RESUMO

Environmentally friendly methods for silver nanoparticles (AgNPs) synthesis without the use of hazardous chemicals have recently drawn attention. In this work, AgNPs have been synthesized by microwave irradiation using only honey solutions or aqueous fresh pink radish extracts. The concentrations of honey, radish extract, AgNO3 and pH were varied. AgNPs presented mean sizes between 7.0 and 12.8 nm and were stable up to 120 days. The AgNPs were employed as co-catalyst (TiO2 @AgNPs) to increase the hydrogen photogeneration under UV-vis and only visible light irradiation, when compared to pristine TiO2 NPs. The prepared photocatalyst also showed hydrogen generation under visible light. Additionally, AgNPs were used to assemble a nanoplasmonic biosensor for the biodetection of extremely low concentrations of streptavidin, owing to its specific binding to biotin. It is shown here that green AgNPs are versatile nanomaterials, thus being potential candidates for hydrogen photogeneration and biosensing applications.

RESUMO

Cruciferous vegetables such as cauliflower and radish contain isothiocyanates exhibiting chemoprotective effects in vitro and in vivo. This research aimed to assess the impact of cauliflower (CIE) and radish (RIE) isothiocyanate extracts on the metabolic activity, intracellular reactive oxygen species (ROS), and LDH production of selected human colorectal adenocarcinoma cells (HCT116 and HT-29 for early and late colon cancer development, respectively). Non-cancerous colon cells (CCD-33Co) were used as a cytotoxicity control. The CIE samples displayed the highest allyl isothiocyanate (AITC: 12.55 µg/g) contents, whereas RIE was the most abundant in benzyl isothiocyanate (BITC: 15.35 µg/g). Both extracts effectively inhibited HCT116 and HT-29 metabolic activity, but the CIE impact was higher than that of RIE on HCT116 (IC50: 0.56 mg/mL). Assays using the half-inhibitory concentrations (IC50) of all treatments, including AITC and BITC, displayed increased (p < 0.05) LDH (absorbance: 0.25-0.40 nm) and ROS release (1190-1697 relative fluorescence units) in both cell lines. BITC showed the highest in silico binding affinity with all the tested colorectal cancer molecular markers (NF-kB, ß-catenin, and NRF2-NFE2). The theoretical evaluation of AITC and BITC bioavailability showed high values for both compounds. The results indicate that CIE and RIE extracts display chemopreventive effects in vitro, but additional experiments are needed to validate their effects.

RESUMO

A great amount of iron ore tailings from the collapse of the Fundão dam in Southeast Brazil was deposited in an extensive agricultural area. The presence of this material creates insecurity for the resumption of agricultural activities, especially the cultivation of vegetables, which can accumulate metals at potentially toxic levels. In this study, two vegetables consumed in the affected area, arugula and radish, were cultivated in tailings and in soil. Productivity, photosynthetic pigment content, photosynthetic performance, metal accumulation, and the possible risk to food safety were analyzed. The productivity of both vegetables, arugula and radish, did not differ between cultivation in tailings and in soil. There were no differences in pigment content nor substantial differences in the photosynthetic parameters of plants grown in the two substrates. Plants grown in tailings had higher Fe, Mn, and Na contents than those grown in soil, reflecting the higher levels of these elements in the former. There were no visual signs of metal toxicity for plants grown in the tailings. The levels of metals potentially ingested through estimated consumption of arugula and radish grown in the tailings were below the maximum allowable limits for human consumption. In addition, calculated risk indices suggest a low potential for harm to the health of consumers of cultivated vegetables in the tailings. The results presented here suggest that agricultural cultivation in the tailings is viable and contribute to the resumption of vegetable cultivation in the region affected by the tailings released with the collapse of the Fundão dam.

