RESUMO

Different levels of metabolizable energy (ME) and the inclusion of guanidinoacetic acid (GAA) in the diet of 53-week-old Lohmann LSL-CLASSIC hens were used to evaluate its effect on reproductive parameters, egg quality, intestinal morphology, and the immune response. Six diets were used in a 3 × 2 factorial design, with three levels of ME (2850, 2800, and 2750 kcal/kg), and with (0.08%) or without the inclusion of GAA. The addition of GAA to diets with low levels of ME increased (p < 0.05) egg production and egg mass. Moreover, hens fed with 2800 kcal/g without GAA had the highest concentration (p < 0.05) of serum interleukin IL-2, while those fed diets with the same amount of ME but supplemented with 0.08% GAA had the lowest concentration. Finally, the inclusion of 0.08% GAA increased (p < 0.05) the concentration of vascular endothelial growth factor (VEGF), regardless of the ME level in the diet. This study highlights the potential role of GAA in decreasing the energy level of ME (50-100 kcal/g) in the feeding of hens and in the modulation of specific immune responses. Further research is recommended to fully understand the mechanisms of action of GAA on the mechanism target of rapamycin and its relationship with the immune response.

RESUMO

Photoplethysmography (PPG) signals have been widely used in evaluating cardiovascular biomarkers, however, there is a lack of in-depth understanding of the remote usage of this technology and its viability for underdeveloped countries. This study aims to quantitatively evaluate the performance of a low-cost wireless PPG device in detecting ultra-short-term time-domain pulse rate variability (PRV) parameters in different postures and breathing patterns. A total of 30 healthy subjects were recruited. ECG and PPG signals were simultaneously recorded in 3 min using miniaturized wearable sensors. Four heart rate variability (HRV) and PRV parameters were extracted from ECG and PPG signals, respectively, and compared using analysis of variance (ANOVA) or Scheirer-Ray-Hare test with post hoc analysis. In addition, the data loss was calculated as the percentage of missing sampling points. Posture did not present statistical differences across the PRV parameters but a statistical difference between indicators was found. Strong variation was found for the RMSSD indicator in the standing posture. The sitting position in both breathing patterns demonstrated the lowest data loss (1.0 ± 0.6 and 1.0 ± 0.7) and the lowest percentage of different factors for all indicators. The usage of commercial PPG and BLE devices can allow the reliable extraction of the PPG signal and PRV indicators in real time.

RESUMO

The generation albumin-based nanocarriers by precipitation from solution has great interest in the formulation of advanced nutritional products. Microfluidic techniques enable the implementation of low energy and continuum processes, with fast mass transfer and homogeneous mixing at the microscale. Here we describe the microfluidic generation of curcumin-loaded alpha lactalbumin nanoparticles in a simple and inexpensive way, by using off-the-shelf devices designed to produce solvent-shifting nanoprecipitation in core-sheath flows driven by gravity, which has not been reported before. Nanoparticles were characterized by dynamic light scattering, electron microscopy, and infrared spectroscopy. The microfluidic operating conditions were defined by theory and experiments, and the critical parameters controlling the nanoparticles diameter were identified. The prepared nanoparticles resulted practically monodisperse, the curcumin entrapment efficiency was about 40 %, and almost 70 % of the bioactive was gradually delivered in release experiments. The proposed methodology is a promising route to scale up the microfluidic elaboration of nanoparticles for the entrapment of active ingredients.

RESUMO

Objective. In cities situated at high-altitude, the measured x-ray spectra are required to calculate the absorbed dose to water in radiobiology and medical dosimetry because of the air-density effect. This work aims to measure, in a high-altitude city, a set of x-ray spectra from an x-ray tube with tungsten anode generated at potentials of the M-series x-ray beams from NIST in the range of 20 and 150 kV as well as other beam qualities that have been characterized and used for dosimetry study in our group called W-series. To also measure some spectra of mammography interest in the energy range of 25 and 35 kV using additional filtration of rhodium (Rh), molybdenum (Mo) and silver (Ag).Approach.A tungsten anode x-ray tube with potential between 10 to and 160 kV was used. A high-purity germanium (HPGe) detector associated with a DSPEC Jr 2.0TMdigital signal processing module has been used for the spectra measurements. Prior the spectra measurements, the HPGe detector has been characterized and calibrated using several radioactive sources. Mainresults. The M80 spectrum was compared to a mathematically filtered beam from Physicalisch Technische Bundesanstalt corresponding to the same potential finding a difference of 0.7% in the average energy. So, the mathematically filtered beam from PTB matches very well our M80 beam. The beams M80, M100 and M120 were also compared with calculations for the corresponding kilovoltage from an executable that calculates x-ray spectra from tungsten anode x-ray tubes in the energy range between 40 to 300 kV called SpekCalc. The SpekCalc reproduces qualitatively the experimental spectra, but not quantitatively, mainly for M100 and M120 beams.Significance.These spectra can be used to simulate the patient dose as well as image quality using Monte Carlo (MC) codes or to evaluate the absorbed dose in dosimetry studies.

