RESUMO

The current era witnesses a highly dynamic development of Artificial Intelligence (AI) applications, impacting various human activities. Medical imaging techniques are no exception. AI can find application in image acquisition, image processing and augmentation, as well as in the actual interpretation of images. Moreover, within the domain of radiomics, AI can be instrumental in advanced analysis surpassing the capacities of the human eye and experience. While several certified commercial solutions are available, the validation and accumulation of sufficient evidence regarding their positive impact on healthcare is currently constrained. The role of AI presently leans towards being assistive, yet further evolution is anticipated. Risks and disadvantages encompass dependency on computational power, the quality of input data, and their annotation for learning purposes. The transparency of algorithmic functioning is lacking, and issues pertaining to portability may arise. The integration and utilization of AI introduce entirely new ethical and legislative aspects. Predicting the future development of AI in imaging methods is challenging, with a further increase in implementation appearing more probable.

RESUMO

Depression is a common mental health disorder. With current depression detection methods, specialized physicians often engage in conversations and physiological examinations based on standardized scales as auxiliary measures for depression assessment. Non-biological markers-typically classified as verbal or non-verbal and deemed crucial evaluation criteria for depression-have not been effectively utilized. Specialized physicians usually require extensive training and experience to capture changes in these features. Advancements in deep learning technology have provided technical support for capturing non-biological markers. Several researchers have proposed automatic depression estimation (ADE) systems based on sounds and videos to assist physicians in capturing these features and conducting depression screening. This article summarizes commonly used public datasets and recent research on audio- and video-based ADE based on three perspectives: Datasets, deficiencies in existing research, and future development directions.

The survival rate of pediatric acute myeloid leukemia (pAML) is currently around 60%. While survival has slowly increased over the past few decades, the development of novel agents likely to further improve survival for this heterogeneous patient population has been limited by gaps in the pAML pre-clinical pipeline. One of the major hurdles in evaluating new agents for pAML is the lack of pAML patient-derived xenograft (PDX) models. Unlike solid tumors and other types of leukemias, AML is notoriously hard to establish in mouse models, likely due in part to the need for specific human microenvironment elements. Our laboratory at TCH/BCM addressed this gap by establishing a systematic PDX workflow, leveraging advanced immunodeficient hosts and capitalizing on our high volume of pAML patients and close coordination between labs and clinical sections. Patients treated at TCH are offered the chance to participate in specimen banking protocols that allow blood and bone marrow collection as well as the collection of relevant clinical data. All patients who consent and have samples available are trialed for PDX development. In addition, samples from the Children's Oncology Group (COG) are also trialed for PDX generation. Serially transplanting PDX models are validated using short tandem repeat (STR) and characterized using both targeted DNA/RNA next generation sequencing and RNAseq. As of March 2023, this systematic approach has resulted in 26 serially transplanting models. Models have been shared with requesting labs to facilitate external pAML pre-clinical studies. Available PDX models can be located through the BCM PDX Portal. We expect our growing PDX resource to make a significant contribution to expediting the testing of promising novel therapeutics for pAML.

Numerous cities are currently grappling with the challenge of ecological transformation, especially those categorized as resource-exhausted cities. In these urban areas, land use change is a highly scrutinized issue, as different land use strategies can lead to varied outcomes, impacting the ecological environment in multiple dimensions. Assessing ecosystem health reflects the quality of the regional ecological environment and serves as a comprehensive indicator for evaluating the sustainability and stability of urban ecosystems. To this end, a multi-objective optimization model was constructed to predict land use changes under four future development scenarios (four ecological transformation modes), using Shizuishan City (China), a resource-exhausted city situated in an ecologically fragile area, as an example. The "vigor-organization-resilience" assessment framework was employed to evaluate the ecosystem health conditions in each scenario from three dimensions. The study results showed: (1) The ranking of the average ecological health levels in Shizuishan City for 2022 and different future development scenarios is as follows: Low-Carbon Economic Development Scenario (0.302) > Ecological-Economic Coordinated Development Scenario (0.291) > Baseline Scenario (0.290) > Economic Development Scenario (0.281) > 2022 (0.248). (2) Compared to 2022, the ecosystem health levels under the four ecological transformation modes had all improved, with improvement areas accounting for over 60 %, highlighting the urgent necessity of ecological transformation in Shizuishan City. Among them, the Low-Carbon Economic Development Scenario exhibited the largest improvement area, reaching 75.81 %. (3) Ecological system vitality was identified as the dominant dimension influencing the ecological health in this region. This study emphasized multi-objective development needs and provided an integrated ecosystem health assessment method for assessing the comprehensive ecological effects of future ecological transformation modes in resource-exhausted cities.

