicamba alone and mixed with glyphosate as a result of simulated drift in a coffee-producing area. The study was conducted in duplicate at two different coffee cherry development stages. The study was performed with a randomized block design and a 2 × 5 + 1 factorial scheme with four replications using two herbicide spray solutions (dicamba and dicamba + glyphosate) and five low doses (0.25; 1; 5; 10; and 20%). Additionally, a control treatment without herbicide application was also employed. In this study, we evaluated the phytotoxic damage and biometric and productive parameters. Visual damages were observed with the use of dicamba and dicamba + glyphosate doses reduced by 0.25% to 5% in the first days after application. The main symptoms were new leaf epinasty, changes in the internodal distance, and plagiotropic branch curvature. Low doses led to reduced plant height and branch length. The treatments did not reduce productivity and performance but altered the physical classifications of grains.

Over the centuries, human society has evolved based on the ability to select and use more adapted species for food supply, which means making plant species tastier and more productive in particular environmental conditions. However, nowadays, this scenario is highly threatened by climate change, especially by the changes in temperature and greenhouse gasses that directly affect photosynthesis, which highlights the need for strategic studies aiming at crop breeding and guaranteeing food security. This is especially worrying for crops with complex phenology, genomes with low variability, and the ones that support a large production chain, such as Coffea sp. L. In this context, recent advances shed some light on the genome function and transcriptional control, revealing small RNAs (sRNAs) that are responsible for environmental cues and could provide variability through gene expression regulation. Basically, sRNAs are responsive to environmental changes and act on the transcriptional and post-transcriptional gene silencing pathways that regulate gene expression and, consequently, biological processes. Here, we first discuss the predicted impact of climate changes on coffee plants and coffee chain production and then the role of sRNAs in response to environmental changes, especially temperature, in different species, together with their potential as tools for genetic improvement. Very few studies in coffee explored the relationship between sRNAs and environmental cues; thus, this review contributes to understanding coffee development in the face of climate change and towards new strategies of crop breeding.

In February 2020, Coffea arabica L. grown on the coast and in the Southern Sierra Madre of the state of Oaxaca, Mexico obtained the denomination of origin. Which does not have data on color and chemical composition, the first associated with the degree of roasting and the second with lipids (17-18%), as the group of compounds responsible, in part, for flavor, consistency, and may contribute to health benefits. In the present work, color was determined on the CIE L*a*b* scale and the unsaturated fatty acids by Nuclear Magnetic Resonance (NMR) of 1H and 13C in samples of medium roasted specialty coffee from the "Pluma" coffee-growing region, Oaxaca, Mexico. The average value of L* luminosity in ground coffee was 42.1 ± 0.1 reported for a light roast. Unsaturated fatty acids were quantified from the lipid fraction of the gr1 ound grain by NMR 1H and 13C, obtaining on average the highest abundance of linoleic (41.7 ± 0.5 by 1 H and 41.24 ± 0.5 by 13C), followed by oleic (9.2 ± 0.2 by 1H and 7.4 ± 0.2 by 13C) and linolenic (1.5 ± 0.1 by H and 1.1 ± 0.2 by 13C). This study indicates that 1H and 13C NMR spectroscopy is a useful tool for the quantification of linolenic, linoleic, and oleic fatty acids by the method of key signal shifts of these acids found in lipid samples in roasted coffee grains.

In this study, arabic coffee infusion was used to produce a fermented beverage known as kombucha. The physicochemical, antioxidant and antimicrobial activities, as well as in vivo toxicity were evaluate throughout 21 days of fermentation. Reduction in pH and sugar levels were observed throughout the fermentation period. There was no significant difference in the content of total phenolic compounds between the unfermented and fermented beverage, nor between the fermentation times, as well as in the antioxidant activity. The 5-caffeoylquinic acid was identified at all fermentation times evaluated, and no significant difference was observed regarding its concentration. It showed antibacterial and antifungal activity against all strains tested. No toxic effect of the beverages was observed in the in vivo model (Galleria mellonella) studied. These results demonstrated that coffee infusion is a possible alternative for kombucha production since the physicochemical changes prove the metabolic activity of Symbiotic Culture of Bacteria and Yeast.

