RESUMO

Branch number is one of the most important agronomic traits of fruit trees such as peach. Little is known about how LncRNA and/or miRNA modules regulate branching through transcription factors. Here, we used molecular and genetic tools to clarify the molecular mechanisms underlying brassinosteroid (BR) altering plant branching. We found that the number of sylleptic branch and BR content in pillar peach ('Zhaoshouhong') was lower than those of standard type ('Okubo'), and exogenous BR application could significantly promote branching. PpTCP4 expressed great differentially comparing 'Zhaoshouhong' with 'Okubo'. PpTCP4 could directly bind to DWARF2 (PpD2) and inhibited its expression. PpD2 was the only one differentially expressed key gene in the path of BR biosynthesis. At the same time, PpTCP4 was identified as a target of miR6288b-3p. LncRNA1 could act as the endogenous target mimic of miR6288b-3p and repress expression of miR6288b-3p. Three deletions and five SNP sites of lncRNA1 promoter were found in 'Zhaoshouhong', which was an important cause of different mRNA level of PpTCP4 and BR content. Moreover, overexpressed PpTCP4 significantly inhibited branching. A novel mechanism in which the lncRNA1-miR6288b-3p-PpTCP4-PpD2 module regulates peach branching number was proposed.

Fresh-cut processing is a good strategy to enhance the commercialization of peaches and nectarines, which easily deteriorate during low-temperature storage mostly due to the occurrence of chilling injury. Although several studies have been performed to improve the shelf-life of fresh-cut stone fruit, the achievement of high-quality fresh-cut peaches and nectarines still constitutes a challenge. The present study aimed to gain insights into the evolution of the postharvest quality of fresh-cut nectarines (Prunus persica L. Batsch) Big Bang, cold-stored at two different storage temperatures (4 and 8 °C) for up to 10 days. Several aspects influencing the quality traits (sensory and postharvest quality parameters; the profile of phenolic and volatile organic compounds (VOCs)) were explored to predict the marketable life of the fresh-cut nectarines. The respiration rate was higher in samples stored at 4 °C, while the browning process was more evident in fruit stored at 8 °C. Partial Least Squares Regression performed on VOCs showed that samples stored at 4 °C and 8 °C presented a different time evolution during the experiment and the trajectories depended on the interaction between time and temperature. Moreover, Multiple Linear Regression analysis discovered that the 17 VOCs affected by the storage conditions seemed to suggest that no chilling injury was detected for nectarines Big Bang. In conclusion, this approach could also be used with other nectarine cultivars and/or different stone fruits.

The brown marmorated stink bug (BMSB; Halyomorpha halys Stål.) is a highly destructive and polyphagous invasive pest that poses a serious threat to more than a hundred reported host plants. In the current study, the metabolic response of peach fruit of two cultivars-'Maria Marta' and 'Redhaven'-to BMSB infestation was studied using high-performance liquid chromatography (HPLC) and mass spectrometry (MS). In general, a strong phenolic response to BMSB infestation in peach flesh in the injury zone was observed, with flavanol content increasing by 2.4-fold, hydroxycinnamic acid content by 5.0-fold, flavonol content by 3.2-fold, flavanone content by 11.3-fold, and dihydrochalcones content by 3.2-fold compared with the undamaged tissue in the cultivar 'Maria Marta'. The phenolic response in the 'Redhaven' cultivar was even stronger. Consequently, the total phenolic content in the injured flesh also increased, 3.3-fold in 'Maria Marta' and 6.9-fold in 'Redhaven', compared with the uninjured flesh. Infestation with BMSB induced the synthesis of cyanidin-3-glucoside, which is not normally present in peach flesh. In comparison, the phenolic response was lower in peach peel, especially in the cultivar 'Maria Marta'. The study showed that both peach cultivars reacted to BMSB infestation with an increase in phenolic content in the peach flesh, but in a limited area of injury.

