Benthic dinoflagellates produce potent toxins that may negatively affect humans and the marine biota. Understanding the factors that stimulate their growth is important for management strategies and to reduce their potential negative impacts. Laboratory cultures have been extensively used to study microalgae physiology and characterize life cycles, nutrition, growth rates, among other processes. A systematic review of the literature on the growth parameters of the benthic dinoflagellates Ostreopsis cf. ovata, Prorocentrum lima species complex and Coolia malayensis obtained in laboratory cultures of strains isolated from all over the world was performed. The effects of temperature, light intensity, photoperiod, salinity and culture media on the growth rate of these species were evaluated using multiple regressions and a model selection approach, based on the Akaike Information Criteria (AIC). The potential effects of the initial culture abundance and the media volume used on the growth of the species were also assessed. Data from 50 articles (25 for O. cf. ovata, 21 for P. lima and 6 for C. malayensis), resulting in 399 growth parameter values (growth rate, doubling time and maximum yield) were compiled in a database. The genetic clades of O. cf. ovata and P. lima species complex were also noted. Growth rate was the most frequently reported growth parameter for the three species, and 127 values were retrieved for O. cf. ovata, 90 for P. lima and 56 for C. malayensis. Temperature was the factor that best explained the growth response of P. lima and C. malayensis, whereas for O. cf. ovata, temperature and salinity were equally important. Light intensity and photoperiod were included among the six best models for the studied species but presented a weaker effect on growth. Given the observed and future projected climate change, increasing ocean temperature will promote the growth of these species, likely leading to an expansion of their impacts on ecosystems and human health. The use of common garden experiments using multiple strains from different geographic domains, particularly addressing underrepresented lineages is recommended, as they will provide more balanced insight regarding the species physiological responses to environmental drivers.

Public awareness about Benthic Harmful Algal Blooms (BHABs) and their negative impacts has increased substantially over the past few decades. Even so, reports of BHABs remain relatively scarce in South America (SA). This paper provides a comprehensive overview of the current state of knowledge on BHABs in the continent, by integrating data from published articles, books, and technical reports. We recorded ∼300 different occurrences of potentially toxic BHAB species over the Caribbean, Atlantic and Pacific coasts, mostly in marine (>95%) but also in estuarine areas located from 12°36' N to 54°53' S. Over 70% of the data was published/released within the past 10 years, and ∼85% were concentrated in Brazil, Venezuela, Ecuador and Colombia. Benthic species were mainly associated with macroalgae, seagrass and sediment. Incidental detection in the plankton was also relevant, mainly in places where studies targeting BHAB species are still rare, like Argentina, Uruguay, Chile and Peru. The study listed 31 infrageneric taxa of potentially toxic benthic dinoflagellates and eight of estuarine cyanobacteria occurring in SA, with the greatest species diversity recorded in the equatorial-tropical zone, mainly in northeastern Brazil (Atlantic), Venezuela and Colombia (Caribbean), and the Galapagos Islands, Ecuador (Pacific). Local strains of Amphidinium, Gambierdiscus, Coolia and Prorocentrum spp. produced toxic compounds of emerging concern. Prorocentrum lima species complex was the most common and widely distributed taxon, followed by Ostreopsis cf. ovata. In fact, these two dinoflagellates were associated with most BHAB events in SA. Whereas the former has caused the contamination of multiple marine organisms and cases of Diarrhetic Shellfish Poisoning in subtropical and temperate areas, the latter has been associated with faunal mortalities and is suspected of causing respiratory illness to beach users in tropical places. Ciguatera Poisoning has been reported in Colombia (∼240 cases; no deaths) and Venezuela (60 cases; two deaths), and may be also a risk in other places where Gambierdiscus spp. and Fukuyoa paulensis have been reported, such as the Galapagos Islands and the tropical Brazilian coast. Despite the recent advances, negative impacts from BHABs in SA are intensified by limited research/training funding, as well as the lack of official HAB monitoring and poor analytical capability for species identification and toxin detection in parts of the continent.

