RESUMO

To evaluate the impact on the quality of filling with of low-fusion and conventional gutta-percha cones. Thirty-six maxillary canines were prepared and divided into three groups: I-conventional cone with Downpack at 200 °C at 4 mm from the WL; II-low-fusion cone with Downpack at 100 °C up to 4 mm from the WL; III-low-fusion cone with Downpack at 100 °C up to 7 mm from the WL. Temperature variations were measured in thirds on the external surface of the root. The bond strength was evaluated using the push-out test. The adhesive interface was analyzed by scanning electron microscopy. The bond strength and the temperature variation data were analyzed using analysis of variance and the failure type using the chi-square test. The low-fusion cone group with 7 mm Downpack showed higher bond strength (4.2 ± 2.7) compared with conventional cones (2.8 ± 1.6) and low-fusion cones with 4 mm Downpack (2.9 ± 1.6) (p < 0.05), with occurrence of a higher number of adhesive failures to the filling material and mixed failures. Relative to temperature variation, there was less temperature change in the apical third, in the low-fusion cone with Downpack 7 mm (1.0 ± 1.0) (p < 0.05). The use of low-fusion cones allowed the continuous wave condensation technique to be performed at a lower depth of Downpack at 100 °C at 7 mm, with less heating in the apical third, without compromising the quality of filling. Using gutta-percha cones with low fusion, which permits a lower condensation temperature and reduced Downpack depth, maintains the quality of filling, in order to minimize possible damage to the periapical tissues.

RESUMO

Context: Gutta-percha (GP) gets contaminated during handling. It becomes imperative to ensure GP is sterile before placement in root canal space. Aims: The aim of the study was to evaluate and compare the tensile strength of GP treated with four different disinfectant solutions: 3% sodium hypochlorite (NaOCl), amla juice (AJ), Aloe vera (AV) juice, and pancha tulsi (PT). Settings and Design: The study design was an in vitro study. Materials and Methods: Fifty GP cones with a size of 30 were procured from sealed packages in five different groups. Experimental groups were disinfected for 1 min with 3% NaOCl, AJ, AV, and PT except the control group. Tensile and Brinell Testing Machine is used to measure the tensile strengths of GP. Statistical Analysis Used: Results were subjected to statistical analysis using the Kruskal-Wallis test followed by Dunn's post hoc test. Results: The mean tensile strength values for Groups A, B, C, D, and E are 9.49 Mpa, 13.33 Mpa, 12.58 Mpa, 12.69 MPa, and 13.56 MPa, respectively. In the herbal disinfectant group, such as AJ, AV, and PT, the tensile strength was not significantly altered, whereas in the 3% NaOCl group, it was reduced considerably. Conclusions: AJ, AV juice, and PT as a GP disinfectant do not alter the tensile strength of GP cones.

RESUMO

AIM: The aim of this ex vivo randomized study is to evaluate the efficiency of gutta-percha cones that match a nickel-titanium instrumentation system and nonmatching greater taper cones, when used with continuous warm vertical condensation technique. MATERIAL AND METHODS: Thirty-six straight canals were prepared using ProTaper Next files, and the apical third was obturated using either ProTaperNext cones (group A), ISO uniform greater taper cones (group B), or nonstandardized cones (group C). Cone adaptation time was quantified by the number of required modifications. Micro-computed tomography was used to measure voids and sealer percentage. RESULTS: There was no significant difference between the groups regarding void volume (p = 0.666), percentage (p = 0.379), and the number of modifications (p = 0.757). Sealer percentage, however, was significantly lower in group B when compared to group A (p = 0.0194). CONCLUSION: In straight canals, matching gutta-percha cones were not associated with significantly better obturation or saving time to fit the cone. CLINICAL SIGNIFICANCE: Using gutta-percha cones that do not match a nickel-titanium instrumentation system to obturate the straight canals with continuous warm vertical condensation technique is as efficient as using matching cones in terms of obturation quality and ease of cone fit.

