RESUMO

The aim of this study was to evaluate the effect of light on anatomy, physiology and essential oil content and composition of Varronia curassavica. They were analysed two light conditions (full sunlight and protected environment with 50% shade screen) and five accessions (VCUR-101, VCUR-102, VCUR-201, VCUR-302, VCUR-802). V. curassavica cultivated in full sun presented a greater development of the leaf blade and palisade parenchyma thickness for all accessions. Chlorophyll levels did not differ according to the two light environments. The leaf area was larger in the protected environment. The essential oil yield of the accessions ranged from 0.26 to 0.87 mL/plant in full sun and from 0.34 to 0.53 mL/plant in the protected environment. The composition of the essential oil was influenced by the light and the accession. All accessions presented (E)-caryophyllene and α-humulene. The influence of light on the evaluated variables is genotype dependent.


El objetivo de este estudio fue evaluar el efecto de la luz sobre la anatomía, fisiología, el contenido y composición de aceites esenciales de Varronia curassavica. Se analizaron dos condiciones de luz (pleno sol y ambiente protegido con 50% de pantalla de sombra) y cinco accesiones (VCUR-101, VCUR-102, VCUR-201, VCUR-302, VCUR-802). V. curassavica cultivada a pleno sol presentó mayor desarrollo del limbo foliar y espesor del parénquima en empalizada para todas las accesiones. Los niveles de clorofila no difirieron según los dos entornos de luz. El área foliar fue mayor en el ambiente protegido. El rendimiento de aceite esencial de las accesiones varió de 0,26 a 0,87 mL/planta a pleno sol y de 0,34 a0,53 mL/planta en el ambiente protegido. La composición del aceite esencial fue influenciada por la luz y la accesión. Todas las accesiones presentaron (E)-cariofileno y α-humuleno. La influencia de la luz sobre las variables evaluadas depende del genotipo.

BACKGROUND: The anatomical traits associated with water deficit are also observed in plants growing in poor soils. The species may resist water deficit through three main strategies: escape, avoid or tolerate. The Pedregal de San Ángel Ecological Reserve (REPSA), Mexico, is an environment with low nutrient soil and low water availability. It is set on the basalt formation derived from the Xitle volcano eruption. The main vegetation type is characterized as xerophytic shrub. Thus we expect that species growing in this community will show leaf xeromorphic traits and may have any of the three response strategies. We analyzed the foliar anatomy of 52 species of the Asteraceae family at the REPSA because it is the most abundant angiosperm family in the site, showing a wide variety of growth forms and anatomical variation. RESULTS: The foliar anatomies of the studied Asteraceae were highly variable as well as their quantitative traits as revealed by principal component analysis. This agrees with previous studies that found great anatomical variation within the family. Leaves have multiple layered palisade parenchyma and parenchyma bundle sheaths and could not be categorized as xeromorphic because they possess mesomorphic leaf features as simple lamina, single-layered epidermis, and soft large-size glabrous leaves with high specific leaf area. CONCLUSIONS: The combination of mesomorphic and few xeromorphic foliar traits with other characters at the genus and tribal level probably has been essential in Asteraceae to colonize various environments, including those with low water and nutrient availability.

ABSTRACT: The aim of the present study was to evaluate the effect of the colchicine and oryzalin anti-mitotic substances on the induction of tetraploid plants and foliar anatomy of two diploids clones of Solanum commersonii ssp. Nodal segments of Solanum commersonii subsp. commersonii Dun. and Solanum commersonii subsp. malmeanum Bitt. were treated with colchicine (3.5, 5.0, and 6.5mM; 72h) or oryzalin (10, 30, and 50µM; 24h). After the treatment with anti-mitotic substances, nodal segments were inoculated in the MS culture medium and cultivated in vitro (60 days). After in vitro cultivation, the plants were transferred to vases with the substrate Plantmax(r) and kept in the greenhouse (45 days). Plant ploidy level was assessed by flow cytometry and leaf anatomy was assessed by anatomic cuts. An increase was observed in the polar and equatorial diameter of stomata ("gigas effect") of the Solanum commersonii subsp. commersonii (SCC) and Solanum commersonii subsp. malmeanum (SCM) clones, which was due to the use of the anti-mitotic substances. Treatments with colchicine (6.5mM; 72h) and oryzalin (50µM; 24h) caused death of the SCC plants cultured in vitro. In the others treatments of the SCC clone, the use of oryzalin and colchicine caused production of chimeric plants. The treatment of nodal segments of SCM with oryzalin (10-50µM; 24h) was effective on induction of tetraploid plants, which can be employed in genetic breeding programs in crossbreeding with cultured potato.


