RESUMO

Iron chelating peptides have been widely utilized as iron supplements due to their excellent absorption capacity, However, the high cost and cumbersome manufacturing process of these peptides significantly limit their industrial application. In this study, fermentation was used for the first time to prepare iron chelating peptides. Bacillus altitudinis 3*1-3 was selected as the most suitable strain from 50 strains. The hydrolysates of fermented scallop skirts showed excellent iron-chelating capacity (9.39 mg/g). Aspartic acid, glutamic acid, and histidine are crucial for the binding of peptides to ferrous ions. The heptapeptide (FEDPEFE) forms six binding bonds with ferrous irons. Compared with ferrous sulfate, peptide-ferrous chelate showed more stability in salt solution and simulated gastrointestinal juice (p < 0.05). Furthermore, the fermentation method could save >50% of the cost compared with the enzymatic method. The results can provide a theoretical basis for the preparation of ferrous-chelated peptides using the fermentation method.

RESUMO

This unprecedented analysis of the factors that determine the performance of Brazil electric energy contradictory policies identifies the irregular path followed by one of the largest national oil companies in the world. As high convenience comparative case study, the increase and decrease of production and investment within the context of the Brazilian electric matrix in the last 20 years can shed light on Petrobras' disparities in relation to itself and in relation to external variables over time such as policies and investment choices, international oil prices and exploration cost.

RESUMO

Laboratory production of recombinant mammalian proteins, particularly antibodies, requires an expression pipeline assuring sufficient yield and correct folding with appropriate posttranslational modifications. Transient gene expression (TGE) in the suspension-adapted Chinese Hamster Ovary (CHO) cell lines has become the method of choice for this task. The antibodies can be secreted into the media, which facilitates subsequent purification, and can be glycosylated. However, in general, protein production in CHO cells is expensive and may provide variable outcomes, namely in laboratories without previous experience. While achievable yields may be influenced by the nucleotide sequence, there are other aspects of the process which offer space for optimization, like gene delivery method, cultivation process or expression plasmid design. Polyethylenimine (PEI)-mediated gene delivery is frequently employed as a low-cost alternative to liposome-based methods. In this work, we are proposing a TGE platform for universal medium-scale production of antibodies and other proteins in CHO cells, with a novel expression vector allowing fast and flexible cloning of new genes and secretion of translated proteins. The production cost has been further reduced using recyclable labware. Nine days after transfection, we routinely obtain milligrams of antibody Fabs or human lactoferrin in a 25 mL culture volume. Potential of the platform is established based on the production and crystallization of antibody Fabs and their complexes.

RESUMO

The development of economical, energy-saving, and efficient metal-organic framework (MOF)-based adsorbents for atmospheric water collection is highly imperative for the rapid advancement of renewable freshwater resource exploitation. Herein, a feasible one-step solvothermal formation strategy of bimetallic MOF (BMOF) is proposed and applied to construct a solar-triggered monolithic adsorbent for enhanced atmospheric water collection. Benefiting from the reorganization and adjustment of topology structure by Al atoms and Fe atoms, the resultant BMOF(3) consisting of Al-fumarate and MIL-88A has a higher specific surface area (1202.99 m2  g-1 ) and pore volume (0.51 cm3  g-1 ), thereby outperforming the parental MOFs and other potential MOFs in absorbing water. Expanding upon this finding, the solar-triggered monolithic adsorbent is further developed through a bottom-up assembly of polyaniline/chitosan layers and hybridized BMOF(3) skeletons on a glass fiber support. The resultant monolithic adsorbent exhibits superior sorption-desorption kinetics, leading to directional water transport and rapid solar-assisted vapor diffusion. As a proof-of-concept demonstration, an exquisite water harvester is constructed to emphasize a high water yield of 1.19 g g-1 per day of the designed monolithic adsorbent. Therefore, the design and validation of bimetallic MOF-derived solar-triggered adsorbent in this work are expected to provide a reference for the large-scale applications of MOF-based atmospheric water harvesting.

