RESUMO

Domestic donkey plays a key role as a draft animal in rural economy of Pakistan where its population is increasing every year. The complete mtDNA control region of forty randomly sampled donkeys was PCR- amplified and sequenced bi-directionally using specific primers. Distinct mtDNA haplotypes obtained in the current study (KY446001−KY446011) were subjected to haplotype (h) and nucleotide diversity (π) measures using DnaS as well as to phylogenetic, Network, and AMOVA analyses. There were a total 27 polymorphic sites present within 11 unique mtDNA haplotypes from the studied 40 animals from different regions. Neighbor-joining network and median-joining network both illustrated the splitting of all these haplotypes into two well-defined Nubian and Somali lineages, confirming African maternal origin of Pakistani domestic donkey. Diversity parameters h (0.967± 0.037) and π (0.02917± 0.00307) were found to reveal high levels of genetic diversity in Pakistani donkeys. AMOVA demonstrated only 1% of genetic differences between two mtDNA maternal lineages, pointing to lack of population substructure in Pakistani donkeys as is the case with worldwide domestic donkey population. Pakistani donkeys have African maternal origin and high levels of mtDNA diversity. High genetic diversity may be due to non-selective breeding and heteroplasmy. We herein provide the first report on mtDNA diversity of control region in Pakistani domestic donkey.

RESUMO

Equine piroplasmosis is a parasitic illness caused by various protozoa of the Babesia and Theileria genera, which parasitize within red blood cells. The transmission of these pathogens occurs through certain genus of ticks, including Amblyomma, Haemaphysalis, Hyalomma, and Rhipicephalus. In recent times, an increase in the identification of new Theileria species and genotypes has been observed. This is further complicated by the presence of mixed Theileria infections in both mammals and tick vectors, particularly in regions where wildlife and livestock share habitats and vectors. Therefore, the objective of this study is to document the occurrence of Theileria cervi in a non-typical host. A total of 88 horses (Equus caballus) and 10 donkeys (Equus asinus) were sampled in three municipalities in Veracruz, Mexico. Molecular techniques were employed to identify Babesia/Theileria through the amplification of a segment of the 18S-rDNA and hsp70 genes. The phylogenetic reconstruction grouped the obtained sequences into a monophyletic cluster alongside sequences of T. cervi. This work represents the first documented occurrence of T. cervi in equids. These findings have significant implications from an epidemiological point of view. In addition, further studies are needed to determine the distribution and pathogenicity of this species for domestic animals and to develop effective control strategies.

RESUMO

In Colombia, works performed with the colombian creole donkey (CCD) have shown a possible danger of extinction. The above, associate with an alarming decrease in its inventory and a reduction in genetic diversity. Thus, morphological characterization could be the first step for future conservation plans to conduct a repopulation. Therefore, this work aimed to characterize the zoometric and ethnological traits of CCD in five natural subregions of the department of Sucre. 20 males from each of the natural subregions from Sucre (Golfo de Morrosquillo: GM, Montes de María: MM, Mojana: MO, San Jorge: SJ, Sabanas: SA) with an age average of 4.7 ± 1.0 years old and weight estimated between 126.0 ± 19.4 kg were recorded and were submitted to 28 morphometric measurements. Descriptive statistics, analysis of variance between subregions, and hierarchical grouping for statistical analysis were performed. Also, five ethnological indices and five productive indices were estimated. The results showed that most of the zoometric variables varied significantly between subregions. The values assessed were similar to other creole donkey breeds. The hierarchical grouping analysis allowed the creation of two groups with geographic proximity. The structural homogeneity and morphostructural harmony found within the subregions were medium-low. However, when analyzing all the CCD measurements together, medium to high morphostructural uniformity and harmony were evidenced. The ethnological indices allow us to classify the CCD as a longlinear, hypermetric, dolichocephalic, and concavilinear animal. In addition, the productive indices indicated that the CCD is a proportionate donkey, with good poise and aptitude for work.

