RESUMO

Crambe meal (CM) is a potential dietary protein source for ruminant, but its effects in diets for lactating dairy cows remains unknown. We evaluated the effects of inclusion of the low-glucosinolates (GIs) CM (450 mg GIs/kg DM) in partial total mixed ration (pTMR) on performance, efficiency of nutrient utilization, and hepatic function of crossbred Holstein × Zebu and Jersey cows. Eight crossbred Holstein × Zebu cows and four Jersey cows were blocked by breed and days in milk, and randomly assigned in a replicated 4 × 4 Latin square design, and distributed in one of four isonitrogenous TMRs (130 g CP/kg DM): 0, 45, 90, and 135 g CM/kg DM pTMR. Crambe meal was included in pTMR replacing soybean meal (SBM) and ground corn grain (GCG). The pTMRs were offered ad libitum between 7:00 a.m. and 5:00 p.m. Cows were kept on pasture of Panicum maximum cv. Mombaça (90.4 g CP/kg DM) between 6:00 p.m. and 6:30 a.m. Pasture intake was measured from external and internal fecal excretion marker. Inclusion of CM did not affect (P ≤ 0.05) the DM intakes of pTMR and pasture, apparent total-tract digestibilities of organic matter, CP and NDF, ruminal outflow microbial protein, milk yield, milk composition, urinary N excretion, milk N efficiency, and level of plasm hepatic enzymes. The effects of CM inclusion in pTMR were independent of breed. Low-glucosinolates CM can be incorporated up to 135 g/kg DM in pTMR in replacement of SBM and GCG without affecting performance, efficiency of nutrient utilization, and hepatic function of crossbred Holstein × Zebu and Jersey cows.

RESUMO

A variety of plant protein sources have been evaluated in aquafeeds. Crambe meal (CM) has potential for inclusion in fish diets because of its nutritional composition. This study evaluated the apparent digestibility coefficient (ADC) of crambe meal and its potential to partially replace soybean meal (SM) protein in Nile tilapia Oreochromis niloticus diets. The ADC for dry matter, crude protein, ether extract, energy, amino acids, calcium and phosphorus of CM were assessed in fish (n = 80; 65.30 ± 5.32 g). Subsequently, an 80-day feeding trial was conducted with Nile tilapia (n = 140; 6.04 ± 0.25 g) randomly distributed in 20 experimental cages (70 L; seven fish cage-1) allocated in five circular tanks (1000 L) in a recirculation water system, to evaluate the effects of replacement of SM by CM (0, 6, 12, 18 and 24% in isonitrogenous and isoenergetic diets) on growth, blood parameters, fillet yield and proximal composition. The CM shows good digestibility of protein (0.824) and amino acids (0.844) by Nile tilapia and its inclusion in the diet does not affect carcass and fillet yield or proximal composition. Fish fed diets with 24.0% of the SM replaced by CM showed the worst weight gain and feed conversion rate. The protein efficiency ratio decreased in fish fed diets with 12.0, 18.0 and 24.0% of the SM replaced by CM. Hemoglobin, mean corpuscular hemoglobin concentration, total plasma protein, glucose and alanine aminotransferase enzyme activity trend to increase at highest levels of CM in the diet. In conclusion, CM has high digestibility of protein and amino acids for Nile tilapia. However, anti-nutritional factors present in untreated CM interfere on the growth and nutrient utilization of Nile tilapia.