RESUMO

Appropriate costing and economic modeling are major factors for the successful scale-up of health interventions. Various cost functions are currently being used to estimate costs of health interventions at scale in low- and middle-income countries (LMICs) potentially resulting in disparate cost projections. The aim of this study is to gain understanding of current methods used and provide guidance to inform the use of cost functions that is fit for purpose. We reviewed seven databases covering the economic and global health literature to identify studies reporting a quantitative analysis of costs informing the projected scale-up of a health intervention in LMICs between 2003 and 2019. Of the 8725 articles identified, 40 met the inclusion criteria. We classified studies according to the type of cost functions applied-accounting or econometric-and described the intended use of cost projections. Based on these findings, we developed new mathematical notations and cost function frameworks for the analysis of healthcare costs at scale in LMICs setting. These notations estimate variable returns to scale in cost projection methods, which is currently ignored in most studies. The frameworks help to balance simplicity versus accuracy and increase the overall transparency in reporting of methods.

RESUMO

Double-stranded RNA (dsRNA) can trigger RNA interference (RNAi) and lead to directed silencing of specific genes. This natural defense mechanism and RNA-based products have been explored for their potential as a sustainable and ecofriendly alternative for pest control of species of agricultural importance and disease vectors. Yet, further research, development of new products and possible applications require a cost-efficient production of dsRNA. In vivo transcription of dsRNA in bacterial cells has been widely used as a versatile and inducible system for production of dsRNA combined with a purification step required to extract the dsRNA. Here, we optimized an acidic phenol-based protocol for extraction of bacterially produced dsRNA at low cost and good yield. In this protocol, bacterial cells are efficiently lysed, with no viable bacterial cells present in the downstream steps of the purification. Furthermore, we performed a comparative dsRNA quality and yield assessment of our optimized protocol and other protocols available in the literature and confirmed the cost-efficiency of our optimized protocol by comparing the cost of extraction and yields of each extraction method.

RESUMO

The demand for lactic acid and lactic acid-derived products in the food, pharmaceutical, and cosmetic industries is increasing year by year. In recent decades, the synthesis of lactic acid by microbials has gained much attention from scientists due to the superior optical purity of the product, its low production costs, and its higher production efficiency compared to chemical synthesis. Microbial fermentation involves the selection of feedstock, strains, and fermentation modes. Each step can potentially affect the yield and purity of the final product. Therefore, there are still many critical challenges in lactic acid production. The costs of feedstocks and energy; the inhibition of substrates and end-product; the sensitivity to the inhibitory compounds released during pretreatment; and the lower optical purity are the main obstacles hindering the fermentation of lactic acid. This review highlights the limitations and challenges of applying microbial fermentation in lactic acid production. In addition, corresponding solutions to these difficulties are summarized in order to provide some guidance for the industrial production of lactic acid.

RESUMO

PURPOSE: Primary Aldosteronism (PA) is increasingly considered as a common disease affecting up to 10% of the hypertensive population. Standard of care comprises laparoscopic total adrenalectomy but innovative treatment such as RadioFrequency Ablation (RFA) constitutes an emerging promising alternative to surgery. The main aim of this study is to analyse the cost of RFA versus surgery on aldosterone-producing adenoma patient from the French National Health Insurance (FNHI) perspective. METHODS: The ADERADHTA study was a prospective pilot study aiming to evaluate both safety and efficacy of the novel use of adrenal RFA on the patients with PA. This study conducted on two French sites and enrolled adult patients, between 2016 and 2018, presenting hypertension and underwent the RFA procedure. Direct medical (inpatient and outpatient) and non-medical (transportation, daily allowance) costs were calculated over a 6-month follow-up period. Moreover, the procedure costs for the RFA were calculated from the hospital perspective. Descriptive statistics were implemented. RESULTS: Analysis was done on 21 patients in RFA groups and 27 patients in the surgery group. The difference in hospital costs between the RFA and surgery groups was €3774 (RFA: €1923; Surgery: €5697 p < 0.001) in favour of RFA. Inpatient and outpatient costs over the 6-month follow-up period were estimated at €3,48 for patients who underwent RFA. The production cost of implementing the RFA procedure was estimated at €1539 from the hospital perspective. CONCLUSION: Our study was the first to show that RFA is 2 to 3 times less costly than surgery. The trial is registered at ClinicalTrials.gov under the number NCT02756754.