RESUMO

In this study, we evaluated the effectiveness of 1 and 5.25% NaOCl solutions, with or without surfactant, in disinfecting gutta-percha cones. Surface changes of the gutta-percha cones after disinfection were also assessed under scanning electron microscopy (SEM). One-hundred and eight gutta-percha cones previously contaminated with Enterococcus faecalis were left in contact for 30 s or 1 min with the following solutions (n = 12): 1% NaOCl; 1% NaOCl + 2% cetrimide; 5.25% NaOCl; 5.25% NaOCl + 2% cetrimide. Positive control group was composed by the remaining contaminated cones (n = 12), and the negative control group by noncontaminated cones (n = 12). The cones were kept in Eppendorf tubes containing Trypticase Soy Broth at 37°C for 24 hr, and then, the presence of turbidity was analyzed. Ten additional cones (n = 2) were observed under SEM to evaluate surface changes after 1 min of disinfection. The data analysis (Kruskal-Wallis test, α = 5%) demonstrated that regardless the presence of surfactant, the NaOCl solutions used were effective in the disinfection when cones were immersed for 1 min. However, at the 30-s period, only the 5.25% NaOCl solution was effective. SEM analysis demonstrated that NaOCl solutions promoted the formation of sodium chloride crystals on cones surface, less visible when the 1% NaOCl associated to surfactant was used. The addition of surfactant did not affect the disinfection capacity of NaOCl solutions. However, the surfactant has apparently inhibited the sodium chloride crystals deposition on the cones surface when associated to 1% NaOCl solution.

RESUMO

This work explored a novel approach to enhance the antibacterial activity of commercial Gutta-percha (GP) cones, the most commonly used core filling materials used in endodontic treatment. The reported procedure involved an argon (Ar) plasma treatment (PT) of the GP cone surface, followed by the deposition of a ZnO thin film by magnetron sputtering. The resulting surfaces were evaluated for surface topography, antibacterial activity against Enterococcus faecalis and Staphylococcus aureus, and cytocompatibility with human osteoblastic cells. GP cones treated with NaOCl, a routine chair-side protocol, were also tested as reference. The deposition of a ZnO film on pristine GP cones increased its antibacterial activity. Cones pre-treated with Ar-plasma (PT) and coated with the ZnO thin film presented significantly higher antibacterial activity than that observed on the pristine and, also, compared to the ZnO coated cones. The higher antibacterial activity of PT + ZnO cones appears related to the major effects induced by the PT pre-treatment on the cone surface endowing the deposited ZnO film with a homogeneous nanostructured topography that greatly improved surface reactivity. The modified GP cones maintained an appropriate cytocompatibility with human cells. This novel approach provides ready-to-use cones with enhanced antibacterial activity, improving a strict asepsis protocol during endodontic treatment and preventing secondary endodontic infections.

Aim: To evaluate in the tugback and apical adaptation at the working length of four different mechanized systems gutta-percha cones in simulated curved canals. Materials and methods: Four Endo Training Blocks (Dentsply Sirona) with simulated curved canals were instrumented up to the working length established at the orifice located in the canal exit. A metal stop was adjusted in the concavity present at the exit of the canal to ensure the working length during instrumentation in all blocks. The metal stop was maintained using a table walrus. Canal patency was achieved with a #10 K file (Dentsply Sirona). Then the instrumentation protocols for the different systems were performed in each Endo Training Block. ProTaper Next (Dentsply Sirona), ProTaper Gold (Dentsply Sirona), WaveOne Gold (Dentsply Sirona) and Reciproc Blue (VDW) were used according to the manufacturers' instructions (groups 1, 2, 3 and 4 respectively). The canals were irrigated with distilled water between instruments and at the end of the preparation and dried with paper points. Then, the tugback and the apical adaptation at the working length of 10 gutta-percha cones of each system were evaluated in the corresponding Endo Training Block. For the tugback analysis, the gutta-percha cones were introduced in the respective simulated canal and both were raised up to 10 cm in height, holding them for 10 seconds. If the cones remained intact in their position, it was considered that the cone tugback was appropriate, if the cones were detached, a lack of tugback was considered. Likewise, a clinical operating microscope at 8.5X was used to confirm the apical adaptation to the working length in each case. For the overall evaluation the following categories were considered: 1, the gutta-percha cone reached the working length with tugback; 2, the gutta-percha cone reached the working length without tugback; 3, the gutta-percha cone did not reach the working length; 4, the gutta-percha cone exceeded the working length. Results: In three groups 1, 3 and 4 they were not found intra-systems variables in the tugback at the working length. In one group there were observed intra-systems variables but the differences between groups were not statistically significant (P>0.05) (AU)

