RESUMO

In order to obtain the best mass spectrometry identification results for using the most appropriate methods in clinical practice, we explore the optimal pretreatment methods for different species and morphologies of filamentous fungi. 98 fungal strains were treated with formic acid sandwich method, dispersion method, extraction method, and other methods using a medium element mass spectrometer (EXS3000) as a platform. Each strain had three targets, and the identification rates and confidence differences under different pre-treatment methods were compared to evaluate the identification effects of these methods. The mass spectrometry identification rates of 98 filamentous fungi obtained after pre-treatment with formic acid sandwich method, dispersion method, and extraction method were 85.71%, 82.65%, and 75.51%, respectively. The identification rate of the formic acid sandwich method was significantly higher than the other two methods (P < 0 005) has the best identification ability and the obtained confidence is also higher than the other two methods. The use of formic acid sandwich method for mass spectrometry identification of filamentous fungi can achieve ideal identification results, which is suitable for mass spectrometry identification of filamentous fungi in conventional laboratories.

In deep penetration laser welding, the behavior of the keyhole has an important influence on the welding quality. As it is difficult to directly observe the keyhole and detect the pressure inside the keyhole during metal laser welding, theoretical analysis and numerical simulation methods are commonly used methods in studying keyhole behavior. However, these methods cannot provide direct real information on keyhole behavior. In this paper, a method of analogy welding is proposed, in which high speed gas is used to blow the liquid to generate the keyhole. Relevant process experiments were conducted to explore keyhole behavior in analogy welding and real deep penetration laser welding. The pressure balance of the keyhole, both in analogy welding and real deep penetration laser welding, were analyzed. The laws obtained in analogy welding and real deep penetration laser welding are similar, which indicates that studying keyhole formation and the maintenance principle using the analogy welding method proposed in this paper may be helpful for deep understanding of the keyhole formation and maintenance mechanisms in real deep penetration laser welding.

Pharmaceutical plants contain several phytochemicals that are sources of myriad biological activities. These biological activities can be explored in multiple fields for the benefit of mankind. Pharmaceutical plants with high ethnobotanical indices (i.e., use value and relative frequency of citation) were reported with the potential to inhibit lettuce elongation through leachates and volatiles. The focus of the study was to assess Chinese pharmaceutical plants for both antioxidants, as well as allelopathic potentials to explore any underlying relationship. The estimation of antioxidative capacity and content of total phenolics (TPC) for the 55 Chinese pharmaceutical plants was conducted by the assays of DPPH radical scavenging activity (DPPH-RSA), oxygen radical absorbance capacity (ORAC) and the means of Folin−Ciocalteu. The estimation of the activity of allelopathy for collected medicinal plants was done by adopting the sandwich method for plant leachates and the dishpack method for volatile constituents, respectively. The fruits of sea buckthorn (Hippophae rhamnoides) had the most remarkable ORAC value (168 ± 7.04 µmol TE/g) and DPPH radical scavenging activity (440 ± 7.32 µmol TE/g) and contained the highest contents of total phenolic compounds (236 ± 7.62 mg GAE/g) in the 55 pharmaceutical plant species according to the results. In addition, sea buckthorn showed dominant allelopathic potential through plant leachates evaluated by using the sandwich method. Star anise (Illicium verum Hook. f.) showed conspicuous allelopathic activity through plant volatiles assessed by the dishpack bioassay method. Among the same plant species, antioxidative ability and total phenolics, in comparison with potential allelopathy of medicinal herbs indicated that volatile allelochemical had a weak active effect (r = 0.407 to 0.472, p < 0.01), with antioxidant capacity by the dishpack method. However, the evaluation by the sandwich method showed a significant positive correlation (r = 0.718 to 0.809, p < 0.001) with antioxidant capacity. Based on these results, a new hypothesis is that the antioxidant activity of plants may have an involvement with the potential allelopathic activity.

