RESUMO

A 57-y-old male yellow-naped parrot (Amazona auropalliata) was presented because of lethargy, inappetence, and weight loss. Hematology and serum biochemistry were unremarkable, and imaging revealed a mass in the distal esophagus at the coelomic inlet. The luminal diameter of the esophagus was reduced in this area, and passage of ingesta was limited. Following gavage feeding, the patient died and was submitted for autopsy. At postmortem examination, the noted mass effect was a thickening of the distal esophagus with adherent, coalescing, soft, pale-tan plaques on the mucosal surface. Additional gross findings included pale-tan, opaque feed material oozing from the dorsum of the lungs and covering the cranial air sacs. Histology of the esophagus, esophageal-proventricular junction, and proximal proventriculus revealed an unencapsulated, infiltrative, transmural neoplasm that extended from the mucosal surface deep into the muscularis, almost to the adventitia. The neoplasm was composed of cuboidal cells arranged in islands and tubules, consistent with an adenocarcinoma, a rarely reported entity in the esophagus of psittacine birds and to our knowledge not reported previously at the esophageal-proventricular junction.

Despite the conservation of genetic machinery involved in eye development, there is a strong diversity in the placement of eyes on the head of animals. Morphogen gradients of signaling molecules are vital to patterning cues. During Drosophila eye development, Wingless (Wg), a ligand of Wnt/Wg signaling, is expressed anterolaterally to form a morphogen gradient to determine the eye- versus head-specific cell fate. The underlying mechanisms that regulate this process are yet to be fully understood. We characterized defective proventriculus (dve) (Drosophila ortholog of human SATB1), a K50 homeodomain transcription factor, as a dorsal eye gene, which regulates Wg signaling to determine eye versus head fate. Across Drosophila species, Dve is expressed in the dorsal head vertex region where it regulates wg transcription. Second, Dve suppresses eye fate by down-regulating retinal determination genes. Third, the dve-expressing dorsal head vertex region is important for Wg-mediated inhibition of retinal cell fate, as eliminating the Dve-expressing cells or preventing Wg transport from these dve-expressing cells leads to a dramatic expansion of the eye field. Together, these findings suggest that Dve regulates Wg expression in the dorsal head vertex, which is critical for determining eye versus head fate. Gain-of-function of SATB1 exhibits an eye fate suppression phenotype similar to Dve. Our data demonstrate a conserved role for Dve/SATB1 in the positioning of eyes on the head and the interocular distance by regulating Wg. This study provides evidence that dysregulation of the Wg morphogen gradient results in developmental defects such as hypertelorism in humans where disproportionate interocular distance and facial anomalies are reported.

Morphological changes in the late-embryo-stage gastrointestinal tract of Nigerian indigenous chicken were investigated using anatomical techniques. The paraffin-embedded sections were stained with haematoxylin and eosin and periodic acid Schiff-Alcian blue (pH 2.5) stains. During the late pre-hatch period, the framework of the stomach was already established, but glands were yet poorly developed. Randomly oriented pre-glandular cells in the lamina propria mucosae at embryonic day (ED) 14 became organized into elongated simple tubular glands at ED 19 and assumed adult morphology at post-hatch day (PD) 1. Acidic and neutral mucin deposits were associated with the glandular cells. In the small intestine, enterocytes of the tunica epithelialis mucosae transformed from cuboidal-shaped cells (at ED 14) to tall columnar cells (at PD 1). Short crypts of Lieberkühn, goblet cells, microfold cells and enteroendocrine cells were evident at ED 19, while the lamina propria mucosae and submucosa contained mesenchymal cells, reticular cells and isolated lymphoblasts. Similarly, the crypts, lymphoblasts, mesenchymal and reticular cells were also associated with the lamina propria mucosae and submucosa of the caecum and colorectum at ED 19. It was inferred from these findings that extensive cellular and tissue modifications occur in the gastrointestinal tract within the narrow window of the late pre-hatch period. The definitive gut tunics, epithelium, glandular tissues, immune-competent cells and tissues are formed as a result. Thus, the embryonic gut of the bird is made capable of assuming its necessary functions of food digestion, nutrient absorption, water and ion re-absorption, immune surveillance, antibody production and immune responses at hatch.

