RESUMO

This work describes the development of a reusable 2D detector based on radiochromic reaction for radiotherapy dosimetric measurements. It consists of a radiochromic gel dosimeter in a cuboidal plastic container, scanning with a flatbed scanner, and data processing using a dedicated software package. This tool is assessed using the example of the application of the coincidence test of radiation and mechanical isocenters for a medical accelerator. The following were examined: scanning repeatability and image homogeneity, the impact of image processing on data processing in coincidence tests, and irradiation conditions-monitor units per radiation beam and irradiation field are selected. Optimal conditions for carrying out the test are chosen: (i) the multi-leaf collimator gap should preferably be 5 mm for 2D star shot irradiation, (ii) it is recommended to apply ≥2500-≤5000 MU per beam to obtain a strong signal enabling easy data processing, (iii) Mean filter can be applied to the images to improve calculations. An approach to dosimeter reuse with the goal of reducing costs is presented; the number of reuses is related to the MUs per beam, which, in this study, is about 5-57 for 30,000-2500 MU per beam (four fields). The proposed reusable system was successfully applied to the coincidence tests, confirming its suitability as a new potential quality assurance tool in radiotherapy.

Primary immunodeficiency (PID) Disorders include a variable group of diseases that are classified according to the functional defects encountered. Chronic granulomatous disease (CGD) is inherited as an X-linked recessive disorder in many cases, and it is the clinical model of disorders of phagocytosis. Skin and solid organs abscesses are the most common presenting symptoms; we will report the case of a four-day-old boy admitted to our hospital for a neck mass with purulent discharges associated with umbilical stump and circumcision site infection; the diagnosis of CGD was later confirmed by the Dihydrorhodamine (DHR) test that turned out to be positive.

Plants generate reactive oxygen species (ROS) during different metabolic processes, which play an essential role in coordinating growth and response. ROS levels are sensitive to environmental stresses and are often used as a marker for stress in plants. While various methods can detect ROS changes, histochemical staining with nitroblue tetrazolium (NBT) and 3,3'-diaminobenzidine (DAB) is a popular method, though it has faced criticism. This staining method is advantageous as it enables both the quantification and localization of ROS and the identification of the enzymatic origin of ROS in plants, cellular compartments, or gels. In this protocol, we describe the use of NBT and DAP staining to detect ROS generation under different stresses such as nitrogen starvation, wounding, or UV-C. Additionally, we describe the use of NBT staining for detecting enzymatic generation of ROS in native and native SDS PAGE gels. Our protocol also outlines the separation and comparison of the origin of ROS generated by xanthine dehydrogenase1 (XDH1) using different substrates.

(1-x)(Na0.5Bi0.5)TiO3-xCaTiO3 Lead-free piezoelectric systems, positioned near the morphotropic phase boundary, were synthesized for varying compositions (x = 0.0, 0.05, 0.10, 0.15, and 0.20) using the solid-state reaction route. This study delves into the comprehensive investigation of the compositional effects on phase, structure, and electrical characteristics. Specifically, a morphotropic phase boundary (MPB) involving rhombohedral (R3c) and orthorhombic (Pnma) structures was seen in a (1-x)NBT-xCT crystal structure close to the composition of x = 0.10. Information on the pure phase formation and grain size of the intended composite system has been obtained using Rietveld refinement of the X-ray diffraction (XRD) diagram as well as scanning electron microscopy (SEM). The impact of the CT phase on the NBT lattice was investigated through an analysis of the charge density distribution. Using Williamson-Hall plots from XRD data, the average particle diameter was estimated to be between 131.87 nm and 136.54 nm. The relative permittivity increases with the addition of Ca2+, according to dielectric measurements. All ceramics exhibit a diffuse phase transition near (Tm) with a diffusivity range of 1.5-1.8, and a downward shift in depolarization temperature (Td). At the morphotropic phase boundary (MPB), excellent dielectric properties were observed at x = 0.10, which are attributed to the presence of both rhombohedral and orthorhombic structures as well as an appropriate particle size. The conduction process at different temperatures is thermally activated, as determined by the frequency-dependent ac conductivity.

