With the widespread use of nanoenabled agrochemicals, it is essential to evaluate the food safety of nanomaterials (NMs)-treated vegetable crops in full life cycle studies as well as their potential impacts on human health. Tomato seedlings were foliarly sprayed with 50 mg/L ZnO NMs, including ZnO quantum dots (QDs) and ZnO nanoparticles once per week over 11 weeks. The foliar sprayed ZnO QDs increased fruit dry weight and yield per plant by 39.1% and 24.9, respectively. It also significantly increased the lycopene, amino acids, Zn, B, and Fe in tomato fruits by 40.5%, 15.1%, 44.5%, 76.2%, and 12.8%, respectively. The tomato fruit metabolome of tomatoes showed that ZnO NMs upregulated the biosynthesis of unsaturated fatty acids and sphingolipid metabolism and elevated the levels of linoleic and arachidonic acids. The ZnO NMs-treated tomato fruits were then digested in a human gastrointestinal tract model. The results of essential mineral release suggested that the ZnO QDs treatment increased the bioaccessibility of K, Zn, and Cu by 14.8-35.1% relative to the control. Additionally, both types of ZnO NMs had no negative impact on the α-amylase, pepsin, and trypsin activities. The digested fruit metabolome in the intestinal fluid demonstrated that ZnO NMs did not interfere with the normal process of human digestion. Importantly, ZnO NMs treatments increased the glycerophospholipids, carbohydrates, amino acids, and peptides in the intestinal fluids of tomato fruits. This study suggests that nanoscale Zn can be potentially used to increase the nutritional value of vegetable crops and can be an important tool to sustainably increase food quality and security.

This study aims to investigate the potential of miRNA measurements in indicating tomato quality during transportation and storage. The impact of storage temperature, duration, and mechanical loading on tomato senescence, carotenoid content, total soluble solids, fruit firmness, and relevant miRNA concentrations were examined. Significant two-way or three-way interactions were observed between storage conditions and physical/physiological characteristics (excluding carotenoids). Remarkably, significant three-way interactions were found between storage conditions and miRNA concentrations. Strong correlations were observed between the physiological characteristics of the tomatoes and their miRNA concentrations. These findings suggest that measuring miRNAs could serve as a convenient and portable method for evaluating postharvest fruit quality, reducing reliance on labor-intensive laboratory techniques.

Betalains, which consist of the subgroups betaxanthins and betacyanins, are hydrophilic pigments that have classically been used for food colorants. Owing to their strong antioxidant property, their usefulness for application for therapeutic use is also expected. In addition, as betalains are mainly naturally available from plants of the order Caryophyllales, including beet (Beta vulgaris), metabolic engineering for betalain production in crops such as vegetables, fruits and cereals may provide new food resources useful for healthcare. Here we conducted metabolic engineering of betacyanins in tomato fruits and potato tubers. The transgenic tomato fruits and potato tubers with coexpression of betacyanin biosynthesis genes, CYP76AD1 from B. vulgaris, DOD (DOPA 4,5-dioxygenase) and 5GT (cyclo-DOPA 5-O-glucosyltransferase) from Mirabilis jalapa, under control of suitable specific promoters, possessed dark red tissues with enriched accumulation of betacyanins (betanin and isobetanin). The anti-inflammatory activity of transgenic tomato fruit extract was superior to that of wild-type fruit extract on macrophage RAW264.7 cells stimulated with lipopolysaccharide (LPS), as a result of decreased LPS-stimulated transcript levels of proinflammatory genes. These findings were in accord with the observation that administration of the transgenic tomato fruits ameliorated dextran sulfate sodium (DSS)-induced colitis as well as body weight loss and disease activity index in mice, via suppression of DSS-stimulated transcript levels of pro-inflammatory genes, including Tnf (encoding TNF-alpha), Il6, and Ptgs2 (encoding cyclooxygenae 2). Intriguingly, given the fact that the transgenic potato tuber extract failed to enrich the anti-inflammatory activity of macrophage cells, it is likely that metabolic engineering of betacyanins will be a powerful way of increasing the anti-inflammatory property of ordinary foods such as tomato.