RESUMO

This was a retrospective observational study to determine the secular trends in osteoporosis hip fractures in Brazil from 2004 to 2013. The fracture rates were stable for both sexes, and there was not a secular trend. Fractures were predominant in the South and Southeast Regions. The hip fracture rate was lower in this study than in other studies. These regional differences may be considered in the FRAX Brazil calibration. PURPOSE: Hip fractures are well-known osteoporotic fractures with high mortality and morbidity. Epidemiological studies in Brazil on hip fractures are scarce, and the great majority have been performed in small populations from a few cities. None of these studies has analyzed the long-term hip fracture secular trends, which are important data for the promotion of public health actions. METHODS: This was a retrospective observational study with a secular trend analysis in patients over 50 years old who were admitted to the Brazilian Public Health System from 2004 to 2013. We collected hospitalization data according to the ICD-10 for low-trauma hip fractures. The fracture rate was calculated when the patients were stratified by sex, age, and geographic region, and linear regression analysis was performed to evaluate the secular trends. RESULTS: The hip fracture rate per 100,000 inhabitants was 59.69; the rate was 74.72 in females and 42.95 in males. The fracture rates were higher in the South and Southeast Regions and steadily increased with age, and the average ratio of women-to-men was 1.74. No secular trend was detected in the overall population. Surprisingly, the secular trend only increased in the South region from 2004 to 2013, and the secular trends were stable in the other regions. CONCLUSION: Although the secular trend was similar to some worldwide studies, the hip fracture rate was lower than that previously observed in regional studies in Brazil. These regional differences may be considered in the FRAX Brazil calibration.

RESUMO

Adjustment to energy starvation is crucial to ensure growth and survival. In Arabidopsis thaliana (Arabidopsis), this process relies in part on the phosphorylation of the circadian clock regulator bZIP63 by SUCROSE non-fermenting RELATED KINASE1 (SnRK1), a key mediator of responses to low energy. We investigated the effects of mutations in bZIP63 on plant carbon (C) metabolism and growth. Results from phenotypic, transcriptomic and metabolomic analysis of bZIP63 mutants prompted us to investigate the starch accumulation pattern and the expression of genes involved in starch degradation and in the circadian oscillator. bZIP63 mutation impairs growth under light-dark cycles, but not under constant light. The reduced growth likely results from the accentuated C depletion towards the end of the night, which is caused by the accelerated starch degradation of bZIP63 mutants. The diel expression pattern of bZIP63 is dictated by both the circadian clock and energy levels, which could determine the changes in the circadian expression of clock and starch metabolic genes observed in bZIP63 mutants. We conclude that bZIP63 composes a regulatory interface between the metabolic and circadian control of starch breakdown to optimize C usage and plant growth.

RESUMO

Abstract In a healthy athlete, the caloric intake is sufficient for sports energy needs and body physiological functions, allowing a balance between energy availability, bone metabolism, andmenstrual cycle.Onthe other hand, an imbalance causedby low energy availability dueto a restrictive diet, eating disorders or long periods of energy expenditure leads to multisystemic deregulation favoring the essential functions of the body. This phenomenon, described as the female athlete triad, occurs in a considerable percentage of high-performance athletes, with harmful consequences for their future. The present review was carried out based on a critical analysis of themost recent publications available and aims to provide a global perception of the topic relative energy deficit in sport (RED-S). The objective is to promote theacquisition ofmore consolidated knowledgeon an undervaluedtheme, enabling the acquisition of preventive strategies, early diagnosis and/or appropriate treatment.