The right ventricle (RV) has a critical role in hemodynamics and right ventricular failure (RVF) often leads to poor clinical outcome. Despite the clinical importance of RVF, its definition and recognition currently rely on patients' symptoms and signs, rather than on objective parameters from quantifying RV dimensions and function. A key challenge is the geometrical complexity of the RV, which often makes it difficult to assess RV function accurately. There are several assessment modalities currently utilized in the clinical settings. Each diagnostic investigation has both advantages and limitations according to its characteristics. The purpose of this review is to reflect on the current diagnostic tools, consider the potential technological advancements and propose how to improve the assessment of right ventricular failure. Advanced technique such as automatic evaluation with artificial intelligence and 3-dimensional assessment for the complex RV structure has a potential to improve RV assessment by increasing accuracy and reproducibility of the measurements. Further, noninvasive assessments for RV-pulmonary artery coupling and right and left ventricular interaction are also warranted to overcome the load-related limitations for the accurate evaluation of RV contractile function. Future studies to cross-validate the advanced technologies in various populations are required.

The continuous decline of traditional fossil energy has cast the shadow of an energy crisis on human society. Hydrogen generated from renewable energy sources is considered as a promising energy carrier, which can effectively promote the energy transformation of traditional high-carbon fossil energy to low-carbon clean energy. Hydrogen storage technology plays a key role in realizing the application of hydrogen energy and liquid organic hydrogen carrier technology, with many advantages such as storing hydrogen efficiently and reversibly. High-performance and low-cost catalysts are the key to the large-scale application of liquid organic hydrogen carrier technology. In the past few decades, the catalyst field of organic liquid hydrogen carriers has continued to develop and has achieved some breakthroughs. In this review, we summarized recent significant progress in this field and discussed the optimization strategies of catalyst performance, including the properties of support and active metals, metal-support interaction and the combination and proportion of multi-metals. Moreover, the catalytic mechanism and future development direction were also discussed.

Genome editing techniques are being used to modify plant breeding, which might increase food production sustainably by 2050. A product made feasible by genome editing is becoming better known, because of looser regulation and widespread acceptance. The world's population and food supply would never have increased proportionally under current farming practices. The development of plants and food production has been greatly impacted by global warming and climate change. Therefore, minimizing these effects is crucial for agricultural production that is sustainable. Crops are becoming more resilient to abiotic stress because of sophisticated agricultural practices and a better understanding of the abiotic stress response mechanism. Both conventional and molecular breeding techniques have been used to create viable crop types both processes are time-consuming. Recently, plant breeders have shown an interest in genome editing approaches for genetic manipulation that use clustered regularly interspaced short palindromic repeats (CRISPR/Cas9). To ensure the security of the food supply in the future, plant kinds with desired traits must be developed. A completely new era in plant breeding has begun because of the revolution in genome editing techniques based on the CRISPR/CRISPR-associated nuclease (Cas9) systems. All plants may effectively target a particular gene or group of loci using Cas9 and single-guide RNA (sgRNA). CRISPR/Cas9 can thereby save time and labor compared to conventional breeding methods. An easy, quick, and efficient method for directly altering the genetic sequences in cells is with the CRISPR and Cas9 systems. The CRISPR-Cas9 system, which was developed from components of the earliest known bacterial immune system, allows for targeted gene breakage and gene editing in a variety of cells/RNA sequences to guide endonuclease cleavage specificity in the CRISPR-Cas9 system. Editing can be directed to practically any genomic site by altering the guide RNA (gRNA) sequence and delivering it to a target cell along with the Cas9 endonuclease. We summarize recent CRISPR/Cas9 plant research findings, investigate potential applications in plant breeding, and make predictions about likely future breakthroughs and approaches to food security through 2050.

Next-generation sequencing (NGS) is present in all fields of life science, which has greatly promoted the development of basic research while being gradually applied in clinical diagnosis. However, the cost and throughput advantages of next-generation sequencing are offset by large tradeoffs with respect to read length and accuracy. Specifically, its high error rate makes it extremely difficult to detect SNPs or low-abundance mutations, limiting its clinical applications, such as pharmacogenomics studies primarily based on SNP and early clinical diagnosis primarily based on low abundance mutations. Currently, Sanger sequencing is still considered to be the gold standard due to its high accuracy, so the results of next-generation sequencing require verification by Sanger sequencing in clinical practice. In order to maintain high quality next-generation sequencing data, a variety of improvements at the levels of template preparation, sequencing strategy and data processing have been developed. This study summarized the general procedures of next-generation sequencing platforms, highlighting the improvements involved in eliminating errors at each step. Furthermore, the challenges and future development of next-generation sequencing in clinical application was discussed.