Flavonoids contribute to fruit sensorial and nutritional quality. They are also highly beneficial for human health and can effectively prevent several chronic diseases. There is increasing interest in developing alternative food sources rich in flavonoids, and nano-enabled agriculture provides the prospect for solving this action. In this study, triiron tetrairon phosphate (Fe7(PO4)6) nanomaterials (NMs) were synthesized and amended in soils to enhance flavonoids accumulation in tomato fruits. 50 mg kg-1 of Fe7(PO4)6 NMs was the optimal dose based on its outstanding performance on promoting tomato fruit flavonoids accumulation. After entering tomato roots, Fe7(PO4)6 NMs promoted auxin (IAA) level by 70.75 and 164.21% over Fe-EDTA and control, and then up-regulated the expression of genes related to PM H+ ATPase, leading to root proton ef-flux at 5.87 pmol cm-2 s-1 and rhizosphere acidification. More Mg, Fe, and Mn were thus taken up into plants. Subsequently, photosynthate was synthesized, and transported into fruits more rapidly to increase flavonoid synthesis potential. The metabolomic and transcriptomic profile in fruits further revealed that Fe7(PO4)6 NMs regulated sucrose metabolism, shi-kimic acid pathway, phenylalanine synthesis, and finally enhanced flavonoid biosynthesis. This study implies the potential of NMs to improve fruit quality by enhancing flavonoids synthesis and accumulation.

Zizyphus spina-christi L. has antimicrobial properties because of the presence of biologically active compounds. Alternaria is an opportunistic pathogen that causes leaf spots, rots, and blights on a variety of plant parts. This study aimed to reduce the usage of synthetically derived fungicides. Identification of the bioactive components present in leaves and fruits methanolic extracts of Z. spina-christi was performed using high-performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC-MS). The efficacy of the two methanol extracts was tested against (a) in vitro fungal growth and (b) pathogenicity control on non-wounded and wounded tomato fruits. The results revealed that gallic acid and ellagic acid were the major components in leaves extract while quercetin was the major component in fruits extract. In addition, Phenol, 2,5-bis(1,1-dimethylethyl) (40.24%) and Decane, 2-methyl-(18.53%) were the most abundant components in the leaf extract, and the presence of D-mannonic acid, 2,3,5,6-tetrakis-o-(trimethylsilyl), and γ-lactone (22.72%) were major components in fruits extract. The methanolic extracts of Z. spina-christi leaves and fruits demonstrated significant antifungal activity against the growth of Alternaria alternata, A. citri, and A. radicina with variable inhibition percentages at different concentrations. Pathogenicity was increased when the skin was injured, as expected. Both extracts reduced the percentage of infected fruits.

Food product nutritional and sensory characteristics are often deeply linked to its territory of origin; therefore, its authentication by means of elemental composition becomes crucial for traceability and fighting food fraud. This study aims to establish a fast and reproducible procedure for origin and quality assessment of Sicilian tomato fruits, including PGI "Pomodoro di Pachino", by using the X-ray fluorescence (XRF) technique. Measurements were performed on different parts of PGI Pachino tomatoes belonging to the same production lot. Principal Component and Cluster Analyses show that the samples cluster accordingly with the production lot, disentangling the different parts of the fruit. This procedure, which uses XRF yield elemental pattern and statistical analysis, establishes a solid basis for characterizing elemental profiles by a fast XRF in-situ campaign, supporting the traceability system. The reliability of XRF results was confirmed by comparing elemental concentrations with ICP-MS measurements, performed for comparison, and tomato literature values.