The pharmacokinetics of butorphanol after intravenous (IVB) and intramuscular (IMB) administration in donkeys were determined in this preliminary study. Healthy male gelded donkeys (n = 5), aged 6-12 years old, were administered 0.1 mg/kg butorphanol IV or IM in a randomized, crossover design. Blood samples were obtained at predetermined intervals for 24 h (IVB) and 48 h (IMB) after administration. Plasma butorphanol concentrations were determined by high performance liquid chromatography and pharmacokinetic parameters were calculated. Following IVB administration, mean (± SE) apparent volume of distribution, elimination half-life, total body clearance, and area under the plasma concentration time curve from time 0 to infinity (AUC0-∞) were 322 ± 50 mL/kg, 0.83 ± 0.318 h, 400 ± 114 mL/h/kg, 370 ± 131 h·ng/mL, respectively. After IMB administration, a maximum plasma drug concentration of 369 ± 190 ng/mL was reached at 0.48 ± 0.09 h. The IMB AUC0-∞ was 410 ± 60 h·ng/mL. Bioavailability of IMB was 133 ± 45%. The pharmacokinetics of butorphanol in healthy donkeys was characterized by faster elimination half-life compared to values from the equine literature.

RESUMO

The overpopulation of donkeys is recognized as a problem in many parts of the world. The main concerns with uncontrolled donkey populations are habitat degradation and competition for feed resources between donkeys and other species. One of the most effective and humane solutions is the use of immunocontraception. Therefore, this study sought to evaluate the stress imposed by the use of two formulations of a zona pellucida (ZP) vaccine, a recombinant (reZP) and a native porcine (pZP) vaccine, both formulated with a Freund's adjuvant. The stress was objectively measured using fecal cortisol concentrations and physical examination parameters at fixed points before and after vaccination. We hypothesized that fewer changes in physical exam parameters and lower fecal cortisol concentrations would be stimulated in jennies treated with the reZP vaccine due to the selection of specific proteins. Twenty-five reproductively sound jennies were randomly assigned to reZP (n = 9), pZP (n = 8) or control (n = 8) groups. The vaccines were administered at five-week intervals. Physical exam parameters and body wall thickness of injection sites were recorded for each jenny for four days post-injections. Fecal samples were obtained every other day from day 0 (first vaccination) through day 6 and on days 35 to 41 after booster. Injection site reactions were common in all groups with the reZP and pZP groups being overrepresented. Lameness was observed in the pZP and reZP groups that were affected by injection site reactions and open abscesses. The present study showed an increase in fecal cortisol concentrations within 4 days after the first vaccination with ZP vaccines and, thereafter, a decrease in cortisol 35 days later after the second vaccination, especially in donkeys with open abscesses. Our results suggest that acute stress (increased cortisol) was induced after the first vaccination, and chronic stress (decreased cortisol) occurred thereafter in association with open abscesses. In conclusion, reZP and pZP formulated with Freund's adjuvant induced local inflammatory reactions with a differential degree of acute and chronic stress in donkeys.

RESUMO

The aim of this study was to test equine semen cryopreservation techniques for the conservation of donkey germplasm. Ejaculates of three male donkeys were used (n = 18; six ejaculates per donkey; six repetitions), collected by the artificial vagina method. To remove the seminal plasma, the ejaculates were split and submitted to filtration or centrifugation methods. To assess the freezing method, each fraction was submitted to the automated system or the conventional system, and groups were formed: automated centrifuge, automated filtrate, conventional centrifuge and conventional filtrate. After thawing (37°C/30 seconds), were analyzed the sperm kinetic parameters, integrity and functionality of the plasma membrane and mitochondrial membrane potential. Highest sperm concentration (P < .05) was observed in the filtrate groups; the conventional filtrate group presented lower (P < .05) progressive motility and curvilinear velocity compared to the other groups; no difference was observed (P > .05) among the groups for the membrane integrity and functionality, and mitochondrial membrane potential. Thus, centrifugation is the most indicated technique to remove donkey seminal plasma and the automated and conventional freezing methods can be used in donkey semen conservation.