Bacterial infections have become a global public health problem. Rapid and sensitive bacterial detection is of great importance for human health. Among various sensor systems, fluorescence sensor is rapid, portable, multiplexed, and cost-efficient. Herein, we reviewed the current trends of fluorescent sensors for bacterial detection from three aspects (response materials, target and recognition way). The fluorescent materials have the advantages of high fluorescent strength, high stability, and good biocompatibility. They provide a new path for bacterial detection. Several recent fluorescent nanomaterials for bacterial detection, including semiconductor quantum dots (QDs), carbon dots (CDs), up-conversion nanoparticles (UCNPs) and metal organic frameworks (MOFs), were introduced. Their optical properties and detection mechanisms were analyzed and compared. For different response targets in the detection process, we studied the fluorescence strategy using DNA, bacteria, and metabolites as the response target. In addition, we classified the recognition way between nanomaterial and target, including specific recognition methods based on aptamers, antibodies, bacteriophages, and non-specific recognition methods based on biological functional materials. The characteristics of different recognition methods were summarized. Finally, the weaknesses and future development of bacterial fluorescence sensor were discussed. This review provides new insights into the application of fluorescent sensing systems as an important tool for bacterial detection.

Probiotics have several health benefits to the host. However, low pH in the stomach, various digestive enzymes and bile salts in the intestine threaten their viability and function. Thus, probiotics need to be protected during gastric transit to address challenges associated with low viability and impaired function. At present, probiotic delivery systems with different trigger mechanisms have been constructed to successfully introduce numerous high-viability probiotics to the intestine. On this basis, the application of non-targeted/targeted probiotic delivery systems in different gut microenvironment and the adjuvant therapeutic effect of probiotic delivery systems on other disease were discussed in detail. It is important to also note that most of the current studies in this area focused on non-targeted probiotic delivery systems. Moreover, changes in intestinal microenvironment under disease state and discontinuous distribution of disease site limit their development. Thus, emphasis were made on the optimization of non-targeted probiotic delivery systems and the necessity of designing more precisely targeted ones.

Cerebral small vessel disease (CSVD) is the most common chronic vascular disease involving the whole brain. Great progress has been made in clinical imaging, pathological mechanism, and treatment of CSVD, but many problems remain. Clarifying the current research dilemmas and future development direction of CSVD can provide new ideas for both basic and clinical research. In this review, the risk factors, biological markers, pathological mechanisms, and the treatment of CSVD will be systematically illustrated to provide the current research status of CSVD. The future development direction of CSVD will be elucidated by summarizing the research difficulties.

Clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 is the third generation of novel targeted genome editing technology after zinc finger nucleases (ZFNs) and transcription activator like effector nucleases (TALENs). It is also one of the most promising techniques for mutating and modifying genes. The CRISPR-Cas9 system has the advantages of simplicity, high efficiency, high specificity, and low production cost, thus greatly promoting the study of gene function. Meanwhile, it has attracted the attention of biologists. After the development and improvement in recent years, CRISPR-Cas9 system has become increasingly mature and has been widely used in crop improvement. Firstly, this review systematically summarizes the generation and advantages of CRISPR-Cas9 system. Secondly, three derivative technologies of the CRISPR-Cas9 system are introduced. Thirdly, this review focuses on the application of CRISPR-Cas9 system in gene knockout, gene knock-in, and gene regulation, as well as the improvement of yield, quality, and biological resistance of important crops such as rice, wheat, soybean, corn, and potato. Finally, this review proposes the potential challenges of CRISPR-Cas9 system, and discusses the future development of CRISPR-Cas9 system.

