Calcium hydroxide represents the most commonly used intracanal dressing between sessions; however, it may not be effective against all types of microorganisms. Several compounds of plant origin have attracted increasing attention from researchers in recent years. The objective of this study was to evaluate the cytocompatibility and antimicrobial activity of calcium hydroxide associated with the essential oil of Cyperus articulatus and the new bioceramic intracanal medicament Bio-C Temp®. Five experimental groups were designed: group Ca-C. articulatus essential oil; group CHPG-calcium hydroxide associated with propylene glycol; group CHCa-essential oil of C. articulatus associated with calcium hydroxide; and group U-UltraCal® XS; group BCT-Bio-C Temp®. The control group was a culture medium. Cytocompatibility was assessed by the methyltetrazolium (MTT) assay after exposure of the Saos-2 human osteoblast-like cell line to dilutions of commercial products/associations for 24 h and 72 h. The antimicrobial activity against mature Enterococcus faecalis biofilm was evaluated by the crystal violet assay. All commercial products/associations showed a cell viability similar to or even higher than the control group (p > 0.05) for both periods evaluated. C. articulatus essential oil associated or not with calcium hydroxide showed better antibiofilm capacity. C. articulatus associated or not with calcium hydroxide showed superior cytocompatibility and antimicrobial capacity, representing a promissory intracanal medicament.

Punica granatum, popularly known as pomegranate, is a fruit tree with wide worldwide distribution, containing numerous phytochemicals of great medicinal value. The aim of the present study was to determine the phytochemical profile and antioxidant potential of a protein fraction (PF) derived from P. granatum sarcotesta which is rich in lectin. In addition, the acute oral toxicity, genotoxicity and antigenotoxicity of this protein fraction (PF) from P. granatum sarcotesta was measured. The phytochemical profile of PF was determined using HPLC. The in vitro antioxidant effect was assessed using the methods of total antioxidant capacity (TAC) and DPPH and ABTS+ radical scavenging. Acute oral toxicity was determined in female Swiss mice administered a single dose of 2000 mg/kg. This PF was examined for genotoxicity and antigenotoxicity at doses of 500, 1000 and 2000 mg/kg, utilizing mouse peripheral blood cells. Phytochemical characterization detected a high content of ellagic acid and antioxidant capacity similar to that of ascorbic acid (positive control). PF was not toxic (LD50 >2000 mg/kg) and did not exert a genotoxic effect in mice. PF protected the DNA of peripheral blood cells against damage induced by cyclophosphamide. In conclusion, this PF fraction exhibited significant antioxidant activity without initiating toxic or genotoxic responses in mice.

Oral diseases are one of the biggest public health problems worldwide, caused by opportunistic pathogens such as Streptococcus mutans and Enterococcus faecalis. Cyperus articulatus (priprioca) is a plant conventionally used in traditional medicine in the Amazon region. However, little is known about the possible dentistry-related uses of extracts from the rhizomes and solid waste generated by the extraction of essential oils from this vegetable. This study aimed to investigate the chemical composition of volatile compounds and antimicrobial activity through the Minimum Inhibitory Concentration test (MIC and assessment of the toxicity by Hens Egg Test-Chorion Allantoic Membrane (HET-CAM) of the ethanolic extracts from Cyperus articulatus intact rhizomes and solid waste. We identified sesquiterpenes as the main constituents, strong antimicrobial activity of the ethanolic extract of intact rhizomes against S. mutans (MIC = 0.29 mg/mL), moderate antimicrobial activity against E. faecalis of the extract obtained from the solid waste (MIC = 1.17 mg/mL), and absence of toxicity for both tested extracts. The absence of irritation and the antibacterial activity of the ethanolic extract from C. articulatus rhizomes and solid waste reveal its potential for use in the alternative control of bacteria that cause oral infections and may present economic viability as a raw material for dental products.

The genus Passiflora (Passifloraceae) comprises about 500 species. The Passiflora edulis stands out because of its economic and medicinal importance. It is widely planted in tropical and subtropical regions worldwide, especially in South America, the Caribbean, South Africa, and Asia. The aqueous extract of Passiflora edulis Sims f. edulis (Gulupa) leaves is used in traditional medicine for its soothing and tranquilizing effects on the central nervous system. Therefore, evaluating its safety for human use is a fundamental requirement to continue the development of new therapies within the framework of regulatory, preclinical, and clinical guidelines. Here, the sub-acute toxicity study was conducted following the Organization for Economic Cooperation and Development (OECD) guideline 407 for 28 days in Wistar albino rats. The study showed that 1000 mg/kg/day of the aqueous extract in 10 adult Wistar rats (five males and five females) was well tolerated. The hematological results are at normal levels. However, monocytopenia and eosinopenia were observed with a significant difference (P < 0,05) for both male and female rats treated with the aqueous extract of Passiflora edulis. The results show that liver and kidney function profiles were conserved. However, an increase in ALT is observed with significant differences between male and female rats treated with the extract compared to the controls. Study findings were limited to non-adverse histopathological results of a slightly increased incidence of focal periportal lymphocytic infiltrate in the liver and focal corticomedullary nephrocalcinosis in the kidney compared to control. Therefore, the aqueous extract of Passiflora edulis has a good safety profile in oral administration, was well tolerated, and did not cause any lethality or adverse effects in the sub-acute toxicity study in male and female rats. The NOAEL (no observed adverse effect level) for the 28-day subacute toxicity study was considered to be 1000 mg/kg.

