RESUMO

OBJEVTIVE: To investigate the effects of extracts from Chuanwu (Aconitum Carmichaelii) and Banxia (Rhizoma Pinelliae) on the excisional wound healing in a rat's model. METHODS: Rats were performed a surgical lesion with a 2.0 cm resected tissue in the dorsal fascia. Following, animals were randomly divided into model group, YNB group and APE group those were respectively treated with saline, 1 mg/mL of Yunnan Baiyao and 1 mg/mL of Chuanwu (Aconitum Carmichaelii) and Banxia (Rhizoma Pinelliae) extracts. Wound contractions in days 0, 3, 7, 14 and 21 were calculated by an image analyzer. Histological analysis was analyzed using hematoxilin and eosin. Levels of tumor necrosis factor α (TNF-α), interleukin-2 (IL-2), IL-4, IL-10, transforming growth factor-ß1 (TGF-ß1) and basic fibroblast growth factor (bFGF) were determined by real-time quantitative PCR. RESULTS: Compared with that of YNB group and APE gtoup, the skin of rats showed poor re-modeling and re-epithelization characterized by a significant decrease of neovascularization, epithelialization and fibroblast in the model group. In the APE group, levels of TNF-α and IL-2 were significantly down-regulated and IL-4 and IL-10 significantly up-regulated in contrasted with that of model group. In addition, levels of TGF-ß1 and bFGF in the APE group were significantly induced compared with that of model group. CONCLUSION: The results suggest that the extracts from Chuanwu (Aconitum Carmichaelii) and Banxia (Rhizoma Pinelliae) promote wound healing in the rats, which is associated with enhancing anti-inflammatory ability and inducing fibroblast formation.

ETHNOPHARMACOLOGICAL RELEVANCE: Rhizoma Pinelliae Pedatisectae (RPP) has been commonly used in traditional Chinese medicine (TCM) for treatment of various neurological related diseases. However, the mechanisms have not been fully clarified yet. The present study was designed to investigate the therapeutic effect of RPP against cerebral ischemic/reperfusion (I/R) injury in rats models, and more importantly, to explore the underlying mechanism. MATERIALS AND METHODS: Cerebral ischemia and reperfusion was established through the classic middle cerebral artery occlusion (MCAO) for 2h, followed by 24h reperfusion. Rats were orally given different doses of n-butyl alcohol extracts (NBES) of RPP or saline for one week before induction of ischemia. Neurological defect scoring, cerebral infarct volume, oxidative stress markers, inflammatory reaction and nerve cell apoptosis were then estimated. RESULTS: It showed that NBES could alleviate in a dose-dependent manner neurological deficit and reduce the infarct volume in vivo. The protective effects of NBES were associated with increased Superoxide dismutase (SOD) activity and decreased malonaldehyde (MDA), tumor necrosis factor-α (TNF-α) and interleukin-1ß (IL-1ß) levels. In addition, ischemia-induced neuron apoptosis was inhibited by NBES pretreatment, and western blot showed NBES upregulated expressions of B-cell leukemia-2 (Bcl-2) and downregulated Bcl-2-associated X (Bax) expression. CONCLUSION: NBES prevent cerebral I/R injury by alleviating neuronal oxidative injury, inflammatory reaction and neuron apoptosis. The research for the traditional use of RPP provided certain theoretical basis.