RESUMO

AIM: This work aimed at determining the pathogenicity, molecular characterization, host range and rapid detection of Pectobacterium carotovorum subsp. brasiliense (Pcb) causing soft rot disease in radish. METHODS AND RESULTS: The four isolated isolates were inoculated to radish, typical soft rot symptoms were observed and Koch's postulates were proved. The most virulent strain RDKLR was morphologically and biochemically distinct. Pcb showed a positive potato soft rot test and elicited hypersensitivity response on Nicotiana tobaccum. The genes Pel2 and pmrA were used for subspecies characterization of Pcb. It has a wide host range and infection was observed on slices of carrot, tomato, radish, potato, cauliflower, cabbage, chilli, knol-khol, bell pepper and cucumber. Infectivity was also seen in seedlings under glasshouse conditions. Pcb produced cell wall degrading enzymes in semi-quantification assay and is a strong biofilm producer. The LAMP technique was standardized to help rapid detection and take prophylactic measures to manage the disease. CONCLUSION: This work reports Pcb as a new soft rot causing organism of radish in India. Pcb is highly virulent with a broad host range. The LAMP technique helps in rapid detection. SIGNIFICANCE AND IMPACT OF THE STUDY: Pcb-induced soft rot causes significant yield loss, decreased market value, damage in transit, storage and the market. Disease characterization and early identification aid in disease management and prevention in the field.

RESUMO

The collapse of the Fundão tailings dam of the Samarco mining complex in Mariana, Brazil, was the largest mining disaster in the world to date with many socio-economic and environmental impacts. Soil affected by mining tailings was severely altered with negative impacts for agriculture. We tested whether diluting mining tailings with organic soil would eliminate or at least attenuate the ecotoxic effects on plant development and performance. We cultivated radish, Raphanus sativus,in substrates containing different proportions of mining tailings and organic soil: pure tailings (T100%); 2) tailings75%+ soil25%(T75%); 3) tailings50%+ soil50%(T50%); 4) tailings25%+ soil75%(T25%), and 5) pure organic soil (Soil100%, control). There were large differences in soil quality parameters between the 100% tailings treatment (T100%) and the control (Soil100%), as well as for some parameters in the most diluted treatment -T25%(Ca2+, Fe, Mn) in relation to the controltreatment. Although dilution of the tailings soil improved radish development, there was lower radish productivity (leaf area, total biomass, and root/tuber biomass) than for pure soil (control). There were also significantly higher amounts of bioaccumulated metals in radish tubers grown with tailings, even when grown in T25%for Fe content and in T75%for Mn content. These results present a worrisome scenario for human communities in the region of the Doce river, as human consumption of crops produced in soil contaminated with tailingsis not recommended due to potential toxicological effects from high metal concentrations.(AU)

RESUMO

In the present work the radish (Raphanus sativus L.) was used as the low-cost alternative source of peroxidase. The enzyme was immobilized in different supports: coconut fiber (CF), calcium alginate microspheres (CAMs) and silica SBA-15/albumin hybrid (HB). Physical adsorption (PA) and covalent binding (CB) as immobilization techniques were evaluated. Immobilized biocatalysts (IBs) obtained were physicochemical and morphologically characterized by SEM, FTIR and TGA. Also, optimum pH/temperature and operational stability were determined. For all supports, the immobilization by covalent binding provided the higher immobilization efficiencies-immobilization yield (IY%) of 89.99 ± 0.38% and 77.74 ± 0.42% for HB and CF, respectively. For CAMs the activity recovery (AR) was of 11.83 ± 0.68%. All IBs showed optimum pH at 6.0. Regarding optimum temperature of the biocatalysts, HB-CB and CAM-CB maintained the original optimum temperature of the free enzyme (40 °C). HB-CB showed higher operational stability, maintaining around 65% of the initial activity after four consecutive cycles. SEM, FTIR and TGA results suggest the enzyme presence on the IBs. Radish peroxidase immobilized on HB support by covalent binding is promising in future biotechnological applications.

