RESUMO

Antimicrobial resistance is a major threat to public health. Antimicrobial use in animal husbandry is a major concern since it can favor an increase in antimicrobial resistance among farms. Herein, we aim to better understand and characterize the main resistome profiles in microbial communities found in pig farms. Sampling of swine manure was performed in two different timepoints (October 2019 and January 2020) in each of the 14 different swine farms, located in the mesoregion of Western Santa Catarina state in Brazil, a pole of swine product production of worldwide importance. Samples were divided into three groups: farms with the opened regimen and no usage of antimicrobials (F1; n = 10), farms with the closed regimen and usage of antimicrobials (F2; n = 16), and farms with the closed regimen and no usage of antimicrobials (F3; n = 2). The metagenomic evaluation was performed to obtain and identify genetic elements related to antimicrobial resistance using nanopore sequencing. We used ResistoXplorer software to perform composition, alpha and beta diversity, and clustering analysis. In addition, PCR reactions were performed to confirm the presence or absence of seven different beta-lactamase family genes and five phosphoethanolamine transferase gene variants clinically relevant. Our findings based on the identification of resistance genes at the mechanism level showed a prevalence of alteration of the drug target (72.3%) profile, followed by drug inactivation (17.5%) and drug efflux (10.1%). We identified predominantly aminoglycosides (45.3%), tetracyclines (15.9%), and multiclass (11,2%) resistance genes. PCoA analysis indicates differences between F1 and F2 profiles. F2 samples showed increased diversity when compared to the F1 group. In addition, herein we first report the identification of mcr-4 in a slurry sample (C1F1.1) in Santa Catarina State. In general, our findings reinforce that many factors on the practices of animal husbandry are involved in the resistome profile at the mechanism and class levels. Further studies to better understand microbiome and mobilome aspects of these elements are necessary to elucidate transmission pathways between different bacteria and environments.

The infection levels were considered mild (1-100 cyst/trophozoite), moderate (101- 300 cyst/trophozoite) and high (>300 cysts/trophozoite). In addition, information about hygienic-sanitary conditions of each farm was collected. The occurrence of B. coli infection in all swine stool samples analyzed was 60.9% (236/387); however, in G1, G2 and G3 was 54.7% (157/287), 91.7% (55/60) and 65% (26/40) respectively. There was significant difference in the occurrence of G1 (P < 0.05) except between farms B and C (67.9% and 56.6% respectively). There was also a significant difference (P < 0.05) between the occurrence of B. coli found in G2 and G3. The infection levels were considered predominantly mild in G1; and mild to moderate in G2 and G3. Introducción: La implementación de modelos animales para el estudio de los tejidos dentales y periodontales de dientes articulados en sus alvéolos sometidos a altas temperaturas permite el establecimiento de parámetros repetitivos que contribuyen con los procesos de identificación. Objetivo: Describir los cambios radiográficos de los tejidos dentales y periodontales de cerdo (Sus domesticus) sometidos a altas temperaturas. Material y métodos: Se realizó un estudio observacional descriptivo de naturaleza pseudo-experimental in vitro para observar los cambios radiográficos de los tejidos dentales y periodontales en 60 dientes de cerdo doméstico sometidos a altas temperaturas (200, 400, 600, 800 y 1,000 ºC). Resultados: Los tejidos dentales y periodontales estudiados presentan gran resistencia a las altas temperaturas sin variar considerablemente su microestructura, de tal manera que los cambios físicos (estabilidad dimensional, fisuras, grietas y fracturas) que ocurren en la medida que aumenta la temperatura pueden describirse a través de radiografía convencional. Conclusiones: El análisis radiográfico de los dientes articulados en sus respectivos alvéolos se constituye en un mecanismo para determinar la temperatura a la cual estuvo sometido un diente, lo que puede ser empleado durante el proceso de identificación odontológica y documentación de la necropsia médico-legal para el caso de cadáveres o restos humanos quemados, carbonizados e incinerados. El cerdo doméstico (Sus domesticus) se constituye en un modelo animal experimental adecuado para estudiar dichos cambios; sin embargo, se recomienda realizar un estudio en dientes humanos articulados en su respectiva unidad alveolar, para determinar si los hallazgos radiográficos descritos se repiten y son extrapolables.


Introduction: The implementation of animal models for the study of periodontal and dental tissues of teeth articulated into their sockets and subjected to high temperatures allows the establishment of repetitive parameters which might contribute to identification processes. Aim: To describe radiographic changes of pig's (Sus domesticus) periodontal and dental tissues subjected to high temperatures. Material and methods: An in vitro pseudo-experimental, descriptive and observational study was undertaken in order to assess radiological changes of periodontal and dental tissues of 60 domestic pig's teeth which had been subjected to high temperatures (200, 400, 600, 800 and 1,000 ºC). Results: The dental and periodontal tissues subject of this research article presented strong resistance to high temperatures without considerable variation of their micro-structure. Thus, physical changes (dimensional stability, fissures, cracks and fractures) which took place as temperature increased, could be described using a conventional X-ray. Conclusions: Radiographic examination of teeth articulated in their sockets can be established as a mechanism to determine the temperature at which the tooth was subjected. This could be used in processes of dental identification and medical-legal autopsy documentation in cases of burned, carbonized or incinerated human remains. Domestic pigs (Sus domesticus) can be regarded as a suitable experimental animal models to study the aforementioned changes. Nevertheless, a study involving human teeth articulated in their own socket is recommended in order to determine whether the radiographic findings herein described are replicated and can be extrapolated.

Currently the pig industry has undergone several adaptations, since the consumer is becoming more selective, especially with regard to food safety and animal welfare. Europe, the major consumer market of Brazilian pork, prohibits the practice of removing or wearing down of the teeth of piglets during the maternity, justifying this practice discourages the welfare of animals. On the other hand, in Brazil, this practice is still being performed routinely in most pig farms, which impairs the exportation of swine products to demanding markets. There is insuficient information about the harms and benefits of that management, so this study aimed to evaluate the influence of dental activity management on the performance of piglets during nursery. Twenty-six litters randomly distributed according to the order of birth of mother were investigated, being 13 litters subjected to dental management (control group), and 13 litters not subjected to the same management (treatment group). Each litter was evaluated for 21 days, undergoing piglets weighing on the day they were born and at 21 days  occurred when weaning, thus to obtain the variables average daily weight gain (ADG) and weaning weight (WEIGHT). Regarding the ADG of piglets, the control group showed an average gain of 0.193 kg/day while the treatment group showed 0.200 kg/day, with no significant difference (P>0.05) between the ex