Zymosan is a fungi-derived pathogen-associated molecular pattern. It activates the immune system and induces the reduction of feed passage rate in the gastrointestinal tract of vertebrates including birds. However, the mechanism mediating the zymosan-induced inhibition of feed passage in the gastrointestinal tract remains unknown. Since the medulla oblongata regulates the digestive function, it is plausible that the medulla oblongata is involved in the zymosan-induced inhibition of feed passage. The present study was performed to identify the genes that were affected by zymosan within the medulla oblongata of chicks (Gallus gallus) using an RNA sequencing approach. We found that mRNAs of several bioactive molecules including neuropeptide Y (NPY) were increased with an intraperitoneal (IP) injection of zymosan. The increase of mRNA expression of NPY in the medulla oblongata was also observed after the IP injection of lipopolysaccharide, derived from gram-negative bacteria. These results suggest that medullary NPY is associated with physiological changes during fungal and bacterial infection. Furthermore, we found that intracerebroventricular injection of NPY and its receptor agonists reduced the feed passage from the crop. Additionally, the injection of NPY reduced the feed passage from the proventriculus to lower digestive tract. NPY also suppressed the activity of duodenal activities of amylase and trypsin. The present study suggests that fungi- and bacteria-induced activation of the immune system may activate the NPY neurons in the medulla oblongata and thereby reduce the digestive function in chicks.

An adult sulphur-crested cockatoo (Cacatua galerita) was unexpectedly found dead and was presented for post-mortem examination. Macroscopically, the proventricular wall was expanded by an infiltrative off-white mass, 10 mm in diameter, with secondary mucosal ulceration. Cytological examination of impression smears revealed a neoplastic epithelial cell population forming occasional well-defined acini with secretory product. Histologically, the neoplastic cells were predominantly arranged in acini and tubules, with fewer in small clusters, supported by a desmoplastic stroma. Neoplastic cells were polygonal, with mostly well-demarcated cell boundaries and a moderate amount of eosinophilic cytoplasm that was multifocally markedly distended by more intensely eosinophilic material. There was an increased nuclear-to-cytoplasmic ratio with moderately sized to large nuclei (approximately 15 µm diameter) with lightly to densely stippled chromatin and single or paired prominent basophilic nucleoli. The mitotic count was 19 in 10 high-power fields (2.37 mm2), with scattered bizarre mitotic figures and atypia including moderate anisocytosis and anisokaryosis and moderate cellular and nuclear pleomorphism. There was frequent intratubular brightly eosinophilic proteinaceous material, which was Alcian blue and periodic acid‒Schiff positive. Proventricular lymphovascular invasion was detected, with metastases within the ventriculus and lungs. The macroscopic, cytological and histological appearance of the neoplasm was consistent with a proventricular adenocarcinoma.

Eusociality is an extreme form of social behavior found in some invertebrates, especially in ants, which allows them to thrive and dominate the environment through their cooperative and organized behavior. Their complex colony structure and social behavior is provided, among other things, by morphological adaptations, such as the unique structure and a huge variety of proventriculi relative to other insect groups. Here, we report the first discovery of proventriculus by X-ray microcomputed tomography (µCT) in the fossil ants †Oecophylla brischkei Mayr, 1868 and †Oecophylla crassinoda Wheeler, 1922. This is a remarkable fossil find, as it opens up new perspectives for research on phylogeny, the evolution of ant sociality and feeding behavior. We present a 3D model of the proventriculus in Oecophylla smaragdina (Fabricius, 1775) and a 3D model of the proventriculus in †Oe. brischkei and compare them. Fossil proventriculi are not fundamentally different in structure from the recent. Apparently, already in the late Eocene, the lifestyle and sociality of ants of the genus Oecophylla were similar to the one we are currently observing.

