RESUMO

Objective To study the relationship between growth hormone,insulin-like growth factor-1 (IGF-1) and immunity during sepsis.Methods A total of 42 sepsis patients with complete data were selected to detect serum growth hormone,IGF-1,T cells in peripheral blood,and humoral immunity.Multifactor repeated measurement variance analysis was applied to compare various markers between sepsis group and the control group.Partial correlation analysis was used to analyze the relationship between growth hormone,IGF-1 and immunity.Results Serum growth hormone of patients in sepsis group was obviously higher than that in the control group (P =0.013 7,0.012 3,0.011 5),while IGF-1 was obviously lower in sepsis group compared with control group (P=0.014 4,0.013 5,0.013 2).Meanwhile,CD3+ (P =0.0095,0.0089,0.0092),CD4+ (P=0.0163,0.013,0.0158),CD8+ (P=0.010 1,0.019 3,0.015 5),NK (P=0.023 4,0.029 3,0.025 5) and CD19+ cell (P=0.011 3,0.010 2,0.010 0),IgA (P =0.019 3,0.017 8,0.015 7),IgG (P =0.047,0.006 9,0.005 9),and complement C3 (P =0.039 3,0.037 8,0.045 6) in peripheral blood of patients in sepsis group were obviously lower than those in control group.There were positive correlations between IGF-1 and CD3+,CD4+,CD8+,CD19+,NK cells,IgA,IgG,complement C3 (P=0.01 3,0.036,0.015,0.008,0.002,0.005,0.009,0.001) in sepsis patients.Conclusions There were abnormalities in serum growth hormone,IGF-1,cell-mediated immunity and humoral immunity found in sepsis patients.IGF-1 had the correlation with immune function affecting the prognosis of patients with sepsis.

RESUMO

Objective To investigate the synergism and attenuation effects of Pini Pollen on cyclophosphamide chemotherapy in model mice.Methods The models of mouse were induced with S180 tumor and cyclophosphamide.The tumor inhibiting rates, hematology, Liver function, giant cell phagocytosis, hemolysin level and indexes of thymus and spleen were determined to evaluate the synergism and attenuation effects of Pini Pollen(150,300,600 mg/kg) on cyclophosphamide chemotherapy.Results Compared with cyclophosphamide chemotherapy group,the tumor inhibiting rates,white blood cell number,giant cell phagocytosis, hemolysin level and indexes of thymus and spleen were increased, at the same time liver function damage were alleviated significantly in cyclophosphamide plus Pini Pollen groups.Conclusions Pini Pollen has a remarkable synergism and attenuation effects on cyclophosphamide chemotherapy through its immune adjustment functions.

OBJECTIVE: To study the therapeutic mechanisms of pseudolaric acid on allergic contact dermatitis in mice. METHODS: A total of 50 BALB/C mice were selected and randomly divided into control group, model group, and treatment A, B, C groups with 10 rats in each group. ACD model was established in model group, and treatment A, B, C groups but not in control group. Model group received no treatment, but treatment A, B, C groups were treated with external application of the concentration of 0.1%, 0.2% and 0.4% of the pseudolaric acid for the lesions of ear skin. And the weight gain and the swelling degree of the mice' ear were recorded, weight of thymus and spleen were measured. Spleen suspension was prepared to test T lymphocyte and B lymphocyte levels of mice in five groups. Changes in serum IFN-γ, IL-4 and IL-10 levels were tested through the enzyme linked immunosorbent assay (ELISA). RESULTS: The weight gain of mice in model group were significant lower than those of mice in the control group and the treatment A, B, C groups (P < 0.05). Weight gain of mice in treatment A, B groups were significant lower than that of control group (P < 0.05), but the difference in weight gain between treatment C group and control group showed no significant difference (P > 0.05). The swelling degree and the weight of mice ears in model group were significant higher than those of mice in control group and treatment A, B, C groups (P < 0.05). Swelling degree and the weight of mice ears of treatment A, B, C groups were obviously higher than that of control group (P < 0.05). The swelling degree and weight of mice' ears in treatment A, B, C groups were decreased with the increase of the drug dosage, but comparison between A, B and C group showed statistically differences (P < 0.05). The thymus and spleen index of mice in model group were significant higher than those of the other four groups (P < 0.05), among the four groups, thymus and spleen index of treatment A and B group were higher than control group and treatment C group (P < 0.05). The stimulation index of T and B cells of mice in model group was significantly higher than the rest four groups (P < 0.05). The serum IFN-γ level of mice in control group and treatment A, B and C group was obviously lower than that of mice in model group (P < 0.05). The serum IFN-γ level of mice in treatment A, B and C group were decreased with the increasing of the drug dosage, and the level of C group was obviously lower than that of A and B group (P < 0.05). CONCLUSION: The pseudolaric acid has anti-inflammation and immune adjustment the effects showing a remarkable therapeutic effects for the ACD mice.

