RESUMO

The extent of surgical resection is an important prognostic factor in the treatment of patients with glioblastoma. Optical coherence tomography (OCT) imaging is one of the adjunctive methods available to achieve the maximal surgical resection. In this study, the tumor margins were visualized with the OCT image obtained from a murine glioma model. A commercialized human glioblastoma cell line (U-87) was employed to develop the orthotopic murine glioma model. A swept-source OCT (SS-OCT) system of 1300 nm was used for three-dimensional imaging. Based on the OCT intensity signal, which was obtained via accumulation of each A-scan data, an en-face optical attenuation coefficient (OAC) map was drawn. Due to the limited working distance of the focused beam, OAC values decrease with depth, and using the OAC difference in the superficial area was chosen to outline the tumor boundary, presenting a challenge in analyzing the tumor margin along the depth direction. To overcome this and enable three-dimensional tumor margin detection, we converted the en-face OAC map into an en-face difference map with x- and y-directions and computed the normalized absolute difference (NAD) at each depth to construct a volumetric NAD map, which was compared with the corresponding H&E-stained image. The proposed method successfully revealed the tumor margin along the peripheral boundaries as well as the margin depth. We believe this method can serve as a useful adjunct in glioma surgery, with further studies necessary for real-world practical applications.

Mitogen-activated protein kinase (MAPK) pathways control the response to intrinsic and extrinsic stimuli. In the budding yeast Saccharomyces cerevisiae, cells undergo filamentous growth, which is regulated by the fMAPK pathway. To better understand the regulation of the fMAPK pathway, a genetic screen was performed to identify spontaneous mutants with elevated activity of an fMAPK pathway-dependent growth reporter (ste4  FUS1-HIS3). In total, 159 mutants were isolated and analyzed by secondary screens for invasive growth by the plate-washing assay and filament formation by microscopy. Thirty-two mutants were selected for whole-genome sequencing, which identified new alleles in genes encoding known regulators of the fMAPK pathway. These included gain-of-function alleles in STE11, which encodes the MAPKKK, as well as loss-of-function alleles in KSS1, which encodes the MAP kinase, and loss-of-function alleles in RGA1, which encodes a GTPase-activating protein (GAP) for CDC42. New alleles in previously identified pathway modulators were also uncovered in ALY1, AIM44, RCK2, IRA2, REG1, and in genes that regulate protein folding (KAR2), glycosylation (MNN4), and turnover (BLM10). Mutations leading to C-terminal truncations in the transcription factor Ste12p were also uncovered that resulted in elevated reporter activity, identifying an inhibitory domain of the protein from residues 491 to 688. We also find that a diversity of filamentous growth phenotypes can result from combinatorial effects of multiple mutations and by loss of different regulators of the response. The alleles identified here expand the connections surrounding MAPK pathway regulation and reveal new features of proteins that function in the signaling cascade.

EEG signals are valuable signals in clinical medicine, brain research, and the study of neurological illnesses. However, EEG signal attenuation may occur at any time from signal generation through BCI device acquisition due to defects in the brain-computer interface (BCI) devices, restrictions in the dynamic network, and individual variations across the subjects. The attenuation of EEG data will alter the data distribution and lead to information fuzziness, substantially influencing subsequent EEG research. A model based on one-dimensional residual convolutional neural networks (1D-ResCNN) and transfer learning is proposed in this article to reduce the negative impacts of EEG attenuation. An end-to-end manner maps an attenuated EEG signal to a normal EEG signal. The structure employs a multi-level residual connection structure with varying weight coefficients, transferring characteristics from the bottom to the top of the convolutional neural network, enhancing feature learning. In addition, we initialize the subsequent denoising model using the transfer learning method. The combination of these two networks can well solve the attenuation problem of EEG signals. Experiments are carried out using the EEG-denoisenet data set. According to the findings, the model can yield a clear waveform with a decent SNR and RRMSE value.