Storage or transportation temperature is very important for preserving the quality of fruit. However, low temperature in sensitive fruit such as peach can induce loss of quality. Fruit exposed to a specific range of temperatures and for a longer period can show chilling injury (CI) symptoms. The susceptibility to CI at low temperature varies among cultivars and genetic backgrounds. Along with agronomic management, appropriate postharvest management can limit quality losses. The importance of correct temperature management during postharvest handling has been widely demonstrated. Nowadays, due to long-distance markets and complex logistics that require multiple actors, the management of storage/transportation conditions is crucial for the quality of products reaching the consumer.Peach fruit exposed to low temperatures activate a suite of physiological, metabolomic, and molecular changes that attempt to counteract the negative effects of chilling stress. In this review an overview of the factors involved, and plant responses is presented and critically discussed. Physiological disorders associated with CI generally only appear after the storage/transportation, hence early detection methods are needed to monitor quality and detect internal changes which will lead to CI development. CI detection tools are assessed: they need to be easy to use, and preferably non-destructive to avoid loss of products.

Numerous biochemical processes are involved in fruit maturation, such as ethylene production, phenolic compounds accumulation, and antioxidant enzymes production. Therefore, the aim of the present work was the evaluation of ethylene production, and the bioactive compounds change in the exocarp and mesocarp of five peach [Prunus persica (L.)] cultivars during three ripening stages, (1) early ripening (ER), (2) commercial maturation, and (3) full ripening (FR) in order to establish the best stage to harvest each peach variety. The experiment was applied to five peach cultivars growing within an arid bioclimatic environment covering the whole peach production season: two early cultivars, Flordastar and Early Maycrest; one variety of mid-season Rubirich; and two late cultivars, Sweet Cap and O'Henry. Ethylene production, phenolic compounds, and oxidative stress through antioxidant enzyme activities (catalase, peroxidases [PODs] Class III, and ascorbate-POD), malondialdehyde (MDA), and hydrogen peroxide (H2 O2 ) production were determined in the exocarp and mesocarp of peach fruits. The results showed a significant increase in ethylene production during fruit ripening. However, a parallel decrease in the level of phenolic compounds as well as in antioxidant enzyme activities was observed. The FR stage was also characterized by an important accumulation of MDA and H2 O2 . In conclusion, important changes in fruit quality associated with the production level of ethylene were observed. Fruits harvested during the ER stage would be more suitable for delivering to distant markets and more appreciated by the peach industries due to their highest phenolic acid content, best antioxidant enzyme activities, and lowest oxidative stress indicator.

The flat peach is a high economic value table fruit possessing excellent quality and a unique aroma. This article investigated the quality characteristics and aroma fingerprinting of flat peaches (Qingpan, QP; Ruipan 2, R2; Ruipan 4, R4; Wanpan, WP) from Xinjiang in terms of taste, antioxidant capacity, and volatile aroma compounds using high-performance liquid chromatography (HPLC) and HS-SPME-GC-MS. The results showed that the flat peaches had a good taste and high antioxidant capacity, mainly due to the high sugar-low acid property and high levels of phenolic compounds. This study found that sucrose (63.86~73.86%) was the main sugar, and malic acid (5.93~14.96%) and quinic acid (5.25~15.01%) were the main organic acids. Furthermore, chlorogenic acid (main phenolic compound), epicatechin, rutin, catechin, proanthocyanidin B1, and neochlorogenic acid were positively related to the antioxidant activity of flat peaches. All flat peaches had similar aroma characteristics and were rich in aromatic content. Aldehydes (especially benzaldehyde and 2-hexenal) and esters were the main volatile compounds. The aroma fingerprinting of flat peaches consisted of hexanal, 2-hexenal, nonanal, decanal, benzaldehyde, 2,4-decadienal, dihydro-ß-ionone, 6-pentylpyran-2-one, 2-hexenyl acetate, ethyl caprylate, γ-decalactone, and theaspirane, with a "peach-like", "fruit", and "coconut-like" aroma. Among them, 2,4-decadienal, 2-hexenyl acetate, and theaspirane were the characteristic aroma compounds of flat peaches. The results provide a theoretical basis for the industrial application of the special aroma of flat peaches.