RESUMO

Recently, there has been an interest regarding the consumption of wild edible plants in modern diets. However, there is still scarce information about several wild vegetables traditionally consumed. Therefore, this work aims on documenting the nutritional and chemical composition of wild radish (Raphanus raphanistrum L.), as well as its bioactive potential. Results showed that wild radish is a potential source of beneficial compounds, including vitamin E, polyunsaturated fatty acid (particularly α-linolenic acid) and different phenolic compounds, in which fourteen phenolics were identified, with kaempferol-3,7-O-di-rhamnoside being the most abundant. The bioactive potential was exploited using hydroethanolic and decoction extracts. Both proved to inhibit several Gram-positive and Gram-negative bacteria and revealed antioxidant activity, while cytotoxicity against non-tumor cell was not observed. In general, results evidence the interest in recovering the use of this wild vegetable as part of a varied diet, which can bring several health benefits.

RESUMO

In this work, fodder radish seed cake (FRSC) was pyrolyzed in a rotary kiln reactor at 0, 3, and 6 rpm, at final temperature of 500 °C. Maximum biochar yield was observed at 0 rpm (≈ 26 wt.%). Increase of the rotary speed decreased the volatile matter content and increased the ash content of the biochars. Biochars exhibited alkaline pH (≈ 9.0), low electrical conductivity (< 105.6 dS m-1), and high cation exchange capacity (69 to 78 cmolc kg-1), as well as high nitrogen contents (≈ 80 g kg-1). FTIR analysis presented biochars with similar spectra, with carboxyl and carbonyl groups within the structure, along with aromatic rings and nitrogen containing functions (amides). Biochar incubation experiments in an acrisol at different biochar doses (5 g L-1 soil to 40 g L-1 soil) were performed in order to evaluate changes in soil fertility parameters caused by FRSC biochar application. Results indicated that most of macro (N, P, K, Ca, Mg) and micronutrients (S, Cu, Zn, Mn, B, Na) increased with increase of the dosage, along with the decrease in Al and H+ Al contents. An increase in pH (from 4.25 to 5.33) was also observed, in electric conductivity (from 30.0 to 45.7 dS m-1), and a decrease in soil real density (from 3.67 to 2.99 kg L-1) at the dosage of 40 g char L-1 soil.

RESUMO

BACKGROUND: Feral radish (Raphanus sativus L.) is a problematic weed that has become resistant to acetohydroxyacid synthase (AHAS) inhibitor herbicides due to the Trp574Leu mutation. An AHAS gene mutation that causes herbicide resistance may have negative pleiotropic effects on plant fitness. This study reports the effects of the Trp574Leu mutation on AHAS activity and reproductive traits of R. sativus. RESULTS: Eight of 17 feral radish accessions presented individuals resistant to metsulfuron-methyl at 0.5% to >90.0% and all the resistant individuals analyzed showed the Trp574Leu mutation. Without herbicide selection, the AHAS activity was 3.2-fold higher in the susceptible accession than in the resistant one. The resistant accession was >9000-fold more resistant to metsulfuron-methyl and imazethapyr than the susceptible accession. Under low intraspecific competition during two growing seasons, AHAS-resistant feral radish accessions showed 22-38% and 21-47% lower seed numbers and yield per plant than the susceptible accession. CONCLUSION: This is the first report of fitness cost associated with the AHAS Trp574Leu mutation in R. sativus populations. This fitness cost could reduce frequency of the resistant allele without herbicide selection. © 2018 Society of Chemical Industry.

RESUMO

The antibacterial activity of chitosan coatings prepared with acetic or lactic acid, as well as of composite chitosan-gelatin films prepared with essential oils, was evaluated in fresh shredded black radish samples inoculated with Listeria monocytogenes ATCC 19115 and L. monocytogenes ATCC 19112 during seven days of storage at 4 °C. The chitosan coating prepared with acetic acid showed the most effective antibacterial activity. All tested formulations of chitosan films exhibited strong antimicrobial activity on the growth of L. monocytogenes on black radish, although a higher inhibition of pathogens was achieved at higher concentrations of chitosan. The antimicrobial effect of chitosan films was even more pronounced with the addition of essential oils. Chitosan-gelatin films with thyme essential oils showed the most effective antimicrobial activity. A reduction of 2.4 log10 CFU/g for L. monocytogenes ATCC 19115 and 2.1 log10 CFU/g for L. monocytogenes ATCC 19112 was achieved in the presence of 1% chitosan film containing 0.2% of thyme essential oil after 24 h of storage.