AIM: To establish a highly sensitive time-resolved fluorescence immunoassay (TRFIA) of kidney injury molecule-1 (Kim-1) and evaluate its clinical value in acute kidney injury (AKI). METHODS: The Kim-1-TRFIA was established by the double-antibody sandwich method, and the method was evaluated. The established Kim-1-TRFIA was used to detect the concentration of Kim-1 in the serum of healthy controls and patients with AKI. RESULTS: The optimal coating antibody concentration and optimal Eu3+ -labeled antibody dilution ratio for Kim-1-TRFIA are 1 µg/ml and 1:140, respectively. The linear range is 42.71-4666.69 pg/ml. The intra- and inter-assay coefficients of variation are <10%. The specificity of our Kim-1-TRFIA is acceptable. The recovery is between 95.14% and 102.84%. The concentration of Kim-1 in the serum of patients with AKI is 126.50 ± 67.99 pg/ml, which is significantly higher than that in the serum of healthy controls (49.72 ± 16.40 pg/ml, p < 0.001). Staging patients with AKI by glomerular filtration rate shows that the serum concentration of Kim-1 increases significantly with increasing disease severity (p < 0.05). CONCLUSION: A highly sensitive Kim-1-TRFIA was established. With this immunoassay, a good differential diagnosis can be made, and healthy people and AKI patients can be differentiated by detecting the concentration of Kim-1 in the serum. Moreover, the severity of AKI patients can be determined.

To establish a rapid and highly sensitive assay for tumor-associated trypsinogen-2 (TAT-2) based on the time-resolved fluorescence immunoassay (TRFIA) and evaluate its potential clinical value in patients with lung cancer. The double-antibody sandwich method was used in detecting TAT-2 antigen concentrations, and two types of TAT-2 antibodies (coating antibodies and Eu3+ labeled antibodies) were used. A TAT-2-TRFIA method was then established, evaluated, and used in detecting the serum TAT-2 levels of healthy subjects and patients with lung cancer. The linear range of the TAT-2-TRFIA method was 1.53-300 ng/mL, the intra-assay coefficient of variation (CV) were between 1.67% and 8.42%, and the inter-assay CV were between 4.29% and 11.44%. The recovery rates of TAT-2-TRFIA were between 99.17% and 107.06%. The cross-reactivities of trypsin and T-cell immunoglobulin mucin 3 were 0.02% and 0.82%, respectively. The serum TAT-2 levels of patients with lung cancer were higher than those of healthy subjects (P < 0.001). Combined with TAT-2, the sensitivity and specificity of CEA and CA-125 for lung cancer improved significantly. Conclusion: We successfully established a highly sensitive TAT-2-TRFIA method, which was able to facilitate the timely diagnosis of lung cancer.

Unable to move, plants are physically restrained to the place where they grow. Remarkably, plants have developed a myriad of mechanisms to perceive the surrounding environment in order to maximize growth and survival. One of those mechanisms is the ability to perceive mechanical stimulus such as touch (thigmomorphogenesis), in order to adjust growth patterns (in different organs) to either attach to or surround an object. Roots are able to perceive several mechanical forces (e.g., gravity, touch). However, being the "hidden part" of a plant, it is difficult to assess their response to mechanical stimulation. In this chapter, our team presents a simple method to evaluate rice (Oryza sativa L.) root mechanosensing response that can be used to test different conditions (e.g., hormones) affecting rice root response to touch stimulus. This method is affordable to any lab and can be upgraded with a fully automated image recording system. We provide a detailed protocol with several notes for a more comprehensive application.

Since April 2021, the plasma aldosterone concentration has been measured by chemiluminescent enzyme immunoassay (CLEIA) in Japan. In the present study, we developed a new CLEIA using a two-step sandwich method to measure the 24-hour urine aldosterone level. We collected 115 urine samples and measured 24-hour urine aldosterone levels employing radioimmunoassay (RIA), CLEIA, and liquid chromatography-tandem mass spectrometry (LC-MS/MS). The results showed that the 24-hour urine aldosterone levels measured using CLEIA and LC-MS/MS were significantly correlated (ρ = 0.992, P < 0.0001). Based on the results of Passing-Bablok regression analysis, the slope was 0.992 and the intercept -19.3. The 24-hour urine aldosterone levels measured using CLEIA and RIA were also significantly correlated (ρ = 0.905, P < 0.0001). However, the aldosterone level measured by CLEIA was lower than that measured by RIA (slope, 0.729; intercept, 120.9). In Japan, a new guideline for primary aldosteronism has been announced, with changes in the aldosterone measurement method. The cutoff values for oral sodium loading test (OSLT) were changed, but clinical verification using real-world urine samples has not been performed. Therefore, we examined the cut-off value of the 24-hour urine aldosterone level after the OSLT. Receiver operating characteristic analysis revealed a cut-off value for primary aldosteronism of 3 µg/day.