SUMMARY: Birds are the most diversified organisms on Earth, with species covering various niches in each major biome, being essential to understand the modern ecosystem. This study concentrates on the diversification of the anatomical structure of the upper digestive tract for 26 species of zoophage-polyphagous birds and the anatomical differences in the digestive system to reveal aspects related to their evolution and diversification. The trophic spectrum of the selected birds includes several categories of food, or, as in the case of strictly carnivorous birds, to a single food category. After performing the dissections, the digestive tract was separated from the carcass and each digestive segment was measured and analysed. In this study, it was demonstrated that the birds' feeding behaviour influence the macroscopic particularities of the digestive system, more visible in the cranial portion (oropharyngeal cavity, esophagus, proventriculus and gizzard), with little descriptive information in the literature. The tongue is poorly developed and immobile in piscivorous birds, while the tongue of insectivorous birds is long and moves considerably away from the tip of the bill. The esophagus was stretchable and presents longitudinal folds on its entire surface in piscivorous species and not extensible in insectivorous birds.


Las aves son los organismos más diversificados de la Tierra, con especies que cubren varios nichos en cada bioma principal, siendo esenciales para comprender el ecosistema moderno. Este estudio se concentra en la diversificación de la estructura anatómica del tracto digestivo superior para 26 especies de aves zoófago-polífagas y las diferencias anatómicas en el sistema digestivo para revelar aspectos relacionados con su evolución y diversificación. El espectro trófico de las aves seleccionadas incluye varias categorías de alimentos o, como en el caso de las aves estrictamente carnívoras, una sola categoría de alimentos. Después de realizar las disecciones, se separó el tracto digestivo de la canal y se midió y analizó cada segmento digestivo. En este estudio se demostró que el comportamiento alimentario de las aves influye en las particularidades macroscópicas del sistema digestivo, más visibles en la porción craneal (cavidad orofaríngea, esófago, proventrículo y molleja), con poca información descriptiva en la literatura. En las aves piscívoras, la lengua está poco desarrollada e inmóvil, mientras que la lengua de las aves insectívoras es larga y se aleja considerablemente de la punta del pico. El esófago era estirable y presentaba pliegues longitudinales en toda su superficie en especies piscívoras y no extensible en aves insectívoras.

Despite vaccine use, novel strains and variants of infectious bronchitis virus (IBV) have emerged continuously, leading to economic losses to the poultry industry worldwide. This study aimed to characterize the IBV isolate CK/CH/GX/202109 from three yellow broilers in Guangxi, China. Recombination was shown to have occurred in regions of the 1ab gene. Compared to the whole genome of ck/CH/LGX/130530, which is genotypically related to tl/CH/LDT3-03, the 202109 strain had 21 mutations. The pathological assessment showed that this variant caused 30% and 40% mortality in 1-day-old chicks infected with oral and ocular inoculum, respectively. Nephritis, enlarged proventriculus, inflammation of the gizzard, and atrophy of the bursa of Fabricius were also observed at both 7 and 14 days post-infection (dpi). Viral loads in the trachea, proventriculus, gizzard, kidney, bursa, and cloacal swabs were higher at 7 dpi than at 14 dpi. Clinicopathological and immunohistochemical analyses revealed that this virus exhibited multiple organ tropisms capable of infecting the trachea, proventriculus, gizzard, kidney, bursa, ileum, jejunum, and rectum. Almost none of the 1-day-old infected chicks seroconverted until 14 dpi. While the virus was found in the ileum, jejunum, and rectum in the 28-day-old ocular group, the majority of 28-day-old infected chickens seroconverted at 10 dpi. These study findings demonstrate that recombination events and mutations during the evolution of IBV may greatly alter tissue tropism and emphasize the need for the continued surveillance of novel strains and variants in order to control this infection.