In September and November 2016, eight marine sampling sites along the coast of the southeastern Gulf of Mexico were monitored for the presence of lipophilic and hydrophilic toxins. Water temperature, salinity, hydrogen potential, dissolved oxygen saturation, inorganic nutrients and phytoplankton abundance were also determined. Two samples filtered through glass fiber filters were used for the extraction and analysis of paralytic shellfish toxins (PSTs) by lateral flow immunochromatography (IFL), HPLC with post-column oxidation and fluorescent detection (FLD) and UHPLC coupled to tandem mass spectrometry (UHPLC-MS/MS). Elevated nutrient contents were associated with the sites of rainwater discharge or those near anthropogenic activities. A predominance of the dinoflagellate Pyrodinium bahamense was found with abundances of up to 104 cells L-1. Identification of the dinoflagellate was corroborated by light and scanning electron microscopy. Samples for toxins were positive by IFL, and the analogs NeoSTX and STX were identified and quantified by HPLC-FLD and UHPLC-MS/MS, with a total PST concentration of 6.5 pg cell-1. This study is the first report that confirms the presence of PSTs in P. bahamense in Mexican waters of the Gulf of Mexico.

Marine phycotoxins are organic compounds synthesized by some species of microalgae, which accumulate in the tissues of filter-feeder organisms such as bivalve mollusks. These toxins can cause acute intoxication episodes in humans, a severe threat to aquaculture and fisheries. In the State of Pará, Brazil, oyster farming has community, artisanal and sustainable bases, using mangroves as cultivation environment and seed banks. In small-scale production, there are often no established methods of safeguarding the health of consumers elevating the potential risks of shellfish poisoning outbreaks. Our study evaluated the presence of phycotoxins in oysters cultivated in five municipalities in the region of the Atlantic Amazon (Pará, Brazil) assessing the quality of the final product. We further evaluated the microalgae, water quality, and the spatio-temporal variation of physicochemical factors in the same area. Diatoms dominated the microalgae composition, followed by dinoflagellates, some of which are reported to be potentially toxic and producers of paralytic shellfish toxins. For the first time, we describe the occurrence of the potentially toxic dinoflagellate Ostreopsis sp. in the Amazon region. Furthermore, for the first time, toxins were detected in oyster farming in the northeast of the State of Pará, namely GTX2,3, STX, and dc-STX nevertheless, with nontoxic values. The identified toxins represent a potential threat to shellfish consumers.

To describe the phytoplankton species composition, spatio-temporal distribution and habitats during different seasons along the central coast of the state of Campeche, Mexico, southeastern Gulf of Mexico, eight shallow-water (ca 1 m) sites from the city of Campeche Southwest to Villamar were monitored monthly from September 2016 to June 2017 by taking water-bottle samples. Average water temperature varied between 22.2 and 30.9°C and average salinity between 26.6 and 35.0. The Kolmogorov-Smirnov test, the Bartlett's test and canonical correspondence analyses were applied. Sixteen potentially harmful microalgal species were found: five species are bloom-forming, nine are potentially toxic to humans, and two affect aquatic organisms. At all sampling sites, massive algal proliferations occurred in June (beginning of rainy season) caused by the dinoflagellates Heterocapsa sp. (2.6 × 105 cells/L) and Blixaea quinquecornis (2.0 × 104 cells/L) and from October to March (windy season) by the cyanobacterium Trichodesmium sp. (2.6 × 105 cells/L).

