The vulnerable status of the Amazon manatee, Trichechus inunguis, indicates the need to seek measures to guarantee its conservation. In this context, the cultivation of cells in vitro is a strategy that should at least guarantee the preservation of their genetic material. Thus, we established for the first time a primary culture of Amazonian manatee fibroblasts (TINsf) from a skin biopsy of a young male. Karyotypic analysis of the 3rd, 7th, and 12th passages confirmed the taxonomic identity of the species T. inunguis (2n = 56/NF = 92) and indicated that this culture presents genomic stability. Gene and protein expression of vimentin at the 13th passage show the predominant presence of fibroblasts in TINsf. To test the cell line's responsiveness to materials and demonstrate a possible application of this culture, it was exposed to andiroba seed oil (ASO), and its viability and proliferative capacity were evaluated. ASO demonstrated toxic effects at the highest concentrations and longest exposure times tested, reproducing results observed in human cultures, indicating the applicability of TINsf in toxicological and biotechnological studies. After cryopreservation, the TINsf line maintained its proliferative potential, indicating the establishment of a new culture available for future studies.

Carapa guianensis (Andiroba, Meliaceae) is considered a multipurpose tree. In Brazil, Indigenous people have used it as insect repellent and in the treatment of various diseases. Most biological activities and popular uses are attributed to limonoids, which are highly oxygenated tetranortriterpenoids. More than 300 limonoids have been described in Meliaceae family. Limonoids from Andiroba oil have shown high anti-inflammatory and anti-allergic activities in vivo, by inhibiting platelet activating factors and many inflammatory mediators such as IL-5, IL-1ß and TNF-α. It also reduced T lymphocytes, eosinophils and mast cells. In corroboration with the wide popular use of Andiroba oil, no significant cytotoxicity or genotoxicity in vivo was reported. This oil promotes apoptosis in a gastric cancer cell line (ACP02) at high concentrations, without showing mutagenic effects, and is suggested to increase the body's nonspecific resistance and adaptive capacity to stressors, exhibit some antioxidant activity, and protect against oxidative DNA damages. Recently, new methodologies of toxicological assays have been applied. They include in chemico, in vitro, in silico and ex vivo procedures, and take place to substitute the use of laboratory animals. Andiroba by-products have been used in sustainable oil production processes and as fertilizers and soil conditioners, raw material for soap production, biodegradable surfactants and an alternative natural source of biodegradable polymer in order to reduce environmental impacts. This review reinforces the relevance of Andiroba and highlights its ability to add value to its by-products and to minimize possible risks to the health of the Amazonian population.

Chitosan comprises polymeric macromolecules with technical and biological properties that have been used in biomedical healing applications requiring anti-microbial and anti-inflammatory capacities worldwide. In the tropical regions, green banana peel extract and andiroba oil are considered natural products with wound healing properties. The present study, for the first time, synthesized chitosan/green banana peel extract/andiroba oil (CGA) membranes and analyzed them using scanning electron microscopy (SEM) and the swelling and moisture tests. The CGA membranes together with control membranes of plain chitosan and chitosan plus green banana peel extract, were characterized by contact angle measurement, X-ray diffraction (XRD) and differential scanning calorimetry (DSC). Macroscopic analysis showed significant differences in color and transparency caused by the number of decoction days used for extract preparation and the oil content. SEM observations disclosed the formation of two phases, lipid and polymer, in the CGA. The number of decoction days and the andiroba oil content were inversely related to the swelling moisture uptake. All membranes were found to be hydrophilic with contact angles less than 90°. The incorporation of plant extract and oil promoted the appearance of related XRD peaks. DSC curves revealed a reduction in the enthalpy of the CGA membranes compared with plain chitosan, which might be attributed to the evaporation of the natural extract and oil. Based on these findings, the studied newly synthesized membranes demonstrated a potential for healing epithelial lesions.

