The deltoid is the preferred site for intramuscular injection (IMI) because of its easy accessibility for drug and vaccine administration. Government immunization advisories, standard anatomy textbooks, and researchers have proposed various injection techniques and sites, but specific guidelines are lacking for the administration of IMIs in the increasingly used deltoid site. This study analyzes the procedures of administering IMIs in the deltoid related to the neurovascular network underlying the muscle and proposes a preferred site with the least chance of injury. The review protocol was submitted with PROSPERO (ID: 319251). PubMed, Google Scholar, and Websites of National Public Health Agencies were searched from 1950 up to 2022 for articles, advisories, and National Immunization Guidelines using Medical Subject Headings (MeSH) terms, including IMIs, deltoid muscle, safe injection sites, to identify recommendations for safer sites and techniques of administering deltoid IMIs. All the authors strictly adhered to a well-developed registered review protocol throughout the study and followed the risk of bias in systematic reviews (ROBIS) guidance tool. The proposed sites and landmark data were tabulated, and each site was analyzed based on the underlying neurovascular structures. Data were depicted by self-generated images. The initial search identified 174 articles. After applying the inclusion and exclusion criteria, 57 articles were shortlisted. Out of the 39 selected articles, 18 focused on the administration of deltoid IMIs, whereas seven focused on the variations in the underlying neurovascular structures in proximity to the deltoid muscle. The remaining 14 articles were the immunization guides issued by the National Public Health Agencies of the Government of India and abroad, whose data was used for comparison. Twelve deltoid IMI sites and techniques were identified. A site 1-3 fingerbreadths/5 cm below the mid-acromion point (7 studies); mid-deltoid site/densest part of the deltoid (1 study); a site at the middle third of the deltoid muscle (1 study); triangular injection site (1 study). Limitations included the unavailability of free access to complete text in many articles resulting in exclusion. The area around the shoulder joint and up to the lower level of the intertubercular sulcus is highly vascularized by the presence of many anomalous arterial patterns. To avoid injury, a safer site is proposed of 5 fingerbreadths/10 cm below the midpoint of the lateral border of the acromion. The authors received no specific funding for this study except for the journal publication charges.

Cre-loxP is an efficient recombination system originated from P1 phage. Its specific recombination patterns based on the locus of X-over P1 make it one of the most commonly used tools for gene editing in recent years. This paper focuses on the practical application of Cre-loxP system. Firstly, the functions and advantages of CRISPR/Cas9 system in Cre sequence insertion and loxP sequence insertion are analyzed. Then, a series of practical application problems of Cre-loxP system are described. For example, in this paper, the selection of Cre recombinase sequence in situ and safe site, the strategy of loxP sequence insertion, the identification of Cre recombinase tag protein, fluorescence identification of "ectopic" expression, primer design of PCR identification and reproductive strategy of mouse were described. At the same time, the optimization of Cre-loxP system in conditional gene knockout is introduced, such as ligand-induced Cre, promoter activated Cre, photo-induced Cre and activity modification of Cre. Through these optimized applications, we can obtain time-controlled conditional gene knockout, regulate the activity of Cre recombinase, and even avoid the toxicity of Cre recombinase itself. Finally, this paper discusses the defects and challenges of Cre-loxP system, and looks into the future development direction of Cre-loxP system. In summary, this paper reviews the practical application of gene knockout based on Cre-loxP system, summarizes the latest research progress and optimization strategies of Cre-loxP system, and prospects the future gene editing based on Cre-loxP system. This paper aims to provide theoretical guidance for solving practical operation problems based on Cre-loxP system, and to provide new research ideas for more accurate, more controllable and more adaptive gene editing in the future.

Semi-natural grasslands are of great importance to European biodiversity. Their restoration in mountain regions often presents specific problems of raw soil stabilisation and difficult climatic conditions. Methods used to tackle these problems include increasing sowing density and fertilisation. However, past studies were performed at only one site or for a short time and do not clearly show how site traits and time affect restoration results. In six field experiments undertaken on raw soils in the Italian Alps, the effects of sowing density, fertilisation, soil, and climate on the development over time of plant density and cover were analysed over three to six years. The seed was collected from nearby grasslands and sown at rates of 335-21,410 seeds m-2. During the first one-two years, the percentage of seeds producing established plants at low seed rates was higher in stonier surfaces; however, at high rates a larger absolute plant number was recorded in finer soils. Two to five years after sowing, climatic conditions became more important. Full vegetation cover was achieved in all experiments. However, at high altitudes, individual plant cover became greater (and plant density decreased), probably because the grasses needed to accumulate more non-structural carbohydrates necessary to survive during the longer winter. Fertilisation accelerated the establishment of full vegetation cover by 1 to 3 years as it increased the mean cover per plant without affecting plant density. Grassland restoration is possible in mountain raw soils by using the seeds collected from nearby donor sites. High sowing densities are not required even in coarse or higher altitude soils because, in the short term, stones on the ground surface increase the percentage of seed producing established plants and, over the long term, plant density is not dependent on the sowing density. In low-fertile soils, fertilisation accelerates the establishment of full vegetation cover.

