RESUMO

The main aim of the current work was to explore the differential metabolites and differentially expressed genes of longissimus dorsi muscle (LDM) between castrated and uncastrated fattening male South Sichuan black goats (Capra hircus). Then, the key genes regulating important differential metabolites (DMs) in castrated male goats were observed by integrated metabolomics and transcriptomics analyses. In addition, we evaluated the effects of castration on blood constituents, dressing percentage, and water holding capacity of LDM in male black goats. The results showed that the concentrations of alkaline phosphatase (ALP), total cholesterol (TC), triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C) were significantly increased and testosterone was significantly decreased in castrated male goats compared with the uncastrated male goats, while dressing percentage of black goats and water holding capacity of longissimus dorsi muscle were not significant differences. Through metabolomics and transcriptomics analyses, 23 important KEGG pathways, 13 important DMs, 32 important differentially expressed genes (DEGs), and 13 key genes related to the "Metabolism" and "Organismal systems" pathways were screened. Lipid accumulation may be elevated in the blood of fattening South Sichuan black goats after castration. Castration might play a positive role in energy provision, intercellular signaling, muscle function, softening of meat, disease reduction, and anti-oxidation of LDM. P4HA2, AKR1B1, GPT2, L2HGDH, ENSCHIG00000021660, ENSCHIG00000023861, DGAT2, ULK1, SLC38A3, PLA2G4A, SLC6A1, ENSCHIG00000026624, and ND2 might be the key genes regulating important DMs in the KEGG pathways related to "Metabolism" and "Organismal systems" of castrated male goats compared with the uncastrated male goats.

Excessive heavy metals in soils and crops will affect the human living environment and health. In order to study the enrichment characteristics and causes of heavy metals in soils and crops in a mountainous area of southern Sichuan, 1699 cultivated soil samples, 30 rice seed samples, 31 corn seed samples, 15 tea samples, and 76 crop root soil samples were collected. The enrichment characteristics and origins of heavy metals in the soil were studied using mathematical statistics, principal component analysis, and correlation analysis. The enrichment characteristics of heavy metals in crops and their correlations with root soil were discussed. The results showed that the content of heavy metals in cultivated soil was higher than the national soil background value, and many sampling points exceeded the risk screening value. Among them, 770 points of Cd, 176 points of Cu, 116 points of Cr, and 106 points of Ni exceeded the standard. Combining the factor analysis results, it was found that Cd, Cr, Cu, Ni, and Zn in cultivated soil mainly originated from the geological background, and Cd was also affected by soil organic matter and pH. Meanwhile, Hg and Pb were mainly related to human activities. The results showed that the contents of heavy metals in the crops were lower than the national standards; the enrichment coefficients of heavy metals in crops were generally small, and were mainly in the following order: tea > rice > corn. The correlation analysis of the heavy metal contents in the root soil and crops showed that there was no significant correlation among them. Thus, the heavy metals in the soil generally had a high content and low activity, which did not affect the quality and safety of the three crops. However, Cu in corn and CaO in root soil, As in corn seed and Fe2O3 in root soil, and Ni in corn seed and Fe2O3 in root soil showed significant correlations, whereas As in rice seed and CaO in root soil and Hg in rice seed and MgO in root soil showed significant correlations, and no significant correlations were found in tea.

OBJECTIVE:To provide reference for development of hospital pharmacy and clinical pharmacy in south Sichuan. METHODS:Questionnaire was used to investigate the general situation of secondary and tertiary hospitals in 4 cities of south Sich-uan in aspects of partition layout,staffing situation and job content. The results were statistically analyzed. RESULTS:A total of 40 questionnaires was sent out,35 were effectively received with the effective rate of 87.50%. The partition layout was unreasonable in most surveyed hospitals,and the rate of reaching the standard about the area of drug dispensing sector and warehouse was below 85%. There were 1042 pharmacists in 35 hospitals,which were mainly female of younger than 45 years old. In secondary and ter-tiary hospitals,the rate of pharmaceutical professional and technical personnel of health technology,full-time undergraduate and vice-high above title accounted for 4.78% and 3.67%,17.37% and 31.27%,3.15% and 10.73%,respectively. The number of full-time clinical pharmacists was 24 and 69 in secondary or tertiary hospitals,mainly primary title,middle title and undergradu-ate,of whom 26(27.96%)passed job training of National Health and Family Planning Commission,4(4.30%)passed teaching qualifications training. The rate of pharmacists mainly engaged in drug dispensing and clinical pharmacy was 6.32% and 11.03% in secondary and tertiary hospitals,respectively. CONCLUSIONS:The research shows irrationality of partition layout,the shortage of highly educated and highly titles personnel,especially the clinical pharmacy professionals. The large differences exist in hospitals of different levels in several aspects,and the main body of hospital pharmacy should turn to clinical pharmacy.