This study aimed to investigate the role of neutrophils in canine leishmaniosis by assessing neutrophil activation and its relationship with different states of L. infantum infection and antibody and IFN-γ production. Dogs were categorized into five groups: healthy-seronegative (n = 25), healthy-seropositive (n = 21), LeishVet-stage I (n = 25), Leishvet-stage II (n = 41), and LeishVet-stage III-IV (n = 16). Results of the nitroblue tetrazolium reduction test (NBT) showed significantly higher neutrophil activation in stage I (median:17.17, range: [7.33-31.50]%) compared to in healthy-seronegative (4.10 [1.20-18.00]%), healthy-seropositive (7.65 [3.98-21.74]%), stage II (6.50 [1.50-28.70]%), and stage III-IV (7.50 [3.00-16.75]%) groups (p < 0.0001). Healthy-seropositive dogs also displayed higher values than all groups except stage I. Stages II and III-IV did not show significant differences compared to healthy-seronegative. Regarding IFN-γ, stage I dogs had higher concentrations (median:127.90, range: [0-3998.00] pg/mL) than healthy-seronegative (0 [0-109.50] pg/mL) (p = 0.0002), stage II (9.00 [0-5086.00] pg/mL) (p = 0.045), and stage III-IV (3.50 [80.00-548.80] pg/mL) (p = 0.02) dogs. Stage II dogs showed increased IFN-γ compared to healthy-seronegative dogs (p = 0.015), while stage III-IV dogs had no significant differences compared to healthy-seronegative dogs (p = 0.12). Healthy-seropositive dogs had elevated IFN-γ concentrations compared to healthy-seronegative dogs (p = 0.001) and dogs in stage III-IV (p = 0.03). In conclusion, neutrophil activation was higher in dogs with mild disease and healthy-seropositive dogs, and a relationship between neutrophil activation and the production of IFN-γ was found.

BACKGROUND: Distinguishing primary and secondary pulmonary neoplasms can be challenging via cytology, and a rapid, inexpensive diagnostic tool to differentiate these neoplasms is unavailable. Alkaline phosphatase cytochemistry (ALP-CC) has been used to identify primary pulmonary carcinomas in human patients, and we hypothesized it could be applied to canine lung aspirates. OBJECTIVE: We aimed to characterize ALP-CC expression in fine-needle aspirate (FNA) samples of canine pulmonary neoplastic and non-neoplastic tumors. METHODS: A retrospective case search was conducted to identify cases with contemporaneous cytology and histopathology reports from pulmonary lesions, including neoplastic and non-neoplastic etiologies. Slides prepared from pulmonary aspirates were stained for ALP-CC activity, and the percentage of ALP-CC-positive primary pulmonary epithelial tumors was determined. To characterize the ALP-CC expression in non-neoplastic cellular constituents of pulmonary FNA samples, mesothelial cells were also evaluated. RESULTS: Forty-eight canine cases met the inclusion criteria. ALP-CC-positive cells were seen in both neoplastic and non-neoplastic lesions. In non-neoplastic lesions, pulmonary epithelial cells were ALP-CC positive. Eighty-nine percent of primary pulmonary epithelial neoplasms were ALP-CC positive, and no ALP-CC positivity was noted in mesothelial cells. ALP-CC-positive neoplastic cells were seen in a metastatic amelanotic melanoma. CONCLUSIONS: Primary pulmonary epithelial neoplasms are frequently ALP-CC positive, but such positivity is not restricted to this tumor type. Non-neoplastic pulmonary epithelial cells can be ALP-CC positive, whereas mesothelial cells are negative.

