RESUMO

The purpose of the work is to evaluate the cultural and morphological properties of colonies of clinically significant corynebacteria on culture mediums for the isolation of corynebacteria. The study used 9 culture mediums for the isolation of corynebacteria: a culture medium for the isolation of corynebacteria (Corynebacagar); Tellurite-containing blood agars on base - Culture medium № 1 GRM, Culture agar for the cultivation of microorganisms (GRM agar), Culture medium for determining the sensitivity of microorganisms to antibacterial preparations - AGV, culture agar for the cultivation of dry microorganisms (SPA), Clauberg medium II, Hoyle Medium agar (Oxoid), Blood agar base (Conda), Columbia Agar Base (Conda). The work used 7 test strains of microorganisms from the State collections of pathogenic microorganisms - C. diphtheriae biovars gravis, mitis, intermedius, belfanti and subspecies lausannense, C. ulcerans and C.pseudotuberculosis. Studies were carried out in accordance with MUK 4.2.3065-13 «Laboratory diagnosis of diphtheria infection¼. We describe culture-morphological properties of strains on all tested culture mediums the isolation of corynebacteria after 24 and 48 hours of incubation. Analysis of the results on the growth properties of culture mediums showed that all culture mediums had high sensitivity - from dilution 10-7 for all test strains. Colonies of corynebacteria were visually detected on culture mediums after 19-20 hours of cultivation. When cultivating a suspension of corynebacteria from breeding 10-6 on culture mediums, the number of colonies ranged from 95±5 to 120±10. Conclusion. All culture mediums had differential diagnostic properties that ensure the growth of corynebacteria after the day of incubation.

ABSTRACT This is a retrospective study of 160 patients treated in private practice in São Paulo from March 2003 to March 2015. We analyzed 171 results of direct mycological examinations and fungal cultures from nail scrapings. The agreement between direct mycological examination results and fungal culture was satisfactory, consistent with the literature (kappa 0.603). The main agent identified was Trichophyton rubrum (51%). We observed an isolation rate of non-dermatophyte filamentous fungi superior to the literature (34%). Determining the agent is key to defining the appropriate onychomycosis treatment, and knowing the epidemiology of patients treated in private practice helps the dermatologist who works in this context.

Objective To evaluate the BacT/ALERT 3D liquid rapid culture system for the rapid detection of mycobacterium tuberculosis(MTB)and early diagnosis of pulmonary tuberculosis.Methods BacT/ALERT 3D liquid culture mediums and lowenstein-jensen(L-J)solid culture mediums were applied to detect Mycobacterium tuberculosis sputum specimens respectively.Analysis of detection results and detection time were also performed.Results Average positive days of BacT/ALERT 3D liquid culture medium was(13 ±2.05)days,which was shorter than that of L-J solid culture mediums(34.7 ±4.76 )d (P<0.05 ).Compared to the L -J solid culture mediums,BacT/ALERT 3D liquid culture medium had higher positive rate for 59 patients whose sputum smear test was positive,and the positive rate were 71.18%(42/59)and 67.80%(40/59)respectively(P<0.05).BacT/ALERT 3D liquid culture medium had higher positive rate than L-J solid culture mediums for 106 patients whose sputum smear test was negative,and the positive rate were 39.62%(42/106)and 26.42%(28/106)respectively(P<0.05).Conclusion Compared to the traditional L-J solid culture system,BacT/ALERT 3D liquid culture system can shorten detection time and improve the positive detection rate of MTB in specimens with low concentration.

RESUMO

Objective To develop a new method for sterilization in agars culture mediums,i.e.Agars culture mediums is sealed to be sterilized and preserved after resolution.Methods The effect of rudimental Ethylene Epoxide was investigated,and the germiculture results by different preserving methods were analyzed.Air sedimentation bacterium was sampled by different methods to analyze the bacterium capture rate.Results Ethylene Epoxide sterilized agars culture mediums bacterium capture rate was similar with regular agars.Conclusion Ethylene Epoxide sterilized agars culture mediums can be used in sampling of clinical treatment and other outburst incident,reducing the preparation time.
