RESUMO

Filarial nematodes of the Dipetalonema lineage include tick-borne filarioids that infect both domestic and wild vertebrate hosts, but they remain understudied in many cases. In this study, we conducted a molecular characterization of a Dipetalonema-like filarioid (DLF) recently identified in two tick species in French Guiana, South America. While the cox1 mitochondrial gene was the sole marker initially sequenced for describing DLF, its classification and phylogenetic relationship with other members of the Dipetalonema lineage were unclear. Therefore, we better characterized DLF through the sequencing of six additional gene markers and conducted phylogenetic analyses. Based on this multi-locus typing scheme, DLF exhibited significant divergence from known genera and species of filarioids, or other sequences available in public databases, suggesting its potential classification as a novel genus within the Dipetalonema lineage. Phylogenetic analyses further unveiled a close evolutionary relationship between DLF and all other filarioids associated with Acari (ticks and mites) within a robust monophyletic subclade in the Dipetalonema lineage. Overall, these findings confirm the existence of a specialized, Acari-borne group of filarioids and underscore the need for comprehensive investigations into their epidemiology and potential impact on animal health.


Title: Une analyse de séquences multi-locus dévoile un nouveau genre de nématodes filaires, associé aux tiques en Guyane française. Abstract: Les filaires de la lignée Dipetalonema comprennent des espèces transmises par les tiques qui infectent à la fois des hôtes vertébrés domestiques et sauvages, mais qui restent sous-étudiées dans de nombreux cas. Dans cette étude, nous avons réalisé une caractérisation moléculaire d'un filarioïde ressemblant à Dipetalonema (FRD) récemment identifié dans deux espèces de tiques en Guyane française. Alors que la séquence du gène mitochondrial cox1 était le seul marqueur génétique initialement séquencé pour décrire FRD, sa classification et sa relation phylogénétique avec d'autres membres de la lignée Dipetalonema étaient incertaines. Par conséquent, nous avons caractérisé plus précisément DLF en séquençant six gènes supplémentaires et en réalisant des analyses phylogénétiques. Sur la base de ce typage multi-locus, FRD présentait une divergence significative par rapport aux genres et espèces connus de filarioïdes, ou à d'autres séquences disponibles dans les bases de données publiques, suggérant sa classification potentielle en tant que nouveau genre au sein de la lignée Dipetalonema. Les analyses phylogénétiques ont en outre révélé une relation évolutive étroite entre FRD et tous les autres filarioïdes associés aux tiques et acariens au sein d'un sous-clade monophylétique dans la lignée Dipetalonema. Dans l'ensemble, ces résultats confirment l'existence d'un groupe spécialisé de filarioïdes transmis par les tiques et acariens et soulignent la nécessité d'études approfondies sur leur épidémiologie et leur impact potentiel sur la santé animale.

Filarial nematodes of the Dipetalonema lineage are widespread parasites and include some species that are transmitted by ticks. In this study, we conducted a large molecular survey of ticks in French Guiana, South America, to understand the overall diversity of tick-borne filarioids in this remote region largely covered by dense tropical forests. Out of 682 ticks belonging to 22 species and 6 genera, 21 ticks (3.1%) of the species Amblyomma cajennense, A. oblongoguttatum, A. romitii, Ixodes luciae and Rhipicephalus sanguineus sensu lato were positive for infection by filarioids. Molecular typing and phylogenetic analysis identified all these filarioids as members of the Dipetalonema lineage. While the filarioid of R. sanguineus sensu lato is a previously described species, the canine worm Cercopithifilaria bainae Almeida & Vicente, 1984, all other filarioids detected in this study are related but distinct to already known species in the genera Cercopithifilaria, Cruorifilaria and Dipetalonema. Their vertebrate host range may include a wide variety of mammals present in French Guiana, but dogs, capybaras, and opossums are the best candidate hosts for some of these filarioids. Although the detection of members of the Dipetalonema lineage in ticks of significant medical or veterinary interest is of concern, the risk of contracting a tick-borne filarial infection is still largely unknown. The pathogenicity of these filarioids, their epidemiology, developmental cycles, and mechanisms of transmission by South American tick species now require further study.


Title: Détection moléculaire des nématodes filaires de type Cercopithifilaria, Cruorifilaria et Dipetalonema chez les tiques de Guyane française. Abstract: Les nématodes filaires de la lignée Dipetalonema sont des parasites répandus dont plusieurs espèces sont transmises par les tiques. Dans cette étude, nous avons mené une vaste surveillance moléculaire des tiques en Guyane française, en Amérique du Sud, afin de caractériser la diversité des filaires transmis par les tiques dans cette région largement couverte de forêts tropicales denses. Sur 682 tiques appartenant à 22 espèces et 6 genres, 21 tiques (3.1 %) des espèces Amblyomma cajennense, A. oblongoguttatum, A. romitii, Ixodes luciae et Rhipicephalus sanguineus sensu lato étaient positives pour la détection des filaires. Le typage moléculaire et l'analyse phylogénétique ont permis d'identifier toutes ces filaires comme des membres de la lignée Dipetalonema. RESUMO

We report two cases of disseminated hyperinfection by filariae in free-living primates Saimiri sciureus and Saguinus niger from eastern Amazon. Histopathological examination showed the presence of Dipetalonema gracile microfilariae in blood, liver, lungs, spleen, small intestine, kidney, brain, liver, and adults in peritoneal thoracic cavity.