Peach (Prunus persica (L.) Batsch) is a fruit tree of economic and nutritional importance, but it is very sensitive to drought stress, which affects its growth to a great extent. Lauric acid (LA) is a fatty acid produced in plants and associated with the response to abiotic stress, but the underlying mechanism remains unclear. In this study, physiological analysis showed that 50 ppm LA pretreatment under drought stress could alleviate the growth of peach seedlings. LA inhibits the degradation of photosynthetic pigments and the closing of pores under drought stress, increasing the photosynthetic rate. LA also reduces the content of O2-, H2O2, and MDA under drought stress; our results were confirmed by Evans Blue, nitroblue tetrazolium (NBT), and DAB(3,3-diaminobenzidine) staining experiments. It may be that, by directly removing reactive oxygen species (ROS) and improving enzyme activity, i.e., catalase (CAT) activity, peroxidase (POD) activity, superoxide dismutase (SOD) activity, and ascorbate peroxidase (APX) activity, the damage caused by reactive oxygen species to peach seedlings is reduced. Peach seedlings treated with LA showed a significant increase in osmoregulatory substances compared with those subjected to drought stress, thereby regulating osmoregulatory balance and reducing damage. RNA-Seq analysis identified 1876 DEGs (differentially expressed genes) in untreated and LA-pretreated plants under drought stress. In-depth analysis of these DEGs showed that, under drought stress, LA regulates the expression of genes related to plant-pathogen interaction, phenylpropanoid biosynthesis, the MAPK signaling pathway, cyanoamino acid metabolism, and sesquiterpenoid and triterpenoid biosynthesis. In addition, LA may activate the Ca2+ signaling pathway by increasing the expressions of CNGC, CAM/CML, and CPDK family genes, thereby improving the drought resistance of peaches. In summary, via physiological and transcriptome analyses, the mechanism of action of LA in drought resistance has been revealed. Our research results provide new insights into the molecular regulatory mechanism of the LA-mediated drought resistance of peach trees.

Peach (Prunus persica L. Batsch) and apricot (Prunus armeniaca L.) are two species of economic importance for fruit production in the genus Prunus. Peach and apricot fruits exhibit significant differences in carotenoid levels and profiles. HPLC-PAD analysis showed that a greater content of ß-carotene in mature apricot fruits is primarily responsible for orange color, while peach fruits showed a prominent accumulation of xanthophylls (violaxanthin and cryptoxanthin) with yellow color. There are two ß-carotene hydroxylase genes in both peach and apricot genomes. Transcriptional analysis revealed that BCH1 expresses highly in peach but lowly in apricot fruit, showing a correlation with peach and apricot fruit carotenoid profiles. By using a carotenoid engineered bacterial system, it was demonstrated that there was no difference in the BCH1 enzymatic activity between peach and apricot. Comparative analysis about the putative cis-acting regulatory elements between peach and apricot BCH1 promoters provided important information for our understanding of the differences in promoter activity of the BCH1 genes in peach and apricot. Therefore, we investigated the promoter activity of BCH1 gene through a GUS detection system, and confirmed that the difference in the transcription level of the BCH1 gene resulted from the difference of the promoter function. This study provides important perspective to understanding the diversity of carotenoid accumulation in Prunus fruits such as peach and apricot. In particular, BCH1 gene is proposed as a main predictor for ß-carotene content in peach and apricot fruits during the ripening process.

