RESUMO

The yield and quality of potato can be severely affected by bacterial ring rot, which is caused by Clavibacter michiganensis subsp. sepedonicus (Cms). Recently, using natural compounds to control bacteria has received more attention. In this study, five antibacterial compounds from ethyl acetate (EtOAc) extract of Fructus Polygoni orientalis (FPO) against Cms were isolated and the most active compound was screened. Five active compounds were identified as 3,3'-di-O-methylellagic acid (1), 3,3'-di-O-methylellagic acid-4-O-ß-D-glucopyranoside (2), dihydroquercetin (3), protocatechuic acid (4) and quercetin (5). Compound 3 (dihydroquercetin, DHQ) was confirmed as the most active compound. The diameter of inhibition zone (DIZ), minimum inhibitory concentration (MIC), protective efficiency and curative efficiency of DHQ were 22.50 mm, 0.313 mg/mL, 84.49% and 79.63%, respectively, which exceeded these of thiophanate-methyl (TM) in antibacterial activity assays; this indicated that DHQ had satisfactory antibacterial activities against Cms in vitro and in vivo. Results of cell membrane damage assessments indicated that DHQ could reduce membrane potential (MP), disrupt the cell membrane integrity, and promote the leakage of nucleic acids and proteins. Overall, these findings suggested that DHQ could serve as a promising lead molecular against Cms, which could provide a basis for its further derivatization.

RESUMO

Objective To develop an method for detecting the characteristic chromatograms of Qihong decoction by high-performance liquid chromatography coupled with diode arry and evaporative light-scattering detectors (HPLC-DAD-ELSD). Methods The determination was carried out with Venusil MP-C18 (250 mm × 4.6 mm, 5 μm) column,using acetonitrile-0.2% formic acid as mobile phase with gradient elution at a flow rate of 1.0 ml/min and detected at the wave length 254 nm, 290 nm, 365 nm. The drift tube temperature for ELSD was set at 70 ℃, and the nebulizing gas flow rate was 2.8 L/min. Results There were 34 chemical compositions in characteristic chromatograms of Qihong decoction. Among them, 16 peaks came from Polygoni Orientalis Fructus, 17 from Astagali Radix, respectively. A total of 18 chemical constituents were identified. Conclusions The method was simple, steady and reliable which could be applied to the quality control of Qihong decoction.

Objective:To explore the immunosuppression effects and tocolysis of fructus polygoni orientalis on abortion mice induced by LPS.Methods:Mice of Kunming (55 mice) were divided into control group (group A ,10 mice) and experimental group. Experimental group were divided into group B ( intraperitoneal injection LPS ) , group C ( ingestion fructus polygoni orientalis ) , and group D (ingestion fructus polygoni orientalis and intraperitoneal injection LPS ),each group of 15 mice.Then the pregnancy results were observed ,the positive of α-NAE+and the varity of CD 14+and CD204+macrophages ,TNF-αin the uteri were identified by enzyme-histochemistry ,immunohistochemistry and ELISA.Results:The abortion rate and the embryo resorbing rate were all 100%( P<0.01 ) in group B.But there was the decreased abortion rate of 13.33% in group D .The embryo resorbing rate decreased to 10.39%.The number and positive cell area of α-NAE+and CD14+macrophages in the uteri of gestation mice of group B was greatly increased comparing with group A ( P<0.01 ) .These effects outside of myometrium of group D were remarkably increased comparing with group A (P<0.01),but there was no distinct difference in the inside of myometrium and function layer.The number and positive cell area of CD204+macrophages in group C and D was greatly increased comparing with group A and B ( P<0.01 ) .The TNF-αcontents in the uteri of mice in group B were greatly increased comparing with group A (P<0.01),but the positive cell area of CD14,CD204 were close to normal levels in group D.Conclusion:The effect of miscarriage induced by LPS is antagonized by fructus polygoni orientalis through inhibiting the phenotype ,activity and function characteristics of macrophages in the uteri of gravidity mice.