The objective of this study was to evaluate the growth and quality of arabica coffee seedlings produced in tubes under different sources of phosphorus (P). The experiment was conducted in a greenhouse in a randomized block design with ten treatments (control, Osmocote®, three doses of pelleted organomineral, three doses of mashed organomineral, MAP, coated MAP) and four replications, three plants per plot. The dose of organomineral, MAP, and MAP coated was based on the recommendation of P2O5for the formation of coffee seedlings.For Osmocote®, the manufacturer's recommendation was used depending on the amount, in dm³, of substrate. After 180 days of transplanting, the growth characteristics of the plants (leaf dry matter, stem, and roots) were evaluated, and the aerial part and root system ratio and Dickson's quality index were calculated, as well as regression equations were adjusted for the evaluated parameters. For the growth and quality of seedlings, it was observed that in fertilizations with pelleted and branched organomineral, coated MAP and Osmocote®, the highest averages were obtained for the contents of dry matter, height, diameter of the collection and quality indexes, when compared to the other sources of P2O5.The dose that provides the maximum growth and quality of coffee seedlings for the treatments with organomineral was calculated, this being equal to 6 g/dm³ of P2O5for bran and pelleted organomineral. According to the results, the sources that promoted the formation of a vigorous seedling, with an adequate supply of nutrients and a good quality standard are pelleted organonineral fertilization and MAP, reducing the risk of losses and consequently the need to discard the seedlings in the nursery.(AU)


O objetivo deste estudo foi avaliar o crescimento e a qualidade de mudas de café arábica produzidas em tubetes sob diferentes fontes de fósforo(P). O experimento foi conduzido em casa de vegetação em delineamento de blocos casualizados com dez tratamentos(controle, Osmocote®, três doses de organomineral peletizado, três doses de organomineral farelado, MAP, MAP revestido)e quatrorepetições, três plantas por parcela. A dose de organomineral, MAP e MAP revestidofoi baseada na recomendação de P2O5para a formação de mudas de café. Para Osmocote®, foi utilizada a recomendação do fabricante emfunção da quantidade, em dm³, de substrato. Após 180 dias do transplante, foram avaliadas as características de crescimento das plantas (matéria seca de folhas, caule e raízes), calculadas a razão parte aérea e sistema radicular e o índice de qualidade de Dickson, bem como ajustadas equações de regressão para os parâmetros avaliados. Para o crescimento e qualidade das mudas, observou-se que nas adubações com organomineral peletizado e farelado, MAP revestido e Osmocote®, as maiores médias para os teores de matéria seca, altura, diâmetro da coletae índices de qualidade, quando comparados as outras fontes de P2O5.Foi possível identificara dose que proporciona o máximo crescimento e qualidade de mudas de café para os tratamentos com organomineral, sendo esta igual a 6 g/dm³ de P2O5para o organomineral farelado e peletizado. De acordo com os resultados, as fontes que promoveram a formação de uma muda vigorosa, com aporte adequado de nutrientes e um bom padrão de qualidade são a adubação organomineral peletizada e MAP, reduzindo o risco de perdas e consequentemente a necessidade de descarte das mudas no viveiro.(AU)

INTRODUCTION: Coffea arabica L. leaves are considered a by-product of the coffee industry however they are sources of several bioactive compounds. OBJECTIVES: This study aimed to evaluate the chemical composition and the in vitro antibacterial activity of the lyophilised ethanol extract of arabica coffee leaves (EE-CaL). MATERIAL AND METHODS: The chemical characterisation of EE-CaL was performed using ultra-performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry (UPLC-Q-ToF-MS/MS). The in vitro antibacterial effect of EE-CaL was evaluated using the broth microdilution method and the adapted drop plate agar method to determine the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC), respectively. RESULTS: The chemical analysis of EE-CaL revealed the presence of compounds from the alkaloid class, such as trigonelline and caffeine, in addition to the phenolic compounds such as quinic acid, 5-caffeoylquinic acid, caffeic acid-O-hexoside, mangiferin, (epi)catechin, (epi)catechin monoglucoside and procyanidin trimer. Regarding the antibacterial potential, EE-CaL was active against Gram-positive and Gram-negative bacteria, being more effective against Escherichia coli (ATCC 25922) (MIC = 2500 µg/mL and bactericidal effect). CONCLUSION: The results of this research suggest that coffee leaves, a by-product, possess compounds with antibacterial properties. Thus, further studies with coffee leaf extracts must be carried out to relate the compounds present in the extract with the antibacterial activity and find the mechanisms of action of this extract against bacteria.