BACKGROUND: Rhizoma Pinelliae is the dried tuber of Pinellia ternata (Thunb.) Breit. Modern pharmacological studies have shown that Rhizoma Pinelliae has antitussive, antiemetic, glandular secretion inhibiting and antitumor effects. OBJECTIVES: To optimize the processing technology of Rhizoma Pinelliae Praeparatum, and to study its anti-tumor effect. METHODS: Orthogonal design method was applied to analyze the effects of factors such as licorice concentration volume, soaking time and processing temperature on processing technology of Rhizoma Pinelliae Praeparatum; MTT assay and flow cytometry were used to determine the inhibitory effect of Rhizoma Pinelliae Praeparatum on Bel-7402 cells. RESULTS: During the processing of Rhizoma Pinelliae Praeparatum, the size of influence of licorice concentration volume, soaking time and processing temperature on processing results of Rhizoma Pinelliae was: B>C>A in descending order, i.e. soaking time>processing temperature>licorice concentration volume, different concentrations of Rhizoma Pinelliae Praeparatum ethanol extracts could all exert inhibitory effect on the growth and proliferation of Bel-7402 cells, and with the increase of drug concentration and the extension of culture time, the cell proliferation inhibitory effect of Rhizoma Pinelliae Praeparatum ethanol extract became more and more evident. Apoptotic rate of 1.5 mg/ml Rhizoma Pinelliae Praeparatum ethanol extract group reached 13.53%, the difference was extremely significant compared with the control group. In conclusion the factor most influential to the processing technology of Rhizoma Pinelliae Praeparatum was soaking time, followed by processing temperature, the factor least influential was licorice concentration volume. CONCLUSION: Rhizoma Pinelliae Praeparatum has inhibitory effect on growth and proliferation of Bel-7402 cells.

RESUMO

Rhizoma Pinelliae Fermentata(RPF)wasoneofthecommonlyusedChinesemateriamedica(CMM). According to the ancient and modern literatures on RPF, the historical evolution, fermentation methods, chemical compositions, efficacy and microbes of RPF were systematically summarized in this paper. Through the analysis on existing problems of fermentation strains, effective components, quality standard and fermentation process, the corresponding solutions were proposed. This work may provide an idea and reference for the further study of RPF.

OBJECTIVE: To establish and compare the HPLC fingerprints of different parts of Rhizoma Pinelliae (RPI), Rhizoma Pinelliae Praeparatum Cum Alumine (RPA), Rhizoma Pinelliae Praeparatum Cum Zingibere Et Alumine (RPZ), Rhizoma Pinelliae Praeparatum (RPP) and Jing Pinelliae (JPI) and provide reliable method and scientific basis for their quality control. METHODS: The HPLC fingerprints of water, 75% ethanol and 95% ethanol extracts of Rhizoma Pinelliae and processed products were established and analyzed with Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2004A). RESULTS: The chromatograms of water, 75% ethanol and 95% ethanol extracts were generated as the representative standard fingerprints. Thirteen common peaks were obtained in the fingerprints of Rhizoma Pinelliae and Rhizoma Pinelliae Praeparatum Cum Alumine; 15 common peaks were obtained in the fingerprints of Rhizoma Pinelliae Praeparatum Cum Zingibere Et Alumine; 17 common peaks were obtained in the fingerprints of Rhizoma Pinelliae Praeparatum and Jing Pinelliae. Among the common peaks, 8 characterized peaks were identified as inosine, guanosine, adenosine, succinic acid, ephedrine hydrochloride, liquiritin, glycyrrhizic acid, and 6-Gingerol. Guanosine, adenosine, succinic acid, and ephedrine hydrochloride existed in Rhizoma Pinelliae and processed products. 6-gingerol was only detected in Rhizoma Pinelliae Praeparatum Cum Zingibere Et Alumine. Liquiritin and glycyrrhizic acid were detected in Rhizoma Pinelliae Praeparatum and Jing Pinelliae. A new peak (peak 8) appeared in the chromatograms of Rhizoma Pinelliae Praeparatum Cum Alumine and Rhizoma Pinelliae Praeparatum Cum Zingibere Et Alumine; Rhizoma Pinelliae Praeparatum and Jing Pinelliae did not show inosine and had a new peak (peak 11) compared to Rhizoma Pinelliae. CONCLUSION: This study established the HPLC characteristic fingerprints of different parts of Rhizoma Pinelliae and processed products. The method is stable, time-saving, reliable and can identify Rhizoma Pinelliae and its four different processed products, which provides a scientific basis for the quality control of Rhizoma Pinelliae.

Outlining the medicinal properties of unprocessed Rhizoma Pinelliae and its general usage nowadays,this paper highlighs the view of veteran TCM practitioner LI Ke on processed Rhizoma Pinelliae and the reasons for the clinical overdose application and its clinical effect.Besides,before proposing a new thinking to the clinical use of herbs in the future,the author concludes his clinical experience in application of unprocessed Rhizoma Pinelliae under the guidance of Master LI,particularly the application in the treatment of difficulty miscellaneous disease.