During our conveying the microbial structures of phycosphere microbiota (PM) derived from diverse marine harmful algal bloom (HAB) dinoflagellates, a new rod-sharped, white-colored cultivable bacterial strain, designated as LZ-15-2, was isolated from the PM of highly toxic Alexandrium catenella LZT09. Phylogenetic analysis of 16S rRNA gene sequence indicated that strain LZ-15-2 belonged to the genus Marivita within the family Rhodobacteraceae, and demonstrated the highest gene similarity of 99.2% to M. cryptomonadis CL-SK44T, and less than 98.65% with other type strains of Marivita. Phylogenomic calculations on average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between the new isolate and M. cryptomonadis CL-SK44T were 99.86% and 99.88%, respectively. Genomic comparison of strain LZ-15-2 with available genomes of Marivita species further verified its taxonomic position within the genus of Marivita. Moreover, comparative genomics analysis showed a proximal similarity of strain LZ-15-2 with M. cryptomonadis CL-SK44T, and it also revealed an open pan-genome status based on constructed gene accumulation curves among Marivita members with 9,361 and 1,712 genes for the pan- and core-genome analysis, respectively. Based on combined polyphasic taxonomic characteristics, strain LZ-15-2 represents a new member of M. cryptomonadis, and proposed as a potential candidate for further exploration of the detailed mechanisms governing the dynamic cross-kingdom algae-bacteria interactions (ABI) between PM and their algal host LZT09.

Most of the shellfish fisheries of Mexico occur in the Gulf of California. In this region, known for its high primary productivity, blooms of diatoms and dinoflagellates are common, occurring mainly during upwelling events. Dinoflagellates that produce lipophilic toxins are present, where some outbreaks related to okadaic acid and dinophisystoxins have been recorded. From January 2015 to November 2017 samples of three species of wild bivalve mollusks were collected monthly in five sites in the southern region of Bahía de La Paz. Pooled tissue extracts were analyzed using LC-MS/MS to detect lipophilic toxins. Eighteen analogs of seven toxin groups, including cyclic imines were identified, fortunately individual toxins did not exceed regulatory levels and also the total toxin concentration for each bivalve species was lower than the maximum permitted level for human consumption. Interspecific differences in toxin number and concentration were observed in three species of bivalves even when the samples were collected at the same site. Okadaic acid was detected in low concentrations, while yessotoxins and gymnodimines had the highest concentrations in bivalve tissues. Although in low quantities, the presence of cyclic imines and other lipophilic toxins in bivalves from the southern Gulf of California was constant.

Dinophytes are widely distributed in marine- and fresh-waters, but have yet to be conclusively documented in terrestrial environments. Here, we evaluated the presence of these protists from an environmental DNA metabarcoding dataset of Neotropical rainforest soils. Using a phylogenetic placement approach with a reference alignment and tree, we showed that the numerous sequencing reads that were phylogenetically placed as dinophytes did not correlate with taxonomic assignment, environmental preference, nutritional mode, or dormancy. All the dinophytes in the soils are rather windblown dispersal units of aquatic species and are not biologically active residents of terrestrial environments.

During the southern summer of 2020, large phytoplankton blooms were detected using satellite technology in Chile (western Patagonia), where intensive salmonid aquaculture is carried out. Some harvesting sites recorded massive fish mortalities, which were associated with the presence of the dinoflagellate species Cochlodinium sp. The bloom included other phytoplankton species, as Lepidodinium chlorophorum, which persistently changed the colour of the ocean to green. These blooms coincided with the government-managed emergency lockdown due to the COVID-19 pandemic. Local in situ sampling was slowed down. However, imagery from the Copernicus programme allowed operational monitoring. This study shows the benefits of both Sentinel-3 and Sentinel-2 satellites in terms of their spectral, spatial and temporal capabilities for improved algal bloom monitoring. These novel tools, which can foster optimal decision-making, are available for delivering early alerts in situations of natural catastrophes and blockages, such as those occurred during the global COVID-19 lockdown.

Ciguatera is a food intoxication caused by the consumption of primarily coral fish; these species exist in large numbers in the seas that surround the Colombian territory. The underreported diagnosis of this clinical entity has been widely highlighted due to multiple factors, such as, among others, ignorance by the primary care practitioner consulted for this condition as well as clinical similarity to secondary gastroenteric symptoms and common food poisonings of bacterial, parasitic or viral etiology. Eventually, it was found that people affected by ciguatoxins had trips to coastal areas hours before the onset of symptoms. Thanks to multiple studies over the years, it has been possible to identify the relation between toxigenic dinoflagellates and seagrasses, as well as its incorporation into the food chain, starting by fish primarily inhabiting reef ecosystems and culminating in the intake of these by humans. Identifying the epidemiological link, its cardinal symptoms and affected systems, such as gastrointestinal, the peripheral nervous system and, fortunately with a low frequency, the cardiovascular system, leads to a purely clinical diagnostic impression without necessitating further complementary studies; in addition, what would also help fight ciguatera poisoning is performing an adequate treatment of the symptoms right from the start, without underestimating or overlooking any associated complications.