RESUMO

Equine infectious anemia (EIA) is listed by the World Organization for Animal Health (OIE) as one of the equine diseases that must be notified. No effective treatment or vaccine is available. EIA control is based on segregation and euthanasia of positive equids. The disease is caused by the equine infectious anemia virus (EIAV), a member of the genus Lentivirus of the Retroviridae family. Despite the importance of this disease in equids, EIA has been poorly studied in donkeys (Equus asinus). We evaluate the sanitary conditions related to EIAV in donkeys from a shelter of abandoned animals captured on the roads of the Ceará. A total of 124 donkeys were randomly selected, and three horses lived at the same shelter. The animals were clinically evaluated, and a group of the 20 animals was submitted to hematological tests. Three diagnostic tests for EIA were used, agar gel immunodiffusion (AGID), enzyme-linked immunosorbent assay (ELISA) using EIAV recombinant protein gp90 (rgp90) and recombinant protein p26 (rp26) ELISA, and polymerase chain reaction (PCR) for detection of the EIAV tat-gag gene. From the donkeys, only 1 animal was positive using AGID 0.81% (1/124), compared to 21.8% (27/124) in the rgp90 and 10.5% (13/124) in the rp26 ELISA. Proviral DNA was detected by PCR tat-gag in 8.8% (11/124), and phylogenetic analysis confirms that the EIAV sequences of donkeys from the Brazilian Northeast grouped with Pantanal Brazilian sequences. Thus, in light of the results, we conclude that donkeys are carriers of EIAV and could be sources of infection.

RESUMO

The present investigation was undertaken to highlight the histomorphometric changes of testicular tissues according to season and age in donkeys (Equus asinus) under northern Algeria conditions. The experiment was conducted from February 2019 to January 2020. A total of 21 sexually mature donkeys were selected randomly. The testis were immediately collected after slaughter for the histological observation. The analysis of the correlation coefficients shows negative correlations between DL and other parameters such as DST, GCEH, TTA and ITSA, ranged between -0.24 and -0.79. Also, there were a high negative correlations between season and the histomorphometric parameters, except DL was revealed positive (r=0.65). On the other hand, there were positive correlations between age and testicular histomorphometrics parameters. The results indicated that DST, GCEH, TTA and ITSA values were significantly higher in winter and autumn seasons than in spring and summer seasons. It is also noteworthy that DST, GCEH, TTA and ITSA values were significantly higher in adult and aged donkeys than in young donkeys. Our comparative analysis of histological parameters, suggests that the sexual activity usually occurs during winter and autumn in local donkeys of Algeria. In addition, our results of histomorphometric of testicular tissues are correlated with age of donkeys.(AU)

RESUMO

Clostridial infections, particularly clostridial myonecrosis, can be fulminant and fatal; they often arise without an obvious history of trauma. The majority of equine clostridial myonecrosis cases are associated with intramuscular injection. This paper presents the first report of clostridial myositis by C. perfringens in a donkey after an injection of flunixin meglumine on the neck, describing the clinical, necroscopic, and histopathological events. Bacterial isolation and biochemical tests confirmed the definitive diagnosis of C. perfringens.

RESUMO

Known as an animal of multiple functions, the Equus asinus has always been used for various purposes, such as entertainment, horseback riding, means of transport, agricultural traction and dairy farming. Although, donkeys are associated with a vast heritage of social, cultural, economic and ecological importance, they have lost their importance in the activities of rural properties, both in the developed economies of European countries and in the Northeast region of Brazil. Specific studies of production systems aimed rational exploitation of the donkey species in Brazil do not exist. New perspectives for the use of donkey in the Brazilian semiarid region through the dairy industry has aroused scientific interest and the interest of investors. The donkey, allied to the low production costs and rusticity of the species, has the capacity to contribute to the economy with products of high biological value and therapeutic characteristics. Thus, this review provided a broad view of the donkey bred in the Northeast of Brazil, observing its characteristics and relevance for the region. The benefits of donkey milk and the need to promote the production and marketing of this milk in the dairy production chain will also be discussed.

The aim of this study was to characterize and determine how days in milk (DIM) affect the milk fatty acid (FA) profile of grazing donkeys. Donkey milk is very similar to human milk, containing bioactive molecules such as FA and proteins. However, there is a lack of scientific and technical information on the changes in the FA profile of asinine milk in pasture-feeding systems. Seven multiparous Pega donkeys, maintained in an exclusively extensive system, were used. Milking was undertaken twice a day, once a week for 16 weeks. Milk samples were composed according to the average lactating days as follows: 55, 110, 165, 220 and 275 DIM. A descriptive analysis of the milk fat composition was performed, and the linear and quadratic effects of DIM on the milk FA profile were tested. The milk FA profile of grazing donkeys is influenced by lactation days and is characterized by high concentrations of polyunsaturated FA (PUFA) with a low n-6 to n-3 ratio (0.66 g/100 g), mainly due to the high level of linolenic acid (16.8 g/100 g). Most of the individual FAs did not change during lactation, but stearic and oleic acid linearly decreased (p < .05), and total polyunsaturated FA (PUFA) and n-3 FA increased (p < .05) with DIM. The milk profile of grazing donkeys is influenced by the day of lactation and is characterized by a high concentration of n-3 PUFA (mainly α-linolenic acid) and a lower n-6 to n-3 ratio. As the number of days of lactation increased, the concentration of n-3 PUFA also increased. Therefore, higher milk quality may be associated with higher days in milk.