Methyl jasmonate hormone can stimulate the production of several metabolites responsible for improving fruit quality and nutritional attributes related to human health. In this context, efforts to manipulate tomatoes, such as using hormonal treatment to increase metabolite levels essential to plant growth and human nutrition, have received considerable attention. The aim of this study was to show the impact of metabolic profile on fruit quality and nutritional properties under exogenous methyl jasmonate during fruit ripening. The treatments were performed using 100 ppm of methyl jasmonate and 100 ppm of gaseous ethylene over 24 h. Ethylene emission, fruit surface color and metabolomics analysis were measured at 4, 10, and 21 days after harvest, considering the untreated fruits as control group. Methyl jasmonate induced the production of amino acids-mainly glutamine, glutamic acid and γ-aminobutyric acid (at least 14-fold higher)-and fatty acids-mainly oleic, linoleic, and linolenic acids (at least three-fold higher than untreated fruits); while exogenous ethylene predominantly affected sugar metabolism, increasing the levels of fructose, mannose and glucose to at least two-fold that levels in the untreated fruits. Additionally, methyl jasmonate significantly affected secondary metabolites, inducing by at least 80% the accumulation of α-tocopherol and ß-sitosterol in fully ripe fruits. Our results suggest that the postharvest application of the hormone methyl jasmonate can contribute to the sensory characteristics and increase the nutritional value of the fruits since important changes related to the tomato metabolome were associated with compounds responsible for the fruit quality and health benefits.

Soluble sugar is known to improve the sweetness and increase tomato sauce yield. Studies have focused on improving the content of soluble sugar in tomato fruits, usually by promoting functional genes. We studied two genes (SlINVINH1 and SlVPE5) that inhibited the accumulation of soluble sugar in tomato fruits and obtained two genes' knocked-out lines (CRISPR-invinh1 or CRISPR-vpe5) using CRISPR/Cas9. Aggregated lines with CRISPR-invinh1 and CRISPR-vpe5 were gained by hybridization and self-pollination. Compared to wild-type lines, the glucose, fructose, and total soluble solid (TSS) contents of CRISPR-invinh1 and CRISPR-vpe5 increased significantly. Glucose, fructose, and TSS levels further improved simultaneously with CRISPR-invinh1 and CRISPR-vpe5 than with single gene knock-out lines. This indicates that these genes have a synergistic effect and will increase the soluble sugar content. Thus, the knock-out SlINVINH1 and SlVPE5 may provide a practical basis for improving the sweetness of tomato fruits and their processing quality.

The purpose of the present study was to investigate the efficacy of the experimental formulations of the metribuzin (MET) and tribenuron-methyl (TBM) herbicides embedded in the matrix of degradable poly-3-hydroxybutyrate blended with wood flour in field-grown tomato and beet crops infested by weeds. There is a necessity to develop environmentally friendly and effective means to protect plants because of the shortcomings of the free herbicide forms such as the environmentally unsafe spray application of solutions and suspensions of the widespread metribuzin and tribenuron-methyl herbicides, removal from soil during watering events and rains, and transport to natural aquatic environments, where the herbicides accumulate in the trophic chains of biota. Free TBM is also rapidly inactivated in soil and metabolized to nontoxic products in plants. The efficacy of experimental formulations of metribuzin and tribenuron-methyl embedded in the matrix of degradable poly-3-hydroxybutyrate blended with wood flour was tested in field-grown tomato and beet crops infested with weeds. Application of metribuzin resulted in the highest productivity of tomatoes (2.3 kg/m2) and table beet (3.4 kg/m2), improved biometric parameters of tomato fruits and beet roots, and caused reduction in nitrate nitrogen concentrations in them. The mode of herbicide delivery did not affect sugar contents, but application of both metribuzin and tribenuron-methyl induced a 1.7-fold and 1.4-fold, respectively, increase in vitamin C concentrations in tomato fruits and beet roots relative to the vegetables grown on the subplots treated with free herbicides and the intact plants. Embedded herbicides can be used as preemergence herbicides in the field. Fig. a Graphical abstract.