The development of rapid and sensitive detection methods for pathogenic bacteria is crucial for the therapy and prevention of related diseases. However, the rapid and ultrasensitive assays are difficult to be realized simultaneously. To solve the problem, a sandwich method based on Ω-shaped fiber optic localized surface resonance (Ω-FOLSPR) was constructed, where poly adenine-tailed aptamer (PolyA-apt) and SH modified gold nanoparticles tags (AuNPs tags) were chosen as the capturing aptamer and amplifying tags, respectively. The small AuNPs were modified on the surface of fiber-optic (FO) rapidly, which saved the preparation time. Then, the PolyA-apt was modified on the AuNPs surface to capture the bacteria effectively due to its ability to adjust the density and conformation of aptamer on the AuNPs surface. Finally, the large AuNPs tags were used to generate intense signal enhancement. It is found that the sandwich method enables the unique characteristic of a time-dependent sensitivity enhancement. Specifically, the LOD of 108.0 CFU/mL and 7.4 CFU/mL was achieved with the analysis time of 10 min and 100 min, respectively. Besides, the Ω-FOLSPR sensor exhibits excellent selectivity against the other bacteria and good performance for detecting the spiked and natural samples. This sandwich method provides a time-flexible strategy due to the combination of effective signal amplification and real-time analysis for bacterial detection, displaying great potential for practical bacterial detection.

Objective:To systematically evaluate the efficacy and safety of tissue adhesive combined with lauromacrogol (modified Sandwich method) for gastric varices.Methods:Literature in the Cochrane Library, PubMed, EMbase, CNKI, VIP and Wanfang were searched by two independent researchers from the establishment of the databases to June 30, 2020, and qualified data from the eligible literature were extracted. Revman 5.3 was used to analyze outcomes including hemostatic efficiency, incidence of postoperative fever, chest and abdominal pain, ulcer, ectopic embolism and complications, and postoperative re-bleeding rate (Mantel-Haenszel method).Results:A total of 8 randomized controlled trials including 898 patients were included in this meta-analysis. The results showed that compared with the classic Sandwich method, the modified version had a better hemostatic effect ( P=0.01, OR=2.07, 95% CI: 1.17-3.68) and a lower incidence of postoperative ectopic embolism ( P=0.001, OR=0.06, 95% CI: 0.01-0.34). There were no significant differences in the incidences of postoperative fever ( P=0.58, OR=0.86, 95% CI: 0.52-1.44), chest and abdominal pain ( P=0.83, OR=0.95, 95% CI: 0.58-1.56), local ulcer ( P=0.31, OR=0.66, 95% CI: 0.30-1.47) , re-bleeding ( P=0.14, OR=0.76, 95% CI: 0.53-1.09) or overall incidence of adverse reactions ( P=0.24, OR=0.66, 95% CI: 0.33-1.32). Conclusion:The modified Sandwich method of tissue adhesive combined with lauromacrogol is an effective and safe method in the treatment of gastric varices.

Deaths due to invasive fungal disease (IFD) have been increasing every year. Early and rapid detection is important to reduce the mortality rate associated with IFD. In this study, we explored a novel diagnostic method for detecting IFD, which involves the G Factor α subunit (GFαSub) from Limulus polyphemus. The GFαSub double-sandwich method was developed to detect (1,3)-ß-D-glucans in human serum using purified GFαSub and horseradish peroxidase-labeled GFαSub. The GFαSub double-sandwich method and the G test were performed and compared. Using GFαSub sequence analysis, the expression plasmid pET30a-GFαSub252-668 was synthesized, and GFαSub252-668 was expressed and purified via isopropyl-ß-d-thiogalactoside induction and nickel-nitrilotriacetic acid affinity. The optimization method was established via the orthogonal method. Using this method, the sera of 36 patients with IFD and 92 volunteers without IFD underwent detection, and the receiver operating characteristic curve of the GFαSub252-668 double-sandwich method was described. The sensitivity and specificity of the GFαSub252-668 double-sandwich method were 91.67 and 82.61%, respectively, and there was good correlation with the G test for the serum specimens of 36 patients with pulmonary IFD (R 2 = 0.7592). In conclusion, our study suggests that the GFαSub252-668 double-sandwich method was satisfactory at detecting IFD cases. This method can be promoted and further developed as a novel method for diagnosing IFD.