In this study, off-line two-dimensional (2D) complexation high speed counter-current chromatography (HSCCC) was developed for the separation of bovine serum albumin (BSA) binders from the ethyl acetate extract of Fructus polygoni orientalis. Target-guided strategy of BSA functionalized iron oxide magnetic nanoparticles coupled with high performance liquid chromatography-tandem mass spectrometry ((BSA-Fe3O4 MNPs)-HPLC-MS/MS) experiment was proposed. In the orthogonal separation system, a Normal-Phase HSCCC with 0.01mol/L copper ion as complexation agent in the aqueous phase was employed for the first dimension and Recycling HSCCC, Reverse-Phase HSCCC with 0.1mol/L copper ion were used for the second dimension in parallel. Including two pairs of cis-trans isomers, seven BSA binders including 3,5,7-Trihydroxychromone (1), taxifolin (2), N-cis-paprazine (3), N-cis-feruloyltyramine (4), N-trans-paprazine (5), N-trans-feruloyltyramine (6) and an unidentified compound (7) were obtained. The purities of these seven compounds were all over 95.0% as determined by HPLC. The complexation HSCCC behaviors of seven compounds were also investigated by studying their relationship with copper ion. Results showed that the combinative method using (BSA-Fe3O4 MNPs)-HPLC and HSCCC is a quick, efficient, and reproductive technique to isolate potentially bioactive compounds from the complex mixture system of natural products. And the usage of off-line 2D-HSCCC and introduction of chelating metal ion into solvent system are effective ways to implement HSCCC separations in complex samples.

OBJECTIVE:To provide references for the quality assessment of traditional Chinese liquid medicine-Fructus Polygoni Orientalis.METHODS:The description and the physicochemical identification of Fructus Polygoni Orientalis were carried out;the contents of the extractives were determined and the chemical composition of which was studied.The quality of commercial Fructus Polygoni Orientalis medicinal substances from15nationwide areas was investigated.RESULTS:The results showed that the content of the preparation was higher than that of the raw material in the water soluble extractive assaying;the content of the alcohol-soluble extractive was the opposite.Meletin was found to be contained in the Fructus Polygoni Orien?talis acetic ether extraction liquid by the TLC identification.There was only half maturing rate for most of the commercial Fructus Polygoni Orientalis.CONCLUSION:This method can be used as a reference for the quality assessment of Fructus Polygoni Orientalis.

AIM:In order to ensure the quality stability of Fructus Polygoni Orientalis,to study the determination method of the fingerprint of Fruetus Polygoni Orientalis and to establish the fingerprint of Fructus Polygoni Orientalis.METHODS:The HPLC assay was used to establish the fingerprint of Fructus Polygoni Orienlalis and 28 pieces of goods were compared.RESULTS:The fingerprint of Fructus Polygoni Orientalis with 7 common peaks was established.The relative retention time and the ranges of relative area of the common peaks were determined.CONCLUSION:The established fingerprint can be used for the quality control of Fructus Polygoni Orientalis.

Objective To study the activities of the alcoholic extract from Fructus Polygoni Orientalis (EFPO) against antioxidation. Methods The peroxidation in homogenate of rat heart, liver and kidney was induced by ?OH generation system Fe2++ascorbic acid. Malondialdehyde (MDA) was determined by TBA colorimetric method. Superoxide (O-2) from zymosan-stimulated neutrophils of rat was measured by NBT reduction method. H2O2-caused hemolysis of erythrocytes was measured by spectrometry. Results MDA in homogenate of heart, liver and kidney of rat that induced by ?OH generation system Fe2++ascorbic acid was inhibited by 6.3, 12.5, 25, 50, 100, 200 ?g/L EFPO; IC50 of EFPO to MDA in heart, liver and kidney was 31.8, 32.5 and 40.2 ?g/L respectively and its dose-effect relation was of negative relativity and the relative coefficient was -0.886, -0.874 and -0.918 (all P