Introducción: La bioluminiscencia es la capacidad de ciertos organismos para transformar la energía química en energía lumínica mediante varios procesos bioquímicos. Objetivo: El aislamiento e identificación por primera vez de bacterias luminiscentes en agua marina superficial y la identificación de dinoflagelados luminiscentes marinos del Parque Nacional Isla del Coco, Costa Rica. Metodología: Se colectaron muestras de agua marina obtenida por buceo a 20 m y a nivel superficial de 13 sitios en la Isla del Coco, Costa Rica. Por otra parte, se analizaron muestras de fitoplancton colectadas desde la superficie hasta los 30 m de profundudad en los alrededores de 8 sitios de la Isla del Coco, y se determinaron varias especies luminiscentes pertenecientes a los géneros Ornithocercus y Ceratocorys. Resultados: Se logró obtener 7 aislados bacterianos luminiscentes, se identificaron y caracterizaron bioquímicamente mediante una plataforma automatizada (Vitek) con altos niveles de confianza, se ubicaron taxonómicamente dentro del género Vibrio,2 especies: V. alginolyticus y V. parahaemolyticus, además, algunos aislados presentaron resistencia al antibiótico ampicilina y 100% capacidad hemolítica. Esta investigación muestra evidencia de la presencia de especies microscópicas marinas en Isla del Coco, Costa Rica, capaces de presentar el fenómeno de la luminiscencia, por lo que profundizar en su estudio sería relevante en cuanto a la importancia de estos microorganismos en la producción de metabolitos secundarios y como indicadores de floraciones algales nocivas, por lo que se hace necesario realizar más investigación científica para determinar su potencial biotecnológico. Conclusiones: De la misma forma, los resultados obtenidos en esta investigación sugieren expandir las localidades de colecta y aislamientos de microorganismos luminiscentes, acompañado de una caracterización bioquímica y molecular, con el fin de explorar la diversidad microbiana asociada a eventos de luminiscencia y determinar los ambientes en el que estas especies se desarrollan.


Introduction: Bioluminescence is the ability of certain organisms to transform chemical energy into light energy through various biochemical processes. Objective: Isolation and identification for the first time of luminescent bacteria of superficial marine water, and the identification of marine luminescent dinoflagellates of Isla del Coco National Park, Costa Rica. Methods: Samples of seawater obtained by diving at 20 m and at a surface level of 13 sites were collected. On the other hand, phytoplankton samples collected from the surface up to 30 m deep were analyzed in the surroundings of 8 sites of Cocos Island, and several luminescent species belonging to the genera Ornithocercus and Ceratocorys were determined. Results: Seven luminescent bacterial isolates were obtained, they were identified and characterized biochemically by means of an automated platform (Vitek) with high levels of confidence, they were taxonomically located within the genus Vibrio, 2 species: V. alginolyticus and V. parahaemolyticus, in addition, some isolates presented resistance to the antibiotic ampicillin and 100% hemolytic capacity. This research shows evidence of the presence of marine microscopic species in Cocos Island, Costa Rica, capable of presenting the phenomenon of luminescence, so that further study would be relevant in terms of the importance of these microorganisms in the production of metabolites secondary and as indicators of harmful algal blooms, so it is necessary to conduct more scientific research to determine their biotechnological potential. Conclusions: In the same way, the results obtained in this investigation suggest expanding the collection and isolation of luminescent microorganisms, accompanied by a biochemical and molecular characterization, in order to explore the microbial diversity associated with luminescence events and determine the environments in which that these species develop.

