RESUMO

An undescribed substituted dihydroxanthene-1,9-dione, named funiculosone, was isolated together with its two analogues identified as mangrovamide J and ravenelin, from the culture filtrates of Talaromyces funiculosus (Thom) Samson, Yilmaz, Frisvad & Seifert (Trichocomaceae), an endolichenic fungus isolated from lichen thallus of Diorygma hieroglyphicum (Pers.) Staiger & Kalb (Graphidaceae), in India. Funiculosone was characterized, essentially by spectroscopic methods, as 4,8,9a-trihydroxy-3,4a-dimethyl-4a,9a-dihydro-4H-xanthene-1,9-dione. Its relative stereochemistry was deduced by single crystal X-ray analysis while the absolute configuration was assigned as 4S,4aS,9aS by ECD spectra in comparison to that of the closely related mangrovamide J. This latter, to which, not being an amide, an inappropriate common name was given, was only recently isolated, together with undescribed and known prenylatedindole alkaloids and chromone derivatives from an unidentified Penicillium sp. X-ray structural analysis of the isolated mangrovamide J, for which no biological activity was previously reported, revealed polymorphism and a new crystalline phase is described. All the compounds displayed antibacterial activity with an IC50 range 23-104 µg/mL when assayed against Escherichia coli Escherich and Staphylococcus aureus Ogston. Funiculosone also showed anticandidal activity against Candida albicans Berkhout with an IC50 35 µg/mL.

Abstract In the frame of an ongoing lichen inventory of Atlantic Rainforest remnants in Northeast Brazil, five new species of Graphidaceae were discovered in a small forest fragment, Mata do Cipó, in Sergipe state, the smallest state of Brazil and among those with the highest deforestation rate in the country. An additional new species had already been collected in Panama before and was now also found in the Mata do Cipó and is described here as well. In total, 40 species of Graphidaceae are reported for this remnant, including a large number of taxa indicative of well-preserved rainforest. The new species are: Fissurina atlantica T.A. Pereira, M. Cáceres & Lücking, sp. nov., Graphis subaltamirensis Passos, M. Cáceres & Lücking, sp. nov., Ocellularia cipoensis L.A. Santos, M. Cáceres & Lücking, sp. nov., O. sosma T.A. Pereira, M. Cáceres & Lücking, sp. nov., O. submordenii Lücking, sp. nov. (also known from Panama), and Pseudochapsa aptrootiana M. Cáceres, T.A. Pereira & Lücking, sp. nov. The findings are discussed in the context of the strong fragmentation of the Atlantic Rainforest, with individual remnants apparently serving as refugia for residual populations of rare species of lichen fungi that were more widely distributed in the past, but currently seem to occur only in isolated fragments.


Resumo No âmbito de um inventário atual de liquens dos remanescentes da Mata Atlântica, no Nordeste do Brasil, cinco espécies novas de Graphidaceae foram descobertas em um pequeno fragmento florestal, Mata do Cipó, no estado de Sergipe, o menor estado do Brasil, o qual está entre aqueles com maior taxa de desmatamento no país. Uma nova espécie adicional já havia sido coletada no Panamá, anteriormente, e agora também foi encontrada na Mata do Cipó, e está sendo descrita aqui também. No total, 40 espécies de Graphidaceae são relatadas para este remanescente, incluindo um grande número de táxons indicadores de floresta bem preservada. As novas espécies são: Fissurina atlantica T.A. Pereira, M. Cáceres & Lücking, sp. nov., Graphis subaltamirensis Passos, M. Cáceres & Lücking, sp.nov., Ocellularia cipoensis L.A. Santos, M. Cáceres & Lücking, sp. nov., O. sosma T.A. Pereira, M. Cáceres & Lücking, sp. nov., O. submordenii Lücking, sp. nov. (também conhecido do Panamá), e Pseudochapsa aptrootiana M. Cáceres, T.A. Pereira & Lücking, sp. nov. Os achados são discutidos no contexto da forte fragmentação da Mata Atlântica, com remanescentes aparentemente servindo de refúgio para populações residuais de espécies raras de fungos liquenizados que, provavelmente, tiveram uma distribuição mais amplia no passado, mas que atualmente parecem ocorrer apenas em fragmentos isolados.

In addition, the study includes Graphis tenoriensis and Phlyctis karnatakana reported for the first time from China.

The species is characterized by a loosely corticate, rough, whitish grey to greyish green thallus, elongate and irregularly branched lirellae with an apically thin complete thalline margin (negrosina morph), laterally carbonized, entire proper exciple, clear hymenium, hyaline, 16~20 transversely locular ascospores, and about 50~95 × 10~15 µm in size. In addition, members of the taxon produce norstictic and stictic acids. Currently, the lichen flora of Vietnam include Arthonia radiata, Brigantiaea tricolor, Coenogonium implexum, Dirina paradoxa, Herpothallon sipmanii, Pertusaria pertusa, and Sarcographina cyclospora.

PREMISE OF THE STUDY: This study elucidates the phylogenetic position of a unique taxon of Graphidaceae occurring on rock in coastal desert areas, assessing its importance for our understanding of the evolution of the largest family of tropical lichenized fungi. • METHODS: We used maximum likelihood and Bayesian approaches to reconstruct a three-gene phylogeny of Graphidaceae and a Bayesian molecular clock approach to estimate divergence dates for major clades, as well as Bayesian ancestral ecogeography state analysis. • KEY RESULTS: The new genus Redonographa represents a new subfamily, Redonographoideae, sister to subfamily Graphidoideae. Redonographa is exclusively saxicolous and restricted to the American Pacific coast from California to central Chile, including Galapagos. It contains four species: Redonographa chilensis comb. nov., R. saxiseda comb. nov., R. saxorum comb. nov., and R. galapagoensis sp. nov. The genus Gymnographopsis, with a similar ecogeography but differing in excipular carbonization and chemistry, is also included in Redonographoideae, with the species G. chilena from Chile and G. latispora from South Africa. Molecular clock analysis indicates that Redonographoideae diverged from Graphidoideae about 132 million years ago (Ma) in the Early Cretaceous. • CONCLUSIONS: The divergence date for subfamilies Redonographoideae and Graphidoideae coincides with the early breakup of Gondwana and ancient origin of the Atacama Desert. However, the common ancestor of Redonographoideae plus Graphidoideae was reconstructed to be tropical-epiphytic. Thus, even if Redonographoideae is subtropical-saxicolous, the hypothesis that Graphidoideae evolved from a subtropical-saxicolous ancestor is not supported.

The taxon is characterized by thick, bulging thallus with many calcium oxalate crystal inclusions; an immersed, round to irregular ascomata with free exciple, ellipsoid to ± roundish submuriform, brown ascospores, and lack of secondary metabolites. A detailed taxonomic description and comments are presented for this taxon. The lichen genus Leucodecton is reported for the first time in South Korea.