Treatment with dopamine agonists in Parkinson's disease (PD) is associated with debilitating neuropsychiatric side-effects characterized by impulsive and compulsive behaviors. The vulnerability to develop such impairments is thought to involve interactions between individual vulnerability traits, types of antiparkinsonian medications, and the neurodegenerative process. We investigated the effect of the dopamine D3/D2 agonist pramipexole (PPX) and selective nigrostriatal degeneration achieved by viral-mediated expression of alpha-synuclein on the expression of repetitive and compulsive-like behaviors in rats. In a task assessing spontaneous food hoarding behavior, PPX increased the time spent interacting with food pellets at the expense of hoarding. This disruption of hoarding behavior was identical in sham and lesioned rats. In an operant post-training signal attenuation task, the combination of nigrostriatal lesion and PPX decreased the number of completed trials and increased the number of uncompleted trials. The lesion led to an increased compulsive behavior after signal attenuation, and PPX shifted the overall behavioral output towards an increased proportion of compulsive lever-presses. Given the magnitude of the behavioral effects and the lack of strong interaction between PPX and nigral degeneration, these results suggest that extra-nigral pathology may be critical to increase the vulnerability to develop compulsive behaviors following treatment with D3/D2 agonists.

This study aimed to compare the radiofrequency (RF) shielding effects of titanium mesh of echo-planar imaging (EPI) versus fast spin-echo (FSE) diffusion-weighted imaging (DWI) to establish a suitable sequence for patients who undergo cranioplasty and for whom titanium mesh was used in brain magnetic resonance imaging (MRI). A 1.5-T MRI scanner with clinical setting sequences was used. A phantom for the examination constructed using a sucrose solution in a plastic container was used to compare the signal attenuation (SA) ratio, area of RF shielding effect (Area), normalized absolute average deviation (NAAD), and apparent diffusion coefficient (ADC) between EPI and FSE-DWI. EPI provided significantly better SA ratio, Area, and NAAD (P < 0.01). When the number of slices increased, the RF shielding became more negative. There was no significant difference in the ADC. Regardless of the k-trajectory, EPI-DWI had a lower RF shielding effect than FSE-DWI in patients undergoing cranioplasty.

PURPOSE: To determine which sequence for frequently used general contrast-enhanced brain MRI shows the least radiofrequency shielding effect of a titanium mesh in cranioplasty using a phantom. METHODS: A 1.5T MRI scanner was used. Frequently used general 2D and 3D spin-echo sequences (SE) and T1 spoiled gradient echo sequences (GRE) used for MRI in clinical settings were adopted in this study. A titanium mesh was placed above a cubic phantom containing manganese chloride tetrahydrate and sodium chloride. The signal attenuation ratio and normalized absolute average deviation (NAAD) were calculated. Moreover, the flip angle (FA) dependency in SE and area of excitation dependency in 3D sequences were analyzed using NAAD. RESULTS: The signal attenuation ratio at the position nearest to the titanium mesh for 2D SE was 71.8% larger than that at the position nearest to the titanium mesh for 3D GRE. With regard to NAAD, 3D GRE showed the highest values among the sequences. When FA was increased, radiofrequency shielding effect was improved. There were no significant differences between the narrow and wide area of excitation. 3D GRE showed the least radiofrequency shielding effect, and it was considered as the optimal sequence for MRI in the presence of a titanium mesh. CONCLUSION: 3D GRE shows the least radiofrequency shielding effect of a titanium mesh after cranioplasty among frequently used general sequences for contrast-enhanced brain MRI.

The specific mechanisms underlying compulsive behavior in obsessive-compulsive disorder (OCD) are unknown. It has been suggested that such compulsivity may have its origin in cognitive dysfunction such as impaired processing of feedback information, received after the completion of goal-directed actions. The signal attenuation (SA) task models such a processing deficit in animals by attenuating the association strength between food reward and audiovisual feedback (signal) presented after performance of an operant response. The compulsive-like responding resulting from SA is well characterized in rats, but was so far not established in mice, a species for which powerful genetic OCD models exist. Thus, first, we demonstrate that the SA task can be implemented in mice and show that attenuation of reward-associated response feedback produces similar behavior in C57BL/6 mice as previously reported in rats. Second, we tested the hypothesis that SAPAP3 knock-out mice (SAPAP3-/-), prone to exhibit several OCD-like abnormalities including excessive grooming, show enhanced compulsive-like behavior in the SA task compared with their wild-type (WT) littermates. However, task-related compulsivity measures in SAPAP3-/- and WT did not yield significant differences, neither following SA nor during "regular" extinction of operant behavior. Thus, compulsive-like instrumental behavior following feedback distortion was not potentiated in compulsively grooming mice, implicating specifically that (1) a general deficit in feedback processing is not related to excessive grooming in SAPAP3-/- and (2) different manifestations of compulsivity may be driven by independent mechanisms.

