Herbicides have been used to control Brachiaria grass in pastures established or in formation given their practicality, however their efficiency is questionable due to the lack of specific graminicides for different forage species. Therefore, the goal of this study was to evaluate the efficiency of glyphosate and fluazifop-p-butyl in the control of Brachiaria decumbens (signalgrass) and the intoxication levels of Panicum maximum cv. Tanzania (Tanzania quinea grass) in pasture establishment. The experiments were designed in randomized blocks, in a 2 × 5 × 4 factorial arrangement of two herbicides (fluazifop-p-butyl and glyphosate), five doses equivalent to the commercial dosage of each herbicide (0.25; 0.50; 1.00; 1.50; 200), and four evaluation times after herbicide application (15, 21, 30 and 45 days). There was interaction between doses and evaluation times. The dose 1.5 L ha-1 fluazifop-p-butyl provides efficient control of signalgrass, however, leads to high intoxication in Tanzania guinea grass. Glyphosate is efficient in the control of signalgrass even at the lowest dose (90 g ha-1 ), however, it causes high intoxication in Tanzania guinea grass, preventing its use in developing pastures. It can be concluded that fluazifop-p-butyl and glyphosate herbicides are not recommended for the control of B. decumbens, cv. Basilisk in developing pastures of Tanzania guinea grass.(AU)

Urochloa decumbens (Stapf) R. D. Webster is one of the most important African forage grasses in Brazilian beef production. Currently available genetic-genomic resources for this species are restricted mainly due to polyploidy and apomixis. Therefore, crucial genomic-molecular studies such as the construction of genetic maps and the mapping of quantitative trait loci (QTLs) are very challenging and consequently affect the advancement of molecular breeding. The objectives of this work were to (i) construct an integrated U. decumbens genetic map for a full-sibling progeny using GBS-based markers with allele dosage information, (ii) detect QTLs for spittlebug (Notozulia entreriana) resistance, and (iii) seek putative candidate genes involved in defense against biotic stresses. We used the Setaria viridis genome a reference to align GBS reads and selected 4,240 high-quality SNP markers with allele dosage information. Of these markers, 1,000 were distributed throughout nine homologous groups with a cumulative map length of 1,335.09 cM and an average marker density of 1.33 cM. We detected QTLs for resistance to spittlebug, an important pasture insect pest, that explained between 4.66 and 6.24% of the phenotypic variation. These QTLs are in regions containing putative candidate genes related to defense against biotic stresses. Because this is the first genetic map with SNP autotetraploid dosage data and QTL detection in U. decumbens, it will be useful for future evolutionary studies, genome assembly, and other QTL analyses in Urochloa spp. Moreover, the results might facilitate the isolation of spittlebug-related candidate genes and help clarify the mechanism of spittlebug resistance. These approaches will improve selection efficiency and accuracy in U. decumbens molecular breeding and shorten the breeding cycle.

Herbicides with long residual period may increase the risk of environmental contamination. Adequate management of forage can reduce the half-life of the picloram, one of the most herbicides used in weed control. This study aims to determine the half-life of picloram, using high-performance liquid chromatography in a cultivated soil with Brachiaria brizantha trimmed or not. Brachiaria brizantha was cultivated in 60 pots filled with samples of oxisol, and 30 others were kept uncultivated with this forage. This plant was cut off close to the ground, after 60 days of emergency on 30 vessels. Picloram was applied in all of the plots. Soil samples were collected at 2, 16, 30, 44, 58, 72, 86, 120, 150, and 180 days after the application of this herbicide. These samples were air-dried and stored at - 20 °C. Picloram was extracted by HPLC/UV-Vis detector. Half-life of this herbicide was calculated using kinetics models. The mere presence of roots in treatment with signalgrass cutoff did not reduce the concentrations of this herbicide, except when the emergence of new leaves occurred. The absence of B. brizantha cultivation in areas with application of picloram increases the risk of environmental contamination and successive crops due to the half-life of this herbicide. Brachiaria brizantha reduced half-life picloram and environmental risk in pastures. The validation method is suitable for determining picloram in low concentrations in soil.

BACKGROUND: Forage grasses of the African genus Urochloa (syn. Brachiaria) are the basis of Brazilian beef production, and there is a strong demand for high quality, productive and adapted forage plants. Among the approximately 100 species of the genus Urochloa, Urochloa decumbens is one of the most important tropical forage grasses used for pastures due to several of its agronomic attributes. However, the level of understanding of these attributes and the tools with which to control them at the genetic level are limited, mainly due to the apomixis and ploidy level of this species. In this context, the present study aimed to identify and characterize molecular microsatellite markers of U. decumbens and to evaluate their cross-amplification in other Urochloa species. FINDINGS: Microsatellite loci were isolated from a previously constructed enriched library from one U. decumbens genotype. Specific primers were designed for one hundred thirteen loci, and ninety-three primer pairs successfully amplified microsatellite regions, yielding an average of 4.93 alleles per locus. The polymorphism information content (PIC) values of these loci ranged from 0.26 to 0.85 (average 0.68), and the associated discriminating power (DP) values ranged from 0.22 to 0.97 (average 0.77). Cross-amplification studies demonstrated the potential transferability of these microsatellites to four other Urochloa species. Structure analysis revealed the existence of three distinct groups, providing evidence in the allelic pool that U. decumbens is closely related to Urochloa ruziziensis and Urochloa brizantha. The genetic distance values determined using Jaccard's coefficient ranged from 0.06 to 0.76. CONCLUSIONS: The microsatellite markers identified in this study are the first set of molecular markers for U. decumbens species. Their availability will facilitate understanding the genetics of this and other Urochloa species and breeding them, and will be useful for germplasm characterization, linkage mapping and marker-assisted selection.