Ostreopsis is a benthic dinoflagellate genus comprising eleven species including seven that are toxigenic. Ostreopsis lenticularis was first described from French Polynesia and is widely distributed in many insular locations from the Pacific Ocean. The current study presents morphological and molecular evidence of the presence of O. lenticularis at the Fernando de Noronha Archipelago, South Atlantic. Light microscopy and scanning electron microscopy of field cells showed morphological features in agreement with the description of O. lenticularis. Cells were broadly oval, 66.0-117.5 µm in dorso-ventral diameter, 50.0-92.5 µm in width, with thecal plates presenting two kinds of pores. Phylogenetic analyses based on LSU (D1-D3) and ITS sequences from field cells from Fernando de Noronha clustered with O. lenticularis sequences from elsewhere. The species was present in low to high abundances. The current study expands the distribution of O. lenticularis to Fernando de Noronha Archipelago in the South Atlantic Ocean.

Coolia is a genus of marine benthic dinoflagellates which is widely distributed in tropical and temperate zones. Toxicity has been reported in selected Coolia species, although the identity of causative compounds is still controversial. In this study, we investigated the taxonomical and toxicological aspects of Coolia species from Brazil. Since light- and electron microscopy-based morphology was not enough to distinguish small-celled species, ITS and LSU D1-D3 phylogenetic analyses were used for species definition. Cultures of Coolia palmyrensis and Coolia santacroce were established from samples collected along the northeastern Brazilian coast, the first record of both species in South Atlantic waters. Cultures of Coolia malayensis and Coolia tropicalis were also established and exhibited acute in vivo toxicity to adults of Artemia salina, while C. palmyrensis and C. santacroce were non-toxic. The presence of 30 yessotoxin analogues, 7 metabolites of Coolia and 44 Gambierdiscus metabolites was screened in 14 strains of Coolia. 44-methyl gambierone (formerly referred to as MTX3) and a new isomer of this compound were detected only in C. tropicalis, using both low- and high-resolution LC-MS/MS. To our knowledge, this is the first report of gambierone analogues in dinoflagellates other than Gambierdiscus; the role of C. tropicalis in ciguatera poisoning thus deserves to be considered in further investigations.

Some benthic dinoflagellates produce toxins that can affect other organisms including humans, and their proliferation seems to be related to the environmental variability. For this reason, the present study aims to compare the structural variation of potentially toxic dinoflagellates associated with the seagrass Thalassia testudinum from two nearby systems, with different environmental characteristics in Colombian Caribbean, corresponding to a brackish water coastal lagoon and an adjacent bay. Between January 2014 and December 2015, leaves of T. testudinum were collected monthly to obtain the dinoflagellates. Salinity, temperature, dissolved oxygen, pH, nutrients and total suspended solids (TSS) were measured, and precipitation data and the Oceanic Niño Index (ONI) were obtained. Dinoflagellates were detached from the leaves, morphologically identified by analyzing their thecal plates arrangements, and quantified using a Sedgewick-Rafter chamber. The information was analyzed using standard statistics and regression models. Fourteen species of potentially toxic epiphytic dinoflagellate belonging to four genera were recorded, being Prorocentrum the most representative in number of species. The maximum density, dominated by P. lima, were found in Bahía Chengue during the rainy season of 2014 (18452 and 20109 cells g-1 w.w.), with salinity of 35.50, high temperatures (>29.60 °C), dissolved oxygen >6 mg L-1, pH close to 8 and TSS >85 mg L-1. Densities at the Lagoon were lower than 80 cells g-1 w.w. with the highest values of Prorocentrum sp.1 under different environmental conditions. With the statistical relationships between the most abundant species and the main environmental variables, fundamental niche models were proposed in which cells could proliferate. The degree of risk to human health due to the presence of these potentially toxic epiphytic dinoflagellates will not be resolved until their toxicity discarded.