The objective of this work was to validate the trunk water potential (Ψtrunk), using emerged microtensiometer devices, as a potential biosensor to ascertain plant water status in field-grown nectarine trees. During the summer of 2022, trees were subjected to different irrigation protocols based on maximum allowed depletion (MAD), automatically managed by real-time soil water content values measured by capacitance probes. Three percentages of depletion of available soil water (α) were imposed: (i) α=10% (MAD=27.5%); (ii) α=50% (MAD=21.5%); and (iii) α=100%, no-irrigation until Ψstem reached -2.0 MPa. Thereafter, irrigation was recovered to the maximum water requirement of the crop. Seasonal and diurnal patterns of indicators of water status in the soil-plant-atmosphere continuum (SPAC) were characterised, including air and soil water potentials, pressure chamber-derived stem (Ψstem) and leaf (Ψleaf) water potentials, and leaf gas exchange, together with Ψtrunk. Continuous measurements of Ψtrunk served as a promising indicator to determine plant water status. There was a strong linear relationship between Ψtrunk vs. Ψstem (R2 = 0.86, p<0.001), while it was not significant between Ψtrunk vs. Ψleaf (R2 = 0.37, p>0.05). A mean gradient of 0.3 and 1.8 MPa was observed between Ψtrunk vs.Ψstem and Ψleaf, respectively. In addition, Ψtrunk was the best matched to the soil matric potential. The main finding of this work points to the potential use of trunk microtensiometer as a valuable biosensor for monitoring the water status of nectarine trees. Also, trunk water potential agreed with the automated soil-based irrigation protocols implemented.

Glycans recently attracted considerable attention as the proposal of cross-reactive carbohydrate determinants for food allergy. Matrix-assisted laser desorption/ionization-mass spectrometry (MALDI-MS) is powerful in analyzing biomolecules, while its applications in glycans are still challenging. Herein, a novel reactive matrix-assisted laser desorption/ionization (MALDI) matrix, 2-hydrazinoterephthalic acid, was rationally designed and synthesized. It provides uniform co-crystallization with glycans and only produces deprotonated ions with high intensities in the negative-ion mode. In combination with sinapic acid, a rapid and high-throughput method was established for on-target analysis of glycans with a superior limit of detection at the femtomole level and a good linearity (R2 > 0.999). Furthermore, the established method was successfully applied to quantify N-glycans in different cultivars and tissues of peach [Prunus persica (L.) Batsch]. Our work suggests the potential role of N-glycans as biomarkers for food-borne allergy and lays a methodological foundation for the elucidation of the possible relationship between carbohydrate epitopes and food allergy.

BACKGROUND: The kinetic parameters of peach rootstock are a major factor contributing to its nitrogen (N) uptake efficiency, which directly affects the yield and quality parameters. However, the impact of grafting on N kinetic parameters is not sufficiently known as rootstocks are grafted with other scion cultivars before being transplanted to the field. The objective of this study was therefore to evaluate whether the grafting of 'Okinawa' rootstock with the scion cultivar 'Chimarrita' can alter the kinetic parameters related to N uptake. RESULTS: The plants were acclimatized in Hoagland solution for 21 days and, sequentially, were tested to assess the depletion of the internal reserves at 15 and 30 days in CaSO4 solution. Morphological and physiological parameters were evaluated, and kinetic parameters were calculated. The depletion period for to reach the minimum concentration (Cmin ) of NO3 - and NH4 + in the plant was 62 h and 56 h, respectively. The optimal time for internal reserve depletion for 'Okinawa' rootstock with and without the 'Chimarrita' scion was 30 days. CONCLUSION: 'Okinawa' rootstock grafted with 'Chimarrita' scion cultivar was more efficient in NO3 - and NH4 + uptake. © 2022 Society of Chemical Industry.