RESUMO

Crambe (Crambe abyssinica H.), a promising species in the production of biodiesel, has low planting cost, high oil content and easy extraction, besides being an alternative for the crop rotation system. The objective of this study was to evaluate the effects of nitrogen fertilization and line spacing on the production components and yield of the crambe crop under semi-arid conditions. The study was conducted under field conditions in the experimental area of the National Semi-Arid Institute-INSA, located in the city of Campina Grande-PB. The experimental design was a randomized complete block design in 4 x 2 factorial scheme, consisting of four nitrogen doses (30, 60, 90, 120 kg ha-1) with two spacings between planting lines (0.20 and 0.40 meters) with three replications, totaling 24 experimental plots. The evaluated parameters were: number of siliques per plant, dry weight of shoots, number of branches (primary and secondary), primary stem length, number of siliques per stem, mass of 1000 grains and productivity. The data were submitted to analysis of variance, in which the nitrogen doses (quantitative factor) were compared by regression analysis. The different spacings were compared by means of a t-test at the 0.05 probability level by using statistical software SISVAR version 5.2. Nitrogen fertilization at 0.40 m spacing between the planting lines led to the best results for the production components, and the highest productivity was obtained at 0.20 m spacing of with nitrogen fertilization of 120 kg ha-1.

The search for vegetal species regarding effectiveness in the phytoremediation of soils is of great importance, mainly in function of the great environmental problems, such as soil contamination with heavy metals, the necessity of producing more food, among others that mankind face today. This work aimed (i) to evaluate phytoremediation capacity of Crambe abyssinica Hochst and its growth in soil artificially contaminated with Cd and Pb, and (ii) to evaluate the possible impacts of crambe cultivation in contaminated soil conditions, in order to evaluate, to test, and to question the Brazilian CONAMA 420, providing important information that can be useful for governmental and environmental purposes. Two simultaneous experiments were developed, one for each metal. The soils were contaminated with salts of CdCl2 and PbCl2H2O in five doses based on the investigation values (IV) of CONAMA Resolution 420, resulting in 0; 1.5; 3; 9 and 30 mg kg-1 for Cd and 33; 90; 180; 540 and 1800 mg kg-1 for Pb. Gaseous exchange, development, nutritional composition and production of plant components, as well as phytoavailability of metals, were evaluated. The contamination with metals reduced photosynthesis, increased breathing as well as leading to a negative effect on the mineral nutrition and productivity in general; Plants cultivated in soil with Cd presented higher phytoavailability when compared to those cultivated in the Pb conditions, being found metals in all parts of the crambe plants from 1.5 mg kg- 1 of Cd in the soil; and Pb was retained only in roots, not being translocated in the plant. Cd showed higher phytoavailability, being found in all parts of the plant and Pb was retained only in the roots. Cd showed a higher phytoavailability when compared to Pb, also being found in all parts of crambe plants from dose 1.5 mg kg-1 of Cd in soil, which is an environmental problem, since in these concentrations the cultivation of crops is allowed by Brazilian legislation CONAMA 420.

RESUMO

This study aimed to characterize the chemical composition of crambe grains produced with plant growth regulators in the 2014 and 2015 harvests and stored for 180 days. During crambe cultivation, two foliar sprayings were carried out with the following treatments: 1) control treatment with distilled water; 2) indole-3-acetic acid 100 mg L-1 (IAA); 3) 3-gibberellic acid P.A. 100 mg L-1 (GA3); 4) Stimulate®. After harvesting, the grains were analyzed initially and after 180 days of uncontrolled storage. We checked water content, lipids in hot extraction (HE), lipids in cold extraction (CE), proteins, acidity and antioxidant activity. The Tukey test was used to compare the averages (p<0.05) and the parameters were grouped using a multivariate clustering technique presented in dendogram. The CE and HE lipid contents were reduced after storage, whereas the protein content and antioxidant activity of the grains were not altered by storage. However, the grains that received IAA application showed an increase in the CE lipid content and their preservation after storage. GA3 application, on the other hand, reduced the protein content. Lipid contents remained constant at the end of storage, and the acidity was reduced due to the application of GA3 and Stimulate® plant growth regulators. The 180-day storage changed crambe quality and regulators can assist in preserving grain quality during storage. The CE and HE lipid content and the grain acidity were good indicators for evaluating effects.