Hystrichis tricolor is a neglected avian enoplid nematode (superfamiliy Dioctophymatoidea) and known to parasitize various species of the Anatidae (Anas spp. and Mergus spp.) from the northern hemisphere, inducing mainly proventriculitis in domestic and wild waterfowl. Here, we focus on the pathological findings of naturally H. tricholor-infected Egyptian geese (Alopochen aegyptiaca) and a neozoan shelduck (Tandorninae) of Germany. Nowadays, this species is considered the fastest-spreading alien waterfowl species within Western Europe. Additionally, molecular sequencing coupled with phylogenetic characterization of H. tricolor is reported. Post mortem analyses unveiled patent gastric H. tricolor infections in eight of twelve infected birds (8/12; 66.7%), inducing proventriculitis resulting in large visible nodular lesions. Histopathological findings point to chronic host pro-inflammatory immune reactions. These results demonstrate the potential role of Egyptian geese as natural reservoir hosts of H. tricholor and highlight their possible role in parasite transmission (i.e., spillback) into endemic waterfowl species. Due to avian health concerns, the occurrence of hystrichiosis should be monitored in native waterfowl in the future, introducing appropriate management practices in conservation strategies of endemic wild birds not only in Germany but elsewhere in Europe.

This study aimed to determine whether the innate immune system in the proventriculus of broiler chicks responds to lipopolysaccharide (LPS) and whether this response is affected by Newcastle disease and infectious bronchitis (ND/IB) vaccination. Chicks were divided into 4 groups: nonvaccinated and injected with PBS or LPS (V-L- and V-L+), and vaccinated and injected with PBS or LPS (V+L- and V+L+). Vaccination was performed on d 1, and LPS was intraperitoneally injected on d 11 of age. The gene expression and protein levels of immune molecules, including toll-like receptors (TLRs), antimicrobial peptides, interleukin-1ß (IL-1B), and immunoglobulin A (IgA) in the proventriculus and serum were analyzed. The results showed that the expression levels of TLR21 were higher in vaccinated (V+L-) group than in nonvaccinated (V-L-) group. Gene expression levels of avian ß-defensin (AvBDs) and cathelicidin1 (Cath1) were not different among the 4 groups. However, the results of LC/MS analysis showed that the levels of AvBD2, 6, and 7 significantly increased after the LPS challenge in nonvaccinated and vaccinated chicks; the levels were higher in V-L+ and V+L+ than in V-L- and V+L-, respectively. Immunohistochemistry analysis revealed the localization of AvBD1 protein in the epithelial cells of the surface glands and AvBD2 and CATH1 in the heterophil-like cells in the lamina propria of surface glands. Although IL-1B gene expression and protein concentration in the proventriculus tissues were not different among the 4 groups, serum IL-1B levels were upregulated by LPS in both the nonvaccinated and vaccinated groups (V-L- vs. V-L+, V+L- vs. V+L+). Moreover, IgA levels in the proventriculus and serum were not affected by vaccination or LPS challenge. Taken together, we conclude that LPS derived from gram-negative bacteria upregulates the innate immune system, including antimicrobial peptide synthesis in the proventriculus. ND/IB vaccination may not significantly affect antimicrobial peptide synthesis in response to LPS; however, TLR21 expression is upregulated by that vaccination. The antimicrobial peptides synthesized in the proventriculus probably prevent pathogenic microbes from entering the intestine.

The current investigation aimed to clarify the histomorphological features, scanning electron microscopy (SEM) and histochemistry of the proventriculus in the adult hooded crow (Corvus cornix). Twenty-two adult birds of both sexes were collected from El-Fayoum and Beni-Suef governorates, Egypt; ten were used for anatomical investigation, two were scanned using SEM, and ten were subjected to routine histological technique. The obtained anatomical results revealed a small-sized spindle-shaped proventriculus obliquely located in the left ventral part of the body cavity with an average body weight, length and diameter of 1.689 ± 0.231 gm, 1.54 ± 0.383cm and 3.51 ± 0.416cm, respectively. The histological examination revealed a typical tubular organ. The mucosa exhibited several plicae and sulci that were lined with simple columnar cells. The lamina propria was composed of collagenous connective tissue infiltrated by lymphocytes, lymphatic aggregations and tubular glands. The submucosa consisted of collagenic fibres and compound tubuloalveolar glandular lobules lined with oxynticopeptic cells and enteroendocrine cells. The oxynticopeptic cells reacted negatively with PAS and Alcian Blue stains, whereas the ductal cells reacted positively with PAS only. Enteroendocrine cells were seen singly in the lamina epithelialis mucosae and were abundant in submucosal glands. The tunica musclaris was arranged in two layers covered by serosa. The proventricular glands' openings were noticed by SEM as raised tubes with regular circular outlines surrounded by anastomosed microscopic folds. The obtained histomorphological structure of the hooded crow's proventriculus revealed some shared morphological features with most species of birds and some variations that might be attributed to their diet and feeding behaviour.