RESUMO: O objetivo deste estudo foi avaliar o efeito das substâncias antimitóticas colchicina e orizalina na indução de plantas tetraploides e na anatomia foliar de dois clones diploides de Solanum commersonii ssp. Segmentos nodais de Solanum commersonii subsp. commersonii Dun. e Solanum commersonii subsp. malmeanum Bitt. foram tratados com colchicina (3,5, 5,0 e 6,5mM; 72h) ou orizalina (10, 30 e 50µM; 24h). Após o tratamento com as substâncias antimitóticas, os segmentos nodais foram inoculados em meio de cultura MS e cultivados in vitro (60 dias). Após o cultivo in vitro, as plantas foram transferidas para vasos contendo o substrato Plantmax(r) e mantidas em casa de vegetação (45 dias). O nível de ploidia das plantas foi avaliado por citometria de fluxo e a anatomia foliar foi avaliada através de cortes anatômicos. Foi observado um aumento no diâmetro equatorial e polar dos estômatos ("efeito gigas") dos clones de Solanum commersonii subsp. commersonii (SCC) e Solanum commersonii subsp. malmeanum (SCM), que foi produzido pelo uso de substâncias antimitóticas. O tratamento com colchicina (6,5mM; 72h) e orizalina (50µM; 24h) causou morte nas plantas de SCC cultivadas in vitro. Nos demais tratamentos do clone de SCC, o uso de orizalina ou colchicina causou a produção de plantas quiméricas. O emprego de orizalina (10-50µM; 24h), nos segmentos nodais de SCM, resultou na obtenção de plantas tetraploides consistentes, as quais podem ser usadas nos programas para melhoramento genético em cruzamentos com a batata cultivada.

Abstract Myrciaria glomerata O. Berg., Myrtaceae, popularly known as "cabeludinha", has high content of ascorbic acid and anti-inflammatory property and is used in folk medicine. The objectives of this study were the morphological, anatomical and histochemical characterization of the leaves. Leaf studies were made with optical, scanning electron and confocal microscopy. The collection of botanical material was held at the Tijuca Forest, Rio de Janeiro, RJ. Histochemical tests aimed the identification of lipids, starch grains, phenolic compounds and crystals. The leaves are simple, opposite, lanceolate, pinnate, hairy, with involute margins, hypostomatic and dorsiventral. The stomata are anomocytic. The epidermis presents simple trichomes. Epidermal cells show uneven thickening of their periclinal outer walls, mainly on the adaxial side of the leaf. Secretory cavities of essential oils are subepidermal and exceed, in height, the palisade parenchyma, formed by one cell layer. Four to five cellular layers, rich in phenolic compounds and lipids form the spongy parenchyma. The bundles are collateral and there are many crystals of calcium oxalate spread throughout the mesophyll. In the midrib and petiole the bundles are bicollateral. Analysis by scanning electron revealed epicuticular wax rod-shaped and as grains. In confocal microscopy, the adaxial epidermis, the fibers and the secretory epithelium of the cavities show autofluorescence. The data obtained are important in quality control exams of samples of this species.

PREMISE OF THE STUDY: Extrafloral nectaries (EFNs) are structures that secrete nectar and protect plants against herbivores and pathogens. In Gentianaceae, these structures have been described in species of Calolisianthus, Fagraea, and Anthocleista and are important morphological markers for taxonomic and phylogenetic studies. To establish a foundation for further studies, we investigated the occurrence, distribution patterns, and anatomy of EFNs on leaves of 27 species belonging to 13 genera and three tribes of neotropical Gentianaceae. • METHODS: Leaf samples were diaphanized, stained with basic fuchsin, and mounted in glycerinated gelatin. Cross sections were obtained from material embedded in methacrylate or paraffin, stained, and mounted in Permount. Polysaccharides were histochemically stained with periodic acid-Schiff stain. Samples were also examined with scanning electron microscopy. • KEY RESULTS: Unusual EFNs, visible only with light microscopy, were formed of modified epidermal cells. Each EFN consisted of 2-5 secretory cells encircling a central cell. The EFNs varied in size and in the shape and arrangement of the adjacent cells surrounding the secretory cells. EFNs occurred in all analyzed species as isolated units distributed throughout the leaf blade or as aggregates; aggregates were generally visible to the naked eye. Based on their occurrence as aggregates or isolated units and on their location on the leaf blade, six distribution patterns were identified. • CONCLUSIONS: This is the first comprehensive study of EFNs on the leaves of neotropical Gentianaceae. The data suggested that NEFs evolved from isolated units for NEFs in aggregates. The results represent a new source of data for future ecological, systematic, and phylogenetic studies in Gentianaceae.

