RESUMO

Chlordane is an organochlorine pesticide (OCP) that is environmentally persistent. Although exposures to OCPs including chlordane have been associated with elevated liver enzymes, current knowledge on OCPs' contribution to toxicant-associated steatotic liver disease (TASLD) and underlying sex-specific metabolic/endocrine disruption are still widely limited. Therefore, the objective of this study was to investigate the sex-dependent effects of chlordane in the context of TASLD. Age-matched male and female C57BL/6 mice were exposed to chlordane (20 mg/kg, one-time oral gavage) for two weeks. Female mice generally exhibited lower bodyfat content but more steatosis and hepatic lipid levels, consistent with increased hepatic mRNA levels of genes involved in lipid synthesis and uptake. Surprisingly, chlordane-exposed females demonstrated lower hepatic cholesterol levels. With regards to metabolic disruption, chlordane exposure decreased expression of genes involved in glycogen and glucose metabolism (Pklr, Gck), while chlordane-exposed females also exhibited decreased gene expression of HNF4A, an important regulator of liver identity and function. In terms of endocrine endpoints, chlordane augmented plasma testosterone levels in males. Furthermore, chlordane activated hepatic xenobiotic receptors, including the constitutive androstane receptor, in a sex-dependent manner. Overall, chlordane exposure led to altered hepatic energy metabolism, and potential chlordane-sex interactions regulated metabolic/endocrine disruption and receptor activation outcomes.

Increasing evidence from animal and epidemiological studies indicates that perinatal exposure to pesticides cause developmental neurotoxicity and may increase the risk for psychiatric disorders such as autism and intellectual disability. However, the underlying pathogenic mechanisms remain largely elusive. This work was aimed at testing the hypothesis that developmental exposure to different classes of pesticides hijacks intracellular neuronal signaling contributing to synaptic and behavioral alterations associated with neurodevelopmental disorders (NDD). Low concentrations of organochlorine (dieldrin, endosulfan, and chlordane) and organophosphate (chlorpyrifos and its oxon metabolite) pesticides were chronically dosed ex vivo (organotypic rat hippocampal slices) or in vivo (perinatal exposure in rats), and then biochemical, electrophysiological, behavioral, and proteomic studies were performed. All the pesticides tested caused prolonged activation of MAPK/ERK pathway in a concentration-dependent manner. Additionally, some of them impaired metabotropic glutamate receptor-dependent long-term depression (mGluR-LTD). In the case of the pesticide chlordane, the effect was attributed to chronic modulation of MAPK/ERK signaling. These synaptic alterations were reproduced following developmental in vivo exposure to chlordane and chlorpyrifos-oxon, and were also associated with prototypical behavioral phenotypes of NDD, including impaired motor development, increased anxiety, and social and memory deficits. Lastly, proteomic analysis revealed that these pesticides differentially regulate the expression of proteins in the hippocampus with pivotal roles in brain development and synaptic signaling, some of which are associated with NDD. Based on these results, we propose a novel mechanism of synaptic dysfunction, involving chronic overactivation of MAPK and impaired mGluR-LTD, shared by different pesticides which may have important implications for NDD.

Amyotrophic lateral sclerosis (ALS) is an incurable neurodegenerative disease characterized by the death of upper and lower motor neurons. While causative genes have been identified, 90% of ALS cases are not inherited and are hypothesized to result from the accumulation of genetic and environmental risk factors. While no specific causative environmental toxin has been identified, previous work has indicated that the presence of the organochlorine pesticide cis-chlordane in the blood is highly correlated with ALS incidence. Never before tested on the motor system, here, we show that cis-chlordane is especially toxic to motor neurons in vitro- and in vivo-independent of its known antagonism of the GABAA receptor. We find that human stem-cell-derived motor neurons are more sensitive to cis-chlordane than other cell types and their action potential dynamics are altered. Utilizing zebrafish larvae, we show that cis-chlordane induces motor neuron and neuromuscular junction degeneration and subsequent motor deficits in a touch-evoked escape response. Together, our work points to cis-chlordane as a potential sporadic ALS exacerbating environmental pollutant.