Carotenoids are essential components of the photosynthetic antenna and reaction center complexes, being also responsible for antioxidant defense, coloration, and many other functions in multiple plant tissues. In tomato, salinity negatively affects the development of vegetative organs and productivity, but according to previous studies it might also increase fruit color and taste, improving its quality, which is a current agricultural challenge. The fruit quality parameters that are increased by salinity are cultivar-specific and include carotenoid, sugar, and organic acid contents. However, the relationship between vegetative and reproductive organs and response to salinity is still poorly understood. Considering this, Solanum lycopersicum cv. Micro-Tom plants were grown in the absence of salt supplementation as well as with increasing concentrations of NaCl for 14 weeks, evaluating plant performance from vegetative to reproductive stages. In response to salinity, plants showed a significant reduction in net photosynthesis, stomatal conductance, PSII quantum yield, and electron transport rate, in addition to an increase in non-photochemical quenching. In line with these responses the number of tomato clusters decreased, and smaller fruits with higher soluble solids content were obtained. Mature-green fruits also displayed a salt-dependent higher induction in the expression of PSY1, PDS, ZDS, and LYCB, key genes of the carotenoid biosynthesis pathway, in correlation with increased lycopene, lutein, ß-carotene, and violaxanthin levels. These results suggest a key relationship between photosynthetic plant response and yield, involving impaired photosynthetic capacity, increased carotenoid-related gene expression, and carotenoid biosynthesis.

The improvement of the postharvest quality of tomato fruits was evaluated using an edible coating functionalized with an Flourensia cernua extract evaluating the antifungal, structural, barrier, and optical properties. The formulation and evaluation of an edible coating and its application on tomato was evaluated using a response surface methodology to determine the ideal concentrations of candelilla wax, whey protein, and glycerol. Edible films showed good barrier properties, with water vapor permeability varying from 0.435-0.404 g mm/m2 day kPa. The addition o F. cernua extract showed significant improvement in the transparency of films. The edible coating applied to tomato reduced weight and firmness loss. The sensory evaluation proved that the product obtained is acceptable for consumers. The edible coating added with F. cernua extract was the most effective in inhibiting the growth of pathogenic fungi and the visual appearance at the end of storage confirmed the beneficial effect of the edible coating.

Small heat shock proteins (sHSPs) increase stress tolerance in a wide variety of organisms and enable them to endure changes in their environment. However, the molecular mechanism by which sHSPs protect plants against cold stress is unknown. Here, the sHSP of tomato named SlHSP17.7 (Solyc06g076540.1.1) has the characteristic of low temperature induced expression in BL21(DE3) E. coli and a molecular chaperone function in vitro. Overexpression of SlHSP17.7 showed a tolerant response to cold stress treatment due to an induce intracellular sucrose and less accumulation of ROS. Yeast two-hybrid assays showed that SlHSP17.7 is a binding partner of the cation/Ca2+ exchanger (SlCCX1-like; Solyc07g006370.1.1). This interaction was confirmed by pull down and bimolecular fluorescence complementation (BiFC) assays. High SlHSP17.7 and low SlCCX1-like levels alleviated programed cell death (PCD) under cold stress. Thus, SlHSP17.7 might be a cofactor of SlCCX1-like targeting endoplasmic reticulum (ER) membrane proteins, retaining intracellular Ca2+ homeostasis, and decreasing cold stress sensitivity. These findings provide a sound basis for genetic engineering of cold stress tolerance in tomato.

Hydrogen sulfide (H2S) could act as a versatile signaling molecule in delaying fruit ripening and senescence. Ethylene (C2H4) also plays a key role in climacteric fruit ripening, but little attention has been given to its interaction with H2S in modulating fruit ripening and senescence. To study the role of H2S treatment on the fruit quality and nutrient metabolism, tomato fruits at white mature stage were treated with ethylene and ethylene plus H2S. By comparing to C2H4 treatment, we found that additional H2S significantly delayed the color change of tomato fruit, and maintained higher chlorophyll and lower flavonoids during storage. Moreover, H2S could inhibit the activity of protease, maintained higher levels of nutritional-related metabolites, such as anthocyanin, starch, soluble protein, ascorbic acid by comparing to C2H4 treatment. Gene expression analysis showed that additional H2S attenuated the expression of beta-amylase encoding gene BAM3, UDP-glycosyltransferase encoding genes, ethylene-responsive transcription factor ERF003 and DOF22. Furthermore, principal component analysis suggested that starch, titratable acids, and ascorbic acid were important factors for affecting the tomato storage quality, and the correlation analysis further showed that H2S affected pigments metabolism and the transformation of macromolecular to small molecular metabolites. These results showed that additional H2S could maintain the better appearance and nutritional quality than C2H4 treatment alone, and prolong the storage period of post-harvest tomato fruits.

