RESUMO

Seventy-three percent of aerial insectivore species of birds breeding in North America have declined in the past five years. This decline is even greater in migratory insectivorous species, which face stressors in both their breeding and non-breeding ranges. The Purple Martin (Progne subis) is an aerial insectivore swallow that overwinters in South America and migrates to North America to breed. Purple Martin populations have declined by an estimated 25% since 1966. The eastern subspecies (P. subis subis) has declined the most and overwinters in the Amazon Basin, a region rich in environmental mercury (Hg) contamination. Previous studies reported elevated levels of Hg in feathers of this subspecies, which correlated negatively with body mass and fat reserves. Given the propensity of Hg to disrupt the endocrine system, and the role of thyroid hormones in regulating fat metabolism, this study quantifies concentrations of Hg and the thyroid hormone triiodothyronine (T3) in the feathers of P. subis subis. To our knowledge, this is the first study to extract and quantify T3 in feathers; thus, we developed, tested, and optimized a method for extracting T3 from feather tissue and validated an enzyme immunoassay (EIA) to quantify T3 in Purple Martin feathers. The developed method yielded acceptable results for both parallelism and accuracy. The observed T3 concentrations were statistically modeled along with total Hg (THg) concentrations, but these variables were not significantly correlated. This suggests that the observed variation in THg concentration may be insufficient to cause a discernible change in T3 concentration. Furthermore, the observed effect of breeding location on feather T3 concentration might have obscured any effect of Hg.

The eastern North American subspecies of Purple Martin (P. subis subis) comprises >90% of all Purple Martins. This subspecies overwinters and molts in the Amazon Basin, a region that is high in mercury (Hg) contamination, which raises the possibility that observed declines in Purple Martins could be linked to Hg exposure. Exposure to Hg results in numerous and systemic negative health outcomes, including endocrine disruption. Corticosterone (CORT) is a primary modulator of the stress and metabolic axes of vertebrates; thus, it is important in meeting metabolic and other challenges of migration. Because feathers accumulate Hg and hormones while growing, quantification of Hg and CORT in feathers provides an opportunity to retrospectively assess Hg exposure and adrenal activity of birds using minimally invasive methods. We evaluated interrelationships among concentrations of total Hg (THg) and CORT in feathers that grew in the Amazon Basin and body condition (mass, fat score) of these birds in North America. Concentrations of THg in Purple Martin feathers ranged from 1.103 to 8.740 µg/g dw, levels associated with negative physiological impacts in other avian species. Concentrations of CORT did not correlate with THg concentration at the time of feather growth. However, we found evidence that THg concentration may negatively impact the ability of Purple Martins to accumulate fat, which could impair migratory performance and survivorship due to the high energy requirements of migration. This finding suggests potential carryover effects of Hg contamination at the wintering grounds in the Amazon to the summer breeding grounds in North America.

The field-based distribution and bioaccumulation factor (BAF) for per- and polyfluoroalkyl substances (PFASs) were determined in residential Black Swans (Cygnus atratus) from an urban lake (Melbourne, Australia). The concentrations of 46 aliphatic and cyclic PFASs were determined by HPLC-MS/MS in serum and excrement from swans, and water, sediment, aquatic macrophytes, soil, and grass samples in and around the lake. Elevated concentrations of ∑46PFASs were detected in serum (120 ng mL-1) and excrement (110 ng g-1 dw) were strongly related indicating a potential noninvasive sampling methodology. Environmental concentrations of PFASs were consistent with a highly impacted ecosystem and notably high concentrations of perfluoro-4-ethylcyclohexanesulfonate (PFECHS, 67584-42-3; C8HF15SO3) were detected in water (27 ng L-1) and swan serum (16 ng mL-1). In the absence of credible putative alternative sources of PFECHS input to the lake, we propose that the use of high-performance motorsport vehicles is a likely source of contamination to this ecosystem. The BAF of perfluorocarboxylic acids increased with each additional CF2 moiety from PFOA (15.7 L kg-1 ww) to PFDoDA (3615 L kg-1 ww). The BAF of PFECHS was estimated as 593 L kg-1 ww, which is lower compared with that of PFOS (1097 L kg-1 ww).

