RESUMO

Twelve different brands of mineral water were collected from the Algerian market and analyzed to determine the initial antimony (Sb) content in both the PET package and mineral water. Experiments were conducted under different time conditions: 1, 10, to 365 days, different temperatures: 6, 25, and 40 °C, and different bottles sizes: 0.33 L and 1.5 L. The Sb in mineral water bottles varies between 0.50 and 1.12 µg/L for 0.33 L bottles, and 0.37 to 0.77 µg/L for 1.5 L ones. All of these values remain below the limit set by the European Union of 5 µg/L in drinking water. The diffusion coefficient of Sb in PET has been experimentally determined at 6, 25, and 40 °C, after the content of Sb in 1.5 L PET bottles had been determined. In the second part of the study, a factorial design 23 enabled a model the migration of antimony (Sb) in the bottled solutions and highlighted the influencing effects, such as temperature (°C), time (h), and thickness (mm) for two different time domains encompassing the entire validity period of the product. A simple polynomial function based on a single parameter has been determined with a precision indicator R2 = 0.98. This model has the advantage of being simple and fast. The Chronic Daily Intake (CDI) of Sb has been calculated, for adults. It does not exceed the Environmental Protection Agency (USEPA) regulated CDI value of 400 ng/kg/day. The CDI values for children increase as the weight of the children decreases. The passage from the maximum child weight to the minimum value in the study increases the CDI of 77%.

RESUMO

Production and consumption of olive oil is very important in Europe, being this product a basic element in the Mediterranean diet since long ago. The project objective is two-fold: a study of the contribution of virgin olive oils (VOOs) usual packaging to the whole life cycle of the product and a study of the environmental consequences of the Spanish Government regulation on VOO packaging. A life cycle assessment (LCA) according to ISO 14044 has been performed using the CML methodology for the impact assessment. The results show that the packaging influence varies from 2 to 300%, depending on the impact category and type of packaging (glass, tin or polyethylene terephtalate). Glass, which is related to higher quality perception by consumers, was found to be the most influencing material (due to its weight); however, this impact may be fairly reduced by applying ecodesign strategies (such as weight reduction and recycled-glass percentage increase). A new Spanish regulation on the mandatory use of non-refillable oilers in HORECA establishments (hotels, restaurants and caterings) aims to provide more quality assurance and better information to consumers; however, it was also found to mean a 74% increase in greenhouse gases emissions. This regulation was deeply discussed at European level and its application was withdraw due to consumers rejection, except for Spain. The findings of the present case study show that LCA and ecodesign should be important tools to be promoted and applied in policy making to reduce non-desirable consequences of regulation.

RESUMO

DNA complexes made with cationic polymers (polyplexes) developed as nonviral vectors for gene therapy must be enabled to cross through vascular endothelium to transfect underlying tissues upon their administration in the blood circulation. Here, we evaluated the transendothelial passage (TEP) of DNA complexes made with histidinylated linear polyethylenimine (His-lPEI) or linear polyethylenimine (lPEI). In vitro studies were performed by using established transwell lung and skeletal muscle vascular endothelial barriers. The models were composed of a monolayer of human lung microvascular endothelial (HMVEC-L) cells and mouse cardiac endothelial (MCEC) cells formed on a PET insert and immortalized human tracheal epithelial (ΣCFTE29o-) cells and mouse myoblasts (C2C12) as target cells cultured in the lower chamber, respectively. When the vascular endothelium monolayer was established and characterized, the transfection efficiency of target (ΣCFTE29o- and C2C12) cells with plasmid DNA encoding luciferase was used to evaluate TEP of polyplexes. The luciferase activities with His-lPEI and lPEI polyplexes compared to those obtained in the absence of endothelial cell monolayer were 6.5% and 4.3% into ΣCFTE29o- cells, and 18.5% and 0.23% into C2C12 cells, respectively. The estimated rate for His-lPEI polyplexes was 0.135 µg/cm(2).h and 0.385 µg/cm(2).h through the HMVEC-L and MCEC monolayers, respectively. These results indicate that His-lPEI polyplexes can pass through the lung and skeletal muscle vascular endothelium and can transfect underlying cells.

INTRODUCTION: D-004 is a new active ingredient under research and development stage, which is obtained from Roystonea regia (Kunth) O. F. Cook fruits. OBJECTIVE: to precise over the D-004 stability under accelerated conditions in amber glass, polyethylene terephtalate and high density polyethylene containers. METHODS: samples from three batches were stored at (40 ± 2) ºC and (75 ± 5) percent relative humidity for 12 months. Organoleptic characteristics, relative density, refractive index (USP 27) and fatty acids content by gas chromatography, were determined. RESULTS: measured parameters were within the set quality specifications , except for fatty acids content that significantly decreased in polyethylene terephtalate and high density polyethylene containers after 3, 4 and a half and 6 months, respectively. CONCLUSIONS: the amber glass protects more D-004 from degradation than the analyzed plastic containers.
