RESUMO

OBJECTIVE: Tumor-specific growth factor (TSGF) is an immune-related factor that offers good performance in the clinical management of human cancers. However, the role of serum TSGF in esophageal squamous cell carcinoma (ESCC) has not been fully clarified. METHODS: A total of 562 ESCC cases were collected in our study, with available information on preoperative serum levels of TSGF at diagnosis. Preoperative serum TSGF was detected using the rate method. We retrospectively analyzed its correlation with clinicopathological features of ESCC and survival. RESULTS: The cut-off value of serum TSGF was determined to be 60.5 U/mL by receiver operating characteristic (ROC) analysis. Serum TSGF was associated with gender (P < 0.001), tumor location (P = 0.022), tobacco use (P < 0.001), alcohol consumption (P < 0.001), lymph node involvement (P = 0.007), and TNM staging (P = 0.004). The survival analysis revealed that ESCC patients with high levels of serum TSGF had poorer prognosis than those with high TSGF (P = 0.006), especially for male ESCC cases (P = 0.001), under 60 year (P = 0.036), male middle location (P = 0.023), tobacco consumption (P = 0.004), G1 + G2 (P = 0.031), advanced T staging (P = 0.033), lymph node involvement (P = 0.003), TNM staging (P = 0.003). Univariate Cox regression analysis indicated that exposure to smoking and drinking, tumor grade, T staging, lymph node metastasis, TNM staging, and serum TSGF level were the prognosis-related factors of ESCC. Multivariate regression analysis revealed that smoking history, higher serum TSGF levels, and advanced T stage enhanced the risk of ESCC-related death. CONCLUSION: In brief, serum TSGF levels had in relation to malignant features of ESCC. It was positively correlated with survival but was identified as an independent risk factor for ESCC.

RESUMO

Despite the significant improvement in the treatment modalities, cancer is one of the fastest-growing chronic disease conditions all over the world. Genetic and Epigenetic alterations in the normal physiology of the cell are the key factor for tumor development. These changes can trigger the production of abnormal protein expressions through stimulation of different signaling pathways and can deeply affect normal cell growth and proliferation. Any altered protein expression, genetic variation, micro-RNA or post-translational protein modifications that indicate tumorigenesis can act as an early signal termed as biomarker. Cancer, being a multistep process with accumulating genetic and epigenetic alterations, could be detected early with suitable biomarkers. There are several proteins such as AFP, CA-125, PSA, troponin, CEA, osteopontin, CA 19-9 that act as biomarkers which help in early detection, prognosis, and monitoring of disease progression, a hunt for newer biomarkers with higher specificity and sensitivity is still ongoing. Tumor-specific growth factor (TSGF) is one such budding and prevailing tumor biomarker used for the early-stage detection of several types of carcinomas. TSGF is a gene that helps in tumor angiogenesis and gets released during the preliminary stages from cancer cells that ensure the vascular proliferation of the same. In this review, the clinical investigations of TSGF in different kinds of malignancy is discussed in detail and suggests the possibility of using TSGF as a biomarker in early diagnosis of cancer.

RESUMO

Extramedullary plasmacytoma (EMP) of the gastrointestinal tract is an extreme rarity. Clinical manifestations of EMPs are varied, depending on the position and progress of tumor. Here we report a case of an EMP involving rectum in an 80-year-old, male patient with a change of bowel habit. Computed tomography scanning confirmed a circumscribed, iso-attenuating mass with the obvious heterogeneous enhancement. Patient received the surgical resection by laparoscope and the plasmacytoma was finally confirmed by the pathology. Furthermore, we made a literature review about the EMP of gastrointestinal tract to get the further study. Finally, we found out there is no specificity in imaging examination. Diagnosis of EMP still depends on the histopathology.