Objective To establish a method for the determination of alum residual amount in Rhizoma Pinelliae Preparata from different companies,and to provide a scientific evidence for quality evaluation of Rhizoma Pinelliae Preparata.Methods Na2EDTA titration method was applied for the quantitative analysis.Results There were obvious differences in residual quantity of alum in Rhizoma Pinelliae Preparata from different companies.Conclusion The method is simple,reproducible and accurate,it can be used for the determination of alum residual amount in Rhizoma Pinelliae Preparata.

OBJECTIVE:To determine the content of ?-aminobutyric acid in Rhizoma Pinelliae Praeparatum.METHO-DS:In alkaline condition,amino acids reacted with phenyl isothiocyanate and resulted in the derivatives of benzene isothiocyanate-amino acid complex.A Venusil-AA amino acid analysis column(250 mm?4.6 mm,5 ?m)was used and the absorbance at 248 nm was measured.The mobile phase A is 0.05 mol?L-1 sodium acetate solution(pH6.5)and the mobile phase B consisted of acetonitrile-water(80∶20).RESULTS:The linear range of ?-aminobutyric acid was from 2.056 ?g?mL-1 to 12.336 ?g?mL-1,and the recovery was 99.0%(RSD=0.92,n=6).CONCLUSION:This method is simple,accurate and reproducible,and it can be used for quality control of Rhizoma Pinelliae Praeparatum.

Objective To purify a Rhizoma Pinelliae protein,determine its inhibiting activity and mechanism to trypsin,and analyze its N-terminal amino acid sequence. Methods Active Rhizoma Pinelliae protein was purified from the 40% (NH_4)_2SO_4 sediment of the crude extracted protein from Rhizoma Pinelliae by affinity chromatography of trypsin-Sepharose CL 4B and gel filtration of Sephadex G-50;12% of SDS-PAGE was used for determining the purity of the purified Rhizoma Pinelliae protein and estimating its molecular weight;N-terminal amino acid sequence of active Rhizoma Pinelliae protein was analyzed by Edman degradation. Results The purified active Rhizoma Pinelliae protein showed a sin-gle band on SDS-PAGE gel,which estimated molecular weight was about 1. 4?10~4,its specific activity was 1 059. 012 U/mg,and the yield was about 24.40%;The N-terminal sequence of the preceding six amino acid residues of active Rhizoma Pinelliae protein was DPVVDG;The quality inhibiting ratio of active Rhi- zoma Pinelliae protein to trypsin was 1:4. 72,and the Ki value ,the inhibition constant was about 7. 17 ?10~(-6) mol/L. Conclusion Active Rhizoma Pinelliae protein is a kind of serine protease inhibitor,a com-petive inhibitor to trypsin.

RESUMO

AIM: To establish HPLC-digitized fingerprint spectrum (HPLC-DFPS) of Rhizoma Pinelliae so that it will be applied in quality control of Rhizoma Pinelliae. METHODS: A gradient separated method was applied. Column: Agilent Zorbax ODS C_ 18 ; Mobile phase: acetonitrile-water; Detection-wavelength: 210 nm; Flow rate: 0.5 mL/min; Column temperature: 25℃. RESULTS: Using above optimum chromatograph condition, the HPLC-DFPS and technic parameters of Rhizoma Pinelliae were established. CONCLUSION: Comparison of a group of characteristic peaks suggests that the HPLC-DFPS technology is feasible in quality control of Rhizoma Pinelliae.

Objective: To compare Succinic Acid of Rhizoma Pinelliae in viarious areas in China.Method: Using the technology of TCL.Result:Succinic Acid in various areas in China are not very close to one another.The highest amount is in Shanxi Province(about 0.0254% )and the lowest amount is in Hunan Province(about 0.0038% ).Conclusion:There are obvious differences of Succinic Acid among various areas in China.Succinic Acid shoule be between 0.0038%~0.0254%