Among the current indoor positioning technologies, Bluetooth low energy (BLE) has gained increasing attention. In particular, the traditional distance estimation derived from aggregate RSS and signal-attenuation models is generally unstable because of the complicated interference in indoor environments. To improve the adaptability and robustness of the BLE positioning system, we propose making full use of the three separate channels of BLE instead of their combination, which has generally been used before. In the first step, three signal-attenuation models are separately established for each BLE advertising channel in the offline phase, and a more stable distance in the online phase can be acquired by assembling measurements from all three channels with the distance decision strategy. Subsequently, a weighted trilateration method with uncertainties related to the distances derived in the first step is proposed to determine the user's optimal position. The test results demonstrate that our proposed algorithm for determining the distance error achieves a value of less than 2.2 m at 90%, while for the positioning error, it achieves a value of less than 2.4 m at 90%. Compared with the traditional methods, the positioning error of our method is reduced by 33% to 38% for different smartphones and scenarios.

This paper presents and applies an inductive directional coupling technology based on spread spectrum time domain reflectometry (SSTDR) for non-intrusive power cable fault diagnosis. Different from existing capacitive coupling approaches with large signal attenuation, an inductive coupling approach with a capacitive trapper is proposed to restrict the detection signal from transmitting to power source and to eliminate the effect of the power source impedance mismatch. The development, analysis, and implementation of the proposed approach are discussed in detail. A series of simulations and experiments on cables with different fault modes are conducted, along with comparison of existing capacitive coupling, to verify and demonstrate the effectiveness of the proposed method.

Opening a chest in an experimental echocardiographic animal study eliminates ultrasound signal attenuation by the chest wall. We developed a scanning technique that involves the use of an attenuative pad created from a mixture of urethane and titanium dioxide. The pad was interposed within transmission gel between the transducer face and cardiac surface in open-chest scans in a porcine model. Comparative measurements of left ventricular echogenicity without and with the pad demonstrate that the pad reproducibly causes ultrasound signal attenuation that closely mimics chest attenuation in clinical transthoracic echocardiographic studies.

Abstract Introduction This study aims to assess the influence of different skinfold thicknesses (ST) and their relation to the attenuation of the mechanomyographic (MMG) signal at different force levels (maximal voluntary contraction – MVC, 40% of MVC and 70% of MVC) of the rectus femoris muscle. Methods Fifteen volunteers were divided in two groups: ST lower than 10mm (G<10) (8 participants) and ST higher than 35mm (G>35) (7 participants). Student t tests were employed to investigate differences between G<10 and G>35 regarding MMG analysis parameters (acceleration root mean square – aRMS, zero crossing – ZC, and median frequency – MDF), for the X, Y and Z axes, as well as for the modulus of these three axes. Results We found that thicker layers of body fat act as attenuator filters for the MMG signal [MDFMVC: X (p = 0.005), Z (p = 0.003); MDF70%MVC: X (p = 0.034); ZCMVC: Z (p = 0.037), modulus (p = 0.005); ZC70%MVC: Z (p = 0.047)]. We found significant correlation between ST values and aRMS in three levels, in the Yaxis (p = 0.591), for the group G<10. For the modulus, the aRMS value showed correlation with ST values for group G>35 in 40%MVC (R2 = 0.610), and 70%MVC (R2 = 0.592). The MDF parameter showed correlation with ST values only in the Yaxis in 70%MVC (R2 = 0.700) for G>35. Conclusions We observed MMG signal attenuation in at least one of the parameters analyzed for each level of the rectus femoris muscle force, indicating that MMG signals are significantly attenuated with increasing thickness of the subcutaneous fat layer.