Objectives: The study aimed to determine the effect of coffee intake on AGEs formation and platelet aggregation in diabetic Wistar rats. Methods: Coffee powder samples were used to prepare a 10% beverage. Diabetes mellitus was induced in the animals by administering 2% alloxan. All animal experiments were approved by the ethics committee for animal experiments under N°. 420/2012 and 536/2013. Diabetic and non-diabetic rats were divided into 6 groups treated and untreated with coffee (7.2 mL/Kg body weight) and aminoguanidine (AGE inhibiting agent) (100 mg/Kg body weight) for 50 days. After 50 days, the animals were fasted for 12 h and anesthetized (40 mg/Kg sodium pentobarbital) intraperitoneally. Blood samples were collected from the abdominal artery puncture. Hematological parameters (red cells, hemoglobin, hematocrit and leukocyte) and glycemic and HbA1c levels were measured. AGEs quantification (spectrofluorometric method) and the platelet aggregation test (aggregation of cuvettes in a four-channel platelet aggregometer) were also conducted. The rats' renal function was evaluated by measuring serum urea and creatinine. Results: Data showed that coffee intake had no effect on the hematological parameters. Fasting glucose and HbA1c dosage were significantly higher in diabetic animals compared to non-diabetic animals (confirmed the effectiveness of inducing and maintaining diabetic status). Results showed that coffee reduced AGE formation and platelet aggregation in our animal model, not altering the animals' renal function. Conclusions: These results suggest beneficial effects on vasculopathy, a common complication in diabetic patients.

BACKGROUND: The occurrence of diseases can alter coffee (Coffea arabica L.) metabolism, causing changes in the composition of coffee beans and beverage quality. However, little is known about which aspects of coffee quality are actually altered by rust (Hemileia vastatrix Berk et Br.) and by its main control methods. The effect of chemical and genetic methods for the control of coffee rust on the quality of coffee beans and beverage was investigated. RESULTS: Both genetic and chemical control reduce the damage caused by the disease in the composition of coffee beans. Genotypes with resistant ancestry, even with resistance breakdown, respond better to chemical control. The combination of genetic and chemical control favors an increase in the sugar content in the beans. CONCLUSIONS: Despite the fact that both genetic and chemical control are effective in reducing disease damage regarding the chemical composition of beans, the quality potential of Timor Hybrid genotypes associated with the cancellation of rust expression through the joint action of genetic and chemical control favors the composition of beans and, consequently, the quantitative assessment of sensory attributes, adding value to the final product. © 2020 Society of Chemical Industry.

For leaves, the critical nutrient ranges are respectively: 2.63 - 2.86, 0.13 - 0.14, 2.13 - 2.33, 1.04 - 1.22, 0.27 - 0.33, 0.15 - 0.18 dag kg-1 of N, P, K, Ca, Mg, and S; and 9 - 14, 15 - 23, 80 - 115, 99 - 148, and 31 - 37 mg kg-1 of Zn, Cu, Mn, Fe, and B. A análise química de flores tem sido estudada em algumas culturas. Para o cafeeiro, a análise do tecido floral possibilitaria a antecipação do diagnóstico nutricional das lavouras. O estudo objetivou (i) comparar a composição mineral de flores e de folhas de cafeeiros (ii) e gerar normas para diagnose nutricional do cafeeiro com base na análise de tecidos de flores e folhas das plantas. Para isso, foram avaliados os teores de nutrientes em flores e folhas e a produtividade de café em 26 lavouras comerciais na região de Manhuaçu, MG, durante três anos. RESUMO: A análise química de flores tem sido estudada em algumas culturas. Para o cafeeiro, a análise do tecido floral possibilitaria a antecipação do diagnóstico nutricional das lavouras. O estudo objetivou (i) comparar a composição mineral de flores e de folhas de cafeeiros (ii) e gerar normas para diagnose nutricional do cafeeiro com base na análise de tecidos de flores e folhas das plantas. Para isso, foram avaliados os teores de nutrientes em flores e folhas e a produtividade de café em 26 lavouras comerciais na região de Manhuaçu, MG, durante três anos. As faixas críticas de nutrientes determinadas em flores são: 2,78 - 3,17; 0,23 - 0,28; 2,80 - 3,12; 0,30 - 0,37; 0,24 - 0,30; 0,15 - 0,18 dag kg-1 de N, P, K, Ca, Mg e S, e 17 - 21; 12 - 18; 52 - 80; 26 - 43 e 28 - 48 mg kg-1 para Zn, Cu, Mn, Fe e B, respectivamente. As faixas críticas de nutrientes em folhas foram: 2,63 - 2,86; 0,13 - 0,14; 2,13 - 2,33; 1,04 - 1,22; 0,27 - 0,33; 0,15 - 0,18 dag kg-1 de N, P, K, Ca, Mg e S, e 9 - 14; 15 - 23; 80 - 115; 99 - 148 e 31 - 37 mg kg-1 para Zn, Cu, Mn, Fe e B, respectivamente. A diagnose nutricional do cafeeiro, quanto aos nutrientes N, P, Ca, Fe, Cu e Mn, pode ser antecipada por meio da análise de flores.