Conifers in northern forests, such as fir and spruce, preferably regenerate on coarse woody debris, including fallen logs, stumps, and snags. In northern Japan, the sub-boreal conifer species Picea jezoensis is completely dependent on coarse woody debris for seedling establishment. To understand the fine-scale spatial genetic structure (FSGS) of this species, a 5-ha plot was established in central Hokkaido, and 531 individual trees were categorized into four life-stages (seedling, sapling, juvenile, and mature) on the basis of age and size. The FSGS of the established seedlings and later growth stages was investigated using 11 nuclear simple sequence repeat loci. A STRUCTURE analysis of seedlings and saplings established on fallen logs revealed that genetically related individuals were spatially localized between adjacent logs. We also found a significant FSGS in early life-stages based on a decline in the kinship coefficient calculated between individuals over shorter to longer spatial distances. Furthermore, the estimation of dispersal kernels indicated the frequent occurrence of short-distance seed dispersal. These results indicated that genetically related seedlings and saplings regenerated on the same or nearby fallen logs. In contrast to the results for the early stages, mature-stage trees showed no significant FSGS. We ran a simulation to examine the hypothesis that the FSGS could be eliminated by demographic thinning during life history processes. We calculated values for simulated offspring generated under three sets of conditions; i.e., by removing (i) inbred individuals, (ii) randomly chosen individuals, and (iii) all individuals on the specific fallen logs. However, the results for the FSGS were significant for all simulated data sets. This indicated that inbreeding depression, stochastic loss, or eradication of establishment sites by local disturbances alone could not explain the lack of FSGS among mature-stage trees. Therefore, it is possible that the colonization history of mature trees present on the study site might differ from that of the current offspring.

Plant stigmas and bee pollinators are competitors for pollen. Pollen placed on a pollinator's body can be picked up by conspecific stigmas or it can be collected by the pollinator as food. Hypothetically, one solution is for pollen to be placed on 'safe sites' on the pollinator's body, sites where the pollinator cannot easily remove it, leaving the pollen for stigmas. We compared 14 sites on the bumblebee body in terms of the ability of the bee to groom off fluorescent power, a dust that may be thought of as analogous to pollen. The safest sites were along the midline of the dorsal thorax, the dorsal abdomen, and the ventral abdomen. Next, we counted how much pollen is borne on the 14 sites by bees visiting one nectariferous and three nectarless Pedicularis species. In the four species, only 7, 26, 28, and 30% of pollen found on the bees were on safe sites. Finally, we observed that the 14 sites were contacted by stigmas of the four Pedicularis species; none of the most contacted sites were safe sites. Across all four Pedicularis species, pollen is mainly positioned on sites of the bee body that were beneficial for both the plant and the bee, not on sites detrimental to either of them. Our analysis showed that the conflict of interest between flowers and bees can be solved by cooperation. Pedicularis pollen is placed where it strengthens the mutualism between plants and pollinators.

It is becoming increasingly important for clinicians to identify a safer intramuscular (IM) injection site in the deltoid muscle because of possible complications following the vaccine administration of IM injections. We herein examined 4 original IM sites located on the perpendicular line through the mid-acromion to establish a safer IM injection site. Thirty healthy volunteers participated in this study and the distances from our 4 IM sites to some anatomical landmarks on their left arms were measured. Ultrasonography (US) was also performed to measure the thickness of the deltoid muscle and identify the posterior circumflex humeral artery (PCHA) along the course of the axillary nerve. Subcutaneous thickness was measured using 2 methods: measuring the skin thickness with caliper after pinching the skin, and with US. The results obtained revealed that the intersection between the anteroposterior axillary line (the line between the upper end of the anterior axillary line and the upper end of the posterior axillary line) and the perpendicular line from the mid-acromion was the most appropriate site for IM injections because it was distant from the axillary nerve, PCHA, and subdeltoid/subacromial brusa. At this site, depth of needle insertions was 5 mm greater than the subcutaneous thickness at a 90° angle, which was sufficient to penetrate subcutaneous tissue in both sexes. Subcutaneous thickness can be assessed with almost the same accuracy by US or measuring with calipers after pinching the skin. The results of the present study support the improved vaccine practice for safer IM injections.

Plants often suffer reductions in fecundity due to insect herbivory. Whether this loss of seeds has population-level consequences is much debated and often unknown. For many plants, particularly those with long-lived seedbanks, it is frequently asserted that herbivores have minimal impacts on plant abundance because safe-site availability rather than absolute seed number determines the magnitude of future plant recruitment and hence population abundance. However, empirical tests of this assertion are generally lacking and the interplay between herbivory, spatio-temporal variability in seed- or safe-site-limited recruitment, and seedbank dynamics is likely to be complex. Here we use a stochastic simulation model to explore how changes in the spatial and temporal frequency of seed-limited recruitment, the strength of density-dependent seedling survival, and longevity of seeds in the soil influence the population response to herbivory. Model output reveals several surprising results. First, given a seedbank, herbivores can have substantial effects on mean population abundance even if recruitment is primarily safe-site-limited in either time or space. Second, increasing seedbank longevity increases the population effects of herbivory, because annual reductions in seed input due to herbivory are accumulated in the seedbank. Third, population impacts of herbivory are robust even in the face of moderately strong density-dependent seedling mortality. These results imply that the conditions under which herbivores influence plant population dynamics may be more widespread than heretofore expected. Experiments are now needed to test these predictions.