The present study was implemented to provide comprehensive information on the developmental sequence of the proventriculus of Muscovy ducks by gross examination, macro-micrometric analysis and by using light microscope, scanning electron microscope (SEM) and transmission electron microscope (TEM). Samples from 55 healthy post-hatching Muscovy ducks of both sexes ranging from 1 to 60 days old. The proventriculus began cranially opposite to the cranial end of the liver at 1-15 days old, but in front this level at 30 and 60 days old. Morphometrically, the length of the proventriculus was increased by about four folds while weight by 19 folds at 60 days old when compared with those at one day old. Scanning electron microscopy of the proventricular lumen at one day old exhibited numerous small mostly rounded irregularly distributed openings of the glands, but uniformly distributed and surrounded by closely packed concentrically arranged mucosal folds resembling a rosette shape at the older ages. Histologically, in all studied stages, rounded, elongated oval or polymorphic shaped lobules of the proventricular glands were occupied within the muscularis mucosa. The tubuloalveolar secretory units lined with secretory (oxyntico-peptic) cells with variable shapes had secretory granules increased by the development. Numerous argyrophilic endocrine cells were demonstrated away from the glandular lumen at older ages. Transmission electron microscopy revealed that the cytoplasm of the secretory cells contained homogeneously electron-dense granules at a young age, but two types of these granules could be recognized at 60 days old. In conclusion, this study provides a wide difference in the morphometric and the structure of the proventriculus from one day to 60 days old. This difference between the examined age-stages may be related to the feeding strategy (behavior) and the functional adaptations from the young to the older ages.

Early feeding post-hatching (EFPH) can impact the immune response and modify the immunity-related gene expression. Therefore, we aimed to examine the effects of EFPH with different diets composition versus fasting during the first 72 h of chick's life on the histomorphological structures of the liver, proventriculus, central and peripheral lymphoid organs, and immunity-related genes in layer-type chicks during the brooding period. A total of 400 chicks were randomly allotted into 4 groups with 4 replicates each. The experimental groups during the first 72 h of life were: feed and water deprivation (control, T1), feeding a starter layer diet (20% CP and 11.84 MJ/kg ME, T2), feeding a starter layer diet contained 3% molasses in its composition (20% CP and 11.81 MJ/kg ME; T3), and feeding a starter broiler diet (23% CP and 12.68 MJ/kg ME, T4). After the first 72 h of chick's life, all chicks were fed ad libitum the T2 diet. EFPH had no negative effect on the development of the lymphoid or digestive organs in chicks. Greater relative weights of the spleen and bursa of Fabricius (p < 0.05) were observed in the early fed chicks compared to control at day 14 of age. Histomorphological examination revealed an increase (p < 0.05) in thymus cortex and cortex:medulla in the T3 and T4 groups compared to the fasted ones at day 28 of age. Pelicae height, follicular width, cortex, and cortex:medulla of bursa were improved (p < 0.01) in the fed groups compared to fasted chicks, with resultant influences on the primary lymphoid organs. Compared to control, higher germinal center areas and white pulp of the spleen (p < 0.05) were recorded in the early fed chicks, implying augmented proliferation and maturation of B cells in the secondary lymphoid organs. In the liver, a strong positive reaction to Best's carmine stain in the early fed groups, indicating that the liver of these chicks had numerous glycogen granules or greater glycogen density in the cytoplasm of hepatocytes. There was a significant enhancement in the proventriculus mucosal and gland thickness, as well as fold height (p < 0.05) in the early fed chicks. The expression levels of splenic Toll-like receptor 2, interleukin 4, tumor necrosis factor α, and interferon gamma were up-regulated (p < 0.01) in most of the early fed chicks (T2, T3, and T4) compared to fasted ones at 14 day of age. In conclusion, EFPH could modify the splenic-immunity related genes and modulate the histomorphology of the digestive (liver and proventriculus) and lymphoid organs in layer-type chicks during the brooding period.

