RESUMO

Neurodevelopmental disorders, such as autism spectrum disorder (ASD) and attention deficit hyperactivity disorder (ADHD), are characterized by persistent changes in communication and social interaction, as well as restricted and stereotyped patterns of behavior. The complex etiology of these disorders possibly combines the effects of multiple genes and environmental factors. Hence, exposure to insecticides such as imidacloprid (IMI) has been used to replicate the changes observed in these disorders. Lutein is known for its anti-inflammatory and antioxidant properties and is associated with neuroprotective effects. Therefore, the aim of this study was to evaluate the protective effect of lutein-loaded nanoparticles, along with their mechanisms of action, on Drosophila melanogaster offspring exposed to IMI-induced damage. To simulate the neurodevelopmental disorder model, flies were exposed to a diet containing IMI for 7 days. Posteriorly, their offspring were exposed to a diet containing lutein-loaded nanoparticles for a period of 24 h, and male and female flies were subjected to behavioral and biochemical evaluations. Treatment with lutein-loaded nanoparticles reversed the parameters of hyperactivity, aggressiveness, social interaction, repetitive movements, and anxiety in the offspring of flies exposed to IMI. It also protected markers of oxidative stress and cell viability, in addition to preventing the reduction of Nrf2 and Shank3 immunoreactivity. These results demonstrate that the damage induced by exposure to IMI was restored through treatment with lutein-loaded nanoparticles, elucidating lutein's mechanisms of action as a therapeutic agent, which, after further studies, can become a co-adjuvant in the treatment of neurodevelopmental disorders, such as ASD and ADHD.

Phelan-McDermid syndrome (PMS) is a rare, heterogeneous, and complex neurodevelopmental disorder. It is generally caused by a heterozygous microdeletion of contiguous genes located in the distal portion of the long arm of chromosome 22, including the SHANK3 gene. Sequence variants of SHANK3, including frameshift, nonsense mutations, small indels and splice site mutations also result in PMS. Furthermore, haploinsufficiency in SHANK3 has been suggested as the main cause of PMS. SHANK3 is also associated with intellectual disability, autism spectrum disorder and schizophrenia. The phenotype of PMS is variable, and lacks a distinctive phenotypic characteristic, so the clinical diagnosis should be confirmed by genetic analysis. PMS is a multi-system disorder, and clinical care must encompass various specialties and therapists. The role of risperidone, intranasal insulin, insulin growth factor 1, and oxytocin as potential therapeutic options in PMS will be discussed in this review. The diagnosis of PMS is important to provide an appropriate clinical evaluation, treatment, and genetic counseling.

BACKGROUND: Hip motion in the transverse plane is coupled with foot motion in the frontal plane during closed kinematic activities, such as gait. Considering that movement patterns and bone alignment might influence passive mechanical properties of joints in the long term, it is possible that hip passive stiffness and foot complex stiffness and alignment are related to each other. OBJECTIVES: To investigate whether hip passive stiffness, midfoot passive stiffness and shank-forefoot alignment are related to each other. METHOD: Thirty healthy adult individuals with a mean age of 25.4 years participated (18 women and 12 men). The Foot Torsimeter was used to measure midfoot stiffness, and hip stiffness and foot alignment were measured using clinical measures. Pearson and Spearman correlation coefficients were calculated to test the associations between each pair of variables, with α = 0.05. RESULTS: Hip stiffness was positively correlated with midfoot absolute stiffness (r = 0.41, p = 0.02), indicating that increased hip stiffness is associated with increased midfoot stiffness. There were no associations between shank-forefoot alignment and the other variables. CONCLUSIONS: In clinical settings, individuals with reduced hip passive stiffness may also have reduced midfoot passive stiffness, and vice versa. Shank-forefoot alignment is not linearly associated with hip or midfoot passive stiffness.

The objective of this research was to evaluate the effect of seeder furrow opening mechanisms on soil physical characteristics, plant growth, and soybean yield in lowland areas with the presence of compacted layer near the surface. An experiment was conducted in the lowland experimental area of the Federal University of Santa Maria-RS, Brazil, during the 2015/16 and 2016/17 growing season. The experimental design was a randomized block with five replications and four treatments: Planter using shank (S) at 0.23 m depth; S at 0.13 m; Planter using double disk at 0.07 m depth and Raised-bed + S at 0.12 m depth. The use of S at 0.23 m depth allowed reduction of penetration resistance and increase of soil macroporosity in the layers 0-0.1 and 0.1-0.2 m depth in the seeding row. Sowing with S at 0.23 m depth and Raised-bed + S at 0.12 m depth allowed a better development of soybean plants in compacted irrigated rice area (higher nodulation, leaf area index and root growth), as well as higher grain yield.(AU)