RESUMO

Increasing the productive lifespan of dairy cows is important to achieve a sustainable dairy industry, but making strategic culling decisions based on cow profitability is challenging for farmers. The objective of this study was to carry out a lifetime cost-benefit analysis based on production and health records and to explore different culling decisions among farmers. The cost-benefit analysis was conducted for 22 747 dairy cows across 114 herds in Quebec, Canada for which feed costs and the occurrence of diseases were reported. Costs and revenues related to productive lifespan were compared among cohorts of cows that left their respective herd at the end of their last completed lactation or stayed for a complete additional lactation. Hierarchical clustering analysis was carried out based on costs and revenues to explore different culling decisions among farmers. Our results showed that the knowledge of lifetime cumulative costs and revenues was of great importance to identify low-profitable cows at an earlier lactation, while only focusing on current lactation costs and revenues can lead to an erroneous assessment of profitability. While culling decisions were mostly based on current lactation costs and revenues and disregarded the occurrence of costly events on previous lactations, there was variation among farmers as we identified three different culling decision clusters. Monitoring cumulative costs and revenues would help farmers to identify low-profitable cows at an earlier lactation and make the decision to increase herd productive lifespan and farm profitability by keeping the most profitable cows.

RESUMO

The economic feasibility of replacing corn grain with by-product from biscuit manufacture (BBM) as an energy source in the diet for purebred and crossbred native lambs raised in feedlot was evaluated. Four production systems were developed based on nutritional management and genetic groups: (PS1) purebred native lambs fed a conventional diet based on corn and soybean meal; (PS2) purebred native lambs fed a diet with BBM replacing 100% of the corn; (PS3) crossbred native lambs fed a conventional diet based on corn and soybean meal; and (PS4) crossbred native lambs fed a diet with BBM replacing 100% of the corn in the diet. The extrapolation was performed for a module of 24 Morada Nova (MN) breed lambs (12 MN red (MNR) and 12 ½MNR × ½MN white lambs)) with an initial body weight of 18.8 ± 0.875 and 15.0 ± 0.838 kg for MNR and crossbred, respectively. Feed, animal acquisition, and labor present the total production costs. The greatest profits were obtained in the production systems that fed the crossbred lambs with BBM. The lesser cost of BBM contributed to greater total factor productivity regardless of genetic group (1.095 vs. 1.015 for BBM and conventional feed, respectively). Systems using BBM (SP2 and SP4) showed an estimated payback of 5.44 and 3.24 years, respectively, while the use of conventional feed contributed to negative economic data (PS1 and PS3) with payback period greater than 10 years. The use of BBM as an energy source in the diet of crossbred sheep was economically feasible and showed better economic indices when compared to the systems using a conventional diet.

RESUMO

The objective was to assess grazing as an element of profitability and competitiveness of a small-scale lamb fattening production system in central México and compare its economic performance by means of an analysis of scenarios. Two scenarios were analysed to assess the contribution of grazing on profitability and competitiveness. The first analysis was when grazing was the feed base, and secondly, costs of opportunity and economic impacts were studied under the assumption that sheep do not graze, and total feed has to be bought from external suppliers. The economic effect of grazing on the profitability was analysed by means of the Policy Analysis Matrix. Differences were found between strata; farmers with more than 70 sheep have the best profitability indices and the least vulnerability under the non-grazing scenario. Grazing had a positive effect reducing the cost of production and increasing competitiveness in the four strata assessed. However, farmers with higher technical level, specialised breeds and larger flocks (strata 3 and 4) have higher economic profits. The conclusion was that the profitability in fattening sheep is linked to taking advantage of grazing. However, to keep and improve the current financial performance, there is a need for the adoption of strategies for an integral improvement of the system and the adoption of better grazing management to further reduce production costs.

RESUMO

Telenomus remus (Nixon) is a dominant egg parasitoid of the destructive agricultural pest Spodoptera frugiperda (J. E. Smith), and so is used in augmentative biocontrol programs in several countries. An optimized mass-rearing system is essential to produce biological control products in a timely and cost-effective manner. In this study, the photoperiod, host egg:parasitoid ratio, and exposure time were evaluated to identify the optimal rearing conditions for T. remus on the alternative host Spodoptera litura (Fabricius) eggs. Results showed that increasing photoperiod above 12L:12D remarkably improved parasitoid progeny yield and life table parameters. Overlong photoperiods shortened female longevity, but within acceptable limits. There was a significant negative correlation between parasitism rate and host egg:parasitoid ratio under exposure times of 12 and 36 h, but not 24 h. Percentage of female progeny increased significantly along with increasing the host egg:parasitoid ratio. A significant negative relationship between the number of emerged adults per egg and the host egg:parasitoid ratio was observed at an exposure time of 36 h. It was concluded that T. remus may be mass-reared most efficiently on S. litura eggs using a photoperiod of more than 12L:12D, a 14-20:1 host egg:parasitoid ratio, and an exposure time of 24 h. These findings can be used to produce T. remus more efficiently and at lower costs.