: Recurrent blooms of Ostreopsis cf. ovata have been reported in Brazil and the Mediterranean Sea with associated ecological, and in the latter case, health impacts. Molecular data based on the D1-D3 and D8-D10 regions of the LSU rDNA and ITS loci, and the morphology of O. cf. ovata isolates and field populations from locations along the Brazilian tropical and subtropical coastal regions and three oceanic islands are presented. Additional ITS sequences from three single cells from the tropical coast are provided. Toxin profiles and quantities of PLTX and their analogues; OVTXs; contained in cells from two clonal cultures and two field blooms from Rio de Janeiro were investigated. Morphology was examined using both light and epifluorescence microscopy. Morphometric analysis of different strains and field populations from diverse locations were compared. Molecular analysis showed that six of the seven sequences grouped at the large "Atlantic/Mediterranean/Pacific" sub-clade, while one sequence branched in a sister clade with sequences from Madeira Island and Greece. The toxin profile of strains and bloom field samples from Rio de Janeiro were dominated by OVTX-a and -b, with total cell quotas (31.3 and 39.3 pg cell-1) in the range of that previously reported for strains of O. cf. ovata.

Monthly phytoplankton samples were collected from January 2013 to December 2015 at a fixed sampling site in Bahía de La Paz, Gulf of California. During this study 26 samplings were Amphidinium cf. carterae positive. The highest densities of A. cf. carterae (754.2 × 103 to 1022.4 × 103 cells L-1) were recorded during a bloom detected in January 2015 when water temperatures were 20-22 °C. This dinoflagellate showed a well-marked seasonal variation, being found mainly from November to April. Blooms of the species were linked to the upwelled water due to the northwesterly wind. Cysts surrounded by a mucilaginous membrane of A. cf. carterae were found. We also observed these hyaline cysts inside zooplankton fecal pellets. Other benthic/tychoplanktonic dinoflagellates and diatoms, including some potentially toxic species were also found. The occurrence of blooms of A. cf. carterae in Bahía de La Paz could represent a risk for aquaculture activities and human health.

Se determinó la composición de ácidos grasos de 54 cepas microalgales colectadas del Perú y mantenidas en el Banco de Germoplasma de Organismos Acuáticos (IMARPE) con la finalidad de determinar su uso nutricional en la acuicultura. Para ello se realizaron cultivos en un volumen de 50 mL y se determinaron los porcentajes relativos de ácidos grasos mediante transesterificación directa y cromatografía de gases. En el grupo Chlorophyta las microalgas que presentaron los mayores valores de porcentaje relativo de ácidos grasos fueron Chlamydomonas reinhardtii (16:0; 41.2%), Scenedesmus obtusus (18:1n-7; 33.6%), Acutodesmus dimorphus (18:1n-9; 37.1%), Desmodesmus armatus (18:3n-3; 32.2%) y Tetraselmis contracta (16:4n-3; 16.5%). En cambio en el grupo Bacillariophyta, los ácidos grasos más abundantes fueron 16:1n-7 en Chaetoceros didymus (20.2%), 18:4n-3 en Navicula sp. (28.3%) y EPA en Asterionellopsis sp. (31.5%). Por otro lado, en el grupo Dinophyta, todas las cepas superaron el 20% de DHA, en particular, la cepa de Akashiwo sanguinea presentó el mayor porcentaje relativo de este ácido graso (29.9%) y de los ácidos grasos 16:0 (24.8%) y EPA (16%). Se discute el uso de estas cepas según su contenido de ácidos grasos.


In This work, we determinate the fatty acids composition and their nutritional value in 54 microalgal strains, collected from Peru and stored in Banco de Germoplasma de Organismos Acuáticos (IMARPE). The cultures were grown to 50 mL and analyzed by direct transesterification and gas chromatography. In the Chlorophyta group, the microalgaes that present the highest relative percentage of fatty acids were Chlamydomonas reinhardtii (16:0; 41.2%), Scenedesmus obtusus (18:1n-7; 33.6%), Acutodesmus dimorphus (18:1n-9; 37.1%), Desmodesmus armatus (18:3n-3; 32.2%) and Tetraselmis contracta (16:4n-3; 16.5%). Moreover in the Bacillariophyta group, the most abundant fatty acids were 16:1n-7 in Chaetoceros didymus (20.2%), 18:4n-3 in Navicula sp. (28.3%) and EPA in Asterionellopsis sp. (31.5%). By the other hand, in the Dinophyta group, all strains exceed the 20% of DHA, in particular Akashiwo sanguinea, it was strain to have the highest percentage of this fatty acid (29.9%) in addition to 16: 0 (24.8%) and EPA (16%). We discussed uses of these strains is according to their fatty acids content.