Textile effluents, although their composition can vary considerably, typically contain high levels of dissolved salts and exhibit wide variations in pH. Ecotoxicological studies regarding the effects of these parameters, however, have been limited owing to the need for sensitive and easy-to-handle bioindicators that require low amounts of sampling, are cost-effective, time-efficient, and ethically endorsed. This kind of study, additionally, demands robust multi-factorial statistical designs that can accurately characterize the individual and combined relationship between variables. In this research, Response Surface Methodology (RSM) was used to calculate the individual and interaction effects of NaCl concentration and pH value of a Simulated Textile Effluent (STE) on the development rate (DR) of the bioindicators: Bacillus subtilis bacteria and Lactuca sativa lettuce. The results demonstrated that the bioindicators were sensitive to both NaCl and pH factors, where the relative sensitivity relationship was B. subtilis > L. sativa. The quadratic equations generated in the experiments indicated that increased concentrations of 50-250 mg L-1 of NaCl caused a perturbance of 1.40%-34.40% on the DR of B. subtilis and 0.50%-12.30% on L. sativa. The pH factor at values of 3-11 caused an alteration of 27.00%-64.78% on the DR of the B. subtilis and 51.37%-37.37% on the L. sativa. These findings suggest that the selected bioindicators could serve as effective tools to assess the ecotoxicological effects of textile effluents on different ecological systems, and the RSM was an excellent tool to consider the ecotoxicological effects of the parameters and to describe the behavior of the results. In conclusion, the NaCl and pH factors may be responsible for disrupting different ecosystems, causing imbalances in their biodiversity and biomass. Before discharge or reuse, it is suggested to remove salts and neutralize pH from textile effluents and, mostly, develop novel, eco-friendlier textile processing techniques.

Historically, the ocular toxicity of manufactured consumer materials has been evaluated using the rabbit in vivo Draize rabbit eye test. The animal data obtained were used by the United Nations Globally Harmonized System of Classification and Labelling of Chemicals (UN GHS) to define the classification and labelling (C&L) for eye damage/irritation endpoint. However, the Draize test, a method which was never formally validated, has been widely criticized because of its technical limitations. In addition, ethical and economic issues and advances in scientific knowledge, and political and public pressures have made animal experimentation unsustainable. This scenario has consequently led to the development of nonanimal testing and protocols/approaches with considerable predictive value and relevance for humans. It is widely accepted that one single nonanimal method cannot cover all the criteria of damage/inflammation assessed by regulatory adopted in vivo animal testing. Thus, integrated testing strategies (ITS) have been proposed, including a tiered testing approach combining different nonanimal testing with different endpoints, which have been used for regulatory purposes, on a case-by-case basis and within integrated approaches to testing and assessment (IATA), to identify materials according to their ability to trigger eye damage. In particular, the top-down and bottom-up approaches have been recommended for the C&L of materials, which cause serious eye damage or eye irritation, respectively. This chapter describes detailed protocols for eye irritation testing based on cells (Short Time Exposure-STE, OECD No. 491/2017), a vascularized membrane (the Hen's Egg Test-Chorioallantoic Membrane-HET-CAM) and corneal tissue (Bovine Corneal Opacity and Permeability-BCOP, OECD No. 437/2017), which can be applied using top-down or bottom-up approaches. In addition, it suggests making a corneal histomorphometric evaluation as an additional parameter in the BCOP method to differentiate materials that cause serious eye tissue damage (UN GHS Cat. 1) from materials that have reversible eye irritation effects (UN GHS Cat. 2).

Despite the risk of significant adverse toxicological effects of Hg to humans and wildlife, Hg use in anthropogenic activities, and artisanal small-scale gold mining (ASGM) in particular, is widespread throughout Latin America. However, there are few research and monitoring studies of Hg toxicity in fish and fish-eating wildlife in Latin America compared to North America. In the present paper, we reviewed the literature from published articles and reports and summarized and assessed data on Hg in fish from 10 391 individuals and 192 species sampled across Latin America. We compared fish Hg levels with toxicity reference values (TRVs) for fish and dietary TRVs for fish-eating wildlife. We determined that fish, piscivorous birds, and other wildlife are at risk of Hg toxicity. We observed a large disparity in data quantity between North and Latin America, and identified regions requiring further investigation. In particular, future biomonitoring and research should focus on exposure of wildlife to Hg in Peru, Chile, Uruguay, the eastern and northern regions of Brazil, Venezuela, Ecuador, and Colombia. We also discuss Hg risk assessment methodological issues and recommend that future evaluations of Hg risk to wildlife must collect key physiological variables, including age, body size, and ideally Hg-to-Se molar ratios. Paraná River, the six largest in the world, is receptor of pollution loads from tributaries traversing urban and industrialized areas, and extensive agriculture, particularly in its middle and low stretch along the Argentinean sector, where most of the productive activities of the country develop. Within the frame of monitoring surveys, the quality of bottom sediments from distal positions of twenty tributaries and three of the main course was evaluated. The assessment covered testing lethal and sublethal effects with the Hyalella curvispina based toxicity test, a benthic macrofauna survey and physicochemical variables of sediment matrix composition. A multivariate statistical analysis approach permitted integrating the obtained data from the different survey lines of evidence, explaining potential causes of the measured biological effects. The main perturbations detected were associated to tributaries in the middle sector of the basin, where anoxic conditions with high sulfide contents prevail mostly related to organic matter inputs of diverse combined activities, where sediments induce high lethality, and a consequent strong reduction of the benthic community population and diversity. The integrated survey approach proved being a robust tool in the assessment of causative-adverse effects relationships.