RESUMO

BACKGROUND: The purpose of this study is to evaluate the expression of tumor-specific growth factor (TSGF) and microRNA-214 (miR-214) in the serum of patients with primary hepatocellular carcinoma (PHC) and their predictive values for the curative effect of transcatheter arterial chemoembolization (TACE). METHODS: A retrospective analysis of the clinical data of 87 PHC patients were treated with TACE. According to the curative effect 1 month after TACE, PHC patients were divided into disease control group (n=56) and disease progression group (n=31). The expression levels of TSGF and miR-214 were detected by qRT-PCR before or after treatment with TACE in disease control group and disease progression group. The predictive value of TSGF and miR-214 for the efficacy of TACE were evaluated by the receiver operating characteristic (ROC) curve. The Kaplan-Meier survival curve was drawn according to the critical value of ROC. The effect of pretreatment levels of TSGF and miR-214 was analyzed on the three-year survival rate of patients. RESULTS: After TACE treatment, the mRNA expressive of serum TSGF were increased in progression group, while the levels of miR-214 were decreased compared with control group (P<0.05). Importantly, at one month after TACE treatment, the levels of TSGF were decreased in control group and progression group, while the expression of miR-214 was increased compared with in both groups before TACE treatment(P<0.05). The result of ROC analysis showed that for predicting the curative effect of TACE, the levels of TSGF and miR-214 expression were (181.32, 0.63) (0.849 and 0.807) and (0.759-0.938, 0.707-0.907) in the cutoff values, AUCs and 95% confidence intervals, respectively. The result of Kaplan-Meier analysis showed that the 3-year survival rate of the low TSGF group was up-regulated than that of the high TSGF group [65.12% (31/44) vs. 30.23% (13/43); χ2=5.014; P=0.025]. The 3-year survival rate of the miR-214 high expression group was up-regulated than that of the miR-214 low expression group [61.70% (29/47) vs. 30.00% (12/40); χ2=6.928; P=0.008]. CONCLUSIONS: Serum TSGF and miR-214 could be used as potential biomarkers for PHC diagnosis and prognosis.

RESUMO

CONTEXT: Osteosarcoma (OS) is a progressive primary bone tumor that originates from immature stromal spindle cells. After chemotherapy, the serum-related indexes which are related to the prognosis. AIMS: The aim of this study is to investigate the correlation between changes in serum cyclooxygenase-2 (COX-2), basic fibroblast growth factor (bFGF), vascular endothelial growth factor (VEGF), transforming growth factor-beta (TGF-ß), and tumor-specific growth factor (TSGF) levels and prognosis of patients with osteosarcoma (OS) before and after treatment. SETTINGS AND DESIGN: Data of 75 patients with OS (observation group) and 55 healthy controls (control group) were retrospectively analyzed. MATERIALS AND METHODS: Chemotherapy was administered to the observation group. Serum lactate dehydrogenase, alkaline phosphatase, and TSGF levels were measured before and after treatment. The observation group patients were classified as normal or abnormal according to the changes in serum COX-2, bFGF, VEGF, TGF-ß, and TSGF levels after chemotherapy. Patients were followed up for 7.5 years, and the survival rate was determined. STATISTICAL ANALYSIS USED: Single-factor influencing prognosis was included in the Cox model, and independent factors influencing prognosis were analyzed. RESULTS: After chemotherapy, the mean serum COX-2, bFGF, VEGF, and TSGF levels decreased significantly in the observation group but were still higher than those in the control group. Furthermore, serum TGF-ß levels increased in the observation group but were still lower than those in the control group. The 5-year survival rate of patients with normal serum COX-2, bFGF, VEGF, and TSGF levels was significantly higher in the normal subgroup than in the abnormal subgroup. Cox analysis showed that the Enneking stage and COX-2 level after chemotherapy were independent prognostic factors. CONCLUSIONS: The serum COX-2, bFGF, VEGF, and TSGF levels of patients with OS significantly changed after chemotherapy, and the short-term survival rate of patients with normal levels of these biomarkers after chemotherapy was high.

RESUMO

Objective: To discuss application of apparent diffusion coefficient (ADC) in diffusion weighted imaging (DWI) combined with serum alkaline phosphatase (ALP) and tumor specific growth factor (TSGF) in evaluating neoadjuvant chemotherapy for osteosarcoma. Methods: A total of 78 patients with osteosarcoma who were admitted to People's Hospital of Gansu from January 2016 to August 2018 were collected as study subjects. All the patients were treated with neoadjuvant chemotherapy. Before chemotherapy, at the end of 4 courses of chemotherapy, before and after surgery, MRI examination and detection of serum ALP and TSGF were performed. According to results of pathological examination, the 78 patients were divided into effective chemotherapy group (n=54) and ineffective chemotherapy group (n=24). ADC values, levels of serum ALP and TSGF, change rates of ADC values and levels of serum ALP and TSGF were compared between the two groups. The value of ADC value combined with serum ALP and TSGF in evaluating curative effect of neoadjuvant chemotherapy for osteosarcoma was analyzed with receiver operating characteristic curve (ROC). Results: After chemotherapy, ADC value in effective chemotherapy group increased significantly, while levels of serum ALP and TSGF decreased significantly (t=7.269, 18.778, 23.237, all P<0.05). Only after surgery, ADC value, levels of serum ALP and TSGF increased or decreased significantly in ineffective chemotherapy group (t=7.316, 15.083, 20.930, all P<0.05). Before and after chemotherapy, change rates of ADC values and levels of serum ALP and TSGF in effective chemotherapy group were all significantly higher than those in ineffective chemotherapy group (t=7.604, 5.482, 5.048, all P<0.05). ROC curve analysis showed that area under the curve (AUC) of ADC value combined with serum ALP and TSGF for evaluating curative effect of neoadjuvant chemotherapy was 0.912, which was higher than that of ADC value, ALP, TSGF, ADC value combined with ALP, ADC value combined with TSGF (0.847, 0.787, 0.701, 0.885, 0.876, respectively). Conclusion: ADC value combined with serum tumor markers ALP and TSGF is reliable in evaluating curative effect of neoadjuvant chemotherapy for osteosarcoma.