INTRODUCTION: The nitro blue tetrazolium (NBT) reduction test (NBTT) has been used for measuring the metabolic activity of phagocytes of mammals. Activated neutrophils transform NBT into formazan in the cytoplasm. The NBTT can detect the activation of neutrophils in peripheral blood and is used to assess neutrophil function in dogs. However, the NBTT is not used frequently in the clinical setting, as samples should be processed after blood collection. OBJECTIVE: The aim of this study was to evaluate the effect of storage on NBTT in dog blood samples. MATERIALS AND METHODS: Residual EDTA blood samples from 22 dogs were included of different ages, breeds, and sex. The buffy coat layer was separated from the blood and incubated with 0.1% NBT. The NBTT was performed at 0, 24, 48, and 72 h after the collection of blood. Blood samples were stored at 4°C until the tests were performed. Blood smears were evaluated by light microscopy, and the NBT reduction rate was reported, which represents the percentage of activated neutrophils. The NBT reduction rate was calculated after counting 300 neutrophils in each slide. RESULTS: The means of NBTT in dog blood samples at 0, 24, 48, and 72 h were 8.3%, 8.5%, 8.7%, and 7.8%, respectively. No significant differences were observed between time points. CONCLUSIONS: This study showed that the NBTT can be performed up to 72 h after the collection of canine blood if correctly refrigerated at 4°C. This finding supports the performance of the NBTT in the clinical setting.

Whole-mount in situ hybridization (WISH) is an extremely useful technique for visualizing specific mRNA targets and solving many biological questions. In planarians, this method is really valuable, for example, for determining gene expression profiles during whole-body regeneration and analyzing the effects of silencing any gene to determine their functions. In this chapter, we present in detail the WISH protocol routinely used in our lab, using a digoxigenin-labelled RNA probe and developing with NBT-BCIP. This protocol is basically that already described in Currie et al. (EvoDevo 7:7, 2016), which put together several modifications developed from several laboratories in recent years that improved the original protocol developed in the laboratory of Kiyokazu Agata in 1997. Although this protocol, or slight modifications of it, is the most common protocol in the planarian field for NBT-BCIP WISH, our results show that key steps such as the use and time of NAC treatment to remove the mucus need to be taken into account depending on the nature of the gene analyzed, especially for the epidermal markers.

Currently, metal carbonitride (MXene) has been identified as a hot research topic in the research area of surface-enhanced Raman scattering (SERS). In this study, Ti3C2Tx/Ag composite was fabricated as SERS substrate with different Ag contents. The fabricated Ti3C2Tx/Ag composites show good SERS behavior by detecting 4-Nitrobenzenethiol (4-NBT) probe molecules. Through calculation, the SERS enhancement factor (EF) of the Ti3C2Tx/Ag substrate was as high as 4.15 × 106. It is worth noting that the detection limit of 4-NBT probe molecules can be achieved ultralow concentration of 10-11 M. In this system, electromagnetic enhancement mechanism and chemical enhancement mechanism have synergistic effects on SERS phenomenon. Meanwhile, the Ti3C2Tx/Ag composite substrate exhibited good SERS reproducibility. In addition, the SERS detection signal hardly changed after 6 months of natural standing, and the substrate showed good stability. This work suggests that the Ti3C2Tx/Ag substrate could be used as a sensitivity SERS sensor for practical application, and could be applied in the field of environmental monitoring.

Solid solutions (1-x-y)(Na0.5Bi0.5)TiO3-xBaTiO3-y(K0.5 Bi0.5)TiO3 with (x (mol.%) = 0, 7 and 100); y(mol.%) = 0, 20 and 100) compositions have been prepared by a conventional solid-state reaction method, and their structure, dielectric properties and depolarization temperature have been examined. At room temperature, X-ray diffraction (XRD) patterns reveal that the crystalline structure of the ceramics was perovskite. The morphotropic phase boundary (MPB) of the ternary system lying between rhombohedral (R3c) and tetragonal (P4mm) phases is in the range of (x (mol.%) = 7 and y (mol.%) = 20). The Raman-active modes for 0.73NBT-0.07BT-0.20KBT were separated and identified under the framework of group theory. SEM micrographs illustrate the quasi-uniform distribution of the grains, which are compact. The dielectric properties of the ceramics were studied in the frequency range of 1 kHz-100 kHz from ambient temperature to 600 °C. Dielectric measurements indicate that all ceramics show a diffuse phase transition near the temperature (Tm) for diffusivity of the order of 1.4-1.7 and a shift of (Tm) towards high temperatures. The resistance and capacitance of the various contributors (grain and grain boundary) in our samples are also discussed using a brick-layer model. Excellent piezoelectric properties for d33 = 146 pCN-1 and electromechanical coupling factors kp = 29.4% were observed at morphotropic phase boundary (MPB), which was assumed to be associated with the coexistence of rhombohedral and tetragonal phases and accurate grain size. This work establishes a new approach for improving lead-free piezoelectric ceramics based on 0.73NBT-0.07BT-0.20KBT.