RESUMO

The aim of this study was to investigate whether resolvin E1 (RvE1) protects against hepatic fibrosis in a murine model of liver fibrosis induced by Schistosoma japonicum infection. A total of 30 pathogen-free Kunming mice were randomly and equally divided into three groups: Control (uninfected, untreated), model (infected, untreated) and RvE1 intervention (infected, RvE1-treated; 100 ng daily). The mice were infected with Schistosoma japonicum by inoculating the abdominal skin with 20±2 cercariae to induce models of liver fibrosis. The area and numbers of the granulomas in the livers were assessed through histopathology after 70 days of treatment. The levels of tumor necrosis factor (TNF)-α and interferon (IFN)-γ were evaluated in the serum by enzyme-linked immunosorbent assay (ELISA). The expression levels of TNF-α were detected in the hepatic tissue by reverse transcription-polymerase chain reaction and western blot analysis. The activity levels of alanine aminotransferase and aspartate aminotransferase were determined in the serum by ELISA. The expression levels of laminin (LN), hyaluronic acid (HA), procollagen type III (PC-III) and type IV collagen (IV-C) were detected in the serum by radioimmunoassays. The results revealed that the mean area of the granulomas was smaller in the RvE1 intervention group compared with that in the model group. Following RvE1 treatment, the serum levels of TNF-α were lower than those in the model group, while the serum levels of IFN-γ were higher compared with those in the model group. The expression levels of TNF-α were lower in the hepatic tissue following RvE1 treatment compared with those in the model group. The indicators of liver fibrosis, the levels of LN, HA, PC-III and IV-C in the serum, were lower following RvE1 treatment than those in the model group. In conclusion, RvE1 treatment may reduce the growth of granulomas, thereby slowing the process of hepatic fibrosis, and this effect may be the result of anti-inflammatory and immune system adjustment.

RESUMO

ObjectiveTo evaluate the clinical efficacy and safety of astragalus injection on the immune function in patients with senile sepsis.Methods Sixty patients with old age sepsis in Critical Care Medicine Department of Guangdong Provincial Traditional Chinese Medicine Hospital were enrolled and randomly assigned into control and treatment groups according to the table of random numbers, 30 cases in each group. According to 2012 sepsis guidelines for treatment, including antibacterial drug, mechanical ventilation, visceral function support, etc., the therapy was given to the control group; besides the treatment in the control group, intravenous drip of 60 mL astragalus injection(10 mL per ampoule) in 250 mL 0.9% normal saline was additionally given in the treatment group, once a day for 7 days. Before and after treatment, the immunological indexes, acute physiology and chronic health evaluationⅡ(APACHEⅡ) score, sequential organ failure assessment(SOFA) score, duration of mechanical ventilation and time of stay in intensive care unit(ICU), 28-day mortality and adverse drug reactions were compared between the two groups.Results Before treatment, there were no statistically significant differences in CD3+, CD3+CD4+, CD3+CD8+ and T helper cells /T suppressor cells(Th/Ts)levels between the two groups(allP>0.05), while CD3-NK+ of the control group was significantly higher than that in the treatment group〔(10.47±6.22)% vs. (6.26±4.13)%,P0.05). After treatment in the treatment group, IgG was increased compared with that in the control group〔IgG(g/L): 13.07±5.43 vs. 10.10±3.96,P 0.05〕. The differences in IgA, IgM, complement(C3,C4) and total serum complement activity(CH50) in the comparisons between the two groups had no statistical significance after treatment(allP>0.05). The differences in APACHEⅡ score(13.83±6.18 vs. 15.90±7.48), SOFA score(7.38±4.66 vs. 6.89±4.19), time of stay in ICU(day: 11.63±5.13 vs. 13.62±8.08), invasive ventilation time(hour: 155.44±119.68 vs. 224.08±174.15) and noninvasive ventilation time(hour: 55.55±42.24 vs. 98.57±43.17) had no statistical significance in comparisons between the treatment group and control group after treatment(allP>0.05). The difference in 28-day mortality had no statistical significance in comparison between the treatment group and control group〔16.7%(5/30) vs. 20.0%(6/30),P>0.05〕. In 60 cases, there were 2 patients with adverse drug reaction, one diarrhea and another little rashes, the rest of the patients did not appear any drug side effect.ConclusionAstragalus injection combined with conventional western medicine therapy possibly has certain effect on adjustment of disturbance of immunologic functions in old patients with sepsis, and its therapeutic safety is well.