BACKGROUND: Soon after the commercial release of sunflower cultivars resistant to imidazolinone herbicides, several uncontrolled feral radish (Raphanus sativus L.) populations were found in south-eastern Buenos Aires, Argentina. These populations were studied in field, glasshouse and laboratory experiments aiming to characterise their resistance profile and to develop management tools. RESULTS: Three feral radish accessions were highly resistant to ten active ingredients of five families of acetohydroxyacid synthase (AHAS)-inhibiting herbicides. Sequence analysis of the AHAS gene detected a Trp574Leu mutation in all resistant accessions. One accession with an intermediate level of resistance was heterozygous for this mutation, probably owing to gene exchange with a susceptible subpopulation located in the field margin. Herbicide-resistant and herbicide-susceptible radish could be controlled in sunflower by alternative herbicides. CONCLUSION: This is the first report of feral radish with resistance to herbicides belonging to all the AHAS-inhibiting herbicide families, conferred by Trp574Leu mutation in the AHAS gene. An appropriate herbicide rotation with alternative herbicides such as fluorochloridone or aclonifen and an increase in the diversity of cropping systems are important for minimising the prevalence of these biotypes.

RESUMO

In this study, the potential for the sorption of Pb(2+) and Cd(2+) from aqueous solutions using HNO3-treated activated carbon (TAC) obtained from radish press cake (Raphanus sativus L.), a solid residue from biodiesel production, was investigated. Activated carbon (AC) was obtained by physical activation with CO2(g). Chemical modification with HNO3 was employed to increase the sorption capability of the AC. The sorption of Pb(2+) and Cd(2+) was studied in monometallic systems in equilibrium with different metal-ion concentrations (10-400 mg L(-1)). The experimental sorption equilibrium data were fit to the Langmuir and Freundlich isotherm models. The maximum sorption capacity (qmax) obtained for AC from the Langmuir isotherm was 45.5 mg g(-1) for Cd(2+) and 250 mg g(-1) for Pb(2+). Moreover, TAC presented qmax of 166.7 mg g(-1) (1.48 mmol g(-1)) for Cd(2+) and 500.0 mg g(-1) (2.41 mmol g(-1)) for Pb(2+)showing the effect of chemical modification. Sorption-desorption studies showed that the interaction between metals and TAC is reversible and this sorbent can be reused for several consecutive cycles. Furthermore, the sorption of Cd(2+) and Pb(2+) by TAC was not affected by the presence of competing ions. The experimental data obtained in this study indicated that this solid residue is viable for the production of sorbents that remove metals, such as cadmium and lead, from wastewaters and thereby contribute to the sustainable development of the production of biodiesel.

RESUMO

The aim of this study was to evaluate the phytotoxic activity of aqueous extracts from mature leaves, stem bark and roots of Drimys brasiliensis Miers on germination and initial seedling development of two target species, Triticum aestivum L. (wheat) and Raphanus sativus L. (radish). The aqueous extract was prepared in a proportion of 10 g of plant powder, dissolved in 100 mL of distilled water, resulting in a 10% extract concentrate. Dilutions were made with distilled water to 7.5; 5.0; 2.5% and 0% (control). Germination and seedling growth bioassays were carried out under controlled laboratory conditions. The different plant parts exerted inhibitory effects on germination and early growth of wheat and radish, however, the extract obtained from the leaves was more effective in inhibiting the germination of radish. All plant parts of D. brasiliensis may constitute a promising source in the search for compounds capable of acting as natural phytotoxins.

