RESUMO

Objective To investigate the expressions of human papillomavirus (HPV) 16E6 and E-cadherin in different cervical tissues. Methods Immunohistochemistry expressions of HPV16E6 and E-cadherin were analyzed in 30 normal cervical tissues,34 cervical intraepithelial neoplasias (CIN),and 60 squamous cervical cell carcinomas. Results The differences in positive expression rates of HPV16E6 and E-cadherin were statistically significant among normal cervical tissues,CIN Ⅰ,CIN Ⅱ-Ⅲ,cervical squamous cell carcinomas (x2 =40.166,P =0.000;x2 =31.295,P =0.000),respectively.The abnormal expressions of HPV16E6 and E-cadherin expression were significantly related to clinical pathological stage, tumor size, histological grade, invasion depth of squamous cervical cancers (P < 0.05),but not correlated with age (P >0.05).The expressions between HPV16E6 and E-cadherin protein were negatively correlated (rs =-0.647,P =0.000).Conclusions There may be relationship between HPV16E6 and E-cadherin,and the abnormal expressions of two proteins may be associated with poor prognosis of cervical carcinomas.

RESUMO

Objective To investigate the effect of over-expressed macrophage migration inhibitory factor (MIF) on epithelial-mesenchymal transition (EMT) in human cervical carcinoma SiHa cells. Methods Recombinant eukaryotic expression plasmid liposome enhanced transfection of green fluorescent protein gene (pEGFP-N1)-MIF was constructed and then transfected into human cervical cancer SiHa cells.Experimental cells were classified into three groups (SiHa-pEGFP-N1-MIF,SiHa-pEGFP-N1,and SiHa).Western blot was used to detect the expression of MIF protein, and the expressions of EMT-related markers such as E-cadherin and vimentin in SiHa cells were determined before and after transfection. Results The eukaryotic expression vector pEGFP-N1-MIF significantly increased the expression of MIF protein in SiHa cells (P < 0.05),and after overexpression of MIF gene in SiHa cells, the expression of E-cadherin protein in SiHa-pEGFP-N1-MIF group was significantly lower than that in control groups (P <0.05),while the expression of vimentin in SiHa-pEGFP-N1-MIF group was significantly higher than that in control groups (P < 0.05).Conclusions Overexpression of MIF in cervical cancer SiHa cells can promote the EMT occurrence.

RESUMO

Objective To detect the expression of Cytokeratin 19 (CK19) mRNA in the peripheral blood of cervical carcinoma patients and evaluate its clinical significance. Methods The expression of CK19 mRNA was evaluated by fluorescent quantitative reverse transcription polymerase chain reaction ( FQRT-PCR ) in the peripheral blood of 138 patients with cervical carcinoma and 36 patients with benign gynecological tumors. In 138 patients, possible correlations between clinical pathological factors were analyzed. Results The positive expression rates of CK19 mRNA was 69.6% in 138 cervical carcinomas in comparison with 13.9% in benign gynecological tumors, and 57.9% in patients with FIGO Stage Ⅰ A to Ⅱ A cervical carcinoma in comparison with 80.6% in patients with FIGO Stage Ⅱb to Ⅳ cervical carcinoma. The expression of CK19 mRNA in patients with FIGO Stage Ⅰ A to Ⅱ A cervical carcinoma were significantly correlated with stage, differentiation and lymph vascular space involvement, but was not associated with prognostic factors including age, bully tumor size, pathological types, deep stromal invasion and lymph node metastasis. In multivariate survival analysis, lymph vascular space involvement was an independent risk factor of CK19mRNA expression in patients with cervical carcinoma. Conclusions Fluorescent quantitative RTPCR can be used to detect the expression of CK19 in the peripheral blood of cervical carcinoma patients, and it is a sensitive and specific technique.CK19 mRNA in peripheral blood may be a potential biomarker for detecting micrometastasis in cervical carcinoma. The results suggest a possible use of this approach for evaluating prognosis.

RESUMO

Objective To investigate the effects of caveolin-1 overexpressing on the growth of cervical squamous cell cancer Hela cell line. Methods Eukaryotic expression vector of human caveolin-1 gene was introduced into Hela cells by Lipofectamine. The clones stably overexpressed caveolin-1 were identiffed by RT-PCR, immunofluorescence cell staining techniques and Westernblotting. Cells proliferation viabihty was tested by MTT assay, and flow cytometry was used to assay the cell cycle and apoptosis, and the relative phosphorylation level of extracellular regulated protein kinases (Erk1/2) were detected by Westernblotting. Results The clones stably overexpressed caveolin-1 were obtained. Compared with the parental Hela cells, the tranfected cells exhibited a slower rate of growth. FAGS analysis results revealed that overexpression of caveolin-1 resulted in the cell cycle arrest in the G0/G1 [ ( 68. 04 ± 2. 57 ) % vs ( 53.41 ±1.01)%] phase and increased the apoptotic cell fraction[ (19. 18 ±2.20)% vs (5.63 ±0.55)%, P <0. 05 ]. Western blotting results showed that overexpression of caveolin-1 reduced the phosphorylation of Erk1/2(0.28 ±0.05 vs 0.81 ±0.07, P <0.05). Conclusions Overexpression of caveolin-1 suppressed the growth of Hela cells and induced apoptosis, down-regulation of Erk1/2 phosphorylation might be involved in its mechanism.

RESUMO

Objective To detect COX-2 mRNA in cervical cancer and normal cervix, and explore the pathogenesis of adenocarcinoma of the uterine cervix. Methods COX-2 expression was detected by RT-PCR to explore the relationship between COX-2 and the adenocarcinogenon of cervical cancer in 36 cases of adenocarcinoma of the uterine cervix and 15 cases normal cervical tissue. Results The positive rate of COX-2 mRNA in adenocarcinoma of the uterine cervix tissues (55.6%) was higher than that of normal tissues ( 20. 0% ), the difference was significant ( P < 0. 05). The expression of COX-2 mRNA was associated with stages and clinical grading of tumor ( P < 0. 05), but no significant difference was found between stages and clinical grading of uterine cervix adenocarcinoma( P > 0. 05). Conclusion The hyper expression of COX-2 mRNA in uterine cervix adenocarcinoma took part in the carcinogenesis of tumor.