This studys objective was to evaluate the efficacy of treating cutaneous wounds in Wistar rats using a therapeutic laser alone or in combination with topical application of andiroba oil. Twenty-four Wistar rats were distributed into three groups (T4, T7, and T14) of eight animals. To prepare the wounds, a total of four skin fragments per animal were removed using an 8-mm cutaneous biopsy punch. Each animal was inflicted with four surgical wounds, and each wound was subjected to one treatment. The treatments were as follows: saline solution (control, Cn); laser therapy (L), using a 660-nm laser wavelength and 10-J/cm² energy density; fresh andiroba oil (An); laser therapy followed by topical andiroba oil administration (LAn). All treatments in all animals were conducted for 4- (T4), 7- (T7), and 14- (T14) day periods. Edema and purulent secretion were observed in three animals in the An group, and the appearance of an exuberant crust was also observed in one animal from the same group. The LAn group presented the worst wound healing rate and contraction velocity (p < 0.05). Microscopically, there was no difference between groups regarding the presence of inflammation, necrosis, formation of granulation tissue, fibroplasia, and the presence of types 1 and 3 collagen at different treatment times. It was concluded that laser treatment of cutaneous wounds in conjunction with andiroba oil application did not present benefits in reference to the 0.9% NaCl.(AU)


O objetivo deste estudo foi avaliar a eficácia do tratamento de feridas cutâneas em ratos Wistar utilizando o laser terapêutico em associação ou não com o óleo de andiroba. Vinte e quatro ratos foram avaliados em três tempos de tratamento (T4, T7 e T17), sendo oito animais em cada tempo. Para a realização das feridas cirúrgicas, quatro fragmentos de pele foram removidos com punch de biopsia de 8 milímetros. Cada animal recebeu quatro feridas e cada ferida foi tratada com um tra-tamento: solução salina (controle, Cn); laserterapia (L), usando um laser com comprimento de onda de 660nm e 10J/cm2 de densidade de energia; óleo de andiroba in natura (An); e laserterapia seguida pela administratação tópica de óleo de andiroba (LAn). Todos os tratamentos foram realizados e avaliados nos dias quatro (T4), sete (T7) e 14 (T14) de pós operatório. Foram observados edema e secreção purulenta em três animais do grupo An, e a presença de crosta exuberante também foi obervada em um animais do mesmo grupo. O grupo LAn apresentou a pior cicatraização e velocidade de contração da ferida (p < 0,05). Não houve diferença entre os grupos à avaliação microscópica da presença de inflamação, necrose, formação de tecido de gra-nulação, fibroplasia e presença de colágeno tipo 1 e tipo 3 nos diferentes tempos de tratamento. Foi possível concluir que o tratamento com laseterapia em associação ao óleo de andiroba não apresentou benefícios em relação a utilização da solução de cloreto de sódio a 0,9%.(AU)

Developing a biomimetic material to wound care is an emerging need for the healing process. Poly (ε-caprolactone) (PCL) is a polymer with the necessary dressing's requirements often used in medicine. Their surface, physic-chemical and biological properties can be modified by adding bioactive compounds, such as andiroba seed oil (Carapa guianensis). This Amazonian natural plant has medicinal and pharmacological properties. For this purpose, PCL polymeric films incorporated with andiroba oil were investigated. The synthesis of hybrids materials was carried out in the solvent casting method. Thermal properties were evaluated using thermogravimetric analysis (TGA/DTGA) and differential scanning calorimetry (DSC). The solvent type on the surface and hydrophilicity of samples was studied using a scanning electron microscope (SEM). Additionally, contact angle measurements, functional groups analysis, fluid absorption capacity, and cell viability were performed. The results demonstrated the influences of andiroba oil under the morphology and thermal properties of the polymeric matrix; the hydrophilicity of the hybrid film obtained by acetic acid was reduced by 13%; the porosity decreased as the concentration of oil increased, but its higher thermal stability. The L929 cell line's proliferation was observed in all materials, and it presented nontoxic nature. It was demonstrated the ability of PCL hybrid film as a matrix for cell growth. Then, the materials were proved potential candidates for biomedical applications.