Different nematodes affect canines, however Acanthocheilonema reconditum was considered mostly a non-pathogenic parasite. Climate change, animal migration, and other factors transformed the dynamics of vector-borne diseases, including filariasis. Since 2016, a sudden increase in the number of dogs with microfilaremia was reported by different veterinary centers in Cali, southwest Colombia. The objective of this study was to molecularly identify the etiologic agent of this filariasis outbreak detected in this city, using PCR−RFLP and evaluating dogs' clinical signs. From 2018−2019, canine filariasis cases were prospectively evaluated after a microscopic test, recruiting 82 cases and 43 healthy controls from 2971 samples. Acanthocheilonema reconditum (Nematoda, Onchocercidae) was identified in 61.3% of the cases (49/82) by PCR−RFLP. Sanger sequencing of the 5.8S ribosomal RNA gene and internal transcribed spacer-2 fragment was additionally performed on seven cases, confirming A. reconditum in all of them. The filariasis cases are statistically associated with male dogs who have clinical signs of anemia, low levels of hemoglobin and hematocrit (p < 0.0001), and high levels of plasma proteins (p < 0.001). This emerging canine disease constitutes an important public health concern among veterinarians and active surveillance is advised to explore its zoonotic potential.

Filarial nematode infections are common in primates, but have received little attention in the Neotropics. Epidemiological data on filarial infections in primates are still too sparse to fully understand the complex of this parasitism, especially because of the difficulty in studying the ecology and epidemiology of wild primates.. We describe natural infections by Dipetalonema parasitizing 211 primates belonging to eight free-living primate genera in Amazonia, and assess the relationships between parasitic indicators and climatic (rainfall and river level), ecological (fruiting periods of plants) and biological (sex, species' body mass, group size and density) factors. The overall prevalence was 64.4% (95% CI: 64.0 - 64.9); parasitic mean abundance (N filariae per individual) and parasitic mean intensity (N filariae per infected host) of infection were 11.9 (95% CI: 8.3 - 15.6) and 18.4 (95% CI: 13.4 - 23.4) filariae/individual, respectively. Although we observed differences in parasitic parameters among primate genera, there was no correlation between parasitic parameters with density, body mass or group size. Sapajus, Cebus and Lagothrix had the highest prevalence and parasitic mean intensity. Using Lagothrix lagotricha poeppigii, the most sampled species (n = 92), as a model, we found that the number of filariae per infected host was associated with fruit production in swamp forests during the dry season, the time of food scarcity. The long periods of food shortage may cause environmental stress on primates, impairing their immune defenses and leading to increased parasite load but not affecting infection prevalence. However, the lack of information on vector ecology, key to understand risk factors associated to infection rate, prevents confirming the existence of an infection pattern dependent on food availability.

Species of the genus Dipetalonema are parasitic nematodes of the family Onchocercidae (Nematoda; Filarioidea) which infect the peritoneal cavity of Neotropical primates. Of these, six species have been taxonomically described, two of these have been reported infecting the black-faced spider monkey (Ateles chamek): Dipetalonema gracile and Dipetalonema yatesi. Description of Dipetalonema species have been based on morphological characteristics, and their phylogenetic relationships remain unresolved. A few molecular studies have been carried out in Dipetalonema spp. infecting Neotropical primates. Seven filarioid nematodes (6 females and one male) recovered from one A. chamek in the Peruvian Amazon rainforest were morphologically identified as D. yatesi and molecularly characterized. A multi-locus genetic analysis of nuclear ribosomal region (18S) and mitochondrial (cox1, 12S, and nad5) gene sequences supported D. yatesi as a distinct lineage and yielded a highly resolved phylogenetic lineage tree for this filarioid genus of Neotropical primates. Our results highlighted that Dipetalonema species are divided in two well-supported clades, one containing D. yatesi and D. caudispina, and the second containing D. robini, D. gracile, and D. graciliformis. Due to sequence ambiguities from GenBank entries, relationships among isolates of D. gracile and D. graciliformis cannot be fully resolved, which requires further investigation. However, this suggests that these could represent a species complex. Our study confirms that D. yatesi is a valid species and constitutes the first molecular phylogenetic analysis of this parasite in black-faced spider monkeys.