The aim of this study was to evaluate the signal intensity and signal attenuation of swept source optical coherence tomography (SS-OCT) for dental caries in relation to the variation of mineral density. SS-OCT observation was performed on the enamel and dentin artificial demineralization and on natural caries. The artificial caries model on enamel and dentin surfaces was created using Streptococcus mutans biofilms incubated in an oral biofilm reactor. The lesions were centrally cross sectioned and SS-OCT scans were obtained in two directions to construct a three-dimensional data set, from the lesion surface (sagittal scan) and parallel to the lesion surface (horizontal scan). The integrated signal up to [Formula: see text] in depth (IS200) and the attenuation coefficient ([Formula: see text]) of the enamel and dentin lesions were calculated from the SS-OCT signal in horizontal scans at five locations of lesion depth. The values were compared with the mineral density obtained from transverse microradiography. Both enamel and dentin demineralization showed significantly higher IS200 and [Formula: see text] than the sound tooth substrate from the sagittal scan. Enamel demineralization showed significantly higher IS200 than sound enamel, even with low levels of demineralization. In demineralized dentin, the [Formula: see text] from the horizontal scan consistently trended downward compared to the sound dentin.

Translational molecular dynamics in various materials can also be studied by diffusion spectra. These can be measured by a constant gradient variant of the modulated gradient spin echo (MGSE) sequence which is composed of a CPMG RF pulse train superimposed to a constant magnetic field gradient. The application of the RF train makes the effective gradient oscillating thus enabling measurements of diffusion spectra in a wide range of frequencies. However, seemingly straightforward implementation of the MGSE sequence proved to be complicated and can give overestimated results for diffusion if not interpreted correctly. In this study, unrestricted diffusion in water and other characteristic materials was analyzed by the MGSE sequence in the frequency range 50-3000Hz using a 6T/m diffusion probe. First, it was shown that the MGSE echo train acquired from the entire sample decays faster than the train acquired only from a narrow band at zero frequency of the sample. Then, it was shown that the decay rate is dependent on the band's off-resonance characterized by the ratio Δω0/ω1 and that with higher off-resonances the decay is faster. The faster decay therefore corresponds to a higher diffusion coefficient if the diffusion is calculated using standard Stejskal-Tanner formula. The result can be explained by complex coherence pathways contributing to the MGSE echo signals when |Δω0|/ω1>0. In a magnetic field gradient, all the pathways are more diffusion attenuated than the direct coherence pathway and therefore decay faster, which leads to an overestimation of the diffusion coefficient. A solution to this problem was found in an efficient off-resonance signal reduction by using only zero frequency filtered MGSE echo train signals.

As the weak link in global navigation satellite system (GNSS) signal processing, the phase-locked loop (PLL) is easily influenced with frequent cycle slips and loss of lock as a result of higher vehicle dynamics and lower signal-to-noise ratios. With inertial navigation system (INS) aid, PLLs' tracking performance can be improved. However, for harsh environments with high dynamics and signal attenuation, the traditional INS-aided PLL with fixed loop parameters has some limitations to improve the tracking adaptability. In this paper, an adaptive INS-aided PLL capable of adjusting its noise bandwidth and coherent integration time has been proposed. Through theoretical analysis, the relation between INS-aided PLL phase tracking error and carrier to noise density ratio (C/N0), vehicle dynamics, aiding information update time, noise bandwidth, and coherent integration time has been built. The relation formulae are used to choose the optimal integration time and bandwidth for a given application under the minimum tracking error criterion. Software and hardware simulation results verify the correctness of the theoretical analysis, and demonstrate that the adaptive tracking method can effectively improve the PLL tracking ability and integrated GNSS/INS navigation performance. For harsh environments, the tracking sensitivity is increased by 3 to 5 dB, velocity errors are decreased by 36% to 50% and position errors are decreased by 6% to 24% when compared with other INS-aided PLL methods.