Food adulteration is an illegal practice performed to elicit economic benefits. In the context of roasted and ground coffee, legumes, cereals, nuts and other vegetables are often used to augment the production volume; however, these adulterants lack the most important coffee compound, caffeine, which has health benefits. In this study, the mid-infrared Fourier transform spectroscopy (FT-MIR) technique coupled with chemometrics was used to identify and quantify adulterants in coffee (Coffea arabica L.). Coffee samples were adulterated with corn, barley, soy, oat, rice and coffee husks, in proportions ranging from 1-30%. A discrimination model was developed using the soft independent modeling of class analogy (SIMCA) framework, and quantitative models were developed using such algorithms as the partial least squares algorithms with one variable (PLS1) and multiple variables (PLS2) and principal component regression (PCR). The SIMCA model exhibited an accuracy of 100% and could discriminate among all the classes. The quantitative model with the highest performance corresponded to the PLS1 algorithm. The model exhibited an R2c: ≥ 0.99, standard error of calibration (SEC) of 0.39-0.82, and standard error of prediction (SEP) of 0.45-0.94. The developed models could identify and quantify the coffee adulterants, and it was considered that the proposed methodology can be applied to identify and quantify the adulterants used in the coffee industry.

Coffee is a mixture of substances with potential beneficial and adverse health effects. Several studies demonstrate the antioxidant effect of the phenolics compounds present in coffee. Neutrophils produce reactive oxygen species (ROS) by activating of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 2 (NOX2), which plays a key role in organism defenseagainst microbial pathogens. Diabetes mellitus patients are more susceptible to bacterial and fungal infections. The present study evaluated the influence of coffee beverage on NOX2 activity and ROS generation and the impact of this effect on phagocytosis and killing of Candida albicansby neutrophils from diabetic and non-diabetic animals. Diabetes mellitus was induced in male Wistar rats using 2% alloxan. Diabetic and non-diabetic animals were divided into groups treated and untreated with coffee drink (7.2 mL/kg/day) or apocyanine (16 mg/kg/day) for 50 days. After 50 days, the animals' glycemic profile was measured by blood glucose and glycated hemoglobin (HbA1c) tests. The generation of ROS in neutrophilic cells was measured by chemiluminescence and cytochrome C reduction assays. C. albicans phagocytosis and death were evaluated by optical microscopy using the May-Grunwald-Giemsa staining method. The coffee drink has not altered the glycemic profile and NOX2 activity of the animals. However, coffee reduced the ROS pool in non-diabetic and diabetic animals, but this activity did not harm the phagocytosis or killing of neutrophils. Treatment with apocyanin decreased ROS production and killing capacity of neutrophils from non-diabetic animals against C. albicans. We suggest that the coffee drink intake prevents oxidative damage and does not impair response of the organism against opportunistic microorganism.