O trabalho teve como objetivo avaliar o efeito de mecanismos sulcadores da semeadora sobre características físicas do solo, crescimento de plantas e produtividade de soja em área de terras baixas com presença de camada compactada. Para isso, foi conduzido um experimento na área de várzea da Universidade Federal de Santa Maria, durante as safras agrícolas 2015/16 e 2016/17. O delineamento experimental utilizado foi o de blocos ao acaso, com cinco repetições e quatro tratamentos, sendo eles: Haste sulcadora (HS) a 0.23 m de profundidade; HS a 0.13 m; Disco duplo a 0.07 m e Microcamalhão+HS a 0.12 m. A utilização da HS a 0.23 m possibilitou o aumento da macroporosidade e a redução da resistência à penetração mecânica do solo nas camadas de 0-0.1 e 0.1-0.2 m de profundidade na linha de semeadura. A semeadura com HS a 0.23 m e Microcamalhão+HS a 0.12 m viabilizam melhor desenvolvimento das plantas de soja em área de terras baixas compactada (maior nodulação, índice de área foliar e crescimento radicular), bem como maior produtividade de grãos.(AU)

BACKGROUND: Phelan-McDermid syndrome (PMS) is a rare genetic disorder characterized by global developmental delay, intellectual disability (ID), autism spectrum disorder (ASD), and mild dysmorphisms associated with several comorbidities caused by SHANK3 loss-of-function mutations. Although SHANK3 haploinsufficiency has been associated with the major neurological symptoms of PMS, it cannot explain the clinical variability seen among individuals. Our goals were to characterize a Brazilian cohort of PMS individuals, explore the genotype-phenotype correlation underlying this syndrome, and describe an atypical individual with mild phenotype. METHODOLOGY: A total of 34 PMS individuals were clinically and genetically evaluated. Data were obtained by a questionnaire answered by parents, and dysmorphic features were assessed via photographic evaluation. We analyzed 22q13.3 deletions and other potentially pathogenic copy number variants (CNVs) and also performed genotype-phenotype correlation analysis to determine whether comorbidities, speech status, and ASD correlate to deletion size. Finally, a Brazilian cohort of 829 ASD individuals and another independent cohort of 2297 ID individuals was used to determine the frequency of PMS in these disorders. RESULTS: Our data showed that 21% (6/29) of the PMS individuals presented an additional rare CNV, which may contribute to clinical variability in PMS. Increased pain tolerance (80%), hypotonia (85%), and sparse eyebrows (80%) were prominent clinical features. An atypical case diagnosed with PMS at 18 years old and IQ within the normal range is here described. Among Brazilian ASD or ID individuals referred to CNV analyses, the frequency of 22q13.3 deletion was 0.6% (5/829) and 0.61% (15/2297), respectively. Finally, renal abnormalities, lymphedema, and language impairment were found to be positively associated with deletion sizes, and the minimum deletion to cause these abnormalities is here suggested. CONCLUSIONS: This is the first work describing a cohort of Brazilian individuals with PMS. Our results confirm the impact of 22q13 deletions on ASD and several comorbidities, such as hypotonia. The estimation of a minimal deletion size for developing lymphedema and renal problem can assist prediction of prognosis in PMS individuals, particularly those diagnosed in early infancy. We also identified one atypical individual carrying SHANK3 deletion, suggesting that resilience to such mutations occurs. This case expands the clinical spectrum of variability in PMS and opens perspectives to identify protective mechanisms that can minimize the severity of this condition.

BACKGROUND: The clinical measure of forefoot-shank alignment (FSA) predicts the amount of foot pronation during weight-bearing tasks. This may be mediated by a relationship between FSA and the mechanical resistance of the midfoot joint complex (MFJC) to forefoot inversion, which is a component of weight-bearing foot pronation. OBJECTIVE: To investigate if the clinical measure of FSA is associated with MFJC mechanical resistance to inversion. DESIGN: Cross-sectional observational study. METHOD: Forty-six healthy individuals (27 males; 19 females) with mean age of 26.4 years (SD 5.3) participated in this study. FSA was measured with photographs. The resistance torque of the MFJC against inversion was measured with a specially designed device. Mean torque, mean torque normalized by body mass, and joint resting position were calculated as variables related to MFJC mechanical resistance. Correlation analyses were carried out to test the association between each MFJC resistance variable and the FSA (α = 0.05). RESULTS: /findings: There were significant moderate correlations of FSA with mean torque (r = -0.44, p = 0.002), mean normalized torque (r = -0.42, p = 0.004) and resting position (r = 0.39, p = 0.007). The clinical measure of FSA is associated to the mechanical resistance of the MFJC: (a) the greater the FSA, the smaller the resistance torques; (b) the greater the FSA, the more inverted the forefoot resting position. CONCLUSIONS: These results showed that the clinical measure of FSA is moderately related to mechanical properties of the MFJC.