As an important industrial feedstock, wax esters (WEs) have been used as lubricants in a number of technical processes. There is however currently no large-scale biological source for WE production and alteration in metabolic pathways of plant oils for producing WEs could be attractive to the commercial markets. Here, we present the breeding results of long-term studies on successful development of new crambe lines producing WEs through genetic engineering and cross breeding. The transgenic crambe lines producing WEs at over 25% of the total seed oil were first generated by introduction of the jojoba WE biosynthetic genes ScFAR and ScWS. Further improvement of the lines aiming at improving oxidative stability of WEs was achieved through introducing the CaFAD2-RNAi gene into these lines by crossing. The hybrid lines possessed similar agronomic traits to the wild type and a stable level of WEs over several generations, suggesting a high potential of crambe as an industrial crop for WE production.

RESUMO

The application of plant growth regulators can manipulate the development of cultivated plants, and their physiological effects may be reflected in the physiological quality of the seeds. This study evaluated the effects of plant growth regulators (PGR) on the physiological quality of crambe seeds produced in the 2014 and 2015 harvests. During crop development, we applied two foliar sprayings of the following treatments: 1) control treatment with distilled water; 2) indole-3-acetic acid 100 mg L-1 (IAA); 3) 3-gibberellic acid P.A. 100 mg L-1 (GA3); 4) commercial PGR Stimulate® 6 mL L-1. The seeds were harvested and evaluated for water content, germination percentage and germination speed index (GSI), electrical conductivity and concentrations of leached Ca, Mg and K ions. Means were compared by Tukey ́s and Dunnett tests (p<0.05). Applying Stimulate® increased the germination percentage and the germination speed index, while IAA application showed an opposite effect, increasing the electrical conductivity of crambe seeds. Calcium leaching decreased with IAA application, and all regulators reduced K leaching. We therefore infer that the application of plant growth regulators influences the physiological quality of crambe seeds.

RESUMO

Raw vegetable oil from Crambe abyssinica was subjected to oxidative treatment to enhance its viscosity. The oxidation processes were carried out in the presence of N-hydroxyphthalimide with or without supercritical CO2 as a solvent. Four spectroscopic techniques (Raman, UV-VIS, FT-IR, NMR) were applied to assess the chemical changes taking place during the oxidation. Raman and NMR spectroscopy proved best in the assessment of the chemical transformations leading to increased viscosity of the modified vegetable oil.

RESUMO

Lubricants that are based on renewable raw materials have drawn increased attention in various applications, especially those related to the food industry. Due to the high requirements of environmental protection, there is a need to develop biodegradable base oils that are environmentally friendly and do not contain harmful components. The objective of the research was to obtain a base oil with a certain viscosity and certain desired lubricating properties. Base oils were obtained from Crambe abyssinica oil by means of blending with synthetic oil and oxidation. The oxidation processes were carried out in the presence of N-hydroxyphthalimide with or without CO2 as a solvent. As a final result of this study, oil bases meeting the viscosity requirements and showing suitable lubricating properties were obtained. The Raman spectra of the obtained oils were evaluated.

RESUMO

We studied the feed intake and milk production of Holstein × Zebu crossbred cows fed sorghum silage diets and concentrates, with and without the addition of crambe meal. Using a change-over design for a total of 120 days, eight cows were fed two diets (concentrates + silages with and without crambe meal) in four 30-day periods, with four replications (animals). The crambe meal diet increased (P < 0.05) the dry matter intake (DMI), crude protein intake (CPI), and non-fibrous carbohydrates intake (NFCI) as well as milk production (4.3%). No difference was noted (P > 0.05) for the milk composition between the treatments. Crambe meal as an additive in sorghum silage (100 g/kg in natural matter) showed a great potential for introduction in dairy farming as it substantially increases CP intake, does not reduce food intake, raises animal productivity, and does not affect milk composition.

