RESUMO

Low temperature stress has adverse effects on fish growth and reproduction, causing huge economic losses to the aquaculture industry. Especially, black porgy (Acanthopagrus schlegelii) farming industry in north of Yangtze River has been severely affected by low temperature for a long time. To explore the tolerance mechanism of black porgy to low temperature stress, the experiment was designed. The liver and gill tissues of black porgy were taken from the water temperature point of 15 °C (control group named as CG), 3.8 °C (cold sensitive group named as CS) and 2.8 °C (cold tolerant group named as CT) with a cooling rate of 3 °C/d from 15 °C for histophysiology, transcriptomics and metabolomics analysis. After cold stress, the histological results showed that the nucleus of the black porgy liver tissue appeared swelling, the cell arrangement was disordered; meanwhile the gill lamellae were twisted and broken, the epidermis was detached and aneurysm appeared. In addition, the expression of antioxidant, glucose metabolism and immune-related enzymes in the liver and gill of black porgy also changed significantly after low temperature stress. By analyzing the transcriptome and metabolome dates of black porgy liver, 3474 differentially expressed genes (DEGs) and 689 differentially expressed metabolites (DEMs) involved in low temperature stress were identified, respectively. The results of the transcriptome and metabolome combined analysis showed that individuals in the CS group mainly supplied energy to the body through lipid metabolism and amino acid metabolism, and meanwhile the apoptosis pathway was activated. While, individuals in the CT group mainly through glucose metabolism and steroid hormone biosynthesis to supply energy for the body. The validation results of qPCR on eight functional genes further demonstrated the reliability of RNA-Seq data. In summary, the results provide molecular information about adaptation to climate change and genetic selection of black porgy.

In the present study, we set out to determine whether melatonin combined with a high-fat diet during pregnancy and lactation can prevent liver disorders in offspring. Forty rats were divided into four groups: DC - pregnant rats submitted to the standard diet; DC + Mel - pregnant rats submitted to the standard diet combined with melatonin; HFD - pregnant rats submitted to a high-fat diet; HFD + Mel - pregnant rats submitted to a high-fat diet combined with melatonin. Morphophysiological and biochemical parameters were analyzed. Melatonin (5 mg/kg) was administered intraperitoneally. The HFD group offspring showed an increase in AST, ALT, alkaline phosphatase, cholesterol, triglycerides, LDL and glucose levels, and a reduction in HDL and lipase levels. In the liver obseved steatosis, hepatocellular ballooning, increased lobular parenchyma and reduced non-lobular parenchyma, beside reduced liver glycogen and fibrosis. These changes were not observed in the HFD + Mel group. In conclusion, melatonin combined with a high-fat diet preserves the liver architecture and function in the offspring.

This commentary highlights a "cerebellar classic" by Heinrich Obersteiner (1847-1922), the founder of Vienna's Neurological Institute. Obersteiner had a long-standing interest in the cerebellar cortex, its development, and pathology, having provided one of the early accurate descriptions of the external germinal layer (sometimes called the "marginal zone of Obersteiner" or "Obersteiner layer"). In his communication before the 81st meeting of the Society of German Natural Scientists and Physicians in Salzburg in September 1909, Obersteiner placed special emphasis on the histophysiology of the granule cell layer of the cerebellum and covered most of the fundamental elements of the cerebellar circuitry, on the basis of Ramón y Cajal's neuronism. Those elements are discussed in a historic and a modern perspective, including some recent ideas about the role of granule cells, beyond the mere relay of sensorimotor information from mossy fibers to the Purkinje cells, in learning and cognition.

Essential oils are a promising alternative to insecticides. We investigated the LD50 of oils extracted from Piper corcovadensis, P. marginatum, and P. arboreum after 48 h topical contact with Spodoptera frugiperda larvae using morphometry, histochemistry and immunohistochemistry of the midgut and fat body. Chromatography revealed that E-caryophyllene was the principal compound common to the Piper species. The essential oils of P. corcovadensis, P. marginatum and P. arboreum caused deleterious changes in the midgut of S. frugiperda larvae. P. corcovadensis oil produced the lowest LD50 and significant histopathological alterations including elongation of the columnar cells, formation of cytoplasmic protrusions, reduction in carbohydrate, increased apoptotic index and decreased cell proliferation. P. arboreum oil caused histopathological alterations similar to P. corcovadensis, but caused the highest rate of cell proliferation and increased regenerative cells, which indicated rapid regeneration of the epithelium. Our findings demonstrated the insecticidal potential of P. corcovadensis for control of S. frugiperda owing to the significant damage it inflicted on S. frugiperda midgut.