Brazil has the largest herd of goats of the American continent, with more than 9 million head. Goat farming is considered a growing and important activity for the economy, mainly in the northeastern region of Brazil .In this research, were determined the lipid content and the composition of fillet and shank fatty acids of young goats, registered in the inspection sector of the Ministry of Agriculture and marketed in the city of Salvador (Bahia). The average percentage of total lipids in the fillet was 1.68±0.04 and in the shank of 4.02±0.09, this is a significant difference between the cuts (P<0.05). Twenty-seven (27) fatty acids were identified in the cuts, most are palmitic acid (C16:0), stearic acid (C18:0) and oleic acid (C18:1ω9) present in 21.32%, 20.39%, 34.49% in the fillet and 21.74%, 21.94%, 33.38% in the shank, respectively. The total sums of saturated and polyunsaturated fatty acids ranged of 42.75 to 45.23% and 14.04 to 12.35% between fillet and shank cuts, respectively. The ratio of PUFA/SFA was 0.35 for fillet and 0.15 for shank, and the ratio ω6/ω3 was 3.07 for fillet and 2.27 for shank. Therefore, is a significant difference in total lipid content and fatty acid composition in commercial cuts of beef fillet and shank.(AU)


O Brasil apresenta a maior população de caprinos do continente americano, com mais de nove milhões de cabeças, a caprinocultura é considerada uma atividade em ascensão e de grande importância para a economia, principalmente da região nordeste. Os somatórios totais de ácidos graxos saturados e poli-insaturados variaram de 42,75 a 45,23% e de 14,04 a 12,35%, entre os cortes de filé e de pernil, respectivamente. A razão entre AGPI/AGS foi de 0,35 para o filé e de 0,15 para o pernil, e a razão ω6/ω3 foi de 3,07 para o filé e de 2,27 para o pernil. Constatou-se que existe uma diferença significativa no teor de lipídios totais e da composição de ácidos graxos em cortes comerciais de filé e pernil de carne caprina.(AU)

The melanocortin 1 receptor (MC1R) gene plays a key role in controlling the deposition of melanin. In mammals, the MC1Rgene is regarded as a major candidate gene in the control of melanin formation. In domestic animals, the MC1R gene mainly controls the expression of coat, skin, and plumage color in mammals and birds. In order to breed chickens with dark-green shank faster, we screened the molecular markers for shank color in a HS chicken population by exploring the relationship between polymorphism of the MC1R gene and three different shank colors (light green, dark green and yellow). Two primer pairs for code region of the MC1R gene were designed in the basic of chicken genomic sequence. DNA sequencing was performed to detect the polymorphisms and PCR was used to amplify DNA fragment. Sequences analysis indicated that 7 SNPs were predominant the three HS chicken populations with different shank color, including g.18,287,945C>T, g.18,288,088T>C, g.18,288,150G>A, g.18,288,303A>G, g.18,288,512G>A, g.18,288,513T>C, and g.18,288,520A>C. Association analysis revealed that the dark-green shank population showed moderate polymorphism, whereas the light-green shank population showed low polymorphism among overall 7 SNPs and that SNP6 (g.18,288,513T>C) may be significantly associated with three different shank colors in HS chickens. The haplotype CTGGACA had the largest haplotype frequencies, accounting for 56.22%, and the haplotype combination H1H1 is mainly distributed in the dark-green shank population, and may be used as molecular maker for marker-assisted selection of shank color in HS chickens.(AU)

The presence of a compacted soil layer near the ground surface in paddy fields may limit the growth and development of soybean roots. The objective of this study was to evaluate different planting management systems and irrigation on growth and development of soybean root systems in lowland area. The experiment was carried out in 2013/14 and 2014/15 crop seasons in randomized complete block design with factorial treatment (3x2), with four replications. The treatments consisted of different planting management systems: sowing with double disc (A1); sowing with shank (A2) and deep tillage + sowing with double disc (A3), and irrigation: irrigated (D1) and non irrigated (D2). Planting management systems and irrigation influenced the growth of soybean roots. When double disc was used, roots have lower growth and increase in diameter. Use of shanks and deep tillage provide increased growth and development of soybean roots and greater depth distribution. A presença de uma camada compactada próxima à superfície do solo em áreas de várzeas pode limitar o crescimento e desenvolvimento das raízes de soja. O objetivo do trabalho foi avaliar diferentes manejos de implantação e irrigação no crescimento e desenvolvimento do sistema radicular de soja em área de várzea. O experimento foi realizado nas safras 2013/14 e 2014/15 no delineamento experimental de blocos ao acaso em esquema fatorial (3x2), em faixa, com quatro repetições. Os tratamentos constaram de diferentes manejos de implantação da cultura: semeadura com disco duplo desencontrado (A1); semeadura com haste sulcadora (A2) e escarificação do solo + semeadura com disco duplo desencontrado (A3), e de irrigação: com irrigação (D1) e sem irrigação (D2). Os manejos de implantação e a irrigação influenciaram no crescimento das raízes de soja. No disco duplo, as raízes apresentam menor crescimento e aumento do diâmetro. Os manejos com haste sulcadora e escarificação do solo proporcionam maior crescimento e desenvolvimento de raízes de soja e maior distribuição em profundidade. Uma irrigação suplementar de 55mm no estádio V4 de desenvolvimento das plantas de soja proporciona aumento da área superficial e do volume de raízes em soja, quando a umidade do solo atinge valores abaixo de 60% da capacidade de campo.(AU)