Lipid nanocarriers have a great potential for improving the physicochemical characteristics and behavior of poorly water-soluble drugs, such as aqueous dispersibility and oral bioavailability. This investigation presents a novel nanostructured lipid carrier (NLC) based on a mixture of solid lipid glycerides, fatty acid esters of PEG 1500 (Gelucire® 44/14), and an oil mix composed of capric and caprylic triglycerides (Miglyol® 812). These NLCs were developed by a simple low-energy method based on melt emulsification to yield highly encapsulating and narrowly distributed nanoparticles (~100 nm, PdI = 0.1, and zeta potential = ~-10 mV). Rhodamine 123 was selected as a poorly water-soluble drug model and owing to its spectroscopic properties. The novel NLCs were characterized by dynamic light scattering (DLS), zeta potential, nanoparticle tracking analysis (NTA), transmission electron microscopy (TEM), differential scanning calorimetry (DSC), and colloidal stability. The drug release was determined through a dialysis bag and vertical Franzs' cells to provide insights about the methods' suitability, revealing similar performance regardless of their different fluid dynamics. Rhodamine 123 followed a characteristic biphasic release profile owing to the swelling of the hydrophilic polymer coating and diffusion process from the lipid core as revealed by the Korsmeyers-Peppas kinetic modeling. Moreover, to elucidate the formation and incorporation of Rhodamine 123 into the NLC core, several molecular dynamics simulations were conducted. The temperature was shown to be an important condition to improve the formation of the nanoparticles. In addition, the liquid lipid incorporation to the formulation forms nanoparticles with imperfect centers, in contrast to nanoparticles without it. Moreover, Miglyol® 812 improves hydrophobic molecule solubility. These results suggest the potential of novel NLC as a drug delivery system for poorly water-soluble drugs.

RESUMO

We present the chemical composition, quality parameters and antioxidant capacity of pumpkin seed oils (PSO) from Cucurbita pepo, Cucurbita maxima, and Cucurbita moschata cultivated in Brazil. In addition, PSO nanoemulsions (nanopepo, nanomax and nanomosc) were developed and their physical stabilities were assessed under long-term storage at two temperatures. Among the PSO, C. pepo presented the highest contents of polyunsaturated fatty acids, total carotenoids, and chlorophylls, but the lowest oxidative stability. Conversely, C. maxima PSO showed highest oxidative stability and total tocopherol content but the lowest chlorophyll content. Nanomax and nanopepo were more stable to droplet growth at 4 °C, while nanomosc was more stable at 25 °C. Nanopepo was the most stable formulation after the heating-cooling cycles, whereas nanomax was the most stable under centrifugation regardless the temperature. Overall, all nanoemulsions presented droplet diameter lower than 200 nm and ζ-potential approaching -30 mV until the end of storage.

RESUMO

The effects of low energy electrons in biological tissues have proved to lead to severe damages at the cellular and sub-cellular level. It is due to an increase in the relative biological effectiveness (RBE) of these electrons with a decrease in their penetration range. That is, lower the range higher will be its RBE.Therefore, accurate determination of low energy electron range becomes a key issue for radiation dosimetry. This work reports on in-water electron tracks evaluated at low kinetic energy (1-50 keV) using isotropic mono-energetic point source approach suitably implemented by different general-purpose Monte Carlo codes. For this aim, simulations were performed using PENELOPE, EGSnrc, MCNP6, FLUKA and Geant4-DNA Monte Carlo codes to obtain the particle range, R,R90,R50. Finally, evaluation of dose point kernel (DPK), as used for internal dosimetry, was carried out as an application example. Scaled dose point kernels (sDPK) were estimated for a range of mono-energetic low energy electron sources. The non-negligible differences among the calculated sDPK using different codes were obtained for energy electrons up to 5 keV. It was also observed that differences of in-water range for low-energy electrons, due to the different general-purpose Monte Carlo codes, affected the DPKs used for dosimetry by convolution approach. Finally, the 3D dosimetry was found to be almost not affected at macroscopic clinical scale, whereas non-negligible differences appeared at the microscopic level. Hence, a thorough validation of the used sDPKs have to be performed before they could be used in applications to derive any conclusions.