RESUMO

This research evaluates the economic sustainability of rabbit farms using different housing systems-bicellular (BI), conventional dual-purpose (DP) and enriched cages designed according to the World Rabbit Science Association guidelines (WRSA)-through a field-based study involving six farms over the course of five years. The cages were compared based on three productivity indices expressed in kg of produced live weight/m2 and on eight cost indices expressed in EUR/kg of produced live weight. The results showed that WRSA significantly reduced the productivity index per walkable cage area in buildings and cages, thanks to the longer platform area included in the cage compared to the other systems. Concerning cost indexes, total variable costs were not different among housing systems, whereas significant differences were observed within costs items. As for the feed costs, DP underperforms compared to BI or WRSA (1.15 vs. 1.02 and 0.99 EUR/kg produced live weight); for drugs costs, BI was less competitive compared to DP and WRSA (0.12 vs. 0.06 and 0.05 EUR /kg). In conclusion, under the conditions of the present study, the economic results of farms that adopted housing systems designed to improve rabbit welfare, such as WRSA enriched systems, were economically sustainable and, comparable to conventional housing systems based on BI or DP cages, also provided a significant reduction in drug use in the tested farms. A comprehensive collection of data from more farms at a European level would be necessary to confirm these results on the economics of farms adopting alternative housing systems for rabbits.

RESUMO

The Arabian Peninsula's advantageous climate, availability of non-arable land, access to seawater and CO2-rich flue gas, make it an attractive location for microalgae biomass production. Despite these promising aspects, the region has seen very few studies into the commercial feasibility of algae-based value chains. This work aims to address this gap through a techno-economic feasibility study of algae biomass production costs, comparing different photobioreactor types, locations, and production scales. Flat panel and raceway pond cultivation systems were found to be the most economically attractive cultivation systems, with biomass production costs as low as 2.9 €·kg-1. Potential cost reductions of up to 42.5% and 25% could be accomplished with improvements in photosynthetic efficiencies and increased culture temperatures, respectively. As of such, efforts to source local thermo- and photo- tolerant strains could be the key to unlock the potential of the region for algae commercialization, linking into food, feed and nutraceutical industries.

RESUMO

Abstract Background: Factors such as herd composition, productivity, milk quality and technology level influence the costs and profitability of milk production. Objective: To evaluate indicators that could predict production performance and financial efficiency in three dairy production systems in southern Brazil. Methods: Monthly records of milk quality, production performance and costs from fifty dairy farms were collected. The farms were classified into grazing, semi-feedlot, or feedlot systems. Total revenues, effective operational cost, total operational cost, total cost, gross margin, net income, total cost leveling point, profitability and final milk price were calculated. Results: The feedlot system resulted in greater herd size and milk production per lactating cow (p<0.05), but greater variable costs compared to semi-feedlot and grazing systems. On the other hand, the grazing system achieved greater profitability per year. Factor II (fat and protein rates), and Factor III (herd size and productivity per area) were associated with milk price per liter paid to the farmer (p<0.05), together accounting for approximately 13% of this price. Conclusion: Dairy production systems are influenced by area, lactating cows, productive performance, productivity per area, milk quality, and use of artificial insemination as well as supplementation (concentrate and minerals). Nearly 13% of milk price can be attributed to its fat and protein content as well as herd size and productivity per area.

The objective of the present study was to evaluate the viability of a mycotoxin adsorbent based on beta-glucans from cell walls of Saccharomyces cerevisiae and bentonites in the diets of confined lambs of the Texel breed. Twenty-four lambs (12 males and 12 females) with an average weight of 18.6 kg ± 1.6 were divided into two groups: treated (with adsorbent) and control. The animal diets contained the mycotoxins aflatoxin B1, fumonisin B1 and B2, zearalenone, and deoxynivalenol in concentrations within guidance levels. The animals were slaughtered with body weight of approximately 26.4 kg. The performance and carcass variables of the lambs were evaluated. The daily weight gain was higher in the treated group (216.24 g) than in the control group (185.90 g). The averages for loin eye area, subcutaneous fat thickness, and marbling were 13.93 cm2, 2.66 mm, and 3.20 in the treated group and 12.16 cm2, 2.6 mm, and 3.25 in the control group, respectively. The true, hot, and cold carcass yield, and the carcass cooling losses did not differ between groups. The carcasses were similar in conformation and fat finishing degree, with averages of 3.95 and 3.83, respectively. To the best of our knowledge, this is the first study to evaluate the effect of a mycotoxin additive in the diet of finishing lambs. The use of mycotoxin adsorbent in confined lambs was feasible, and its use resulted in greater daily weight gain in lambs and average net profit.