ABSTRACT: The presence of a compacted soil layer near the ground surface in paddy fields may limit the growth and development of soybean roots. The objective of this study was to evaluate different planting management systems and irrigation on growth and development of soybean root systems in lowland area. The experiment was carried out in 2013/14 and 2014/15 crop seasons in randomized complete block design with factorial treatment (3x2), with four replications. The treatments consisted of different planting management systems: sowing with double disc (A1); sowing with shank (A2) and deep tillage + sowing with double disc (A3), and irrigation: irrigated (D1) and non irrigated (D2). Os tratamentos foram arranjados em parcelas subdivididas. As parcelas principais foram: área de corte (A1) e área de aterro (A2). As subparcelas foram os tipos de preparo do solo e de semeadura: com escarificação do solo e semeadura utilizando disco duplo na semeadora (D1); sem escarificação do solo e semeadura utilizando haste sulcadora na semeadora (D2); sem escarificação do solo e semeadura utilizando disco duplo na semeadora (D3). A cultivar utilizada foi a BMX Tornado. O tratamento com escarificação e haste sulcadora reduziu a resistência do solo à penetração e proporcionou maior teor de Mg e S no tecido foliar na safra 2013/14; e de N, P, K, Ca, Mg e S, em 2014/15. Esse tipo de preparo de solo possibilita maior rendimento de grão de soja sem diferenças entre as áreas de corte e de aterro.(AU)

RESUMO

Introdução: As pontas diamantadas são instrumentos rotatórios abrasivos utilizados principalmente na odontologia restauradora para realização de preparos dentários e acabamentos de restaurações. Estes instrumentos são formados por uma haste metálica, eixo intermediário e ponta ativa. Objetivo: Mensurar a resistência flexural da haste e intermediário de pontas diamantadas de seis marcas comerciais diferentes disponíveis no mercado brasileiro. Materiais e Métodos: utilizou- se 60 pontas diamantadas, sendo 30 pontas 2200 (n=5) e 30 pontas 2130 (n=5), das marcas KG Sorensen, Option, Microdont, FAVA, Zeep, Vortex. Os instrumentos rotatórios foram posicionados em dispositivo que permitiu que suas extremidades fossem apoiadas sobre bases metálicas; submetidas à Máquina Universal de Ensaio (EMIC), para mensurara força máxima aplicada em um deslocamento de 3 mm. Para o padrão de falha, verificou-se se as pontas sofreram dobras ou fraturas totais. Resultado: os instrumentos rotatórios da marca KG Sorensen destacaram-se em todos os testes, apresentando elevados valores de resistência flexural tanto para haste quanto para intermediário; não apresentando nenhuma falha do tipo fratura total. A marca Option apresentou o maior índice de instrumentos com falha do tipo fratura total.Conclusão: não existe padronização da resistência flexural entre as marcas comercias, sendo que a utilização de instrumentos com baixa resistência pode ocasionar maiores taxas de falha, danificando o equipamento ou causar injurias aos tecidos do paciente.


Introduction: The diamond burs are rotary abrasives instruments primarily used in restorative dentistry to perform dental preparations and finishing restorations. These instruments consist of shank, intermediate shaft and head. Objective: To measure the flexural strength of the shank and intermediate shaft diamond burs of six different commercial brands available in the Brazilian market. Methods: 60 diamond burs, 30 burs 2200(n = 5) and 30 burs 2135 (n = 5), of brands KG Sorensen, Option, Microdont, FAVA, Zeep, Vortex were used. The rotary instruments were placed in the device that allowed its ends were supportedon metal bases; submitted to Universal Testing Machine (EMIC), to measure the maximum force applied a displacement of 3mm. For the standard fault, it was found that the burs suffered bends or total fractures. Results: It was observed that the rotary instruments KG Sorensen, stood out all tests, showing high values of flexural strength for shank and intermediate shaft; showing no fault of type fracture total. Option had the highestrate of failure type instruments with total fracture. Conclusion: There is no standardization between the flexural trademarks, and the use of instruments with low resistance may cause higher rates of failure, damaging the equipment or cause injuries to the tissues of the patient.