RESUMO

Indoor Positioning Systems (IPSs) are used to locate mobile devices in indoor environments. Model-based IPSs have the advantage of not having an exhausting training and signal characterization of the environment, as required by the fingerprint technique. However, most model-based IPSs are done using fixed model parameters, treating the whole scenario as having a uniform signal propagation. This might work for most small scale experiments, but not for larger scenarios. In this paper, we propose PoDME (Positioning using Dynamic Model Estimation), a model-based IPS that uses dynamic parameters that are estimated based on the location the signal was sent. More specifically, we use the set of anchor nodes that received the signal sent by the mobile node and their signal strengths, to estimate the best local values for the log-distance model parameters. Also, since our solution depends highly on the selected anchor nodes to use on the position computation, we propose a novel method for choosing the three best anchor nodes. Our method is based on several data analysis executed on a large-scale, Bluetooth-based, real-world experiment and it chooses not only the nearest anchor but also the ones that benefit our least-square-based position computation. Our solution achieves a position estimation error of 3 m, which is 17% better than a fixed-parameters model from the literature.

RESUMO

We studied the effects of chronic exposure to metals on energy reserves and reproduction in the clam Megapitaria squalida in two nearby populations exposed to different levels of pollution from mining operations in the Gulf of California, Mexico. Female M. squalida from San Lucas beach had good reproductive health status, whereas Santa Rosalia specimens consistently showed low energy reserves, massive oocyte resorption throughout the year, high frequencies of undifferentiated individuals, low proportions of ripe and spawning organisms, smaller and fewer oocytes per follicle, and significantly lower follicular areas. Ovarian levels of Co, Cu, Pb, Mn, and Zn were consistently higher in clams from Santa Rosalia. The poor reproductive health of clams inhabiting this site may be attributed to their long-term exposure to high Co, Cu, and Pb concentrations, as these have been shown to cause toxicity and reproductive impairments in other marine organisms.

RESUMO

PURPOSE: The most common sites of low-energy trauma fractures are the femur, vertebra, humerus, and forearm. Ankle fractures have significant morbidity and high costs for surgical procedure. Forearm fractures are common nonvertebral fractures. Forearm fractures are classified as fragility fractures and predictive for fractures at other sites, although do not allow osteoporosis diagnosis. It is controversial whether ankle fractures are osteoporosis fractures. METHODS: Retrospective observational study, with secular trend analysis, in patients over 50 years old admitted in the Brazilian Public Health System, from 2004 to 2013. We collected hospitalization data according to the ICD-10 for low-trauma ankle and forearm fractures. Fracture rate was calculated according to gender, age, and geographic region, performed linear regression analysis, and estimated fracture rates for 2030. Comparison of ankle and forearm rates was also performed, grouping them in 3-year block. ANOVA test was used to compare each block. RESULTS: Ankle fracture rate was 21.39 fractures per 100,000 inhabitants, 23.98 in females and 18.49 in males. Fracture rates were higher in the South and Southeast regions. In absolute numbers, although ankle fracture rate increased with age, there was a significant decrease in the population over 80 years old. Data showed stabilization in ankle fractures from 2004 to 2013, in women and men. In 3-year block analysis, men had higher ankle fracture rates than forearm. However, in women, forearm rates were higher than ankle. CONCLUSION: Our data suggest that ankle fractures in men would be considered as a sentinel fracture with a similar clinical impact of forearm fracture.

RESUMO

The management of Lisfranc injuries is challenging considering the broad spectrum of energy involved and highly variable clinical presentation. Despite the advances in surgical techniques, subtle Lisfranc injuries can lead to chronic pain and permanent disability. Surgical treatment is mandatory for all the unstable injuries; however, the best surgical technique remains controversial. The most predictive factor for a successful outcome is the maintenance of anatomic alignment; therefore, the selection of the appropriate surgical technique is of paramount importance. This article reviews the current treatment options and describes the selection of the surgical technique based on the different clinical presentations.