RESUMO

In this paper, an ultrasensitive electrochemical immunosensor was developed for the detection of tumor specific growth factor (TSGF). Reduced graphene oxide-tetraethylene pentamine (rGO-TEPA) was used to modify the surface of glassy carbon electrode (GCE). Meanwhile, Ag@CeO2 nanocomposite was synthesized and applied as secondary-antibody (Ab2) labels for the fabrication of the immunosensor. The amperometric response of the immunosensor for the reduction of H2O2 was recorded. Simultaneously, electrochemical impedance spectroscopy (EIS) and Cyclic voltammetry (CV) were used to characterize the fabrication process of the immunosensor. The anti-TSGF primary antibody (Ab1) was immobilized onto the rGO-TEPA modified GCE via cross-linking with glutaraldehyde (GA). And then the TSGF antigen and Ab2-Ag@CeO2 were modified onto the electrode surface in sequence. Under the optimal conditions, the immunosensor exhibited a wide linear range (0.500-100pg/mL), a low detection limit (0.2pg/mL), good reproducibility, acceptable selectivity and excellent stability. The proposed sensing strategy may provide a potential application in the detection of other cancer biomarkers.

RESUMO

OBJECTIVE: To investigate the effect of direct moxibustion at Ganshu (BL18) on the serum concentrations of tumor specific growth factor (TSGF) and tumor necrosis factor α (TNF-α) in a rat model with precancerous lesion of primary hepatocellular carcinoma (HCC), so as to explore the mechanism of moxibustion underlying improvement of HCC. METHODS: Sixty male Wistar rats were randomly divided into control group (n=10), model group (n=20), prevention group 1 (n=15) and prevention group 2 (n=15). The normal rats were injected with physiological saline as blank control. At the same time, the rats of other three groups were injected with diethylnitrosamine to establish the HCC model. Direct moxibustion with grain-sized moxa was applied to bilateral Ganshu acupoint of the rats in the prevention group 1 (1 treatment course, 20 days) and prevention group 2 (2 treatment courses, 40 days), 5 doses for each acupoint, 0.5 mg/dose, once every other day. At each time point (before model establishment, the end of 1st course prevention, the end of 2nd course prevention and the end of model establishment), serum levels of TSGF and TNF-α were detected using enzyme-linked immunosorbent assay. RESULTS: Compared with the control group, there was a remarkably increase of serum TSGF and TNF-α contents in the model group at the end of the experiment (P<0.05). At the end of the 1st course of direct moxibustion, the contents of serum TSGF and TNF-α of rats in the prevention group 1 were significantly increased compared with that of the model group (P<0.05). At the end of the 2nd course of direct moxibustion, serum TSGF and TNF-α levels of rats in the model group were higher than the normal group with significantly difference (P<0.05), and the levels of TSGF and TNF-α in the prevention group 2 were significantly reduced in comparison with the model group (P<0.05). CONCLUSION: It was possible that direct moxibustion could inhibit precancerous lesion and postpone hepatocarcinogenesis, and the therapeutic effect of two courses were better than one course.