In the present study, we developed a new chemiluminescent enzyme immunoassay (CLEIA) using a two-step sandwich method to measure aldosterone concentrations. We investigated serum and plasma aldosterone concentrations in 75 blood samples from 27 patients using a radioimmunoassay (RIA) and the CLEIA (with current and newly improved reagents) as well as liquid chromatography-tandem mass spectrometry (LC-MS/MS). Based on the results of the Passing-Bablok regression analysis, the aldosterone levels measured using CLEIA with the new reagents and those measured by LC-MS/MS were found to be significantly correlated (slope, 0.984; intercept, 0.2). However, aldosterone levels varied depending on the measurement method (i.e., CLEIA with the new reagent, CLEIA with the current reagent, and RIA). Aldosterone levels were lower with the improved CLEIA method than with RIA and CLEIA using the current reagent. Therefore, the cutoff values of the screening test as well as those of the confirmatory test for primary aldosteronism (PA) should be adjusted to follow current clinical practice guidelines for PA. The formula that can be used to obtain the aldosterone level (pg/mL) when using CLEIA with the new reagent is 0.765 × RIA (pg/mL) - 33.7. This formula will enable PA cutoff values to be set for provisional screening and confirmatory tests.

Gluten is an insoluble protein produced when glutelins and prolamins, which are found in grains such as wheat, barley, and oats, combine to form an elastic thin film. This dietary gluten can cause severe contraction of the intestinal mucous membrane in some people, preventing nutrient absorption. This condition, called celiac disease (CD), affects approximately 1% of the world's population. The only current treatment for patients with CD and similar diseases is lifelong avoidance of gluten. To analyze the gluten content in food, various enzyme-linked immunosorbent assay (ELISA) tests are currently used. In this study, the gluten content in various food products was analyzed using different kinds of ELISA test kits. For gluten-free food, three different ELISA test kits mostly yielded values below the limit of detection. However, gluten was detected at 24.0-40.2 g/kg in bread, 6.5-72.6 g/kg in noodles, and 23.0-86.9 g/kg in different powder food samples. A significant difference (p < 0.05) in gluten content was observed for these gluten-containing food products. Reproducibility issues suggest that it is necessary to use several ELISA kits for the accurate detection and quantification of gluten in various food products rather than using one ELISA kit.

Electron crystallography is a unique tool to study membrane protein structures and lipid-protein interactions in their native-like environments. Two-dimensional (2D) protein crystallization enables the lipids immobilized by the proteins, and the generated high-resolution density map allows us to model the atomic coordinates of the surrounding lipids to study lipid-protein interaction. This protocol describes the sample preparation for electron crystallographic studies, including back-injection method and carbon sandwich method. The protocols of data collection for electron crystallography, including electron imaging and diffraction, of the 2D membrane crystal will be followed.

To date, how to select an appropriate strategy to treat advanced prostate cancer (PCa) patients with tumor progression after abiraterone acetate (AA) intervention is still confusing. Here, we conducted a case report and review of the literatures focused on this issue. A 63-year-old man presented initially with dysuria. Multiple pelvic bone metastases but no other lesions were detected. He was subsequently diagnosed with PCa after biopsies, the initial clinical staging was cT2cNxM1. This patient was treated with two periods of AA (first treatment: 16-month; secondary treatment: 19-month) combined with other therapeutic regimens [i.e., goserelin acetate, bicalutamide, docetaxel, and radical prostatectomy (RP), etc.], followed by a significant biochemical response during over 5 years of follow-up. In the present case report with 63 months of the treatment course, we found that patient with metastatic PCa who early received AA as the initial regimen could gain benefit from the retreatment of AA: minor progression and gain long-time survival. This "sandwich method" with AA administered initially, withdrawal, and sequentially following Bicalutamide, Docetaxel, RP, and retreatment of AA may prolong the response time of AA retreatment and delay the progression of metastatic PCa, which reminds clinicians should pay attention to this phenomenon and explore the underlying mechanisms.