Large amounts of tomato fruits and derived products are produced in China and may be contaminated by Alternaria mycotoxins, which may have the potential risks for human health. There is thus an increasing interest in reducing the mycotoxins. In the present study, 26 Alternaria strains isolated from tomato black rots were identified according to morphological and molecular grounds, and their mycotoxigenic abilities for alternariol (AOH), alternariol monomethyl-ether (AME) and tenuazonic acid (TeA) were also investigated. The results showed that A. alternate was the predominant species with incidence values of 65.4% (17/26), followed by A. brassicae (7/26) and A. tenuissima (2/26). A. alternate isolates showed the highest capacity for AOH, AME and TeA production among the studied isolates either in vitro or in vivo, suggested that A. alternata may be the most important mycotoxin-producing species in tomato fruits. Thus, UV-C irradiation was used to reduce the mycotoxin produced by A. alternata in our study. The results showed that low dose of UV-C irradiation (0.25 kJ/m2) could effectively inhibit mycotoxins production and penetration in tomatoes. Upon treatment with UV-C, there was 79.6, 76.4 and 51.4% of reduction in AOH, AME and TeA penetration when compared to untreated fruits. This may be associated with the enhanced phenolics by UV-C irradiation. In fact, the induced phenolics were including p-coumaric, ferulic and pyrocatechuic acids, of which p-coumaric acid (1.0 mM) displayed the highest reduction of TeA with 60.2%, whereas ferulic acid (1.0 mM) showed strong inhibitory effects on the AOH and AME production by 59.4 and 79.1%, respectively. Therefore, the application of UV-C irradiation seems to be a promising method for reducing the potential risk of Alternaria mycotoxins in fruits and also for enhancing phenolics of processing products.

A selective liquid chromatographic analytical method was studied for determination of two neonicotinoids, acetamiprid and imidacloprid, in tomato fruits under greenhouse conditions in Egypt. The fruits were extracted and cleaned up by QuEChERS method followed by HPLC determination. The method showed a good linearity with a determination coefficient (R2) of higher than 0.99 for the 0.0125-0.15 µg/mL concentration range. The method was validated using a blank tomato spiked at 5, 25 and 50 mg/kg and the recovery percentages were 83.71, 94.52 and 97.49% for acetamiprid and 88.59, 89.63 and 90.18% for imidacloprid, respectively. The rates of dissipation of both pesticides were studied and the preharvest intervals (PHIs) were calculated. Imidacloprid dissipated faster than acetamiprid and half-life periods were 1.30 and 2.07 days, respectively. Acetamiprid and imidacloprid residues were below the already established European maximum residue limits (EU MRLs) (0.5 mg/kg) 3 and 5 days after application, respectively.

In the present study, the influence of production systems (net-house and open-field) on volatile profiles of three Texas A&M University (TAMU) and five commercial tomato varieties was investigated. Forty metabolites were determined using headspace solid phase microextraction (HS-SPME) equipped with gas chromatography and mass spectrometry (GC-MS). The data was evaluated by multivariate analyses to discriminate the effects of genotype and production system, and to identify potential biomarker(s). The levels of hexanal, p-cymene, and (E)-2-hexenal from TAMU varieties were distinct from those of commercial tomato varieties. Similarly, 16 metabolites were considerably affected by the production systems, and majority of these volatiles were significantly higher in the net-house-grown tomatoes. Multivariate analysis also allowed identifying geranylacetone and d-limonene as potential biomarkers to classify tomatoes according to production systems. These findings underline the importance of the selection of variety and production system to preserve or improve desirable aroma traits in tomatoes.