Brown rot caused by the pathogen of the genus Monilinia is the most destructive disease in peaches worldwide. It has seriously reduced the economic value of the peach (Prunus persica (L.) Batsch) in Nyingchi and Qamdo, Tibet, China. Monilinia fructicola, Monilia mumecola, and M. yunnanensis have been reported as the causal agents of brown rot disease on stone fruits in China. In this study, we report on the identification of M. yunnanensis in peach orchards in Nyingchi and Qamdo, Tibet. From twenty-three isolates with the same characteristics, we identified the representative single-spore isolates T8-1, T8-8, and T8-20 as M. yunnanensis and confirmed that the Tibet brown rot disease was caused by M. yunnanensis based on the morphological characteristics and molecular analyses. The phylogenetic analysis of the glyceraldehyde-3-phosphate dehydrogenase (G3PDH) and ß-tubulin (TUB2) nucleotide sequences and the multiplex PCR identification revealed that the representative isolates T8-1, T8-8, and T8-20 were more closely related to M. yunnanensis than other Monilinia species. Furthermore, the biocontrol strain of Trichoderma T6 presented significant antagonistic activity on the M. yunnanensis T8-1 isolate (T8-1) among the five Trichoderma strains. The highest inhibitory rates for Trichoderma T6 and its fermentation product against T8-1 mycelial growth were 72.13% and 68.25%, respectively. The obvious inhibition zone displayed on the colony interaction area between the colony of T8-1 isolate and Trichoderma T6 and the morphological characterization of the T8-1 hyphae were enlarged and malformed after inoculation with the Trichoderma T6 fermentation product at 20-fold dilution. Our results indicate that the strain of Trichoderma T6 could be considered as a beneficial biocontrol agent in managing brown rot of peach fruit disease.

Water shortage is a key factor that can restrict peach tree growth. Plants produce fatty acids and the fatty acid derivatives lauric acid (LA) and 12-hydroxylauric acid (LA-OH), which are involved in abiotic stress responses, but the underlying stress response mechanisms remain unclear. In this study, physiological examination revealed that in Prunus persica (L.) Batsch, pretreatment with 50 ppm LA-OH and LA reduced drought stress, efficiently maintained the leaf relative water content, and controlled the relative conductivity increase. Under drought stress, LA-OH and LA treatments prevented the degradation of photosynthetic pigments, increased the degree of leaf stomatal opening and enhanced the net photosynthetic rate. Compared with drought stress, LA-OH and LA treatment effectively increased the net photosynthetic rate by 204.55% and 115.91%, respectively, while increasing the Fv/Fm by 2.75% and 7.75%, respectively, but NPQ decreased by 7.67% and 37.54%, respectively. In addition, the level of reactive oxygen species increased under drought stress. The content of O2 - in LA-OH and LA treatment decreased by 12.91% and 11.24% compared to CK-D, respectively, and the content of H2O2 decreased by 13.73% and 19.94%, respectively. At the same time, the content of malondialdehyde (MDA) decreased by 55.56% and 58.48%, respectively. We believe that the main reason is that LA-OH and LA treatment have improved the activity of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). The application of exogenous LA increased the levels of soluble sugars, soluble proteins, proline and free amino acids under drought stress, and maintained the osmotic balance of cells. Compared with CK-D treatment, it increased by 24.11%, 16.89%, 29.3% and 15.04%, respectively. At the same time, the application of exogenous LA-OH also obtained similar results. In conclusion, exogenous LA-OH and LA can alleviate the damage to peach seedlings caused by drought stress by enhancing the photosynthetic and antioxidant capacities, increasing the activities of protective enzymes and regulating the contents of osmotic regulators, but the molecular mechanism is still in need of further exploration.

In plants, a family of terpene synthases (TPSs) is responsible for the biosynthesis of terpenes and contributes to species-specific diversity of volatile organic compounds, which play essential roles in fitness of plants. However, little is known about the TPS gene family in peach and/or nectarine (Prunus persica L.). In this study, we identified 40 PpTPS genes in peach genome v2.0. Although these PpTPSs could be clustered into five classes, they distribute in several gene clusters of three chromosomes, share conserved exon-intron organizations, and code similar protein motifs. Thirty-five PpTPSs, especially PpTPS2, PpTPS23, PpTPS17, PpTPS18, and PpTPS19, altered their transcript levels after inoculation with Botryosphaeria dothidea, a cause of peach gummosis, compared to the mock treatments, which might further affect the contents of 133 terpenoids at 48 hours and/or 84 hours post inoculations in the current-year shoots of 'Huyou018', a highly susceptible nectarine cultivar. Moreover, about fifteen PpTPSs, such as PpTPS1, PpTPS2, PpTPS3, and PpTPS5, showed distinct expression patterns during fruit development and ripening in two peach cultivars, yellow-fleshed 'Jinchun' and white-fleshed 'Hikawa Hakuho'. Among them, the transcription level of chloroplast-localized PpTPS3 was obviously related to the content of linalool in fruit pulps. In addition, elevated concentrations (0.1 g/L to 1.0 g/L) of linalool showed antifungal activities in PDA medium. These results improve our understanding of peach PpTPS genes and their potential roles in defense responses against pathogens.