The present study is designed to monitor the spatio-temporal changes in forest cover using Remote Sensing (RS) and Geographic Information system (GIS) techniques from 1990 to 2017. Landsat data from 1990 (Thematic mapper [TM]), 2000 and 2010 (Enhanced Thematic Mapper [ETM+]), and 2013 to 2017 (Operational Land Imager/Thermal Infrared Sensor [OLI/TIRS]) were classified into the classes termed snow, water, barren land, built-up area, forest, and vegetation. The method was built using multitemporal Landsat images and the machine learning techniques Support Vector Machine (SVM), Naive Bayes Tree (NBT) and Kernel Logistic Regression (KLR). According to the results, forest area was decreased from 19,360 km2 (26.0%) to 18,784 km2 (25.2%) from 1990 to 2010, while forest area was increased from 18,640 km2 (25.0%) to 26,765 km2 (35.9%) area from 2013 to 2017 due to "One billion tree Project". According to our findings, SVM performed better than KLR and NBT on all three accuracy metrics (recall, precision, and accuracy) and the F1 score was >0.89. The study demonstrated that concurrent reforestation in barren land areas improved methods of sustaining the forest and RS and GIS into everyday forestry organization practices in Khyber Pakhtun Khwa (KPK), Pakistan. The study results were beneficial, especially at the decision-making level for the local or provincial government of KPK and for understanding the global scenario for regional planning.

RESUMO

Genetic modification of living organisms has been a prosperous activity for research and development of agricultural, industrial and biomedical applications. Three decades have passed since the first genetically modified products, obtained by transgenesis, become available to the market. The regulatory frameworks across the world have not been able to keep up to date with new technologies, monitoring and safety concerns. New genome editing techniques are opening new avenues to genetic modification development and uses, putting pressure on these frameworks. Here we discuss the implications of definitions of living/genetically modified organisms, the evolving genome editing tools to obtain them and how the regulatory frameworks around the world have taken these technologies into account, with a focus on agricultural crops. Finally, we expand this review beyond commercial crops to address living modified organism uses in food industry, biomedical applications and climate change-oriented solutions.

BACKGROUND: Glycation of immunoglobulin-G (IgG) molecules with monosaccharides may cause significant structural disability, thus resulting in their loss of function. The accumulation of AGEs formed from glycation plays an important role in the aliments associated with metabolic diseases. Therefore, excess sugar in plasma interferes with the functioning of IgG and may contribute to a wide range of diabetes-associated complications. The long-term formation of these heterogeneous AGEs may accumulate and affect plasma proteins, especially long-lived proteins. In this study, we analyze immunoglobulin-G (IgG) glycation with 2'-deoxyribose (deoxyribose) instigated modification in IgG structure and AGEs formation. METHODS: This study aims to glycate IgG from varying concentrations of pentose sugar, 2'-deoxyribose (deoxyribose). Various physicochemical methods and techniques characterized post glycation of IgG, both the native and its glycated analogue. The glycated protein will be assessed for its stability and perturbations by UV-VIS., fluorescence and FT-IR spectroscopic techniques. Moreover, the early glycation product will be done by NBT assay, and other biochemical parameters like HMF, carbonyl content and thioflavin-T assays were also performed to see the biochemical changes induced in the glycated IgG macromolecule. RESULTS: Glycation of protein macromolecules generates stable early glycation products (Amadori products). Later, these Amadori products involved a series of chemical reactions to form more stable advanced glycation end products (AGEs). Our experimental study results could validate the modification in IgG structure and AGEs formation. CONCLUSION: The formation of IgG-AGEs from glycation of IgG with deoxyribose could exert cellular toxicity, and might initiates secondary complications of diabetes. Therefore, this study emphasized the glycation reaction of IgG from deoxyribose, which has not been reported yet.