RESUMO

Background: Obstructive urolithiasis is a rare but potentially serious condition in equids. In the reviewed literature, there are several case reports of urolithiasis in horses and donkeys, but the only mention of this condition in mules occurred as incidental findings at a slaughterhouse. Therefore, this work aims to describe the first report and successful treatment of obstructive urethrolithiasis in a mule (Equus asinus x Equus caballus). Case: A 10-year-old castrated male mule weighing 380 kg was referred for hospital care. Tachycardia (64 beats per min), mild dehydration (7%), increased capillary filling time (3 s), slightly congested mucous membranes, and dysuria were observed. During its attempts to urinate, the mule was able to expose the penis, resulting in only dribbling of urine with reddish coloration. Urethral catheterization failed to reach the urinary bladder and revealed an obstruction at the ischial arch (7 x 4 cm), as confirmed by palpation and ultrasonography. Additionally, rectal ultrasound examination showed urine sedimentation and a single 2.36 mm vesical calculus. After sedation, local anesthesia, and surgical preparation, urethrotomy in the standing position was performed over the urethral obstruction at the ischial arch, reaching the urethrolith that fragmented during removal. Urethral catheterization from the urethrotomy site to flush the urinary bladder and urethra were performed, but the remaining vesical calculus was not retrieved. Considering the presence of a vesical calculus, severe urethral damage caused by the spiculated calculus and catheterization attempts, permanent perineal urethrostomy was performed. Laboratory tests revealed unremarkable hematological parameters, while serum biochemistry showed increased creatinine level. Urinalysis revealed cloudiness, amber appearance, countless red blood cells and bacteria, and calcium carbonate crystals. The urethrolith composition included ammonia, carbonate, and oxalate. Twelve months after surgery, the mule was healthy, the urethrostomy was viable, and no complications were recorded during this period. Discussion: Although uncommon, there are reports describing calculi of different sizes and weighing up to 803 g, causing mild to severe clinical signs according to the degree of obstruction in horses and donkeys. In the mule described here, the urethrolith did not completely obstruct the urethra, but the spiculated calculus caused dysuria and hematuria. In fact, most animals are usually referred for acute abdominal signs or hematuria and pollakiuria, but other unusual signs, such as rectal prolapse, may also be present. In the present report, the diagnosis of obstructive urethrolithiasis was established based on clinical signs and transcutaneous ultrasound of the subischial area, allowing visualization of the urethrolith. To the best of our knowledge, this is the first report of obstructive urethrolithiasis affecting a mule in Brazil. We reiterate that this condition must be included in the differential diagnosis of mules and hinnies with hematuria and dysuria, especially when associated with abdominal pain. Additionally, urethrostomy associated with urethrotomy performed on this mule in the standing position was a low-cost procedure with good results. Due to the lack of specificity regarding the food management of the mule on the previous farm, an assessment cannot be made regarding the effects of its food on urolith composition.(AU)

RESUMO

Donkeys have a long history in the development of human societies. Typically referred to as a beast of burden, traditional uses for donkeys have included the transportation of goods and people, use in agricultural and forestry activities, to access water, and provide citizens in low- and middle-income countries a means of making an income for communities. However, the rise of mechanization, the development of modern farming techniques, and the increasing availability of motorized vehicles have led to donkeys and mules becoming redundant from traditional roles in many parts of the world. We provide examples of where donkeys have successfully transitioned from traditional roles to new, non-traditional roles in Europe and North America, and demonstrate that, although the roles and use of donkeys and mules are changing in a rapidly developing world, we can learn lessons from the past and apply them to current challenges. As the need for working equids declines in transport and agriculture, they still hold great value for recreational, therapeutic, and environmentally friendly methods of animal traction.(AU)