Ostreopsis cf. ovata is a toxic marine benthic dinoflagellate responsible for harmful blooms affecting ecosystem and human health, mostly in the Mediterranean Sea. In this study we report the occurrence of a summer O. cf. ovata bloom in Currais, a coastal archipelago located on the subtropical Brazilian coast (~25° S). This bloom was very similar to Mediterranean episodes in many aspects: (a) field-sampled and cultivated O. cf. ovata cells aligned phylogenetically (ITS and LSU regions) along with Mediterranean strains; (b) the bloom occurred at increasing temperature and irradiance, and decreasing wind speed; (c) cell densities reached up to 8.0 × 104 cell cm-2 on fiberglass screen and 5.6 × 105 cell g-1 fresh weight on seaweeds; (d) and toxin profiles were composed mostly of ovatoxin-a (58%) and ovatoxin-b (32%), up to 35.5 pg PLTX-eq. cell-1 in total. Mussels were contaminated during the bloom with unsafe toxin levels (up to 131 µg PLTX-eq. kg-1). Ostreopsis cells attached to different plastic litter, indicating an alternate route for toxin transfer to marine fauna via ingestion of biofilm-coated plastic debris.

Ciguatera poisoning is caused by the consumption of reef fish or shellfish that have accumulated ciguatoxins, neurotoxins produced by benthic dinoflagellates of the genera Gambierdiscus or Fukuyoa. Although ciguatera constitutes the primary cause of seafood intoxication in Cuba, very little information is available on the occurrence of ciguatoxins in the marine food web and the causative benthic dinoflagellate species. This study conducted on the south-central coast of Cuba reports the occurrence of Gambierdiscus and Fukuyoa genera and the associated benthic genera Ostreopsis and Prorocentrum. Gambierdiscus/Fukuyoa cells were present at low to moderate abundances depending on the site and month of sampling. This genus was notably higher on Dictyotaceae than on other macrophytes. PCR analysis of field-collected samples revealed the presence of six different Gambierdiscus and one Fukuyoa species, including G. caribaeus, G. carolinianus, G. carpenteri, G. belizeanus, F. ruetzleri, G. silvae, and Gambierdiscus sp. ribotype 2. Only Gambierdiscus excentricus was absent from the eight Gambierdiscus/Fukuyoa species known in the wider Caribbean region. Eleven clonal cultures were established and confirmed by PCR and SEM as being either G. carolinianus or G. caribaeus. Toxin production in each isolate was assessed by a radioligand receptor binding assay and found to be below the assay quantification limit. These novel findings augment the knowledge of the ciguatoxin-source dinoflagellates that are present in Cuba, however further studies are needed to better understand the correlation between their abundance, species-specific toxin production in the environment, and the risk for fish contamination, in order to develop better informed ciguatera risk management strategies.

Two novel azaspiracids (AZA) with a molecular mass of 869 Da were found in Pacific strains of Azadinium poporum and characterized by tandem mass spectrometry and high resolution mass spectrometry (HRMS). One compound, AZA-42, was found in Az. poporum strains AZFC25 and AZFC26, both isolated from the South China Sea. AZA-42 belongs to the 360-type AZA that in comparison to AZA-1 has an additional double bond in the F-I ring system of AZA comprising C28-C40. The other compound, AZA-62, was detected in Az. poporum strain 1D5 isolated off Chañaral, Northern Chile. Mass spectral data indicate that AZA-62 is a variant of AZA-11 with an additional double bond in the C1-C9 region of AZA. In addition to the description of the two novel AZA, a comprehensive list of all AZA known to be produced by species of the genera Azadinium and Amphidoma comprising 26 AZA variants is presented.