Endocrine-disrupting chemicals can cause transcriptomic changes that may disrupt biological processes associated with reproductive function including metabolism, transport, and cell growth. We investigated effects from in ovo and dietary exposure to 17ß-trenbolone (at 0, 1, and 10 ppm) on the Japanese quail (Coturnix japonica) hepatic transcriptome. Our objectives were to identify differentially expressed hepatic genes, assess perturbations of biological pathways, and examine sex- and developmental stage-related differences. The number of significantly differentially expressed genes was higher in embryos than in adults. Male embryos exhibited greater differential gene expression than female embryos, whereas in adults, males and females exhibited similar numbers of differentially expressed genes (>2-fold). Vitellogenin and apovitellenin-1 were up-regulated in male adults exposed to 10 ppm 17ß-trenbolone, and these birds also exhibited indications of immunomodulation. Functional grouping of differentially expressed genes identified processes including metabolism and transport of biomolecules, enzyme activity, and extracellular matrix interactions. Pathway enrichment analyses identified as perturbed peroxisome proliferator-activated receptor pathway, cardiac muscle contraction, gluconeogenesis, growth factor signaling, focal adhesion, and bile acid biosynthesis. One of the primary uses of 17ß-trenbolone is that of a growth promoter, and these results identify effects on mechanistic pathways related to steroidogenesis, cell proliferation, differentiation, growth, and metabolism of lipids and proteins. Polycyclic aromatic hydrocarbons (PAHs) impaired pre-migratory fueling in 49 orally dosed Sanderling (Calidris alba). In the present study, 8 genes related to fat deposition and PAH exposure were measured in liver subsamples from these same shorebirds. At the highest dose (1260 µg total PAH [tPAH]/kg body wt/day), PAH exposure decreased liver basic fatty acid binding protein 1 (Lbfabp) and hepatic lipase (Lipc) expression. We utilized lead (Pb) stable isotopes to identify the source of acute Pb poisoning in a Tundra Swan (Cygnus columbianus) and evaluated overall Pb exposure. Upon necropsy, we obtained samples of blood, liver, kidney, heart, thigh, breast, femur, and metallic objects (i.e., fishing sinker, spring and swivels) from the gizzard for Pb isotopic analysis. Pb isotope ratios of blood and soft tissues were essentially identical to the Pb ratios of the sinker, the likely source of acute poisoning. The spring and swivels had lower Pb content and ratios distinct from tissue, suggesting no significant contribution to poisoning. Femur Pb isotopic composition was the most distinct biological sample and indicative of a combination of sources. These results demonstrate isotopic analysis as a viable method for determining the source of acute Pb poisoning, and that Pb isotope ratios in bone most likely record a lifetime-averaged metric of Pb exposure.

Following large crude oil spills, oil from feathers of brooding birds and oiled nesting material can transfer to eggs, resulting in reduced embryonic viability for heavily oiled eggs. Eggs may also be subjected to trace or light oiling, but functional teratogenic effects from sublethal crude oil exposure have not been examined. We assessed whether sublethal application of weathered Deepwater Horizon crude oil to the eggshell surface alters heart rate and metabolic rate in Zebra Finch (Taeniopygia guttata) embryos. We first determined sublethal applications with a dosing experiment. Embryo viability for eggs exposed to 5 µL or more of crude oil decreased significantly. We conducted a second experiment to measure heart rate and metabolic rate (CO2 production) 5 and 9 d after 1 sublethal application of crude oil to eggshells on day 3 of incubation. One application of 1.0 or 2.5 µL of crude oil reduced embryonic heart rate and metabolic rate on day 12 of incubation. Using unfertilized eggs, we measured the transfer of polycyclic aromatic hydrocarbons (PAHs) from the eggshell surface to egg contents 9 d after a single application of sublethal crude oil. Our results suggest avian eggs externally exposed to small amounts of crude oil may exhibit protracted embryonic development and impaired postnatal cardiac performance.