RESUMO

Siparuna guianensis (Laurales: Siparunaceae) has a terpene-rich essential oil with great potential for larvicides. The poor water miscibility of their compounds makes nano-emulsions of great interest for novel bioactive systems, including for control of Aedes aegypti (Diptera: Culicidae). This species is adapted to urban environments with important role in the epidemiology of some arboviruses such as dengue, chikungunya fever, zika, and urban yellow fever. The aim of the present study was to evaluate the feasibility of nano-emulsification to affect Ae. aegypti larvae. An optimal system was achieved by using a nonionic single surfactant, highlighted by its satisfactory size distribution profile. Moreover, improved larvicidal activity in comparison to bulk essential oil can be observed for the nano-emulsions. The estimated LC50 and LC90 values after 24 h of treatment of larvae with the essential oil were, respectively, 86.5232 and 134.814 µg/ml, while the estimated LC50 and LC90 value after treatment with the nano-emulsion were 24.7572 and 75.2452 µg/ml, respectively. The utilization of a simple technique to produce a fine nano-emulsion opens perspective for further integrative practices of mosquito control and giving value to this Amazon plant species may encourage its sustainable use and contribute to conservation policies.

RESUMO

Four Arabica coffees (Brazil, Colombia, Ethiopia, and Guatemala) yield highly variant odours, attesting to the complexities of coffee aroma that command advanced analytical tools. In this study, their volatiles were extracted using solvent-assisted flavour evaporation (SAFE) and headspace solid-phase microextraction (HS-SPME). Due to matrix complexity, some trace odourants were detected in SAFE extracts by aroma extract dilution analysis (AEDA) but remained difficult to quantify by gas chromatography-mass spectrometry (GC-MS). This prompted the application of low energy electron ionisation (EI) coupled with GC-quadrupole time-of-flight (GC-QTOF). Optimal low EI GC-QTOF parameters (EI energy: 15 eV, acquisition rate: 3 Hz) were applied to achieve improved molecular ion signal intensity and reproducibility (relative standard deviation < 10%) across five compounds, which resulted in good linearity (R2 ≥ 0.999) and lowered detection levels (e.g. 0.025 ±â€¯0.005 ng/mL for 4-hydroxy-5-methyl-3(2H)-furanone). Therefore, this method potentially improves the measurement of trace odourants in complex matrices by increasing specificity and sensitivity.

RESUMO

Making Elliptic Curve Cryptography (ECC) available for the Internet of Things (IoT) and related technologies is a recent topic of interest. Modern IoT applications transfer sensitive information which needs to be protected. This is a difficult task due to the processing power and memory availability constraints of the physical devices. ECC mainly relies on scalar multiplication (kP)-which is an operation-intensive procedure. The broad majority of kP proposals in the literature focus on performance improvements and often overlook the energy footprint of the solution. Some IoT technologies-Wireless Sensor Networks (WSN) in particular-are critically sensitive in that regard. In this paper we explore energy-oriented improvements applied to a low-area scalar multiplication architecture for Binary Edwards Curves (BEC)-selected given their efficiency. The design and implementation costs for each of these energy-oriented techniques-in hardware-are reported. We propose an evaluation method for measuring the effectiveness of these optimizations. Under this novel approach, the energy-reducing techniques explored in this work contribute to achieving the scalar multiplication architecture with the most efficient area/energy trade-offs in the literature, to the best of our knowledge.

RESUMO

High-energy methods for the manufacturing of nanomedicines are widely used; however, interest in low-energy methods is increasing due to their simplicity, better control over the process, and energy-saving characteristics during upscaling. Here, we developed a novel lipid-core micelle (LCM) as a nanocarrier to encapsulate a poorly water-soluble drug, nifedipine (NFD), by hot-melt emulsification, a low-energy method. LCMs are self-assembling colloidal particles composed of a hydrophobic core and a hydrophilic shell. Hybrid materials, such as Gelucire 44/14, are thus excellent candidates for their preparation. We characterized the obtained nanocarriers for their colloidal properties, drug loading and encapsulation efficiency, liquid state, stability, and drug release. The low-energy method hot-melt emulsification was successfully adapted for the manufacturing of small and narrowly dispersed LCMs. The obtained LCMs had a small average size of ~ 11 nm and a narrow polydispersity index (PDI) of 0.228. These nanocarriers were able to increase the amount of NFD dispersible in water more than 700-fold. Due to their sustained drug release profile and the PEGylation of Gelucire 44/14, these nanocarriers represent an excellent starting point for the development of drug delivery systems designed for long circulation times and passive targeting.
