Internet of Things (IoT) is being widely developed in various fields, and its penetration rate in daily life is continuously increasing. The nature of the social function of objects and giving them an identity has made it possible to successfully integrate this technology into many traditional systems and improve their performance by automation. Libraries are one of the most obvious examples of smartening by IoT architecture. So far, various architectures have been presented for smartening libraries through IoT technology. However, a low-cost and ideal architecture that can cover all the requirements in a wide range of smart library applications has not been provided. This article attempts to fill some of the existing research gaps in this field by presenting a new architecture for smartening libraries. In the proposed method, Software Defined Networking (SDN) is used to reduce implementation costs and improve the management process of network components. In this architecture, the communication platform of network active objects is formed based on a cluster-based topology. Also, passive Radio Frequency IDentification (RFID) tags are utilized to manage books and library property. Two stages of evaluation have been conducted for the suggested method's performance: actual deployment and computer simulations. Based on the findings, it can be concluded that this study has succeeded in creating an effective and affordable design for smart libraries, which is a major advancement over conventional libraries.

RESUMO

Enzyme engineering allows to explore sequence diversity in search for new properties. The scientific literature is populated with methods to create enzyme libraries for engineering purposes, however, choosing a suitable method for the creation of mutant libraries can be daunting, in particular for the novices. Here, we address both novices and experts: how can one enter the arena of enzyme library design and what guidelines can advanced users apply to select strategies best suited to their purpose? Section I is dedicated to the novices and presents an overview of established and standard methods for library creation, as well as available commercial solutions. The expert will discover an up-to-date tool to freshen up their repertoire (Section I) and learn of the newest methods that are likely to become a mainstay (Section II). We focus primarily on in vitro methods, presenting the advantages of each method. Our ultimate aim is to offer a selection of methods/strategies that we believe to be most useful to the enzyme engineer, whether a first-timer or a seasoned user.

Halohydrin dehalogenase from Agrobacterium radiobacter AD1 (HheC) is a key enzyme in preparing ß-substituted alcohols and optically pure epoxides. However, the weak thermostability of HheC greatly limits its industrial application and needs to be improved. In this study, a combinatorial smart library was constructed and screened for enhancing the thermostability of HheC. Mutation sites Lys203 and Lys204 located at the protein surface were selected and simultaneously randomized with charged amino acids (Arg, Asp, Glu, Lys), resulting in a combinatorial smart library. After screening a total of only 48 clones, three positive variants CSL1 (Lys203Arg), CSL2 (Lys204Arg) and CSL3 (Lys203Arg/Lys204Arg) were obtained. Compared to the wild type HheC, variant CSL2 exhibited an increase in thermal stability with a nearly 6.9-fold improvement of half-life (τ1/2, 55°C) at 55 °C without compromising catalytic activity. In addition, the double mutant CSL3 displayed approximately 3.4-fold higher τ1/2, 55°C value and 1.8-fold higher kcat value toward 1,3-dicholoro-2-propanol (1,3-DCP) than that of the wild-type HheC. Homology modeling analysis highlights that the newly gained hydrogen bonds and optimizing charge-charge interactions at the protein surface might contribute to the overall conformational stabilization, leading to improving the thermostability of HheC.

The ability to stabilize enzymes and other proteins has wide-ranging applications. Most protocols for enhancing enzyme stability require multiple rounds of high-throughput screening of mutant libraries and provide only modest improvements of stability. Here, we describe a computational library design protocol that can increase enzyme stability by 20-35 °C with little experimental screening, typically fewer than 200 variants. This protocol, termed FRESCO, scans the entire protein structure to identify stabilizing disulfide bonds and point mutations, explores their effect by molecular dynamics simulations, and provides mutant libraries with variants that have a good chance (>10%) to exhibit enhanced stability. After experimental verification, the most effective mutations are combined to produce highly robust enzymes.

Public library, academic library and special library are different kinds of library. The aim, objects, contents of service and the different characteristics, needs, problems of smart construction depend on their different functional positions. The problems and challenges faced by smart construction were pointed out based on the analy-sis of the origin, concept and characteristics of smart construction, domestic studies on smart construction, in com-bination with the different functional positions of public library, academic library and special library, and the needs of their smart construction. Suggestions were thus put forward for their solution.

NFC is characterized by its short transfer distance, low energy consumption, multiple operation model, high safety, portability, good interconnection, and compatibility with RFID.It can thus be considered as the updated edition of RFID.The significance to innovate the service of library using combined NFC and RFID was therefore elaborated with some suggestions put forward for constructing the smart library.