This study focused on the potential allelopathy of 50 species of Chinese medicinal plants, which are mainly distributed in the Xinjiang Uyghur Autonomous Region, Inner Mongolia, and Yunnan Province. The "sandwich method" was adopted and used for the screening for allelopathic potential among these plant species. Further phytotoxic evaluation of the candidate species was conducted by applying plant extracts to crops and weed species. The results of this study indicated that among the 50 medicinal plant species evaluated, the fruits of Illicium verum Hook. f. (star anise) showed the most significant allelopathic potential through the leaf leachates. Shikimic acid was identified to be the main bioactive compound (about 7% dry weight) in star anise by reversed-phase High Performance Liquid Chromatography (RP-HPLC) analysis. The phytotoxic bioassay indicated that both the crude extract of the Chinese star anise and the synthetic shikimic acid showed strong inhibitory activity on the radicle and hypocotyl growth of lettuce. The radicle growth inhibition of lettuce caused by the crude extract of star anise could be explained by the contribution of the biological activity of shikimic acid. In conclusion, shikimic acid could be a putative allelochemical in the fruits of Illicium verum and could be utilized in sustainable weed management.

The inhibitory activities of the leachates and volatiles from 53 plant species (spices and herbs) were evaluated against lettuce (Lactuca sativa "Great Lakes 366") seedling growth using the sandwich and dish pack methods, respectively. With the sandwich method, parsley (Petroselinum sativum) showed the strongest inhibitory effect on lettuce radicle growth (77%), followed by tarragon (Artemisia dracunculus) (72%). However, caraway (Carum carvi), dill (Anethum graveolens) (seed), laurel (Laurus nobilis), rosemary (Rosmarinus officinalis), and sage (Salvia officinalis) were the most inhibitory species (100% inhibition of lettuce radicle and hypocotyl growth inhibition at all distance wells) in the dish pack method. Cardamom (Elettaria cardamomum) and thyme (Thymus vulgaris) also showed strong inhibitory activity (100% for radicle and hypocotyl growth inhibition at all 41 and 58 mm distance wells). The headspace sampling and gas chromatography-mass spectrometry (GC-MS) analysis identified the main inhibitory active compounds as carvone in caraway and dill (seeds), 1,8-cineole in laurel and cardamom, and borneol in thyme. Both camphor and 1,8-cineole were detected in rosemary and sage, and the total activity evaluation showed that camphor was the major inhibitory compound in rosemary, although both compounds played equal roles in sage.

Introduction: Use of allelochemicals as inexpensive, biodegradable and eco-friendly substitutes for the management of weeds in crops is a central topic nowadays. Objective: The current study represented the comprehensive screening of allelopathic activity of 73 weeds in Pakistan by the Sandwich method and dish pack method. To investigate the allelochemical compound in top most allelopathic weed. Methods: Allelopathic effects of selected weeds leaves leachate were determined through sandwich technique, while volatile allelopathic effects through dish pack techniques. Qualitative and quantitative phytochemical techniques were applied to investigate allelochemical potential of Melilotus indicus. The statistical analysis of the data described the allelopathic effect of 73 weed plants on lettuce seedling growth in terms of radicle and hypocotyl elongation. Results: Elongation percentage of radicle and hypocotyl ranged 0-74 % to 0-148 % and 0-75 % to 0-84 % respectively at 10 mg concentration of dry plant powder in sandwich method while it was 2-234 % and 7-150 % at in Dish Pack method. Among the plants screened for phytotoxic activity, M. indicus contained the strongest allelochemicals. Qualitative and quantitative analysis of M. indicus showed the presence of flavonoids and phenolic compound along with other allelochemical. Conclusions: Selected weeds may have strong allelochemical potential that can help in the development of bioactive compounds from plant species to be used as natural herbicides and pesticides for sustainable management of weeds and pest.