Foi feito o estudo anatômico da folha de Eugenia florida DC., espécie arbórea da família Myrtaceae, coletada no Campus da Fundação Oswaldo Cruz, Rio de Janeiro, RJ. A espécie apresenta importantes propriedades farmacológicas, incluindo-se atividade antiviral. O presente estudo teve como objetivo fornecer dados, revelados através da microscopia óptica e da microscopia eletrônica de varredura, que possam contribuir para o conhecimento da espécie e, conseqüentemente, para a segurança em sua identificação. Anatomicamente, a folha é hipostomática, com organização dorsiventral do mesofilo. Apresenta tricomas simples apenas sobre a nervura mediana da face adaxial. As células epidérmicas apresentam contorno sinuoso em vista frontal e cutícula estriada. O parênquima paliçádico destaca-se pela grande quantidade de cristais prismáticos de oxalato de cálcio. Em posição subepidérmica ocorrem cavidades secretoras de óleos essenciais, pouco numerosas, nas duas faces da lâmina foliar. As células epidérmicas situadas sobre as estruturas secretoras constituem característica de valor diagnóstico e são reconhecíveis pela célula de topo, que é reniforme, circundada pelas adjacentes, que apresentam disposição radiada. A comparação entre folhas de sol e de sombra revela que, nas primeiras, as estruturas secretoras são completamente diferenciadas, ao contrário das folhas de sombra, além de apresentarem maior concentração de compostos ergásticos.


A study of the foliar anatomy of Eugenia florida DC., a tree of Myrtaceae family collected at Fundação Oswaldo Cruz, Rio de Janeiro, RJ, was accomplished. This species presents important pharmacological properties, including antiviral activity. The aim of this research was to furnish data, revealed by optical microscopy and scanning electronically microscopy, in order to contribute to the knowledge of the species and to its safe identification. Anatomically, the leaf is hipostomatic, with dorsiventral mesophyll. Simple unicellular hairs are present only on the adaxial side of the leaf in the midvein region. Anticlinal walls of epidermal cells are undulate and the cuticle is striate. Palisade parenchyma presents a large amount of prismatic oxalate crystals. In subepidermical position there are secretory cavities bearing ethereal oils. These structures occur in a low frequency and are scattered along the both sides of the foliar blade. The epidermal cells that overlap the secretory cavities form an important characteristic anatomical feature. They are recognizable by the top cell, which presents a kidney shape surrounded by cells in a radiate way. The comparison between sun and shade leaves shows a complete development of the secretory cavities and a higher concentration of ergastic compounds in the first ones.

O presente trabalho apresenta a anatomia foliar de Eugenia brasiliensis Lam., cujo óleo essencial possui propriedades anti-reumática, diurética, anti-inflamatória, além de ser ativo contra o Tripanosoma cruzi. O estudo foi feito, comparando-se folhas coletadas em áreas de restinga e de floresta, registrando-se as diferenças estruturais resultantes da influência dos fatores ambientais. Anatomicamente, a folha de E. brasiliensis é hipostomática, glabra, dorsiventral, com tendência à isobilateralidade. A epiderme apresenta notável espessamento da cutícula e seus estratos. Em posição subepidérmica, ocorrem numerosas cavidades secretoras de óleos essenciais, nas duas faces da lâmina foliar. No parênquima lacunoso situam-se drusas de oxalato de cálcio. As folhas provenientes da restinga apresentam, em relação àquelas coletadas na floresta, maior quantidade de estruturas secretoras de óleos essenciais, maior concentração de substâncias ergásticas, maior espessamento da cutícula e da lâmina foliar, maior quantidade de estômatos, mesofilo mais compacto, fibras esclerenquimáticas e elementos xilemáticos com maior grau de lignificação e maior densidade da rede vascular. Os dados obtidos são indicativos que a escolha do local de coleta de E. brasiliensis é relevante, tendo em vista que, na área da restinga, observa-se maior produção de óleos essenciais e demais substâncias associadas aos seus efeitos terapêuticos.


The present paper presents the foliar anatomy of Eugenia brasiliensis Lam. The essential oils produced by this species present anti-reumathic, diuretic and anti-inflammatory properties and show activity against Tripanosoma cruzi. This study was carried out by comparison between leaves developed in restinga and forest areas, in order to register the main differences in the structure of this organ related to environmental conditions. Anatomically, the leaf of E. brasiliensis is hipostomatic, glabrous, with dorsiventral mesophyll, whit tendency towards the isobilateral type. Thickness of the cuticle and cutinized wall is noteworthy. In sub-epidermical position, there are many secretory cavities that produce essential oils, in both sides of the foliar blade. In the spongy parenchyma there are calcium oxalate druses crystals. E. brasiliensis leaves developed in restinga area present, in relation to that proceeding from forest, a larger amount of secretory structures, a higher ergastic substances concentration, thicker cuticle and foliar blade, more abundant stomata, a more compact mesophyll, sclerenchyma fibers and xilem elements with a deeper lignification and a greater development of the vascular system. The data obtained in this research suggest that the choice of the area where E. brasiliensis should be collected is important, because in restinga area it presents a greater production of essential oils and other substances associated with therapeutic effects.