Na0.5Bi0.5TiO3 (NBT) and Fe- and Mn-modified NBT (0.5 and 1 mol%) ceramics were synthesized by the solid-state reaction method. The crystal structure, dielectric and thermal properties of these ceramics were measured in both unpoled and poled states. Neither the addition of iron/manganese to NBT nor poling changed the average crystal structure of the material; however, changes were observed in the short-range scale. The changes in shapes of the Bragg peaks and in their 2Θ-position and changes in the Raman spectra indicated a temperature-driven structural evolution similar to that in pure NBT. It was found that both substitutions led to a decrease in the depolarization temperature Td and an increase in the piezoelectric coefficient d33. In addition, applying an electric field reactivated and extended the ferroelectric state to higher temperatures (Td increased). These effects could be the result of: crystal structure disturbance; changes in the density of defects; the appearance of (FeTi'-), (Mn'Ti-V••O) and (Mn″Tii-V••O )-microdipoles; improved domain reorientation conditions and instability of the local polarization state due to the introduction of Fe and Mn into the NBT; reinforced polarization/domain ordering; and partial transformation of the rhombohedral regions into tetragonal ones by the electric field, which supports a long-range ferroelectric state. The possible occupancy of A- and/or B-sites by Fe and Mn ions is discussed based on ionic radius/valence/electronegativity principles. The doping of Fe/Mn and E-poling offers an effective way to modify the properties of NBT.

A novel therapeutic strategy for cancer treatment is to target altered tumor metabolism. Glucose- 6-phosphate dehydrogenase (G6PD) has been recently discovered to be implicated in apoptosis and angiogenesis, making it an excellent target in cancer treatment. The current study aimed to screen the plant extracts library to find potent hits against G6PD through enzymatic assay. Protein expression was induced by IPTG and purified using Ni-NTA columns after transformation of the pET-24a-HmG6PD plasmid into E. coli BL21-DE3 strain. An enzymatic assay was established by using purified rG6PD protein, for the screening of G6PD inhibitors. Out of 46 plant extracts screened, the sixteen plant extracts have shown inhibitory activity against the G6PD enzyme. At doses from 1 to 4 µg/ml, this extract demonstrated concentration-dependent inhibition of G6PD with an IC50 value of I.397 µg/ml. Moreover, the anticancer activity evaluation against HepG2 cells determined Smilax china as a potent inhibitor of cancer cells (IC50 value of 16.017 µg/ml). The acute and subacute toxicities were not observed in mice with various concentrations (50, 100, 200 and 2000 mg/kg). Furthermore, to identify the compounds from Smilax china as G6PD inhibitors, a literature-based phytochemical investigation of Smilax china was conducted, and sixty compounds were docked against the NADP+ and G6P binding sites of G6PD. The results of this study showed that three compounds were Scirpusin A, Smilachinin and Daucosterol with MolDock Score of -156.832, -148.215, and -145.733 respectively, against NADP+ binding site of G6PD. Conclusively, Smilax china root extract could be a safer drug candidate for the treatment of hepatocellular carcinoma.

