BACKGROUND: The species of genus Ageratum (family Asteraceae) are distributed in various parts of the world. Ageratum conyzoides and A. houstonianum are the most commonly occurring species in India. These species are quite similar in their morphology thus creating a challenge in identification during the field survey and taxonomic validation. The accurate identification of the species is highly significant especially when those are of medicinal interest. To overcome the barriers in morphological based identification, DNA barcoding has been employed during the present investigation. METHODS AND RESULTS: Morphological and DNA barcodes matK and ITS genes, were employed to differentiate between Ageratum conyzoides and A. houstonianum. The obtained matK and ITS gene sequences were submitted to GenBank and BOLD system to obtain accession numbers. The DNA sequences were aligned with database sequences using BLAST and phylogenetic trees were constructed through neighbor-joining algorithm in MEGA 11 software. The distinguish features of A. conyzoides include ovate to elliptic-oblong leaves with a cuneate base and inflorescence heads forming domed to flat-topped clusters. However, A. houstonianum has triangular to ovate leaves with a cordate to truncate base, cymose clusters in the inflorescence and stipulate glandular involucre bracts. The matK gene has shown the highest identity percentages (100%) for A. houstonianum and 99.87% for A. conyzoides. The phylogenetic tree analysis has demonstrated a close association of A. conyzoides and A. houstonianum with their respective species, supported by bootstrap values in the matK and ITS trees. CONCLUSION: This study revealed that morphological and molecular data can be successfully utilized in the identification of A. conyzoides and A. houstonianum. The matK and ITS barcodes provide promising results in the identification of Ageratum species, with their phylogeny supporting classification within the family asteraceae.

Este trabalho objetivou avaliar a divergência genética entre nove acessos de mentrasto (Ageratum conyzoides) por métodos multivariados utilizando caracteres morfológicos, em quatro épocas de colheita. Os nove acessos de mentrasto foram obtidos de plantas que ocorrem naturalmente nas cinco regiões climáticas do Estado do Tocantins. Em cada época de colheita foram avaliadas as características como biomassa fresca da parte aérea, biomassa seca, área foliar, número de folhas, floração e altura. Pela análise de agrupamento (método de Tocher) foram formados dois grupos na primeira e quarta épocas de colheita, três grupos na segunda época e quatro grupos na terceira época de colheita. Observou-se variação na constituição dos grupos pelos acessos nas épocas de colheita. A divergência genética entre acessos de mentrasto, avaliada por caracteres morfológicos, foi influenciada pelo estádio de desenvolvimento da espécie. A existência de variabilidade entre os acessos coletados em diferentes localidades fornece subsídio para a coleta sistematizada de germoplasma em A. conyzoides no Estado do Tocantins.


This study aimed to analyze the genetic divergence among nine mentrasto (Ageratum conyzoides) accessions by multivariate methods using morphologic traits for four harvesting periods. The nine accessions of mentrasto were obtained from plants that occur naturally in five climatic regions of the state of Tocantins. At each harvesting period, the following traits were evaluated: fresh biomass of the aerial part, dry biomass, leaf area, number of leaves, flowering and height. The grouping analysis (Tocher's method) identified two groups, in the first and fourth harvest periods, three groups in the second harvest period and four groups in the third harvest period. Variation was observed for the group constitutions among the different harvesting periods. The genetic divergence among mentrasto accessions, evaluated by morphologic traits, was influenced by developmental stage of specie. The existence of genetic variability among accessions from different regions supports the systematic collection of A. conyzoides germplasm in the state of Tocantins.

We previously identified changes in gene expression in Ageratum conyzoides plant cells inoculated with Agrobacterium tumefaciens by using cDNA-AFLP. Here, we show that a subset of defense-related genes is differentially regulated by an Agrobacterium attachment-deficient mutant. The expression pattern triggered by this mutant is similar to that induced by inoculation with non-pathogenic bacteria. We also observed that the expression level of the defense genes was inversely correlated with the efficiency of transformation by Agrobacterium. We propose that the plant defense system has an important role in controlling infection and transformation and that Agrobacterium may dampen some plant defense responses.