Primate-parasite interactions are often investigated via coprological studies given ethical and conservation restrictions of collecting primate hosts. Yet, these studies are inadequate to recover adult helminths for taxonomic identification and to accurately assess their prevalence, intensity, abundance, and site of infection. Fresh carcasses found in anthropogenic landscapes come as informative and reliable alternatives. In this study, we identified the helminths of brown howler monkeys (Alouatta guariba clamitans) and their sites of infection, and measured their prevalence, intensity, and abundance of infection. We necropsied 18 adult males, 11 adult females, and 7 juvenile males that died in conflicts with the anthropogenic environment (domestic dog attacks, n = 11; electrocutions and road-kills, n = 10 each; unknown, n = 5) in periurban landscapes of southern Brazil between 2013 and 2019. We found three nematodes (Trypanoxyuris minutus, Dipetalonema gracile, and Parabronema bonnei) and one cestode (Bertiella cf. studeri), a diversity estimated to account for a sampling completeness of 99%. Prevalence ranged from 3% for P. bonnei to 100% for T. minutus. Mean abundance ranged from 2 (D. gracile and B. cf. studeri) to 55,116 (T. minutus) and mean intensity of infection ranged from 4 (B. cf. studeri) to 55,116 (T. minutus). Trypanoxyuris minutus sex ratio was strongly male-biased. The intensity of infection with T. minutus was higher in juvenile males and adult females than in adult males. The low parasite diversity and the helminths' mode of transmission are compatible with howlers' arboreality and folivorous-frugivorous diet. The howlers were not infected with soil-transmitted helminth parasites of humans and domestic animals on the ground and probably did not eat invertebrates to complement the diet. Given the lack of evidence of howler health problems, we suggest that the causes of death of the necropsied howlers are the major threats to the long-term conservation of the species at the study periurban landscapes.

Six Dipetalonema species have been reported from Neotropical monkeys, Dipetalonema gracile, Dipetalonema graciliformis and Dipetalonema caudispina being the dominant species found in French Guiana primates. Adult filarioids isolated from the abdominal cavity of tamarins (Saguinus midas) in French Guiana were morphologically and molecularly identified as D. graciliformis. Phylogenetic analysis based on DNA and amino acid sequences of the cox1 gene as well as the concatenated sequences of the cox1 and the 18S genes indicated that D. graciliformis belongs to the clade 4 (ONC4) of Onchocercidae. Blast analysis of the 18S rDNA revealed that D. graciliformis in the studied tamarins is conspecific with the filarioid circulating in howler monkeys (Alouatta macconnelli) in French Guiana, previously referred to as unidentified Onchocercidae species.

RESUMO

The necropsy of 24 dogs with positive blood serum test for Dipetalonema sp microfilarias demonstrated that 93.9% of the parasites were located in the subcutaneous tissue and 6.1% in general visceral organs. The helminths were localized in different anatomical regions, the chest was found with the highest frequency.


Foram necropsiados 24 cães com exame de sangue positivo para microfilarias de Dipetalonema sp. Verificou-se que 93,9% dos parasitos foram encontrados no tecido subcutâneo e 6,1% nas vísceras. A localização dos helmintos variou com as regimes anatômicas, sendo o tórax o local de maior freqüência.

Three distinctive canine populations, constituted, respectively, by 451 stray dogs (group I), 136 kennel dogs (group II) and 226 domiciliated dogs (group III), were studied concerning the prevalence of circulating microfilariae, through the blood serum technique. The genus of the parasites (Dipetalonema sp and/or Dirofilaiia immitis) was identified through its morphologic characteristics. In group I, a 36.8% prevalence was obtained to microfilariae of the Dipetalonema sp; from the 27 positive dogs (19.8%) in group II, 26 (19.1%) were infested by the Dipetalonema sp and only one (0.7%) by the Dirofilaria immitis. In finally, the 3Jd group, 7.0% of the animals harboured the Dipetalonema sp and 0.5% the Dirofllaria immitis.


Três populações caninas distintas, constituídas, respectivamente, de 451 animais errantes (grupo I), 136 animais domiciliados em grupo (grupo II) e 226 cães domiciliados individualmente (grupo III), foram estudadas para avaliação da prevalência de microfilárias circulantes, através da técnica do soro sanguíneo. Pelo exame das características morfológicas das microfilárias realizou-se a identificação do género dos parasitas (Dirofilaria immitis e/ou Dipetalonema sp.) . No grupo I obtiveram-se 166 casos (36,8%) positivos de Dipetalonema sp. Dos 27 casos positivos (19,8%) do grupo II, 26 (19,1%) foram identificados infestados por Dipetalonema sp e apenas 1 (0,7%) por Dirofilaria immitis; no grupo III obtiveram-se 16 casos (7%) de parasitismo por Dipetalonema sp e 1 (0,5%) por Dirofilaria immitis.