Objective: The aim of the present study was to verify if the Protaper Next instruments X1, X2, and X3 and the Protaper Universal gutta-percha cones F1, F2, and F3 meet the ANSI/ADA 101 and 78 recommendations. Material and Methods: The instruments' dimensions were compared with those obtained by the gutta-percha cones. For this, ten instruments and ten cones of each type were used. Micromorphometric images were obtained and analyzed through a stereomicroscope (OptiCam) and measurements were obtained with TSView software, version 7.2.1.7. The D3, D6, and D9 diameters were compared with the nominal diameters using the Student's t-test. After this, the real diameters of the instruments D3, D6, and D9 were compared with the gutta-percha cone diameters by Anova supplemented with SNK test for multiple comparisons. A p ≤0.05 was considered significant. Results: The results showed statistically significant differences between the real diameter and nominal diameter of the instruments (p <0.05). The X3 instrument showed a statistically significant difference when compared to the F1 cone (p <0.05), and the cone values were lower than the instrument values. Conclusion: Within the limitations of this study, it can be concluded that: 1) the X1 instrument and the gutta-percha points did not meet the recommendations, and 2) the F1 cone can be an option for filling the modeled channels with the X3 instrument, but termoplastification is required.

RESUMO

PURPOSE: To evaluate the alterations on the surface of gutta-percha cones (GPCs) on exposure to the different irrigation solutions and their possible antibacterial effect against Enterococcus faecalis. (E. faecalis). MATERIALS AND METHODS: Disinfection ability of different solutions (5.25% sodium hypochlorite, 2% chlorhexidine, 1% peracetic acid, and QMix) were tested with 96 GPCs and the time of exposure to each solution was 5 and 10 minutes, respectively. GPCs used in this study were contaminated with E.faecalis. After disinfection, GPCs were placed in tubes containing the medium and incubated at 37˚C for 7 days. All tubes were visually checked for turbidity at 24-hour intervals. About 92 new GPCs were analyzed by means of SEM/EDS to assess the topography and chemical elements present on their surface. The data generated was analyzed using Pearson chi-square test, p<0.05. RESULTS: There were no significant statistical differences in disinfection quality between the irrigation solutions used on GPCs contaminated with E. faecalis (p>0.05). SEM/EDS analyses showed no alteration in the superficial features of GPCs after treating with various irrigation solutions. CONCLUSION: QMix was found to be an effective agent for rapid disinfection of GPCs as well-known irrigation solutions. Irrigation solutions were found to have sterilized the GPCs after both 5 and 10 minutes of exposure.

RESUMO

AIM: To evaluate and compare the tensile strength values and influence of taper on the tensile strength of Gutta-percha (GP) cones after disinfection with sodium hypochlorite (SH) and Aloe vera gel (AV). MATERIALS AND METHODS: Sixty GP cones of size 110, 2% taper, 60 GP cones F3 ProTaper, and 60 GP of size 30, 6% taper were obtained from sealed packs as three different groups. Experimental groups were disinfected with 5.25% SH and 90% AV gel except the control group. Tensile strengths of GP were measured using the universal testing machine. RESULTS: The mean tensile strength values for Group IA, IIA and IIIA are 11.8 MPa, 8.69 MPa, and 9.24 MPa, respectively. Results were subjected to statistical analysis one-way analysis of variance test and Tukey post-hoc test. 5.25% SH solutions decreased the tensile strength of GP cones whereas with 90% AV gel it was not significantly altered. CONCLUSION: Ninety percent Aloe vera gel as a disinfectant does not alter the tensile strength of GP cones.

