RESUMO

Anaerobic membrane bioreactors (AnMBRs) have demonstrated an excellent capability to treat domestic wastewater. However, biofouling reduces membrane permeability, increasing operational costs and overall energy demand. Soluble microbial products (SMPs) that build up on the membrane surface play a significant role in the biofouling. In this study, the production of SMPs in a 32 L submerged AnMBR operated at three different organic loads (3.0, 4.1 and 1.2 kg chemical oxygen demand (COD)/m3d for phases 1, 2 and 3, respectively) during long-term operation of the reactor (144, 83 and 94 days) were evaluated. The samples were taken from both the permeate and the sludge at three different heights (0.14, 0.44 and 0.75 m). Higher production of SMPs was obtained in phase 2, which was proportional to the membrane fouling. There were no statistically significant differences (p > 0.05) in the SMPs extracted from sludge at different heights among the three phases. In the permeate of phases 1, 2 and 3, the membrane allowed the removal of 56%, 70% and 64% of the SMP concentration in the sludge. SMPs were characterized by molecular weight (MW). A bimodal behavior was obtained, where fractions prevailed with an MW < 1 kDa, associated with SMPs as utilization-associated products (UAPs) caused fouling by the pore-blocking mechanism. The chemical analysis found that, in the SMPs, the unknown COD predominated over the known COD, such as carbohydrates and proteins. These results suggest that further studies in SMP characterization should focus on the unknown COD fraction to understand the membrane fouling in AnMBR systems better.

Mollusk shell mineralization is a tightly controlled process made by shell matrix proteins (SMPs). However, the study of SMPs has been limited to a few model species. In this study, the N66 mRNA of the pearl oyster Pinctada mazatlanica was cloned and functionally characterized. The full sequence of the N66 mRNA comprises 1,766 base pairs, and encodes one N66 protein. A sequence analysis revealed that N66 contained two carbonic anhydrase (CA) domains, a NG domain and several glycosylation sites. The sequence showed similarity to the CA VII but also with its homolog protein nacrein. The native N66 protein was isolated from the shell and identified by mass spectrometry, the peptide sequence matched to the nucleotide sequence obtained. Native N66 is a glycoprotein with a molecular mass of 60-66 kDa which displays CA activity and calcium carbonate precipitation ability in presence of different salts. Also, a recombinant form of N66 was produced in Escherichia coli, and functionally characterized. The recombinant N66 displayed higher CA activity and crystallization capability than the native N66, suggesting that the lack of posttranslational modifications in the recombinant N66 might modulate its activity.

Mollusk biomineralization is a process controlled by a complex interplay of proteins, ions and external regulators. In spite of several studies, there is a lack of knowledge of who (molecules involved), how (mechanism) and why (evolution and adaptation) mollusk are designed as we know them. In this study, a shell matrix protein, N66, has been purified and characterized biochemically from the shell of Pteria sterna. Two protein bands with carbohydrates associated were separated with a molecular weight of ~60 and 64 kDa. It has carbonic anhydrase activity and it is able to form crystal polymorphs of calcium carbonate in vitro. The mRNA N66 was obtained from the mantle tissue of Pteria sterna and the deduced amino acid sequence contained a carbonic anhydrase (CA) domain and a Asn/Gly-rich domain (aa243-439). The CA domain contained three His residues acting as zinc ligands and the gate-keeper residues present in all α-CAs (Glu166-Thr525), being thus similar to the human isoform hCAVII. Also, to test whether the posttranslational modifications present on the native N66 affects the CA activity and its crystallization capability in vitro, a recombinant N66 was overexpressed in Escherichia coli and functionally characterized. Our results show that recombinant N66 has higher CA activity and produce larger size crystals in vitro than the native N66 protein, suggesting that intrinsic properties of the native N66, such as glycosylations and/or phosphorylations, might regulate its activity.

Quantum dots exhibit extraordinary optical and mechanical properties, and the number of their applications is increasing. In order to investigate a possible effect of coating on the inhalation toxicity of previously tested non-coated CdS/Cd(OH)2 quantum dots and translocation of these very small particles from the lungs, rats were exposed to coated quantum dots or CdCl2 aerosol (since Cd(2+) was present as impurity), 6h/d for 5 consecutive days. Cd content was determined in organs and excreta after the end of exposure and three weeks thereafter. Toxicity was determined by examination of broncho-alveolar lavage fluid and microscopic evaluation of the entire respiratory tract. There was no evidence for translocation of particles from the respiratory tract. Evidence of a minimal inflammatory process was observed by examination of broncho-alveolar lavage fluid. Microscopically, minimal to mild epithelial alteration was seen in the larynx. The effects observed with coated quantum dots, non-coated quantum dots and CdCl2 were comparable, indicating that quantum dots elicited no significant effects beyond the toxicity of the Cd(2+) ion itself. Compared to other compounds with larger particle size tested at similarly low concentrations, quantum dots caused much less pronounced toxicological effects. Therefore, the present data show that small particle sizes with corresponding high surfaces are not the only factor triggering the toxic response or translocation.

RESUMO

During sessions of physical therapy, a tetraplegic patient with incomplete spinal cord section at C5 level was found to have changes in the posture post intervention. The therapy consisted of tactile stimulation in specific points of his body to generate reflex patterns of muscle activation (Vojta therapy principies). To quantify these postural changes in the subject, the center of pressure displacement in sitting posture was measured. An analysis of the changes observed in postural control through fourteen months of follow-up is presented and discussed in the report bellow.


Durante las sesiones de terapia física, en un paciente tetrapléjico con sección incompleta de la médula espinal a nivel C5, se encontró que hubo cambios en la postura, posterior a la intervención. La terapia consistió en estimulación táctil en puntos específicos del cuerpo para generar patrones reflejos de activación muscular (principios de la terapia Vojta). Para cuantificar estos cambios de postura en el sujeto, se midió el desplazamiento del centro de gravedad en la posición sentada. Un análisis de las variaciones observadas en el control postural, a través de catorce meses de seguimiento, se presentan y discuten en el siguiente informe.