In the present study, we investigated whether the immortalized chicken hepatocellular carcinoma cell line, leghorn male hepatoma (LMH), had a comparable aryl hydrocarbon receptor (AhR) response to primary chicken embryonic hepatocytes (CEHs) when used in a well-established assay for chemical screening and prioritization. The LMH cells were grown as 2-dimensional (2D) confluent cells and 3D spheroids to determine the optimal cell culture states for chemical screening. Cytochrome P450 1A4 and 1A5 (CYP1A) activity and gene expression were compared between CEHs and LMH cells grown in 2 culture states following exposure to the dioxin-like compound 3,3',4,4',5-pentachlorobiphenyl (PCB-126). The CYP1A activity was measured using the ethoxyresorufin-O-deethylase (EROD) assay, and changes in mRNA expression associated with the AhR pathway were determined using a custom-designed polymerase chain reaction array. Among LMH cell culture states (i.e., 2D vs 3D), EROD induction was observed only in 3D LMH spheroids. Similarly, 3D spheroids had the greatest number of changes in AhR-related genes compared with confluent cells. Overall, these results suggest that LMH cells grown as 3D spheroids have a metabolic and gene expression profile that is comparable to that of CEH, and may represent a suitable animal-free alternative for in vitro screening of chemicals. Bird eggs are often used to monitor levels and trends of mercury (Hg) in the environment. Logistical issues in such studies sometimes require the use of eggs at different stages of development. Therefore, it is important to understand how embryonic age may affect egg Hg concentrations. Studies using dry weight Hg concentrations in eggs assume that dry mass (e.g., lipids) does not change through embryonic development and thus expressing concentrations on a dry weight basis adjusts for any changes that may occur in egg mass throughout development. That assumption is tested in the present study by injecting chicken eggs with Hg and measuring Hg concentrations in embryonic contents at different stages of artificial incubation. The results indicate that dry weight Hg concentrations in eggs at later stages of development are statistically greater than in undeveloped eggs. To address this issue, we developed a method of standardizing egg Hg concentrations for embryonic age based on egg contents density (egg contents mass/egg contents volume). A combined dataset (n = 225) from precocial and semi-precocial birds was used to demonstrate that egg contents density declines with increasing embryonic development. Based on this finding, we applied a formula to adjust egg dry weight Hg concentrations for stage of development: adjusted egg [Hg] = unadjusted [Hg] × (egg contents density). The application of this approach to adjust data from the chicken egg Hg injection study resulted in there being no statistically significant difference in dry weight Hg levels in eggs at different stages of embryonic development. The adjustment approach described in the present study is a straightforward method to account for stage of development when assessing Hg levels in avian eggs and should be widely applicable to studies of this nature. Organophosphorous flame retardants (OPFRs) are contaminants of emerging concern. There is growing evidence of environmental contamination and exposures to both humans and wildlife. Here, the objective was to increase understanding of the potential neurodevelopmental effects of two relevant OPFRs, TMPP (tri (methylphenyl) phosphate; a non-halogen-containing OPFR) and TDCIPP (tris (1,3-dichloro-isopropyl) phosphate; a halogen-containing OPFR) in an avian embryo/chick model. We injected white leghorn chicken eggs with a range of TMPP (0, 10, 100, and 1000 ng/g) or TDCIPP (0, 10, 100, 1000, 50,000 ng/g) concentrations at incubation day 0 exposing embryos throughout the ~21-day in ovo period. Hatching success was unaffected by TMPP, but TDCIPP-exposed chicks had higher early-incubation mortality in 100 and 50,000 ng/g groups. On 7-9-day-old chicks, we assessed behavior via tests concerning righting reflex, angled balance beams, gait patterns, wing flap reflex, and open field movements. Chicks exposed to 100 ng/g TDCIPP achieved 40% lower maximum velocity in the open field test than vehicle-exposed controls, while those exposed to 1000 ng/g TDCIPP achieved 20% higher maximum velocity than vehicle-exposed controls. Chicks exposed to 50,000 ng/g TDCIPP showed reduced righting response success. There were no dose- or treatment-related differences in angled beam, gait analysis, or wing flap reflex tests. Cerebrum hemispheres from 10-day-old chicks were examined for neurochemistry (acetylcholinesterase [AChE] activity and both nicotinic [nACh] and muscarinic [mACh] acetylcholine receptor levels) and cerebellums were examined for histopathology. TDCIPP-exposed chicks had reduced number of degenerate Purkinje cells (TDCIPP, 1000 ng/g), possibly indicating disruption of neurodevelopment. No neurochemical effects were found in TMPP- or TDCIPP-exposed chicks. In general this study shows some possible neurodevelopmental effects in chicks exposed to TDCIPP when levels greatly exceeded those measured in wild bird eggs and no clear changes in TMPP-exposed chicks. This study builds upon previous in vitro studies as well as work on adult birds showing that toxic responses in avian models can vary among species and OPFRs.

Environmental safety assessments for exposure of birds require the provision of acute avian toxicity data for both the pesticidal active substance and formulated products. As an example, testing on the formulated product is waived in Europe using an assessment of data for the constituent active substance(s). This is often not the case globally, because some countries require acute toxicity tests with every formulated product, thereby triggering animal welfare concerns through unnecessary testing. A database of 383 formulated products was compiled from acute toxicity studies conducted with northern bobwhite (Colinus virginianus) or Japanese quail (Coturnix japonica) (unpublished regulatory literature). Of the 383 formulated products studied, 159 contained only active substances considered functionally nontoxic (median lethal dose [LD50] > highest dose tested). Of these, 97% had formulated product LD50 values of >2000 mg formulated product/kg (limit dose), indicating that no new information was obtained in the formulated product study. Furthermore, defined (point estimated) LD50 values for formulated products were compared with LD50 values predicted from toxicity of the active substance(s). This demonstrated that predicted LD50 values were within 2-fold and 5-fold of the measured formulated product LD50 values in 90% and 98% of cases, respectively. This analysis demonstrates that avian acute toxicity testing of formulated products is largely unnecessary and should not be routinely required to assess avian acute toxicity. In particular, when active substances are known to be functionally nontoxic, further formulated product testing adds no further information and unnecessarily increases bird usage in testing. A further analysis highlights the fact that significant reductions (61% in this dataset) could be achieved by using a sequential testing design (Organisation for Economic Co-operation and Development test guideline 223), as opposed to established single-stage designs.