Objetivo: En el presente estudio se midió la actividad alelopática de 73 hierbas en Pakistán a través de los métodos "sandwich" y "dish pack", con el fin de investigar los compuestos aleloquímicos más importantes en las hierbas analizadas. Métodos: A través del método "sandwich" se determinaron los efectos alelopáticos de los lixiviados de hojas en las hierbas analizadas, mientras que el efecto alelopático volátil a través de la técnica "dish pack". Para investigar el potencial aleloquímico de Melilotus indicus se aplicaron técnicas fitoquímicas tanto cualitativas como cuantitativas. El análisis estadístico de los datos describió el efecto alelopático de 73 especies de hierbas sobre el crecimiento radicular y del hipocótilo en plántulas de lechuga. Resultados: El porcentaje de elongación radicular y del hipocotilo varió entre 0-74 % hasta 0-148 % y 0-75 % hasta 0-84 % respectivamente, con una concentración de 10 mg de polvo de planta seca con el método "sandwich". Además, fue entre 2-234 % y 7-150 % con el método "dish pack". Entre las plantas analizadas para actividad fitotóxica, M. indicus presentó los aleloquímicos más fuertes. Los análisis cualitativos y cuantitativos de M. indicus mostraron la presencia de flavonoides y compuestos fenólicos, junto con otros aleloquímicos. Conclusiones: Las hierbas seleccionadas pueden tener un potencial aleloquímico fuerte que ayude al desarrollo de compuestos bioactivos de plantas, para usar como hierbicidas y pesticidas naturales en el manejo sostenible de plagas y malas hierbas.

Turkey has one of the richest plant diversities in the Mediterranean region. In the current literature, no broad screening has been conducted on the potential allelopathy of plants from Turkey. This study aimed to evaluate the allelopathic activity of a large number of plants from Turkey for the first time and to determine the species with significant plant growth inhibitory potentials by bioassay. Dried samples of different plant parts were collected from local herbalists. The sandwich method was used to evaluate the potential allelopathy of 126 medicinal plants belonging to 55 families. The results of lettuce radicle and hypocotyl growth for 10 and 50 mg sample treatment conformed to normal distribution. Significant inhibition on lettuce radicle elongation with 10 mg sample was observed in 40 species, out of which 27 species showed over 50% inhibitory activity. The results suggested that these species could contain potential inhibitory compounds against lettuce radicle or hypocotyl growth. The calyxes of Hibiscus sabdariffa (3.2% of control) and the seeds of Prunus dulcis (5.7% of control) showed the most potent growth inhibitory activity on lettuce radicle elongation. The potential plant growth inhibitory effects of these plants, together with the fruits of Rhus coriaria and seeds of Prunus mahaleb, have been reported in this study for the first time. All these plants are medicinal, and the results hereby presented provide essential information about the allelopathic effects of medicinal plants from Turkey.

To improve viral antibody detection and disease control, laboratories need faster and simpler methods for direct detection of H9AIV in clinical samples. In this study, test strips were developed for rapid detection of H9AIV antibodies in poultry serum by a sandwich method with double-line detection. The hemagglutinin protein was labeled with colloidal gold as a detection reagent and was blotted on the test lines, while goat anti-rabbit IgG was utilized on the control line of the nitrocellulose membrane. The test strips have high specificity, sensitivity, and stability, with a correlation coefficient of 0.9656 and coefficients of variation below 10%. Application of the kit to quantitative detection of H9AIV antibodies in 504 samples collected from chickens showed a coincidence rate of 80.56% with previously run HI assays.

Objective:To report surgical techniques, outcomes, clinical experience and application key point of transcanal endoscopic ear surgery for tympanosclerosis. Method: Data from 25 patients (13 males and 12 females; aged 27-61 years, mean=48.8 years) who underwent transcanal endoscopic ear surgery for tympanosclerosis in our department from January 2016 to June 2017 were retrospectively analyzed. Removal of tympanosclerosis and tympanoplasty were individualized. Patients were followed up at postoperative 1, 3 and 6 month. Result: There were no failure of tympanal reconstruction by sandwich. The mean preoperative air conduction threshold was higher than that of postoperation ï¼»(51.12±16.03)dB HL and (31.64±12.76)dB HL, respectivelyï¼½(P<0.01). The mean preoperative air-bone gap was significantly increased compared to that of postoperation ï¼»(30.33±10.12)dB and (14.19±8.97)dB,respectivelyï¼½ (P<0.01). There were no complication, recurrence, residual tympanosclerosis or secondary surgery needed. Conclusion: Stand alone transcanal endoscopic ear surgery was a safe and effective method for tympanosclerosis.