The water resource carrying capacity (WRCC) is a carrying capacity of natural resources. It affects the application and expansion of the carrying capacity of water resources. This subject involves various elements, such as water resources, the ecological environment system, humans and their economic and social systems, and a wider range of biological groups and their survival needs. Based on the objective recognition of the complex relationship between the water resource system, ecological environment system, and economic and social system, the support scale of water resources and the ecological environment for economic and social development is studied. Current research on the carrying capacity of water resources has mostly shifted from the previously limited support capacity of water resources to include factors such as the population, economy, and ecology, establishing the internal relationships between the economics, water resources, and ecological environment. This reflects the comprehensive carrying capacity of the entire region (or river basin) of water resources and the ecological environment system on an overall economic and social scale. Based on the conceptual connotation of the WRCC and the actual problems facing water resources in Henan Province, the paper uses a system dynamics method to develop information feedback between the four subsystems of Henan Province: economic, population, water resource, and water environment subsystems. The index system of the WRCC in Henan Province is also determined. The weight of each index is comprehensively determined by a combination weighting method of the analytic hierarchy process and an entropy weight method, and then a fuzzy comprehensive evaluation method is used to evaluate the WRCC of Henan Province under four different development models. The validation period of the model is 2010-2020, and the forecast period is 2021-2030. The results indicate that during the period 2021-2030, the WRCC of Henan Province showed a slight upward trend overall under the four models, but the increase rates were different under the different models. Among the four models, the comprehensive model's benefit was the best, which not only maintained the healthy and stable development of the economy and society but also improved the pressure on the water resources and the quality of the water environment.

The balance and optimization of ecosystem services (ESs) are the basis of spatial planning and ecological landscape design. Selecting suitable ESs and developing integrated, quantitative, and spatially explicit assessment models is the key to balance research. In Xilinhot in eastern Inner Mongolia, China, grain production, animal husbandry production, and habitat quality are key ESs that affect the livelihoods of local farmers and herders and the regional ecological balance. Based on GlobeLand30 data for 2000-2020, we designed six future land scenarios for the region and used the future land use simulation (FLUS) model to simulate the land use/cover scenarios in 2030. Then, we analyzed staple grain production (SGP), sustainable stocking capacity (SSC), and habitat quality (HQ) under each scenario, and constructed a multi-ES comprehensive trade-off method, using the comprehensive trade-off score (CTS) to measure their overall development quality. The results show the following. (1) Under various scenarios, the SGP is negatively correlated with SSC and HQ, and the SSC is positively correlated with HQ. (2) In the inertial development scenario, the economic development priority scenario, and the quality habitat protection scenario, the SGP will increase, the SSC and HQ will decrease; among these scenarios, the economic development priority scenario is the most significant; their CTSs are 0.97, 0.95, and 0.98, respectively. In the ecological comprehensive governance scenario, the SSC will increase, SGP and HQ will decrease, and CTS is 0.98. (3) Based on the comprehensive trade-off analysis, the economic and ecological coordination scenario is the most beneficial for regional sustainable development. It achieves the stability of the SGP; although the SSC decreases slightly, the negative impact can be offset by a larger improvement in HQ. This paper provides clear policy suggestions for regional development, and the methodological framework we have constructed provides a reference for the study of complex land scenario simulations and multi-ecosystem service comprehensive trade-offs.

Objective:To evaluate the present situation and future development for undergraduate major construction of health service and management.Methods:One hundred and twenty-three domestic universities with undergraduate health service and management major were selected as the research subjects. This paper analyzed the present situation of major distribution and aim setting, faculty, curriculum system, construction of teaching material, enrollment number, graduate destination, among other areas, using network data collection, questionnaire survey, and descriptive analysis methods. It also analyzed the development trend of this major with regards to the national policy guidance and discipline development dynamics.Results:The results showed that the number of domestic universities with undergraduate major for health service and management had increased from 5 in 2016 to 123 at present, with an average annual growth rate of 472.0%. The curriculum system of various universities covered 13 core courses, such as management and introduction to basic medicine. The total credits required for graduation ranged from 149 to 224, and the proportion of practical teaching in the teaching plan ranged from 12.1% to 45.1%. The credit requirements varied greatly; moreover, the proportion of practical teaching in most universities was not high and the content was narrow. Among the 21 universities participating in the questionnaire survey, 47.6% of them focused their education on cultivating versatile talents, 52.4% had 4-6 full-time teachers, 47.2% of all full-time teachers were 31-40 years old, 42.9% universities had teachers with senior professional titles accounting for 30%-40% of the faculty, and 28.6% universities had teachers with doctoral degrees comprising ≥50% of the faculty. The aim setting of most universities was more or less vague, the number of full-time teachers couldn′t meet the needs of major development, and the connotation construction lagged relatively.Conclusions:The inconsistency of the rapid increasing number of universities with the connotation construction of major has become a key barrier to the connotative development of undergraduate major of health service and management. In the future, building the quality of teaching according to national standards; advancing the integration of government, industry, university, research, and implementation; improving faculty quality; and optimizing the curriculum system will be part of the development trend.