RESUMO

AIM: The aim of this study was to evaluate the antimicrobial efficacy of Aloe vera and to determine its effectiveness in decontaminating gutta percha cones. MATERIALS AND METHODS: A concentrated extract of Aloe vera was used to check for the antimicrobial efficacy using the agar well diffusion method. Presence of zones' of diffusion was identified against three common GP contaminants namely, E.coli, E.faecalis and Staph. aureus. New GP Cones, freshly taken out of the packet were then decontaminated for 1minute using Aloe vera gel and then placed in thioglycolate broth to check for the presence of turbidity. RESULTS: The zones of inhibition on the agar plate were measured as 24mm,21mm and 24mm respectively. The broth remained clear even after 48 hours of incubation. CONCLUSION: We conclude that Aloe vera is indeed effective as a GP decontaminant and it holds a promising future as a medium for storage of GP cones.

RESUMO

This study investigated the effectiveness of detergent and aqueous solutions of 2 percent chlorhexidine digluconate in decontaminating gutta-percha cones (gpc) contaminated with bacteria, yeast, or bacterial spores. Gutta-percha cones were contaminated with 10(7)-10(8) colony-forming units per milliliter (cfu/ml) of the following test organisms: Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, or Candida albicans. Spores of Bacillus subtilis were also tested. Contaminated gpc were treated with the chlorhexidine solutions for 1, 5, 10, or 15 min. Each cone was then transferred to a tube containing saline and the micoorganisms were recovered after homogenization for cfu determination. Both detergent and aqueous chlorhexidine solutions were effective in eliminating S. aureus, E. faecalis, and C. albicans cells adhered on the surface of gpc within 1 min of exposure. E. coli was eliminated in 5 min with detergent solution. The Bacillus subtilis spores were eliminated by chlorhexidine solutions within 5 min. The results of this study demonstrated that both aqueous and detergent solutions of 2 percent chlorhexidine digluconate were effective in decontaminating gpc within 5 minutes of exposure.

The cones of gutta-percha are the main root canal filling materials. The aim of this study was to evaluate the effectiveness of some products in the decontamination of these cones. Eighty (80) cones of gutta-percha #40, separated in 8 groups with 10 cones each one, have been used as sampling. Seventy cones have been contaminated with Enterecoccus faecalis for immersion in saline solution contends 105 to 108 bacterial cells/mL. The groups have suffered decontamination for G1: alcohol 70%; G2: alcohol 70% + iodine 1%; G3: alcohol 70% + chlorexidine 4%; G4: chlorexidine 4%; G5: NaOCl 2,5%; G6: NaOCl 5,25%; G7: saline solution; G8: it was not contaminated and it did not suffer decontamination (control). After 1 minute in contact with each product, five (5) cones have been removed, washed in sterile saline solution and introduced individually in tubes containing BHI solution. The other cones have been removed after 5 minutes in contact with the solutions, repeating the same process of the previous ones. The set of tubes have been agitated per 1 minute and taken to the sterilizer and kept there at 37ºC for 48 hours. The tubes that presented turbidity of BHI solution have been considered positive. It could be observed that in the time of 1 minute there was a bacterial growth in groups 1, 2, 5 and 7 and in the time of 5 minutes only groups 5 and 7 had presented bacterial growth. Group 8 presented one contaminated cone. With the used methodology, we concluded that the alcohol 70% + chlorexidine 4%, chlorexidine 4% and NaOCl 5,25% solutions did not allow the development of E. faecalis, promoting therefore good disinfection of the gutta-percha cones in an adjusted time for clinic practice