Problems related to oil authenticity make it difficult to obtain the benefits associated with each type of vegetable oil. Fraudulent practices have been revealed by several targeted and nontargeted methods. In this paper, spectroscopic techniques (FT-IR, 1H NMR, and 13C NMR) were applied to determine the chemical profiles of 23 Brazilian commercial vegetable oils obtained from five different high-value aggregated matrices (andiroba, babassu, baru, castor, and sweet almond oils) and investigate their adulteration, by comparison with the corresponding reference samples. Each technique is useful for the particular information it provides: differences in free fatty acids by FT-IR; adulteration with omega-3-enriched oils by 1H NMR, and adulteration of unsaturated-enriched oil with another unsaturated oil without linoleic acid by regiospecific analysis. Our findings highlight the importance of fusion-based methods in providing precise information for use in oil quality authentication.

Polyembryony is the differentiation and development of multiple embryos in a single seed. This characteristic can provide advantages, as more than one embryo is produced with the same amount of resources, and the probability of establishment of at least one seedling increases. However, sibling seedlings may also increase competition, affecting development and survival. In the present study, the possible advantages and disadvantages of polyembryony were analyzed in the initial establishment of seedlings of Carapa surinamensis (Meliaceae), a tree species that produces monoembryonic or polyembryonic seeds. In this regard, the development of single seedlings was compared with a pair of seedlings emerging from polyembryonic seeds. We compared the development of seedlings attached to or detached from each other and to the seed resources. We observed two levels of competition: (a) for the seed reserves during germination and initial development, as multiple embryos of C. surinamensis share the same reserves, and (b) for external factors, mostly space for root and shoot development, and also for light. Reducing the competition for external factors by separating the siblings was not enough to reduce the effects of competition for seed reserves in the first six months of development. Nevertheless, viable seedlings were produced in all treatments. Thus, depending on sprout management in the nursery, the number of seedlings per seed can be significantly increased by detaching the seedlings, or more vigorous seedlings can be obtained when only one seedling is maintained.(AU)

Esta característica pode proporcionar diversas vantagens, como aumentar o número de embriões produzidos com a mesma quantidade de recursos, e aumentar a probabilidade de estabelecimento de pelo menos uma plântula de uma única semente. Por outro lado, a competição entre plântulas pode aumentar, afetando seu desenvolvimento e sobrevivência. Neste estudo, foram analisadas as possíveis vantagens e desvantagens da poliembrionia em sementes de andiroba, Carapa surinamensis (Meliaceae), uma espécie arbórea que produz sementes monoembriônicas ou poliembriônicas. Comparamos o desenvolvimento de plântulas únicas com o de pares de plântulas provenientes de sementes poliembriônicas. As plântulas foram mantidas unidas ou separadas entre si e ligadas à ou destacadas da semente. Os resultados revelaram dois níveis de competição: (a) pelas reservas da semente durante a germinação e desenvolvimento inicial da plântula, quando embriões múltiplos de C. surinamensis compartilham as mesmas reservas, e (b) por fatores externos, principalmente espaço para o desenvolvimento da raiz e da parte aérea, e luz. A redução da competição por fatores externos, através da separação das plântulas, não foi suficiente para reduzir os efeitos da competição pelas reservas das sementes nos primeiros seis meses de desenvolvimento. Apesar disso, plântulas viáveis foram produzidas em todos os tratamentos. Abstract Species of the Carapa spp. complex, occurring in the Neotropics, Africa and India, have multiple uses, including timber, with the seed oil being used in phyto-pharmaceutical products and cosmetics. This study aimed to determine the thermal ranges of the germination process, comparing germination criteria used by seed physiologists and seed technologists, and to suggest recommendations for seed quality assessment. Germination was assessed at constant temperatures between 10 ─ 40 °C using three germination criteria: (1) radicle length ≥ 0.5 cm (physiological criterion); (2) epicotyl length ≥ 1 cm; and (3) epicotyl length ≥ 5 cm (criterion for seed quality tests). The base temperature was similar for the three criteria and ranged between 10 ─ 2 °C. The Maguire's Speed Index indicated 30 °C as most adequate. However, the upper temperature limit differed: for radicle protrusion it was above 40 ºC; and for both epicotyl lengths, it was between 35 ─ 40 °C. Seed coat removal accelerated the germination process of these recalcitrant seeds, and is recommended for seed quality assessment, which allows completion of the germination trial in approximately one month.