RESUMO

Recombinant proteins play an important role in many areas of our lives. For example, recombinant enzymes are used in the food and chemical industries and as high-quality proteins for research, diagnostic and therapeutic applications. The production of recombinant proteins is still dominated by expression systems based on microbes and mammalian cells, although the manufacturing of recombinant proteins in plants - known as molecular farming - has been promoted as an alternative, cost-efficient strategy for three decades. Several molecular farming products have reached the market, but the number of success stories has been limited by industrial inertia driven by perceptions of low productivity, the high cost of downstream processing, and regulatory hurdles that create barriers to translation. Here, we discuss the technical and economic factors required for the successful commercialization of molecular farming, and consider potential future directions to enable the broader application of production platforms based on plants.

RESUMO

In the olive production sector, which is increasingly expanding beyond the borders of the Mediterranean basin, harvesting is the most demanding phase, from both an economic and organisational point of view. Traditional olive orchards are still predominant, with centuries-old and large plants, and are characterised by the gradual ripening of drupes and irregular planting patterns. Even though the structural conversion of these olive orchards into more modern cultivations may be difficult owing to their historical, monumental, and landscaping importance, as well as the existing legal restrictions, supporting a "modernisation" process aimed at mechanising the main farming operations remains a priority. Technological innovation is, therefore, a primary objective for Mediterranean olive growing, as well as for the enhancement of its strengths. The present study aimed at assessing different olive harvesting sites, considering the technical, economic, and environmental aspects, to develop a better version of the "olive harvesting database". The applied methodology, also called the "modular approach", represents a useful tool to apply in unitary process assessment to obtain a comprehensive database of diverse agricultural operations. Eight olive harvesting systems were compared: six highly mechanised scenarios, one based on mechanical-aided harvesting, and the final one based on fully manual harvesting. The mechanised systems obtained a better performance in terms of working capacity, as only 3.5 h ha-1 were needed to harvest 12 t using a self-propelled trunk shaker. In addition, the economic results revealed that mechanical harvesting, diversely from manual or aided harvesting, is the only way to decrease production costs. From an environmental point of view, manual and mechanical-aided harvesting showed the best performance in terms of impact per hour. However, using the mass-based unit (1 kg of harvested olives), the results were the opposite and this could be very relevant for the ecoprofile of olive oil.

ABSTRACT In order to reduce the feeding costs in cuniculture, the inclusion of wilted and silaged elder was evaluated on the productive and economic performance of rabbits in the fatting phase. 27 rabbits (crosses between Chinchilla, Californian and New Zealand) were selected with an average initial weight of 1467,6 ± 271,1g. They were divided and randomly distributed in each of three treatments, control (100% balanced feed), silage (85% balanced feed + 15% elder silage) and wilted (88% balanced feed + 12% wilted elder). During 28 days, weight gain, feed conversion ratio (FCR), carcass yield and viscerosomatic index were evaluated and analyzed through a variance analysis in a complete random design. Gross margin of partial income (GMPI) was calculated through a partial budget analysis for the sale of the product in vivo and carcass. The group that received the silage showed greater weight gain (1040.4 ± 75.2g) and higher intake (12.5 ± 0.3kg MS / cage), than control group (P˂0,05), but FCR was not different between these groups (P˂0.05). No statistical differences were observed in the remaining productive parameters (P>0,05). Silage diet presented the highest cost, but at the same time it produced the highest GMPI, for sale in vivo (13%) and carcass (9%). Silage elder can be considered as a feeding alternative, improving the productive parameters and the GMPI.

ABSTRACT The potato is affected by the attack of Tecia solanivora that causes losses of up to 80%. Genetically modified varieties and biocontrol agents, can be used for its control. This study aimed to determine the socioeconomic potential of small potato producers in the Cundiboyacense region for the adoption of these biotechnological strategies, through face-to-face surveys and the use of methodologies such as partial budgets and logistic regression model. The results reveal that the type of seed defines adoption, there being an interest in technologies that allow pest control, so they would be willing to pay up to 30% more of the current value for this. The methodology of partial budgets showed a positive economic effect in the different scenarios proposed. It was concluded that small potato producers in the municipalities analyzed have a high socioeconomic potential for the adoption of the biotechnology package.