The effects of nCeO2 on food quality are not well known yet. This research was performed to determine the impact of nCeO2 on radish (Raphanus sativus L.). Plants were cultivated to full maturity in potting soil treated with nCeO2 at concentrations of 0, 62.5, 125, 250, and 500 mg/kg. Germination, growth, photosynthesis, ionome, and antioxidants were evaluated at different growth stages. Results showed that at 500 mg/kg, nCeO2 significantly retarded seed germination but did not reduce the number of germinated seeds. None of the treatments affected gas exchange, photosynthesis, growth, phenols, flavonoids, and nutrients' accumulation in tubers and leaves of adult plants. However, tubers' antioxidant capacity, expressed as FRAP, ABTS(•-) and DPPH, increased by 30%, 32%, and 85%, respectively, in plants treated with 250 mg nCeO2kg(-1) soil. In addition, cerium accumulation in tubers of plants treated with 250 and 500 mg/kg reached 72 and 142 mg/kg d wt, respectively. This suggests that nCeO2 could improve the radical scavenging potency of radish but it might introduce nCeO2 into the food chain with unknown consequences.

The objective of this research was to evaluate crop yield and some characteristics and yield components of transgenic soybean cultivars sown after different winter cover crops in the first year under no tillage system. The experimental design was the completely randomized block with split plots and four replications. The main plots consisted of five winter cover crops, white oat (Avena sativa L.), forage turnip (Raphanus sativus L.), barley (Hordeum vulgare L.), wheat (Triticum aestivum L.) and ground pea (Pisum sativum L.) and an area under fallow (spontaneous vegetation). The subplots consisted of six soybean cultivars (BRS 243 RR, BRS 245 RR, BRS 247 RR, BRS 255 RR, BRS 256 RR and BRS 244 RR). Variance analysis for agronomic characteristics showed that soybean yield components were influenced by the interaction between winter crop and soybean cultivar. Thus, final population, number of nodes and pods per plant, nodes dry matter per plant, number of grains per pod and grain yield were affected significantly. When soybean nodulation was evaluated, the treatment with the area under fallow showed lower values. There was difference among winter crops for BRS 243 RR grain yield, white oat showed the highest values.

RESUMO

The black oats use (Avena strigosa) as species of soil covering in the winter, cause immobilization of the nitrogen (N), that reduces the plant development and grain yield of maize cultivated in succession. Thus, the black oat intercropped systems with leguminous as common vetch (Vicia sativa) and brassicas as oilseed radish (Raphanus sativus) is aimed at increasing nitrogen (N) disponibility in the system and the permanence timing of its residues in the soil. Two experiments were carried out in the growth seasons of 2001/2002 and 2002/2003, in Rio Grande do Sul, Brazil. The first one was aimed at evaluating the effect of three winter species of soil covering, grown as a single culture and as intercropped crops on maize grain yield, with and without nitrogen side-dressed. The second one was aimed at determining the most adequate seed ratio of oilseed radish and black oat in intercropped systems, as soil covering crops in the winter preceding maize, under different nitrogen levels side-dressed. In Experiment I, treatments were composed by N application of 180kg ha-1, a control without N side-dressed and seven winter soil covering systems. In the Experiment II, treatments consisted of two levels of N side-dressing application in maize, a control without N side-dressed, and of three seed ratio of oilseed radish and black oat, as single and as intercropped crops and a control without crop in the winter. In all intercropped systems, independently of seed ratio used, the oilseed radish was mostly responsible for the yield of dry mass of the systems. The intercropped systems of common vetch or oilseed radish with black oat minimize the negative effect of oat on maize grain yield cultivated in succession in systems with low N availability and, even with high N supply, maize grain yield also increases when grown after common vetch.

The effect of three levels of shading (0; 30 and 50%) on the development and tuberous root yield of radish (Raphanus sativus L.) cv. "Vermelho Redondo" was studied under field conditions, at the São Manuel County, State of São Paulo, Brazil. The plants were evaluated at 7, 14, 21 and 28 days after the emergency. The 50% level of shading increased the life cycle and foliar area, reduced the leave chlorophryll content and the tuberous root yield. The 30% level of shading did not reduce the size and weight of the roots.