Donkeys (Equus asinus) and mules represent approximately 50% of the entire domestic equine herd in the world and play an essential role in the lives of thousands of people, primarily in developing countries. Despite their importance, donkeys are currently a neglected and threatened species due to abandonment, indiscriminate slaughter, and a lack of proper sanitary management. Specific knowledge about infectious viral diseases that affect this group of Equidae is still limited. In many cases, donkeys and mules are treated like horses, with the physiological differences between these species usually not taken into account. Most infectious diseases that affect the Equidae family are exclusive to the family, and they have a tremendous economic impact on the equine industry. However, some viruses may cross the species barrier and affect humans, representing an imminent risk to public health. Nevertheless, even with such importance, most studies are conducted on horses (Equus caballus), and there is little comparative information on infection in donkeys and mules. Therefore, the objective of this article is to provide a brief update on viruses that affect donkeys and mules, thereby compromising their performance and well-being. These diseases may put them at risk of extinction in some parts of the world due to neglect and the precarious conditions they live in and may ultimately endanger other species' health and humans.

RESUMO

SUMMARY: The objective of this present investigation was undertaken to study the testicular and epididymal biometrical characteristics in Algerian donkeys throughout the year according to age, body weight and seasonal changes. The study was conducted from February 2019 to January 2020. A total of 24 sexually mature donkeys (Equus asinus) were selected randomly. The testis and epididymis were collected after slaughter of donkeys and separated from the conjunctive and adherent tissues. The epididymis has been carefully removed at the testicular junction. In total, 10 biometric measures were selected and performed. Our results revealed that there are significant differences (P<0.05) between groups in most biometrics values. All biometric parameters varied throughout the year and were affected by the season. Significant differences of the GSI and SC values (P<0.05) were observed in different age groups and seasons. On the other hand, no significant differences were observed between the body weight categories of donkeys. The analysis of the correlation coefficients between the biometric values shows high positive correlations, ranged between 0.98 and 0.72 (P<0.001). There was a high positive correlation between age and all the parameters, ranged from 0.85 to 0.61 (P<0.001). However, there were low negative correlations between season and; testicular and epididymal biometrics. It is the first investigation that describes the male reproductive organs in donkeys of the Algerian race (Equus asinus), on the basis of biometric testicular and epididymal measurements. Our results showed that the essential differences were noted between some biometric parameters and the age, season and body weight of donkeys. In addition, the correlation coefficients were supported between biometric measurements and these factors. However, other approaches are necessary to undertake, such as histology of reproductive organs and hormone measurement, for a deeper understanding of the physiology of reproduction in donkeys.

The application of morphofunctional measurements allows the selection of suitable animals to perform working activities and ensure that animal maximum capacities are not exceeded. Mules are commonly used by small farmers for agricultural work in mountain areas where the access is extremely difficult. We aimed to estimate the functionality indexes of working mules in mountain areas of the Colombian Central Andes. A total of 94 adult mules were evaluated for withers height, thoracic perimeter, body length, neck length, and body weight, which were used to determine the body index (BI), proportionality index (PI), and load carriage index (LCI). Descriptive statistics, analysis of correlations, and principal component analysis were performed. Males presented higher morphometric measurements than females. There was a negative correlation between BI with PI and LCI. The principal component analysis was able to merge characteristics in two components explaining 81.78% of the variance in the indexes. These results demonstrated that working mules in the studied area have morphometric characteristics that define them as mesolinear animals with a low frame and broad chest, and these characteristics can function as a guide to identify desirable conformation indices for working mules, determining values of adequate load, respecting animal welfare.(AU)

RESUMO

To investigate the mtDNA variation and origin of maternal lineages in South American donkeys and to reassess the domestication of donkeys in northeast Africa, we analyzed sequences (489 bp of the D-loop) from 323 domestic donkeys sampled from Peru, Brazil, Ethiopia and Egypt. Altogether, the 323 sequences displayed 53 different haplotypes (45 in Ethiopia, 14 in Egypt, eight in Peru and six in Brazil). Among the four populations, Egyptian donkeys possessed the highest haplotype diversity (0.910 ± 0.032), followed by Brazilian donkeys (0.879 ± 0.060). The Clade I haplotypes dominated in Peruvian donkeys (65%), whereas Clade II haplotypes dominated in Brazilian donkeys (67%). Estimates of FST values showed a high genetic differentiation between Peruvian and Brazilian donkey populations (FST  = 0.4066), which could be explained by the complex introduction history of South American donkeys. Phylogeographic analysis indicates that northeast Africa could be the most probable domestication center for Clade I donkeys. Analysis of molecular variance confirmed a weak genetic structure in domestic donkey populations among four continents (Europe, Asia, Africa and South America).