RESUMEN La adopción de tecnologías generadas para el cultivo del café depende, en gran medida de factores, como la disponibilidad de recursos y el nivel de escolaridad de los productores, lo cual, determina la importancia de analizar las condiciones socioeconómicas, en la estructuración de los sistemas productivos de café. El objetivo de esta investigación fue analizar las principales características sociales y económicas de los cafeteros en los ecotopos 220A y 221A, departamento de Nariño. Con base en un marco muestral de 16.767 predios cafeteros, se seleccionaron aleatoriamente 159 productores (86, del ecotopo 220A y 73, del 221A), con el objeto de aplicar el formulario de encuesta. En el análisis estadístico, 58 variables categóricas fueron sometidas a un análisis multivariado, mediante el método de correspondencias múltiples y análisis de clasificación jerárquica. El ecotopo 220A, se caracterizó por tener áreas de café entre 1 y 3 hectáreas, viviendas con techos de eternit y zinc, pisos de cemento, energía eléctrica, acueducto y unidad sanitaria. El rendimiento está entre 1.001 y 2.000kg.ha-1, costos de producción menores a COP1.500.000 (USD444,83). Los caficultores asisten a jornadas de capacitación y su núcleo familiar está compuesto por 4-7 personas. En el ecotopo 221A predominan las casas con techo de teja, pisos en cemento, energía eléctrica, acueducto y unidad sanitaria; las aguas servidas se disponen en pozos sépticos. No se aplican las buenas prácticas agrícolas (BPA), los rendimientos son menores a 1.500kg.ha-1 cps (café pergamino seco) y los costos de producción son inferiores a COP1.500.000ha-1. año-1 (USD444,83).


ABSTRACT The adoption of technologies generated for the coffee crop, depends to a great degree on factors such as the availability of resources and the producer´s scholarship which determines the importance of socioeconomic conditions in the coffee productive system structure. The objective of this investigation was to analyze the main coffee grower´s social and economic characteristics in the 220A and 221A Nariño Department ecotopes. 159 producers (86 from the ecotype 220A and 73 from the ecotope 221A), were selected randomly based on a sampling frame of 16.767 coffee farms, with the purpose of applying the survey questionnaire. In the statistical analysis, 58 categorical variables were used, they were subjected to a multivariate analysis using the multiple correspondence method and hierarchical classification analysis. The 220A ecotope was characterized by having coffee areas between 1 and 3 hectares, houses with eternit and zinc roofs, cement floors, electric power, aqueduct and sanitary unit. The performance is between 1.001 and 2.000kg.ha1, the cost of production is less than COP1.500,000 (USD44,83); the coffee growers attend training sessions and their family nucleus is made up of 4 - 7 people. In the ecotope 221A the houses with tile roof, cement floors, electricity, aqueduct and sanitary unit predominate; wastewater is disposed of in septic tanks. Good agricultural practices (GAP) aren't used, the yields are less than 1.500kg.ha-1 (dried parchment coffee) and the cost of production is less than $1.500.000 ha-1. year-1

When it comes to the sensory analysis of specialty coffees, it is necessary to discuss the protocol of tests of the Specialty Coffee Association (SCA) and the formation of Q-Graders as reliable parameters in the sensory analysis. However, the training of these Q-Graders and the use of the SCA protocol has generated discussions in the scientific community and demonstrated the importance of talking about the level of professional precision as well as the use of the protocol. This study sought to understand the relation of accuracy and efficiency of the Q-Graders and protocol in the sensory analysis of coffees. Three experiments were carried out to evaluate and test the hypotheses regarding the level of precision in relation to the shift, the number of samples and the judgment abilities of Q-Graders due to the quality of the specialty coffee and nonspecialty coffee. These three experiments were performed by seven Q-Graders in an isolated environment during three consecutive days with 137 specialty coffee samples and 7 nonspecialty coffee samples as defined by the SCA. The results indicate that the Q-Graders present high precision when evaluating excellent and outstanding coffees, as defined by the SCA. In addition, the effect of the shift did not exert fatigue on the Q-Graders. However, evaluation errors were made by the Q-Graders on very good coffee (77.00-80.00 points), which are considered as nonspecialty by the SCA, thus allowing a more in-depth discussion on what would be the boundary between a specialty and nonspecialty coffee. PRACTICAL APPLICATIONS: The article deals with the sensory evaluation process of coffees, and it has great importance to teaching and research institutions in Brazil, the largest producer, largest exporter and the country with the highest concentration of coffee scholars in the world. The article focuses on a new approach and a new way of looking at the sensory evaluation process of coffees, using the methodology of the Specialty Coffee Association to analyze the accuracy and efficiency of the Q-Graders.