Brachiaria (signalgrass) is now the most widely used tropical grass genus in Central and South America. However, Brachiaria spp. can cause hepatogenous photosensitization in livestock. Steroidal saponins, specifically protodioscin, present in Brachiaria spp. may be responsible for liver injury and subsequent photosensitization. The objective of this study was to determine the effect of ensiling Brachiaria decumbens and Brachiaria brizantha or making hay from Brachiaria decumbens on the concentrations of steroidal saponin in these grasses. Brachiaria grass had no detectable levels of the saponin protodioscin after 24 days of ensiling. In addition, in Brachiaria decumbens, the concentration of the protodioscin decreased 48% over the first three days after haymaking and then remained constant. These results suggest that livestock consuming Brachiaria either as silage or hay may have reduced risk of intoxication by protodioscin. .(AU)

Entretanto, as espécies de Brachiaria podem causar fotossensibilização hepatógena em animais de produção. Saponinas esteroides, especificamente protodioscina, presentes em Brachiaria, podem ser responsáveis pelo dano hepático e consequentemente fotossensibilização. O objetivo deste estudo foi determinar o efeito da ensilagem de Brachiaria decumbens e Brachiaria brizantha e fenação de Brachiaria decumbens sobre a concentração de saponina esteroide. The experiment was carried out aiming to classify and quantify tiller categories of Brachiaria decumbens cv. Basilisk from different heights found on the same pasture. Four different plant heights (10; 20; 30 and 40 cm) living at the same pasture were evaluated. Randomized block design with three replications was used. Tillers were classified according to their stage of development, growth origin, defoliation level and size. The number of vegetative tillers reduced linearly, while the number of reproductive tillers and dead tillers increased linearly according to the plant height. The number of aerial tillers showed a quadratic response in relation to the plant height. The pastures having grasses of 10, 20, 30 and 40 cm in height had respectively 292, 408, 496 and 344 aerial tillers/m2. The population density of defoliated tillers reduced linearly according to the plant height. The population density of tillers without the apical shoot showed quadratic response to the signal grass height, being the smallest values found for places having grasses of 10 cm (176 tillers/m2) and 40 cm (180 tillers/m2) in height. The number of tillers without defoliation quadratic responded to the plant height, being the highest values found for places having grasses extremely high. The areas with the highest grasses also showed the highest numbers of tallest tillers. There is indeed a spatial variability of the vegetation and a high morphological diversityof tillers on Brachiaria decumbens cv. Basilisk pastures managed under the continuous stockingsystem.


O experimento foi conduzido com o objetivo de classificar e quantificar as categorias de perfilhos de Brachiaria decumbens cv. Basilisk existentes em locais com alturas variáveis em um mesmo pasto. Os tratamentos foram quatro alturas de plantas (10, 20, 30 e 40 cm) avaliadas dentro de uma mesma pastagem. O delineamento experimental foi em blocos casualizados, com três repetições. Os perfilhos foram classificados quanto ao estádio de desenvolvimento, à origem de crescimento, ao nível de desfolhação e ao tamanho. O número de perfilho vegetativo reduziu linearmente, enquanto que os números de perfilhos reprodutivos e mortos incrementaram linearmente com a altura da planta no mesmo. O número de perfilho aéreo apresentou padrão de resposta quadrático em função da altura da planta, sendo que os locais do pasto com 10, 20, 30 e 40 cm de altura possuíram, respectivamente, 292, 408, 496 e 344 perfilhos aéreos/m2. A densidade populacional de perfilhos desfolhados reduziu de forma linear com a altura da planta. A densidade populacional de perfilhos sem o meristema apical apresentou resposta quadrática à altura da planta de capim-braquiária, observando-se os menores valores nos locais com 10 cm (176 perfilhos/ m2) e com 40 cm (180 perfilhos/m2) de altura. O número de perfilhos sem desfolhação respondeu de forma quadrática à altura da planta, com maiores valores nos locais do pasto com as alturas extremas. Os locais mais altos do pasto possuíram maior participação de perfilhos mais compridos. Em pasto de Brachiaria decumbens cv. Basilisk manejada em lotação contínua existe variabilidade espacial da vegetação e grande diversidade morfológica de perfilhos.