RESUMO

Crambe is an important crop grown worldwide for industrial oil and seed meal. Besides the fungal and bacterial diseases, the crop is reported to be infected by tobacco mosaic virus, beet western yellows virus and turnip mosaic virus under experimental condition. Till now, there was no report of natural infection of any begomovirus in this crop. In the present study, a leaf curl disease was observed in germplasm accessions of three species of Crambe (C. abyssinica, C. glabrata and C. hispanica). Based on the symptoms and presence of whitefly population in the field, begomovirus infection was suspected. Molecular characterization through RCA approach, indicated presence of croton yellow vein mosaic virus (CYVMV, KJ747958) and croton yellow vein mosaic betasatellite (CroYVMB, KM229762). Co-agroinoculation of partial dimeric construct of CYVMV with complete dimeric construct of CroYVMB, produced typical leaf curl symptoms in C. abyssinica, whereas, agroinoculation of partial dimeric construct of CYVMV alone could not produce symptoms in the same plant. In contrast, the CYVMV construct alone could produce symptom in Nicotiana benthamiana, a model host for plant virus studies. In N. benthamiana co-inoculation of CroYVMV with CYVMV construct develop more severe symptoms. However, neither the CYVMV construct alone nor the co-inoculation with CroYVMB produce any symptom in Arabidopsis thaliana even with different methods of inoculation. Inoculated Arabidopsis thaliana also did not yield any amplification of the virus as assessed through PCR and rolling circle amplification (RCA). Thus it confirmed that for successful infection in crambe, CYVMV requires betasatellite, while in N. benthamiana, it does not require betasatellite for symptom induction and in Arabidopsis thaliana CYVMV alone or in presence of betasatellite did not replicate and produce any symptom. This study constitutes the first confirmed record of natural infection of a begomovirus in crambe and further confirmed that cognate betasatellite of CYVMV has differential role in infectivity in different hosts.

RESUMO

Low-calorie structured lipids (SLs) rich in behenic and oleic acids were produced by enzymatic (EI) and chemical interesterification (CI) of high oleic sunflower oil and fully hydrogenated Crambe abyssinica oil in proportions of 60:40, 50:50, 40:60 and 30:70 (w/w), respectively. The immobilized lipase from Thermomyces lanuginosus (Lipozyme TL IM) was used for 3 h at 70 °C at a dosage of 7% (w/w) free of organic solvents. The original blend (BE) and the reaction products were evaluated for their fatty acid (FA) and triacylglycerols (TAG) compositions, solid fat contents, thermal analysis, regiospecific distribution of the FA, microstructure and polymorphism. The tendency was towards a reduction in the C52, C54, C60, C62 and C64 TAG contents and an increase in the C56, C58 e C66 TAG contents, therefore the solid fat content was reduced. There was lower acyl migration at the sn-2 position of the TAGs in EI as compared to CI in all the blends ratios. Needle-like crystals were predominant in the IE while large symmetrical spherulites were observed in the BE. Reduction in the mean crystal diameter was observed in all the blends, in addition to modifications of the crystal morphology. X-ray diffraction analysis showed a predominance of the ß' form crystals in both the IE and CI. The SLs were produced for application in the food industry as bakery/confectionery fats in the proportions of 60:40/50:50 and as additive in the crystallization of lipids in the proportions of 40:60/30:70.

RESUMO

Biotechnological production of fatty alcohols, important raw materials in the chemical industry, has been receiving considerable attention in recent years. Fatty alcohols are formed by the reduction of fatty acyl-CoAs or fatty acyl-ACPs catalyzed by a fatty acyl reductase (FAR). In this study, we introduced genes encoding FARs from Arabidopsis thaliana (AtFAR5) and Simmondsia chinensis (ScFAR) into Crambe abyssinica hairy roots via Agrobacterium rhizogenes-mediated transformation. The efficiency of the transformation ranged between 30 and 45%. The fatty alcohols were only detected in the transgenic hairy root lines expressing ScFAR gene. In all tested lines stearyl alcohol (18:0-OH), arachidyl alcohol (20:0-OH), and behenyl alcohol (22:0-OH) were produced. The content of 18:0-OH varied from 1 to 3% of total fatty acids and fatty alcohols, while the amount of either 20:0-OH and 22:0-OH did not exceed 2%. The transgenic hairy root lines produced from 0.98 to 2.59 nmol of fatty alcohols per mg of dry weight. Very low activity of ScFAR was detected in the microsomal fractions isolated from the selected hairy root lines. To our knowledge, this is the first report on the fatty alcohol production in the hairy root cultures. Biotechnol. Bioeng. 2017;114: 1275-1282. © 2016 Wiley Periodicals, Inc.