We investigated the cytoarchitecture and surface ultrastructural characteristics of different cells lining the epithelium of the saccus vasculosus of Notopterus chitala (Hamilton, 1822). The saccus vasculosus, the richly vascularized reddish, sac like organ is situated on the ventral aspect of the diencephalon just behind the pituitary gland. Histologically, the saccus vasculosus consisted of several loculi lined with a heterogeneous population of a large number of specialized coronet cells and a smaller number of alternately arranged supporting cells. The loculi were densely surrounded by blood sinusoids. The loculi opened into several collecting channels that united and ultimately drained into the third ventricle of the brain. The coronet cells possessed central or basal nuclei and an apical globular protrusion. Under scanning electron microscopy, the coronet cells were variable in shape with prominent nuclei. The supporting cells were triangular in shape and placed in between the coronet cells. The apical parts of the coronet cells had different shapes of globular protrusions with distinct stalks. Intense silver stain was noted in the terminal end of the coronet cells and nerves were attached with blood vessels. Under scanning electron microscopy observation, nerve fibers of different caliber were attached to the coronet cells and blood vessels. The various cells of the saccus vasculosus in N. chitala were correlated with their functional significance.

This commentary highlights a "cerebellar classic" by a pioneer of neurobiology, Christfried Jakob. Jakob discussed the connectivity between the cerebellum and mesencephalic, diencephalic, and telencephalic structures in an evolutionary, developmental, and histophysiological perspective. He proposed three evolutionary morphofunctional stages, the archicerebellar, paleocerebellar, and neocerebellar; he attributed the reduced cerebellospinal connections in humans, compared to other primates, to the perfection of the rubrolenticular and thalamocortical systems and the intense ascending pathways to the red nucleus in exchange for the more elementary descending efferent pathways. Jakob hypothesized the convergence of cerebellar pathways in associative cortical regions, insisting on the intimate collaboration of the cerebellum with the frontal lobe. The extensive lines of communication between regions throughout the association cortex substantiate Jakob's intuition and begin to outline the mechanisms for substantial cerebellar involvement in functions beyond the purely motor domain. Atop a foundation of anatomical and phylogenetic mastery, Jakob conceived ideas that were noteworthy, timely, and have much relevance to our current thinking on cerebellar structure and function.

This paper described the architecture of the dog aortic arterial wall focused in the ascendant; thoracic descendent, and abdominal parts of the aorta. Furthermore, in this study we analyzed the wall structure of the right external iliac artery and the left vertebral artery of dog aiming to compare their architectural patterns with the aortic wall mainly in abdominal aortic part, which was structured by a mixed muscular and connective matrix equilibrated elements. The variables analyzed were the thickness of each mural layer; the number of elastic lamellae, mainly localized into the medial layer structure, and the diameters of each aortic segment. The aortic structure showed a typical elastic pattern in the ascendant and descendant thoracic parts, but in its abdominal segment a lesser presence of elastic lamellae permitted to classify it with a mixed mural composition. The aortic segments mainly presented variable amounts of smooth muscle cells and connective matrix assuming a meshwork pattern. Furthermore, smooth muscle cells were seen intermingled with connective lamellae and fibers forming the complex meshwork related, which certainly helped to guarantee the aortic mural integrity during the aortic blood flow.


El objetivo de este trabajo fue describir la estructura histológica de la aorta del perro adulto, a través del análisis morfométrico. Fueron estudiados los tres segmentos de la aorta, ascendente y descendente torácicos, y el segmento abdominal inferior a los riñones. Esta última porción presentó un patrón estructural mixto, con tejido muscular liso y conectivo, con equilibrio entre las células musculares lisas y el componente mural tipo estroma, con elementos fibrosos y laminillas elásticas. En las otras partes de la aorta, se observó el clásico patrón elástico en la estructura mural, con una mayor proporción de los componentes activos (musculares lisos) que los pasivos, o sea, laminillas y fibras conectivas. El número de las laminillas elásticas fue decreciente desde la aorta ascendente en dirección a la parte abdominal de la aorta. Los diámetros de los tres segmentos tiñeron de manera similar. La comparación entre la estructura mural de la aorta y la histología de la arteria iliaca interna derecha y arteria subclavia izquierda, ambas musculares, fueron analizadas de manera objetiva, encontrando diferencias en el patrón mural aórtico, principalmente en el segmento aórtico terminal.