Microtubules are essential components of the cytoskeleton of all eukaryotic cells and consist of α- and ß-tubulin heterodimers. Several tissue-specific isotypes of α- and ß-tubulins, encoded by distinct genes, have been described in vertebrates. In the African clawed frog (Xenopus laevis), class II ß-tubulin (tubb2b) is expressed exclusively in neurons, and its promoter is used to establish different transgenic frog lines. However, a thorough investigation of the expression pattern of tubb2b has not been carried out yet. In this study, we describe the expression of tubb2b-dependent Katushka fluorescence in the forebrain of premetamorphic Xenopus laevis at cellular resolution. To determine the exact location of Katushka-positive neurons in the forebrain nuclei and to verify the extent of neuronal Katushka expression, we used a transgenic frog line and performed several additional antibody stainings. We found tubb2b-dependent fluorescence throughout the Xenopus forebrain, but not in all neurons. In the olfactory bulb, tubb2b-dependent fluorescence is present in axonal projections from the olfactory epithelium, cells in the mitral cell layer, and fibers of the extrabulbar system, but not in interneurons. We also detected tubb2b-dependent fluorescence in parts of the basal ganglia, the amygdaloid complex, the pallium, the optic nerve, the preoptic area, and the hypothalamus. In the diencephalon, tubb2b-dependent fluorescence occurred mainly in the prethalamus and thalamus. As in the olfactory system, not all neurons of these forebrain regions exhibited tubb2b-dependent fluorescence. Together, our results present a detailed overview of the distribution of tubb2b-dependent fluorescence in neurons of the forebrain of larval Xenopus laevis and clearly show that tubb2b-dependent fluorescence cannot be used as a pan-neuronal marker.

This work reports on radiochromic dosimeters for 1D UV light measurements. The dosimeter is composed of a 25% Pluronic F-127 that forms a physical gel matrix and nitro blue tetrazolium chloride (NBT) as a radiation-sensitive compound. This dosimeter was exposed to UVA, UVB and UVC radiation, and the radiochromic reactions were followed with reflectance spectrophotometry including changes in light reflectance and color coordinates in the CIELAB color system. The exposition of dosimeters to all UV radiation caused color changes from pale yellow to dark violet, and its intensity increased with increasing absorbed dose. The effects of NBT concentration and UV radiation type on the dose-response of the dosimeters were also examined. The results obtained reveal that the dosimeters are the least sensitive to irradiation with UVC and the most sensitive to irradiation with UVB (e.g., dosimeter with 2 g/dm3 of NBT was characterized by the following parameters: the threshold dose 0.1 J/cm2; the dose sensitivity -5.97 ± 0.69 cm2/J; the linear dose range 0.1-2.5 J/cm2; the dynamic dose range was equal to 0.1-3 J/cm2). The results obtained reveal that the NBT-Pluronic F-127 dosimeters can be potentially useful as 1D sensors for artificial UV radiation sources measurements.

Hemichordates are benthic marine invertebrates closely related to chordates. Several species, including Ptychodera flava in the phylum Hemichordates, can undergo whole body regeneration from a small fragment. P. flava is widely distributed in the warm Indo-Pacific region and is easily collected in the lower tidal zone of a shallow beach with a coral reef. Here, we describe the methods for animal collection and preparation of regenerating tissues. The prepared tissues can be used for various molecular and/or histological experiments. We also demonstrate how to examine gene expression patterns in the tissues using whole mount in situ hybridization.

BACKGROUND: The National Benchmark Test (NBT) that determines academic readiness is widely used by Faculties as an additional measure to select students for the study of medicine. Despite this, many students continue to experience academic challenges that culminate in delayed graduation and sometimes academic exclusion or discontinuation of studies. AIM: This study aimed to understand academic and non-academic variables linked with academic difficulties in the first three years of medical education. METHODS: The study sample consisted of six cohorts of medical students for the period 2011 to 2016 (n = 1392). Only the first three of the six-year medical programme were selected for analysis. Survival analysis and Cox Proportional Hazard (CPH) was used to identify academic and non-academic variables associated with academic difficulties. RESULTS: A total of 475 students (34%) experienced academic difficulty; 221 (16%) in the first year of study, 192 (14%) in the second year and 62 (5%) in the third year of study. The results show that Intermediate Upper, Lower and Basic levels for all NBT domains, living in university residence, rurality and male gender were risk factors for academic difficulty. CONCLUSION: In mitigating these factors, the NBT must inform the type of support programmes to augment the students' skills and promote academic success. Additionally, existing support programmes should be evaluated to ascertain if they reach students at risk and whether participating in these programmes yield positive academic outcomes.