The peach (Prunus persica L.) is one of Tunisia's major commercial fruit crops and is considered one of the biggest water consumers of all crops. In warm and arid areas of southern Tunisia, irrigation is necessary to ensure orchard longevity and high yield and fruit quality. Nevertheless, under water-scarcity conditions and low water quality, water management should rely on efficient deficit irrigation strategies. In this study, sustained deficit irrigation (DI) and partial root-zone drying (PRD50) at 50% of crop evapotranspiration (ETc) were evaluated for their impact on the primary and secondary metabolites of the peach fruit of early cultivar Flordastar grown in the Tataouine region. A full irrigation (FI) treatment at 100%, etc., was used as a control treatment. Color, dry-matter content, firmness, organic acids, sugars, phenolic compounds, vitamin C, ß-carotene and minerals were assessed on harvested mature fruits. Dry-matter content and firmness increased significantly under DI and PRD50 (13% and 15.5%). DI fruit had the highest soluble-solid content (SSC), reaching Brix values of 14.3°. Fruit sorbitol and sucrose contents were not affected by Di and PRD50. Higher glucose in fruit juice was observed in PRD50 (23%) and DI (21.5%) compared to FI, which had the highest malic acid content (33.5-37%). Quinic and citric acids decreased with DI and PRD50, while almost all individual phenolic compounds increased with deficit irrigation. Hydroxycinnamates and anthocyanins were significantly higher in fruits harvested from DI and PRD50 treatments. Proanthocyanidins (catechin and epicatechin) were only improved by DI, while flavone compounds and vitamin C were not affected by irrigation restrictions. ß-carotene was higher in fruits yielded under FI (0.71 mg/100 g DM) than DI and PRD50 (0.21-0.43 mg/100 g DM). Macro- and micronutrients significantly increased in DI and PRD50 fruit. A significant difference between DI and PRD50 fruits was observed for Zn and Fe concentrations. This research highlights the positive impact of reduced irrigation on bioactive-fruit quality attributes and the suitability of PRD50 and DI as tools for irrigation management in arid areas of southern Tunisia, contributing to water-saving in orchards and the improvement of fruit commercial value.

BACKGROUND: With the domestication of ornamental plants, artificial selective pressure favored the propagation of mutations affecting flower shape, and double-flower varieties are now readily available for many species. In peach two distinct loci control the double-flower phenotype: the dominant Di2 locus, regulated by the deletion of the binding site for miR172 in the euAP2 PETALOSA gene Prupe.6G242400, and the recessive di locus, of which the underlying factor is still unknown. RESULTS: Based on its genomic location a candidate gene approach was used to identify genetic variants in a diverse panel of ornamental peach accessions and uncovered three independent mutations in Prupe.2G237700, the gene encoding the transcript for microRNA miR172d: a ~5.0 Kb LTR transposable element and a ~1.2 Kb insertion both positioned upstream of the sequence encoding the pre-miR172d within the transcribed region of Prupe.2G237700, and a ~9.5 Kb deletion encompassing the whole gene sequence. qRT-PCR analysis confirmed that expression of pre-miR172d was abolished in di/di genotypes homozygous for the three variants. CONCLUSIONS: Collectively, PETALOSA and the mutations in micro-RNA miR172d identified in this work provide a comprehensive collection of the genetic determinants at the base of the double-flower trait in the peach germplasms.