The realisation of manned space exploration requires the development of Bioregenerative Life Support Systems (BLSS). In such self-sufficient closed habitats, higher plants have a fundamental role in air regeneration, water recovery, food production and waste recycling. In the space environment, ionising radiation represents one of the main constraints to plant growth. In this study, we explore whether low doses of heavy ions, namely Ca 25 Gy, delivered at the seed stage, may induce positive outcomes on growth and functional traits in plants of Solanum lycopersicum L. 'Microtom'. After irradiation of seed, plant growth was monitored during the whole plant life cycle, from germination to fruit ripening. Morphological parameters, photosynthetic efficiency, leaf anatomical functional traits and antioxidant production in leaves and fruits were analysed. Our data demonstrate that irradiation of seeds with 25 Gy Ca ions does not prevent achievement of the seed-to-seed cycle in 'Microtom', and induces a more compact plant size compared to the control. Plants germinated from irradiated seeds show better photochemical efficiency than controls, likely due to the higher amount of D1 protein and photosynthetic pigment content. Leaves of these plants also had smaller cells with a lower number of chloroplasts. The dose of 25 Gy Ca ions is also responsible for positive outcomes in fruits: although developing a lower number of berries, plants germinated from irradiated seeds produce larger berries, richer in carotenoids, ascorbic acid and anthocyanins than controls. These specific traits may be useful for 'Microtom' cultivation in BLSS in space, in so far as the crew members could benefit from fresh food richer in functional compounds that can be directly produced on board.

Numerous studies have been focusing on breeding tomato plants with enhanced lycopene accumulation, considering its positive effects of fruits on the visual and functional properties. In this study, we used a bidirectional strategy: promoting the biosynthesis of lycopene, while inhibiting the conversion from lycopene to ß- and α-carotene. The accumulation of lycopene was promoted by knocking down some genes associated with the carotenoid metabolic pathway. Finally, five genes were selected to be edited in genome by CRISPR/Cas9 system using Agrobacterium tumefaciens-mediated transformation. Our findings indicated that CRISPR/Cas9 is a site-specific genome editing technology that allows highly efficient target mutagenesis in multiple genes of interest. Surprisingly, the lycopene content in tomato fruit subjected to genome editing was successfully increased to about 5.1-fold. The homozygous mutations were stably transmitted to subsequent generations. Taken together, our results suggest that CRISPR/Cas9 system can be used for significantly improving lycopene content in tomato fruit with advantages such as high efficiency, rare off-target mutations, and stable heredity.

Here, we reported that multiwalled carbon nanotubes (MWCNT) added to hydroponics system can enhance fruit production of exposed tomato plants. We quantified the exact amount of MWCNT accumulated inside of fruits collected by MWCNT-exposed plants using an advanced microwave induced heating technique (MIH). We found that absorption of MWCNT by tomato fruits significantly affected total fruit metabolome as was confirmed by LC-MS. Our data highlight the importance of comprehensive toxicological risk assessment of plants contaminated with carbon nanomaterials.

Little is known about the effects of surface modification on the interaction of nanoparticles (NPs) with plants. Tomato (Solanum lycopersicum L.) plants were cultivated in potting soil amended with bare and citric acid coated nanoceria (nCeO2, nCeO2+CA), cerium acetate (CeAc), bulk cerium oxide (bCeO2) and citric acid (CA) at 0-500 mg kg-1. Fruits were collected year-round until the harvesting time (210 days). Results showed that nCeO2+CA at 62.5, 250 and 500 mg kg-1 reduced dry weight by 54, 57, and 64% and total sugar by 84, 78, and 81%. At 62.5, 125, and 500 mg kg-1 nCeO2+CA decreased reducing sugar by 63, 75, and 52%, respectively and at 125 mg kg-1 reduced starch by 78%, compared to control. The bCeO2 at 250 and 500 mg kg-1, increased reducing sugar by 67 and 58%. In addition, when compared to controls, nCeO2 at 500 mg kg-1 reduced B (28%), Fe (78%), Mn (33%), and Ca (59%). At 125 mg kg-1 decreased Al by 24%; while nCeO2+CA at 125 and 500 mg kg-1 increased B by 33%. On the other hand, bCeO2 at 62.5 mg kg-1 increased Ca (267%), but at 250 mg kg-1 reduced Cu (52%), Mn (33%), and Mg (58%). Fruit macromolecules were mainly affected by nCeO2+CA, while nutritional elements by nCeO2; however, all Ce treatments altered, in some way, the nutritional quality of tomato fruit. To our knowledge, this is the first study comparing effects of uncoated and coated nanoceria on tomato fruit quality.