The avian alimentary tract has evolved into different histologic structures to accommodate the physical and chemical features of several food types and flight requirements. We compared the esophagus, proventriculus, and gizzard of the domestic fowl, Gallus gallus domesticus (GGD) and kestrels, Falco tinnunculus (FT) using immunohistochemistry and scanning electron microscopy with various stains and lectins [Dolichos biflorus agglutinin (DBA) and Ricinus communis agglutinin I (RCA120)], and α-smooth muscle actin (α-SMA). The esophagus of GGD demonstrated thickened epithelium, muscularis mucosae, and inner circular longitudinal tunica muscularis layers; moderate outer longitudinal tunica muscularis layers; and a true crop. In contrast, the esophagus of FT showed a thin epithelium, no muscularis mucosae, moderate inner longitudinal and thick outer circular tunica muscularis layers, and no true crop. In the proventriculus, the nature of the secretion in GGD was neutral, but that of FT was acidic and neutral. In the gizzard, the muscle coat of GGD by α-SMA had no muscularis mucosae, unlike FT, which had muscularis mucosae. In summary, there are many histologic differences between GGD and FT to meet their different physiologic needs, such as feeding.

Lamins interact with the nuclear membrane and chromatin but the precise players and mechanisms of these interactions are unknown. Here, we tested whether the removal of the CaaX motif from Lamin B disrupts its attachment to the nuclear membrane and affects chromatin distribution. We usedDrosophila melanogaster LamA25  homozygous mutants that lack the CaaX box. We found that the mutant Lamin B was not confined to the nuclear periphery but was distributed throughout the nuclear interior, colocalizing with chromosomes in salivary gland and proventriculus. The peripheral position of Lamin C, nuclear pore complex (NPC), heterochromatin protein 1a (HP1a), H3K9me2- and H3K27me3-associated chromatin remained intact. The fluorescence intensity of the DAPI-stained peripheral chromatin significantly decreased and that of the central chromatin significantly increased in the proventriculus nuclei of the mutantflies compared to wild-type. However, the mutation had little effect on chromatin radial distribution inside highly polytenized salivary gland nuclei.

The anatomy of the workers' digestive tube is essential in taxonomical studies of soil-feeding Apicotermitinae termites, especially in soldierless lineages. Two structures, the mesenteric-proctodeal junction and the enteric valve, have long been important to distinguish genera and species. By contrast, the gizzard (proventriculus) has been almost ignored by taxonomists because of its generally regressed state in soil-feeding termites. In this study, we document in detail for the first time the sclerotized structures and ornamentations in the gizzard in the Apicotermitinae subfamily. We identified two main clusters of species: those without ornamentations and those exhibiting a sclerotized pulvillar armature, which may include spicules or spines of diverse sizes, numbers and dispositions. The latter group comprises the majority of African soldierless species, a widely diverse and dominant group in tropical forests and savannas. We outline the potential role of the anatomy of the gizzard in the taxonomy of Apicotermitinae based on the interspecific anatomical variation of the pulvillar armatures. We suggest that sclerotized ornamentations regulate the flow of food particles and break or lacerate aggregates to facilitate the access of enzymes in the midgut.

BACKGROUND: Trypanosoma brucei exhibits a complex life-cycle alternating between tsetse flies and mammalian hosts. When parasites infect the fly, cells differentiate to adapt to life in various tissues, which is accompanied by drastic morphological and biochemical modifications especially in the proventriculus. This key step represents a bottleneck for salivary gland infection. METHODS: Here, we monitored flagellum assembly in trypanosomes during differentiation from the trypomastigote to the epimastigote stage, i.e. when the nucleus migrates to the posterior end of the cell, by using three-dimensional electron microscopy (focused ion beam scanning electron microscopy, FIB-SEM) and immunofluorescence assays. RESULTS: The combination of light and electron microscopy approaches provided structural and molecular evidence that the new flagellum is assembled while the nucleus migrates towards the posterior region of the body. Two major differences with well-known procyclic cells are reported. First, growth of the new flagellum begins when the associated basal body is found in a posterior position relative to the mature flagellum. Secondly, the new flagellum acquires its own flagellar pocket before rotating on the left side of the anterior-posterior axis. FIB-SEM revealed the presence of a structure connecting the new and mature flagellum and serial sectioning confirmed morphological similarities with the flagella connector of procyclic cells. We discuss the potential function of the flagella connector in trypanosomes from the proventriculus. CONCLUSIONS: These findings show that T. brucei finely modulates its cytoskeletal components to generate highly variable morphologies.