Resumo As espécies do complexo Carapa spp. ocorrem na região Neotropical, na África e na Índia, têm usos múltiplos, fornece madeira de valor comercial e o óleo extraído das sementes tem uso fitoterápico e cosmético. O objetivo deste trabalho foi determinar a faixa térmica tolerável do processo germinativo, comparando os critérios de germinação utilizados pelos fisiologistas e os tecnólogos de sementes, e sugerir recomendações para a avaliação da qualidade das sementes. A germinação foi avaliada em temperaturas constantes entre 10 e 40 °C utilizando três critérios de germinação: (1) formação da radícula ≥ 0,5 cm (critério fisiológico); (2) alongamento de epicótilo ≥ 1 cm; e (3) alongamento de epicótilo ≥ 5 cm (critério para testes de qualidade de sementes). A temperatura de base foi semelhante para os três critérios entre 10 e 12 °C. O índice de velocidade de Maguire indicou 30 °C como a temperatura mais adequada. O limite superior de temperatura diferiu entre os critérios, sendo acima de 40 ºC para protrusão da radícula e para ambos os alongamentos de epicótilo entre 35 e 40 °C. A remoção do tegumento de semente acelerou o processo de germinação dessas sementes recalcitrantes sendo recomendada para a avaliação da qualidade da semente, o que permite concluir o teste de germinação em aproximadamente um mês.

Plant products are an important source of bioactive agents against parasitic diseases, including leishmaniasis. Among these products, vegetable oils have gained ground in the pharmaceutical field. Here we report the development of nanoemulsions as a delivery system for copaiba and andiroba oils (nanocopa and nanoandi) in order to test their effects on Leishmania infantum and L. amazonensis. The nanocopa and nanoandi had an average particle size of 76.1 and 88.1, respectively with polydispersity index 0.14 to 0.16 and potential zeta -2.54 to -3.9. The data indicated toxic activity of nanocopa and nanoandi against promastigotes of both Leishmania species ultrastructural analyses by scanning electron microscopy revealed that exposition to nanoemulsions induced oval cell shape and retracted flagella. The treatment with nanocopa and nanoandi led to a reduction in L. infantum and L. amazonensis infection levels in macrophage cultures. The nanoemulsions treatment have significant beneficial effects on all the parameters evaluated in lesions induced by L. amazonensis (lesion size, parasite burden and histopathology) on BALB/c mice. The treatment of L. infantum-infected BALB/c mice with nanoemulsions also showed promising results reducing parasite burden in spleen and liver and improving histopathological features.

Seed coat removal accelerated the germination process of these recalcitrant seeds, and is recommended for seed quality assessment, which allows completion of the germination trial in approximately one month.

O objetivo deste trabalho foi determinar a faixa térmica tolerável do processo germinativo, comparando os critérios de germinação utilizados pelos fisiologistas e os tecnólogos de sementes, e sugerir recomendações para a avaliação da qualidade das sementes. O objetivo deste trabalho foi estudar a morfologia polínica das três espécies de Carapa (Meliaceae) registradas na Amazônia brasileira. Os grãos de pólen foram obtidos de botões florais, flores pistiladas e estaminadas de Carapa guianensis, C. surinamensis e C. vasquezii. A fim de explorar as implicações taxonômicas do estudo de pólen para estas espécies de Carapa, o material foi preparado de acordo com o método de acetólise láctico, permitindo a utilização de estudos de microscopia de luz. Além disso, o material não-acetolisado foi utilizado no estudo da microscopia eletrônica de varredura. O estudo mostra que as três espécies de Carapa apresentam pequenas variações na morfologia polínica. Comparada com as outras espécies deste estudo, Carapa vasquezii têm valores mais elevados para o comprimento e largura da endoabertura, entretanto, tem valores inferiores para diâmetro polar, equatorial, diâmetro equatorial em vista polar e lado do apocolpo.(AU)