RESUMO

This study aimed to compare the changes and determine the clinical significance of carbohydrate antigens CA242, CA199, CA125, carcinoembryonic antigen (CEA), and tumor-specific growth factor (TSGF) before and after cryoablation by Cryocare system. Thirty-one pancreatic cancer patients were selected to receive cryoablation by Cryocare system. The serum expression levels of CA242, CA199, CA125, CEA, and TSGF before and 1 month after treatment were determined. Meanwhile, the serum level of these factors was also determined in 31 healthy volunteers. The parameter changes were analyzed with the clinical pathological data. The serum levels of CA242, CA199, CA125, CEA, and TSGF in the pancreatic cancer group were significantly higher than those of the control group both before and after the cryoablation treatment (P < 0.05). The serum CA199, CEA, and TSGF dramatically decreased 1 month after the treatment, which were statistically different (P < 0.05). The positive rates of serum CA242, CA199, CA125, and CEA in the pancreatic cancer group were much higher than those in the control group both before and after treatment (P < 0.05), and the positive rate of TSGF was significantly higher than that of the control group before the treatment (P < 0.05). The positive rate of CA199, CEA, and TSGF after the treatment was significantly lower than that before the treatment (P < 0.05). Serum level of CA242 was correlated with the tumor diameter, clinical staging, tumor differentiation, lymph node, and liver metastasis (P < 0.05). Except gender, CA199 was correlated with all the other clinical pathological parameters (P < 0.05). The serum levels of CA125 and CEA were correlated with all the other clinical pathological parameters (P < 0.05). The serum level of TSGF was only correlated with tumor differentiation (P < 0.05). Cryoablation treatment by Cryocare system can decrease the serum levels of CA199, CEA, TSGF, and the positive rate. Serum CA199, CEA, and TSGF can be important index for pancreatic cancer treatment assessment. Serum levels of CA242, CA199, CA125, and CEA are of great clinical value for metastasis assessment and prognosis in pancreatic cancer patients.

SCC and TSGF amount were measured by MEIA and biochemistry olorimetry method respectively, and were compared. Results The mean level of serum SCC and TSGF in the cervical cancer group was (6.95 ± 1.23) ng/mL and (81.0 ± 16.3)U/mL respectively, significantly higher than that in the CIN group and control group. The sensitivity and accuracy of combining SCC with TSGF in diagnosing cervical carcinoma were 91.1%and 87.4% respectively, both higher than allying SCC or TSGF alone. Conclusions SCC and TSGF levels are helpful in the diagnosis of cervix carcinoma. The combined determination can improve the sensitivity and accuracy of cervical carcinoma diagnose.

RESUMO

Objective To investigate the clinical value of combined detection of the serum levels of CA125,CA72-4 and tumor specific growth factor(TSGF)in the diagnosis of ovary cancer and the curative effect. Methods Serum levels of CA125,CA72-4 and TSGF in 68 patients with ovary cancer,53 patients with benign ovary rumor and 50 normal controls were measured by chemiluminescent immunoassay(CLIA)and chemical colorimetry. Results and clinical data were statisticaly analyzed. Results The levels of CA125,CA72-4 and TSGF in ovary cancer group were higher than that in the benign ovarian tumor group and healthy controls(all P <0.01).When use three tumor markers united detection, sensitivity, specificity and accuracy were 91.2%,83.0% and 87.4% respectively. The sensitivity and accuracy was higher than any single detection. The specificity with single CA125 testing was consistently. The levels of CA125,CA72-4 and TSGF in patients with ovary cancer were significantly different after surgical treatment for five days(all P <0.05).Conclusion The combined detection of the three markers may increase the positive rate in the early diagnosis of ovarian cancer, and help to differentiate the benign and malignant ovarian tumor, but also is valuable to observe the curative rate and postoperative monitor of the ovary cancer.

RESUMO

Objective To investigate the clinical value ofcombined detection of the serum levels of CA125,TSGF and DD in the diagnosis of ovarian cancer. MethodsThe serum CA125 and DD were determined by enzyme linked immunosorbent assay and TSGF was determined by biochemistry shade selection in all 252 patients(including 84 cases of ovarian cancer,84 cases of ovarian benign diseases,84 cases of controls). ResultsThe concentration of CA125,TSGF and DD in the diagnosis of ovarian cancer group were higher than those in ovarian benign diseases group and the controls(all P<0.05).There was no statistical significance in the concentration of CA 125,TSGF and DD between ovarian benign diseases group and the controls(all P > 0.05).In combined assay of CA 125,TSGF and DD,the sensitivity rate was 94.05%,the specificity rate was 90.48%,and the accuracy rate was 92.26%.Comparing with the single assay, combined analysis had much higher diagnostic sensitivity(all P<0.05). ConclusionThe assay of CA125,TSGF and DD had significant dinical value in diagnosis of breast cancer, combined detection could increase the positive detection of ovarian cancer, and compensate the use of only one tumor marker.