RESUMO

BACKGROUND: The allohexaploid Crambe abyssinica (crambe) is an oilseed crop that has been recognized for its potential value in the chemical industry, particularly in terms of producing high-erucic acid content vegetable oil. However, as an understudied crop, improvement of crambe has been hampered by the lack of genetic and genomic information to enhance its yield, oil quality and resistance against biotic and abiotic stress. Development of molecular markers is therefore of great significance to facilitate genetic improvement of crambe. RESULTS: In this study, high-throughput sequencing was performed to generate sequences for the transcriptome and genome of a widely planted crambe cultivar, Galactica. A total of 186,778 expressed sequence tag (EST) contigs as 8,130,350 genomic contigs were assembled as well. Altogether, 82,523 pairs of primers were designed in the flanking sequences of the simple sequence repeat (SSR) within these contigs. Virtual PCR analysis showed that a fraction of these primers could be mapped onto the genomes of related species of Brassica, including Brassica rapa, B. oleraceae and B. napus. Genetic diversity analysis using a subset of 166 markers on 30 independent C. abyssinica accessions exhibited that 1) 95 % of the designed SSRs were polymorphic among these accessions; 2) the polymorphism information content (PIC) value of the markers ranged from 0.13 to 0.89; 3) the genetic distances (coefficient NEI72) between accessions varied from 0.06 to 0.36. Cluster analysis subsequent on the accessions demonstrated consistency with crambe breeding history. F-statistics analysis revealed a moderate level of genetic differentiation in C. abyssinica (Gst = 0.3934) and a accordingly low estimated gene flow (Nm = 0.7709). CONCLUSION: Application of high-throughput sequencing technology has facilitated SSR marker development, which was successfully employed in evaluating genetic diversity of C. abyssinica as demonstrated in our study. Results showed these molecular markers were robust and provided powerful tools for assessing genetic diversity and estimating crambe breeding history. Moreover, the SSR primers and sequence information developed in the study are freely available to the research community.

RESUMO

ABSTRACT: Crambe is a rapeseed with high oil content and can be used as a winter cover or as a source of raw material for the production of biodiesel, however espite the growing interest in the culture, research on the subject is still incipient, especially concerning the seed production and analysis technology. The purpose of this study is to evaluate the physiological quality of crambe seeds, 'FMS Brilhante' cultivar, by testing the pH of exudate. Five seed lots were submitted to the determination of water content and the tests of germination and vigor (first count, emergence and tetrazolium). In the conduction of pH exudate test, temperatures (25 and 30oC), and periods of seed imbibition in water (15, 30 and 45 minutes) were tested. The experiment was conducted in a completely randomized manner, with four replicates, and the mean values were compared by the Tukey test at 5% probability; Pearson correlation between the pH of the exudate and initial tests was also made. Testing the pH of exudate is promising for separating lots of crambe seeds and the following combinations of 25°C/30 minutes or 30°C/45 minutes can be used.

The aim of this study was to evaluate the effects of using crambe cake in diets for sheep on intake and digestibility. The experimental design was a 4x4 Latin square with four animals, four periods and four treatments. The treatments were diets with 30% sorghum silage and 70% crambe cake (30:70) and a diet with 70% sorghum silage and 30% crambe cake (70:30) in lambs under the voluntary and controlled intake. The animals who fed a diet with a 30:70 ratio consumed more dry matter (DM), crude protein (CP), ether extract (EE) per kg/day, %BW/day and g/kgPV0,75. The ration provided higher digestibility of DM and NDF. The animals in the voluntary regime presented higher consumption of DM, CP, EE per kg/day, %BW/day and g/kgPV0,75. The cake crambe presented digestibility coefficient higher than sorghum silage, regardless of the regime intake. The 30:70 ration presented a difference in N intake and N absorbed. The cake crambe has high nutrient digestibility and acceptance by animals when associated with sorghum silage.(AU)