African trypanosomes are naturally transmitted by bloodsucking tsetse flies in sub-Saharan Africa and these transmission cycles can be reproduced in the laboratory if clean tsetse flies and suitable trypanosomes are available for experiments. Tsetse transmission gives access to more trypanosome developmental stages than are available from in vitro culture, albeit in very small numbers; for example, the sexual stages of Trypanosoma brucei have been isolated from infected tsetse salivary glands, but have not yet been reported from culture. Tsetse transmission also allows for the natural transition between different developmental stages to be studied.Both wild-type and genetically modified trypanosomes have been successfully fly transmitted, and it is possible to manipulate the trypanosome environment inside the fly to some extent, for example, the induction of expression of genes controlled by the Tet repressor by feeding flies with tetracycline.

The black proventriculus is a necropsy finding sign observed during post-mortem examinations of dead young chicks up to 10 days of age and due to variation in color of the affected proventriculus organ, it may be ignored in some cases, in particular when the disease occurs simultaneously with other well-recognized infections of two weeks old chicks. In late January 2018, several live and dead chicks with a history of an unusual increase in mortality showing no clinical signs but sudden dead were referred to the poultry clinic of Faculty of Veterinary Medicine, Urmia University, Urmia, Iran. General routine postmortem examination revealed color changes (light green to dark green color) in the wall of proventriculus together with some degrees of congestion in the lungs and liver of the dead chicks. Microbiology tests revealed that the causative agent was Escherichia coli strain O142 and histopathological examination indicated that the color changing of the organ was due to necrosis of glandular epithelium with locally extensive or diffuse hypercellularity by lymphocytes and macrophages with hemorrhages. In conclusion, black proventriculus due to E. coli strain O142 was responsible for the sudden death of young chicks and based on antibiogram, selective antibiotics should be used for the treatment of the affected flocks.

Motilin and ghrelin were identified in the pheasant by molecular cloning, and the actions of both peptides on the contractility of gastrointestinal (GI) strips were examined in vitro. Molecular cloning indicated that the deduced amino acid sequences of the pheasant motilin and ghrelin were a 22-amino acid peptide, FVPFFTQSDIQKMQEKERIKGQ, and a 26-amino acid peptide, GSSFLSPAYKNIQQQKDTRKPTGRLH, respectively. In in vitro studies using pheasant GI strips, chicken motilin caused contraction of the proventriculus and small intestine, whereas the crop and colon were insensitive. Human motilin, but not erythromycin, caused contraction of small intestine. Chicken motilin-induced contractions in the proventriculus and ileum were not inhibited by a mammalian motilin receptor antagonist, GM109. Neither atropine (a cholinergic receptor antagonist) nor tetrodotoxin (a neuron blocker) inhibited the responses of chicken motilin in the ileum but both drugs decreased the responses to motilin in the proventriculus, suggesting that the contractile mechanisms of motilin in the proventriculus was neurogenic, different from that of the small intestine (myogenic). On the other hand, chicken and quail ghrelin did not cause contraction in any regions of pheasant GI tract. Since interaction of ghrelin and motilin has been reported in the house musk shrew, interaction of two peptides was examined. The chicken motilin-induced contractions were not modified by ghrelin, and ghrelin also did not cause any contraction under the presence of motilin, suggesting the absence of interaction in both peptides. In conclusion, both the motilin system and ghrelin system are present in the pheasant. Regulation of GI motility by motilin might be common in avian species. However, absence of ghrelin actions in any GI regions suggests the avian species-related difference in regulation of GI contractility by ghrelin.