Remote measurement of product core temperature is an important prerequisite to improve the cool chain of food products and reduce losses. This paper examines and shows possible solutions to technical challenges that still hinder practical applications of wireless sensor networks in the field of food transport supervision. The high signal attenuation by water-containing products limits the communication range to less than 0.5 m for the commonly used 2.4 GHz radio chips. By theoretical analysis of the dependency of signal attenuation on the operating frequency, we show that the signal attenuation can be largely reduced by the use of 433 MHz or 866 MHz devices, but forwarding of messages over multiple hops inside a sensor network is mostly unavoidable to guarantee full coverage of a packed container. Communication protocols have to provide compatibility with widely accepted standards for integration into the global Internet, which has been achieved by programming an implementation of the constrained application protocol for wireless sensor nodes and integrating into IPv6-based networks. The sensor's battery lifetime can be extended by optimizing communication protocols and by in-network pre-processing of the sensor data. The feasibility of remote freight supervision was demonstrated by our full-scale 'Intelligent Container' prototype.

OBJECTIVES: To obtain perfusion as well as diffusion information in normal breast tissues and breast lesions from intravoxel incoherent motion (IVIM) imaging with biexponential analysis of multiple b-value diffusion-weighted imaging (DWI) and compare these parameters to apparent diffusion coefficient (ADC) obtained with monoexponential analysis in their ability to discriminate benign lesions and malignant tumors. MATERIALS AND METHODS: In this prospective study, informed consent was acquired from all patients. Eighty-four patients with 40 malignant tumors, 41 benign lesions, 30 simple cysts and 39 normal breast tissues were imaged at 1.5 T utilizing contrast-enhanced magnetic resonance imaging (MRI) and DWI using 12 b values (range: 0-1000 s/mm(2)). Tissue diffusivity (D), perfusion fraction (f) and pseudo-diffusion coefficient (D*) were calculated using segmented biexponential analysis. ADC (b = 0 and 1000 s/mm(2)) was calculated with monoexponential fitting of the DWI data. D, f, D* and ADC values were obtained for normal breast tissues, simple cysts, benign lesions and malignant tumors. Receiver operating characteristic analysis was performed for all DWI parameters. RESULTS: There was good interobserver agreement on the measurements between the 2 observers. D values were significantly different among malignant tumors, benign lesions, simple cysts and normal breast tissues (P = 0.000) and it was the same result for f, D* and ADC values. Further comparisons of these 4 parameters between every single pair were as the following. D and ADC values of malignant tumors were significantly smaller than those of benign lesions, simple cysts and normal tissues (P = 0.000, respectively). The f value of malignant tumors was significantly higher than that of benign lesions, simple cysts and normal breast tissues (P = 0.001, P = 0.000, and P = 0.000). D and ADC values demonstrated higher sensitivity and specificity in differentiating benign lesions and malignant tumors, with area under the curve (AUC) of 0.952 and 0.945, respectively, while f and D* with the lower AUC of 0.723 and 0.630, respectively. Combining f and D values had a sensitivity up to 98.75%. CONCLUSION: DWI response curves in malignant tumors, benign lesions and normal fibroglandular tissues are found to be biexponential fit in comparison with the monoexponential fit for simple cysts. IVIM provides separate quantitative measurement of D for cellularity and f and D* for vascularity and is helpful for differentiation between benign and malignant breast lesions.

RESUMO

The zebrafish is a well-established model system used to study and understand various human biological processes. The present study used OCT to investigate growth of the adult zebrafish brain. Twenty zebrafish were studied, using their standard lengths as indicators of their age. Zebrafish brain aging was evaluated by analyzing signal attenuation rates and texture features in regions of interest (ROIs). Optical scattering originates from light interaction with biological structures. During development, the zebrafish brain gains cells. Signal attenuation rate, therefore, increases with increasing zebrafish brain age. This study's analyses of texture features could not identify aging in zebrafish brain. These results, therefore, indicated that the OCT signal attenuation rate can indicate zebrafish brain aging, and its analysis provides a more effective means of observing zebrafish brain aging than texture features analysis. Using OCT system could further increase the technique's potential for recognition and monitoring of zebrafish brain development.