With the development of precision medicine, molecular targeted therapy has been widely used in the field of cancer, especially in non-small-cell lung cancer (NSCLC). Epidermal growth factor receptor (EGFR) is a well-recognized and effective target for NSCLC therapies, targeted EGFR therapy with EGFR-tyrosine kinase inhibitors (EGFR-TKIs) has achieved ideal clinical efficacy in recent years. Unfortunately, resistance to EGFR-TKIs inevitably occurs due to various mechanisms after a period of therapy. EGFR mutations, such as T790M and C797S, are the most common mechanism of EGFR-TKI resistance. Here, we discuss the mechanisms of EGFR-TKIs resistance induced by secondary EGFR mutations, highlight the development of targeted drugs to overcome EGFR mutation-mediated resistance, and predict the promising directions for development of novel candidates.

Based on early experiences and current conditions, a future time perspective influences college students' behaviors, while psychological violence critically threatens college students' health. This study explored the relationship between a future time perspective and the psychological violence of perpetrators based on an online investigation of 1424 college students (87.1% women) aged 17 to 31 in China. The results showed that a future time perspective is significantly positively associated with psychological violence. Positive future orientation is negatively associated with psychological violence. Negative and confused future orientations are positively associated with psychological violence. These findings support the need to introduce an intervention regarding a future time perspective to reduce psychological violence among college students.

Botulinum toxin (BT) drugs were introduced in the late 1980s. They are now used worldwide in a large number of indications. This huge market and its future opportunities have attracted a number of companies about to enter the competition with projects to develop new BT drugs. We want to outline features of BT drugs that are relevant for their therapeutic use and - with that - are also relevant to compare and to evaluate new BT drugs and to guide their further development. BT drugs may vary in their content of botulinum neurotoxin, complexing proteins and excipients. Their manufacturing is complex and directly influences core features of the final drug. It includes breeding, purification, botulinum neurotoxin activation, stabilisation, potency control, labelling and testing, specific biological activity and packaging. The manufacturer's support concerning product documentation and support, reliability of drug supply, counterfeit protection and - last but not least - competitive pricing is also important. Further developments include the indication spectrum, the market penetration, the drug's duration of action, liquid preparations, transdermal applications, improving antigenicity and a bio-similarity registration process. Most projects, however, will try to produce Botox® analogs at reduced sale prices.

The general public's attitudes, demands, and expectations regarding medical AI could provide guidance for the future development of medical AI to satisfy the increasing needs of doctors and patients. The objective of this study is to investigate public perceptions, receptivity, and demands regarding the implementation of medical AI. An online questionnaire was designed to investigate the perceptions, receptivity, and demands of general public regarding medical AI between October 13 and October 30, 2018. The distributions of the current achievements, public perceptions, receptivity, and demands among individuals in different lines of work (i.e., healthcare vs non-healthcare) and different age groups were assessed by performing descriptive statistics. The factors associated with public receptivity of medical AI were assessed using a linear regression model. In total, 2,780 participants from 22 provinces were enrolled. Healthcare workers accounted for 54.3 % of all participants. There was no significant difference between the healthcare workers and non-healthcare workers in the high proportion (99 %) of participants expressing acceptance of AI (p = 0.8568), but remarkable distributional differences were observed in demands (p < 0.001 for both demands for AI assistance and the desire for AI improvements) and perceptions (p < 0.001 for safety, validity, trust, and expectations). High levels of receptivity (approximately 100 %), demands (approximately 80 %), and expectations (100 %) were expressed among different age groups. The receptivity of medical AI among the non-healthcare workers was associated with gender, educational qualifications, and demands and perceptions of AI. There was a very large gap between current availability of and public demands for intelligence services (p < 0.001). More than 90 % of healthcare workers expressed a willingness to devote time to learning about AI and participating in AI research. The public exhibits a high level of receptivity regarding the implementation of medical AI. To date, the achievements have been rewarding, and further advancements are required to satisfy public demands. There is a strong demand for intelligent assistance in many medical areas, including imaging and pathology departments, outpatient services, and surgery. More contributions are imperative to facilitate integrated and advantageous implementation in medical AI.

RESUMO

The rapid development of medical rehabilitation services in China over the last 30 years has progressed from minimal to the present various models of rehabilitation services in general hospitals, specialized rehabilitation hospitals, and community-based and home-based rehabilitation. Contributing factors such as rapid economic growth, rehabilitation needs after natural disasters, changes in insurance policies, an aging population, and new government policies have accelerated the growth in rehabilitation services. Persons with disability in China include 85 million permanently disabled persons, more than 200 million with disabilities from chronic diseases, and more than 240 million elderly individuals.