RESUMO

The objective of this study was to evaluate the dry matter (DM) yield and nutrient accumulation in cover crops and the effect of these, as well as mechanical management of crop residues on growth and grain yield of crambe (Crambe abyssinica). The experiment was conducted in Santa Helena, Paraná, Brazil in Rhodic Hapludox (Red Latosol in the Brazilian classification) in experimental design of randomized complete blocks with four replications in a split-plot. The plots were: Pearl millet (Pennisetum glaucum), grass brachiaria (Brachiaria brizantha), forage sorghum (Sorghum bicolor) and sunn hemp (Crotalaria juncea) and an area fallow (spontaneous weed) and the sub-plots, the mechanical or not management of crop residues on the soil surface (Triton©). Grass brachiaria produced less amount of DM and showed less accumulation of nutrients. Other cover crops did not differ in DM production, but the sunn hemp showed the highest concentrations of N and Ca, while pearl millet P. The mechanical management of crop residues on the soil surface not influence the yield components of crambe, however provides higher final plant density. The crambe grown after sunn hemp showed higher DM accumulation (1,826 kg ha1) and grain yield (1,066 kg ha1).

The objective of this work was to determine the sorption isotherms of crambe fruits by dynamic method for several conditions of temperature and water activity and adjust different mathematical models to experimental data, and obtain the values of isosteric heat of desorption versus the equilibrium moisture content of the product. The equilibrium moisture content of crambe fruits was determined by dynamic-gravimetric method to temperatures of 25, 30, 35, 40 and 45 °C and water activity for each temperature, from 0.270 to 0.825. From the results obtained it was calculated for each isosteric heat of moisture content equilibrium. The experimental data were adjusted several models available in the literature (Chung-pfost, Copace, GAB, Modified Halsey, Sabbah, Sigma Copace, Cavalcanti Mata, Modified Henderson, Henderson, BET, Oswin e Modified GAB). It was observed that the equilibrium moisture content decreases with increasing temperature for a given water activity like the hygroscopic products. The model Sigma Copace was best described what the hygroscopicity of the crambe fruits. It was observed that the isosteric heat increases with decreasing moisture content of the product, indicating an increase in energy required for water removal. The values of isosteric heat of crambe fruits in the range of moisture content from 3.85 to 11.86 (d.b.%) varied from 4731.73 to 2698.35 kJ kg-1.

Crambe abyssinica is a hexaploid oil crop for industrial applications. An increase of erucic acid (C22:1) and reduction of polyunsaturated fatty acid (PUFA) contents in crambe oil is a valuable improvement. An increase in oleic acid (C18:1), a reduction in PUFA and possibly an increase in C22:1 can be obtained by down-regulating the expression of fatty acid desaturase2 genes (CaFAD2), which code for the enzyme that converts C18:1 into C18:2. We conducted EMS-mutagenesis in crambe, followed by Illumina sequencing, to screen mutations in three expressed CaFAD2 genes. Two novel analysis strategies were used to detect mutation sites. In the first strategy, mutation detection targeted specific sequence motifs. In the second strategy, every nucleotide position in a CaFAD2 fragment was tested for the presence of mutations. Seventeen novel mutations were detected in 1100 one-dimensional pools (11 000 individuals) in three expressed CaFAD2 genes, including non-sense mutations and mis-sense mutations in CaFAD2-C1, -C2 and -C3. The homozygous non-sense mutants for CaFAD2-C3 resulted in a 25% higher content of C18:1 and 25% lower content of PUFA compared to the wild type. The mis-sense mutations only led to small changes in oil composition. Concluding, targeted mutation detection using NGS in a polyploid was successfully applied and it was found that a non-sense mutation in even a single CaFAD2 gene can lead to changes in crambe oil composition. Stacking the mutations in different CaFAD2 may gain additional changes in C18:1 and PUFA contents.