Enzymes play a fundamental role in degradation of molecules during seed germination, development, and deterioration. Endo--mannanase is one of the main enzymes responsible for hydrolysis of mannans in the endosperm during germination of coffee seeds through its action in hydrolytic degradation of cell walls and in weakening the structures of the endosperm that surround the embryo, allowing radicle emergence. The aim of this study was to determine the activity of the endo--mannanase enzyme in the structures of coffee seeds for the purpose of assessing the relationship between this activity and the physiological quality of the seeds under different processing and drying methods. Coffea arabica L. fruit in the cherry maturity stage was subjected to three different types of processing: natural (seeds maintained in the fruit itself), fully washed (fruit pulped mechanically and the seeds demucilaged by fermentation in water), and semi-washed or demucilaged (both fruit pulp and mucilage removed mechanically); and two methods of drying: slow drying (suspended screen) in the shade, and rapid drying in mechanical dryer at 35°C to a moisture content of 11±1%. After processing and drying, the physiological quality of the seeds was evaluated through the germination test, and endo--mannanase enzyme activity was quantified. Coffee seeds submitted to natural processing have lower physiological performance, as well as greater deterioration and greater activity of the endo-ß-mannanase enzyme. Removal of mucilage during fully washed and semi-washed processing of coffee seeds reduces the activity of the endo-ß-mannanase enzyme and lowers deterioration, especially after faster drying. The enzyme endo-ß-mannanase is efficient in studying of the effects of processing and drying on coffee seeds, and can be evaluated in whole seeds, endosperms or embryos.(AU)

Sementes de café submetidas ao processamento natural tem menor desempenho fisiológico, assim como maior deterioração e maior atividade da enzima endo-ß-mananase, principalmente após secagem mais rápida. A remoção da mucilagem durante o processamento despolpado e desmucilado de sementes de café reduz a atividade da enzima endo-ß-mananase e também a deterioração. A secagem rápida em secador afeta negativamente a qualidade das sementes. ABSTRACT The development of coffee plant root system changes when subjected to drip irrigation and fertigation. This work aimed to evaluate the effects of different levels of fertigation on the development of coffee root system by drip irrigation in western Bahia. The experiment was carried out with Catuaí Vermelho IAC 144 coffee plants, of about 3.5 years of age, in the "Café do Rio Branco" farm, located in Barreiras - BA, and consisted of a complete randomized blocks design with 3 replicates. Treatments consisted of three levels of nitrogen and potassium fertilization (900/800, 600/500 and 300/250 kg ha-1 year-1 N and K2O), weekly distributed, by means of fertigation, throughout the process. After the fourth harvest, coffee root system was evaluated, and root length density (RLD) and root density (RD) were determined at different sampled layers. The highest root concentration, root length density (RLD), and root density (RD) were observed in the superficial layers of soil (0-20 cm), and under the dripline (30 and 70 cm from the orthotropic branch). Results showed that the lower the N and K2O levels, the higher was the development (RLD and RD) of the coffee root system.


RESUMO O desenvolvimento do sistema radicular do cafeeiro é alterado quando submetido à irrigação localizada e fertirrigação. Este trabalho foi desenvolvido com o objetivo de avaliar os efeitos de diferentes doses de fertirrigação sobre o desenvolvimento do sistema radicular do cafeeiro irrigado por gotejamento na região Oeste da Bahia. Realizou-se o trabalho na fazenda Café do Rio Branco, localizada em Barreiras - BA, em cafeeiros de aproximadamente 3,5 anos de idade, da variedade Catuaí IAC 144. O experimento foi montado em um esquema fatorial 3 x 3, sendo três níveis de adubação nitrogenada e potássica (900/800, 600/500 e 300/250 kg ha-1 ano-1 de N e K2O), em 3 parcelamentos mensais de fertirrigação (2, 4 e 8 vezes). Após a quarta safra, o sistema radicular do cafeeiro foi avaliado e a densidade de comprimento radicular (DCR) e densidade radicular (DR) em diferentes camadas amostradas foram determinadas. A maior concentração de raízes, densidade de comprimento radicular (DCR) e densidade radicular (DR) foi obtida nas camadas superficiais (0-20 cm) e sob a linha lateral (30 e 70 cm de distância do ramo ortotrópico). As menores doses de N e K2O propiciaram maior desenvolvimento (DCR e DR) do sistema radicular do cafeeiro.