Over time, much knowledge has been accumulated about the active role of the urothelium, principally in rodents and human. Far from being a mere passive barrier, this specialized epithelium can alter the ion and protein composition of the urine, is able to sense and respond to mechanical stimuli such as pressure, and react to mechanical stimuli by epithelial cell communication with the nervous system. Most of the specialized functions of the urothelium are linked to a number of morpho-physiologic properties exhibited by the superficial umbrella cells, including specialized membrane lipids, asymmetric unit membrane particles and a plasmalemma with stiff plaques which function as a barrier to most substances found in urine, thus protecting the underlying tissues. Moreover, the entire mucosa lining the low urinary tract, composed of urothelium and sub-urothelium, forms a functional transduction unit, able to respond to eso- and endogenous physical and chemical stimuli in a manner assuring an adequate functional response. This review will summarize the available information on each area of inquiry from a morpho-functional point of view. Possible considerations pertaining to species of veterinary interest are reviewed as well. The review was prepared consulting the electronic databases PubMed and Cab Abstracts and retrieving all pertinent reports and the relative reference lists, in order to identify any potential additional studies that could be included. Full-length research articles and thematic reviews were considered. Information on the urothelium of some domestic animal species was also included.

RESUMO

As queimaduras são lesões desencadeadas por agentes físicos, químicos, elétricos e térmicos que resultam em níveis variados de perda tecidual. O grau com que estas lesões danificam a pele depende de muitas variáveis, incluindo a duração e intensidade de contato com o agente agressor, a espessura da pele da região anatômica acometida, tamanho da área exposta, vascularização local e idade. Ainda, a perda tecidual é um dos fatores prognósticos destas lesões fornecendo clinicamente a base para a classificação destas lesões, o que faz com que a compreensão da histomorfologia da pele seja fundamental para o entendimento da fisiopatologia das queimaduras. O presente trabalho aborda os mecanismos fisiopatológicos envolvidos nas queimaduras e os vários critérios de classificação destas lesões, buscando fornecer subsídios teóricos para tal compreensão através de uma revisão de literatura abordando a histofisiologia da pele.


Burns are injuries triggered by physical, chemical, electrical and thermal result in varying levels of tissue loss. The degree to which these lesions damage the skin depends on many variables, including duration and intensity of contact with the offending agent, the skin thickness of the anatomical region affected, the size of the exposed area, local vascularization and age. In addition, tissue loss is one of the prognostic factors of these lesions clinically providing the basis for the classification of these lesions and the understanding of the morphology of skin is essential to understanding the pathophysiology of burns. The present paper discusses the pathophysiological mechanisms involved in burns and various criteria for classifying these lesions and to provide theoretical understanding for such a thorough review of literature dealing with histophysiology skin.

The surface epithelium of the vas deferens of Agouti paca, a wild and large South American rodent, was basically formed by principal and basal cells being only the principal cells related to endocytosis processes and also secretion taking base on their cytoplasmic ultrastructural features. Principal cell of vas deferens epithelium were characterized mainly by presence of vesicles with several shapes, sizes and internalized content at their apical cytoplasm occurring smaller pits and pale small vesicles seen next to the apical brush border of microvillus. Moreover, coated vesicles, smooth surface vesicles and great vesicles; multivesicular bodies, endosomes and lysosomes were seen. Presence of an apocrine secretory apparatus was also viewed, showing apical cytoplasmic expansions protruding into the vas deferens luminal compartment. The basal flattened cells, without luminal surface contact, occurred next to the basement membrane of the ductus, and did no exhibit special ultrastructural features.


El epitelio que recubre la luz del conducto deferente de la laca (Agouti paca), un roedor silvestre de Sudamérica, está formado por células principales (P) y básales (B), en donde las células principales están asociadas a los procesos de endocitosis y secreción, teniendo una base en su característica ultraestructural citoplasmática. Las células principales de los vasos deferentes del epitelio se caracteriza principalmente por la presencia de vesículas con variadas formas, tamaños y contenido interior en su citoplasma apical produciendo pequeñas invaginaciones y pequeñas vesículas pálidas ubicadas continuas al borde en cepillo de las microveilosidades apicales. Además, fueron observados, vesículas de superficie lisa y vesículas de gran tamaño; órganos multivesiculares, endosomas y lisosomas. También se observó la presencia de un aparato secretor apocrino, con expansiones citoplasmáticas apicales que se protruyen en el compartimiento luminar del conducto deferente. Células básales aplanadas, sin contacto con la superficie luminal, se encuentran junto a la membrana basal del conducto, las que no presentan características ultraestructurales especiales.