This study aimed to evaluate the insecticidal and repellent effects of tea tree, Melaleuca alternifolia (Myrtales: Myrtaceae), and andiroba, Carapa guianensis (Sapindales: Meliaceae), essential oils on two species of fly. For in vitro studies, free-living adult flies were captured and reared in the laboratory. To evaluate the insecticidal effects of the oils, adult flies of Haematobia irritans (L.) and Musca domestica L. (both: Diptera: Muscidae) were separated by species in test cages (n = 10 per group), and subsequently tested with oils at concentrations of 1.0% and 5.0% using a negative control to validate the test. Both oils showed insecticidal activity. Tea tree oil at a concentration of 5.0% was able to kill M. domestica with 100.0% efficacy after 12 h of exposure. However, the effectiveness of andiroba oil at a concentration of 5.0% was only 67.0%. The insecticidal efficacy (100.0%) of both oils against H. irritans was observed at both concentrations for up to 4 h. The repellency effects of the oils at concentrations of 5.0% were tested in vivo on Holstein cows naturally infested by H. irritans. Both oils demonstrated repellency at 24 h, when the numbers of flies on cows treated with tea tree and andiroba oil were 61.6% and 57.7%, respectively, lower than the number of flies on control animals. It is possible to conclude that these essential oils have insecticidal and repellent effects against the species of fly used in this study.

An integrative approach in mass spectrometry (MS) comprising gas chromatography coupled to MS (GC-MS), ultra-efficiency liquid chromatography coupled to MS (UPLC-MS) and easy ambient sonic-spray ionization MS (EASI-MS) is proposed for the comprehensive characterization of Amazonian oils. Coconut, andiroba and castor seed oils, which are vastly sold in markets of the Amazonian region of Brazil, were selected as a representative test set. These oils were found to contain several lipids such as triacylglycerides (TAGs), fatty acids (FAs), phytosterols and limonoids. In the analyzed samples 30 different TAGs, 11 FAs, 6 phytosterols and 7 limonoids were identified. The antioxidant capacity (AOC) of the oils, as measured by their oxygen radical absorbance capacity (ORAC), was also used to evaluate their potential biological properties as well as their possible consumption as food. Edible virgin coconut oil was the most active (0.720±0.001 Trolox eq./mmol), whereas considerable lower activity was observed for andiroba and castor seed oils. The antimicrobial activities of the oils were also recorded against a panel of pathogenic bacteria and fungi in which andiroba oil was the only one that was active, solely against Enterococcus aeruginosa.

Today, one of the trends of the pharmaceutical, cosmetic and food market is the development of products with components of natural origin, rationally exploiting biodiversity. Brazilian population makes secular use of medicinal plants including andiroba, whose oil is used in folk medicine as febrifuge, pain-relieving, anti-parasitic, anti-allergic as well as insect repellant. The present study attempts to evaluate the profile of utilization of andiroba by analyzing the patenting trends based on information collected on the databases of the World Intellectual Property Organization, European Patent Office and Brazilian National Institute of Industrial Property in the period from 1990 to 2011. The following parameters were analyzed: chronological aspect of the applications, countries of priority, international patent classification, technologies and actors in the technological platform. The temporal analysis of the applications shows an evident increase despite a discontinuous evolution of the number of applications. Pharmaceutical, chemical and cosmetic areas were identified as the main areas for commercial application of the plant. Brazil is the country with the largest number of applications even though the majority of the patent technologies are already in public domain, indicating that the technological information contained in these documents could be used for research and investment in several areas.

This study was perfomed to assess the in vitro effect of oil from the seed of andiroba (Carapa guianensis Aubl.) on Felicola subrostratus (Burmeister, 1838) (Mallophaga: Trichodectidae). Six hundred specimens of F. subrostratus from neighborhood of Jordão, Recife-PE, Brazil, were collected by hand directly from the fur of cats infested naturally. The lice were transported in plastic recipients to the Laboratory of Parasitic Diseases of Domesticated Animals of the Department of Veterinary Medicine of the Universidade Federal Rural de Pernambuco (Brazil) for the immersion test. Four dilutions of andiroba oil (100, 50, 25 and 10%) in distilled water were tested, using Tween 80 as dispersant; two control groups (one with distilled water and the other with monosulfiram) were also formed; four replicates were performed, with 25 specimens for each dilution, totaling 100 lice per treatment. After the test, the insects were kept at room temperature and observed for mortality rates for 72 h. The biological activity of the product achieved 100% mortality of the insects in the first hour at concentrations of 100 and 50% and in the third hour at concentrations of 25 and 10%. The results demonstrate the possibility of controlling F. subrostratus throught the use of oil extracted from andiroba seeds.