The roles of methyl jasmonate (MeJA) and prohydrojasmon (PDJ) in postharvest color development and anthocyanins biosynthesis in the skin of peach fruit remain unclear. In this study, peach fruit were infiltrated with MeJA (200 µM) or PDJ (40 µM) and stored at 22°C for 7 days. The results showed that treatment with MeJA or PDJ had a positive effect on red color formation in peach fruits due to anthocyanins accumulation (∼120% increase). This was attributed to increased enzyme activities, and enhanced transcript abundance of the genes associated with anthocyanins biosynthesis, induced by MeJA or PDJ. Both MeJA and PDJ promoted sucrose biosynthesis, and the subsequently elevated levels of the sucrose during storage were positively correlated with anthocyanins accumulation (0.49) and the activities of key biosynthesis enzymes (0.42-0.79). Based on these findings, we proposed that MeJA or PDJ treatments promote anthocyanins biosynthesis by regulating sucrose metabolism during the postharvest storage of peach fruit.

Pectin is the major component in the primary cell wall and middle lamella, maintaining the physical stability and mechanical strength of the cell wall. Pectate lyase (PL), a cell wall modification enzyme, has a major influence on the structure of pectin. However, little information and no comprehensive analysis is available on the PL gene family in peach (Prunus persica L. Batsch). In this study, 20 PpePL genes were identified in peach. We characterized their physicochemical characteristics, sequence alignments, chromosomal locations, and gene structures. The PpePL family members were classified into five groups based on their phylogenetic relationships. Among those, PpePL1, 9, 10, 15, and 18 had the higher expression abundance in ripe fruit, and PpePL1, 15, and 18 were upregulated during storage. Detailed RT-qPCR analysis revealed that PpePL1 and PpePL15 were responsive to ETH treatment (1 g L-1 ethephon) with an abundant transcript accumulation, which suggested these genes were involved in peach ripening and softening. In addition, virus-induced gene silencing (VIGS) technology was used to identify the roles of PpePL1 and PpePL15. Compared to controls, the RNAi fruit maintained greater firmness in the early storage stage, increased acid-soluble pectin (ASP), and reduced water-soluble pectin (WSP). Moreover, transmission electron microscopy (TEM) showed that cell wall degradation was reduced in the fruit of RNAi-1 and RNAi-15, which indicated that softening of the RNAi fruit has been delayed. Our results indicated that PpePL1 and PpePL15 play an important role in peach softening by depolymerizing pectin and degrading cell wall.

Abstract: Nectarine fruit is highly perishable due to its high moisture content (89%) and susceptibility to decay. Continuous degradation in quality attributes due to physiological responses and ripening result ultimately in post-harvest losses. Drying of fruit offers the possibility to minimize losses and add value to fresh produce. Thus, this study evaluated the impacts of sodium metabisulphite (SMB; 10 g/kg) and characterized the influence of hot air (50 °C) drying on the kinetics, fruit tissue microstructure, and the physicochemical properties of dried 'Sunectwentyone' nectarines (Super star®). Out of the nine mathematical models, Logarithmic and Henderson, and Pabis models were the most suitable to predict the drying behaviour of sliced nectarines (R 2 = 0.94). Based on the microstructural analysis, prolonged drying led to higher tissue displacement/disruption in dehydrated nectarine slices. Results showed that SMB treatment was more effective in maintaining both the freshness and the color of 'Sunectwentyone' nectarine than the untreated. The volatile compounds of eight peach varieties (Prunus persica L.)-"Filina", "Gergana", "Ufo-4", "July lady", "Laskava", "Flat Queen", "Evmolpiya", and "Morsiani 90"-growing in Bulgaria were analyzed for the first time. Gas chromatography-mass spectrometry (GC-MS) analysis and the HS-SPME technique revealed the presence of 65 volatile compounds; the main identified components were aldehydes, esters, and fatty acids. According to the provided principal component analysis (PCA) and hierarchical cluster analysis (HCA), the relative quantities of the identified volatile compounds depended on the studied peach variety. The results obtained could be successfully applied for the metabolic chemotaxonomy of peaches.