In birds, incubation-related behaviors and brood patch formation are influenced by hormonal regulation such as prolactin secretion. Brood patch provides efficient heat transfer between the incubating parent and the developing embryo in the egg. Importantly, several environmental contaminants are already known to have adverse effects on avian reproduction. However, relatively little is known about the effect of contaminants on incubation temperature (Tinc ) in wild birds. By using temperature thermistors placed into artificial eggs, we investigated whether the most contaminated parent birds are less able to provide appropriate egg warming and thus less committed to incubating their clutch. Specifically, we investigated the relationships among 3 groups of contaminants (organochlorines, perfluoroalkyl substances [PFASs], and mercury [Hg]) with Tinc and also with prolactin concentrations and brood patch size in incubating Arctic black-legged kittiwakes (Rissa tridactyla). Our results reveal that among the organochlorines considered, only blood levels of oxychlordane, the main metabolite of chlordane, a banned pesticide, were negatively related to the minimum incubation temperature in male kittiwakes. Levels of PFASs and Hg were unrelated to Tinc in kittiwakes. Moreover, our study suggests a possible underlying mechanism: since we reported a significant and negative association between blood oxychlordane concentrations and the size of the brood patch in males. Finally, this reduced Tinc in the most oxychlordane-contaminated kittiwakes was associated with a lower egg hatching probability. Environ Toxicol Chem 2018;37:2881-2894. © 2018 SETAC.

The Atlantic spotted dolphin (Stenella frontalis) is an endemic species of the tropical-temperate Atlantic Ocean with widespread distribution. Although this species has been the subject of a large number of studies throughout its range, it remains in the "data deficient" category of the International Union for Conservation of Nature (IUCN). Chemical pollution by persistent organic pollutants (POPs) has been listed as one of the major threats to this species, however, there is no information on a wide scale. Thus, the aim of the present study was to investigate the contamination status of spotted dolphins on the metapopulation level as well as determine spatial and temporal variations in POP concentrations and bio-accumulation. A total of 115 blubber samples collected from a large part of the Atlantic basin were analysed for PCBs, DDTs, PBDEs, chlordanes, HCB and mirex. Although PCBs and DDTs were the predominant compounds in all areas, inter-location differences in POP concentrations were observed. Dolphins found at São Paulo, southeastern coast of Brazil, had the highest PCB concentrations (median: 10.5 µg/g lw) and Canary Islands dolphins had the highest DDT concentrations (median: 5.13 µg/g lw). Differences in PCB patterns among locations were also observed. Dolphins from the Azores and São Paulo demonstrated a similar pattern, with relatively highly contributions of tetra- (6.8 and 5.2%, respectively) and penta-CBs (25.6 and 23.8%, respectively) and lower contributions of hepta-CBs (20.8 and 23.5%, respectively) in comparison to other areas. Moreover, the sex of the animals and the year in which sampling or capture occurred exerted an important influence on the majority of the POPs analysed. Comparisons with toxicity thresholds available in the literature reveal that the São Paulo and Canary Island dolphins are the most vulnerable populations and should be considered in future conservation and management programs for the Atlantic spotted dolphin.

This study was conducted to make clear the major intake route of DDT in Bangladesh people to develop strategy and policy that could lead to a reduction in body burden especially in the reproductive age women. The concentrations of several POPs (DDT, PCBs, chlordanes, HCHs, HCB, and PeCB) were quantified in food items, human breast milk and house dust collected in Bangladesh in 2011-2012. Among the POPs analyzed in this study, DDT and its metabolites (ΣDDT) showed the highest concentration. The highest median ΣDDT concentration was found in meat (1.3-1100 ng g-1 wet weight) and house dust (30-1100 ng g-1 dry weight), and followed by human breast milk (20-55 ng g-1 wet weight). Estimated daily intake (EDI) was calculated using the DDT concentrations in food items. The highest intake of DDT was found in an infant and 99% of it was via breast milk feeding. DDT intake via consumption of beef accounted for 69% and 72% of that found in children and adults, respectively. The total EDI of DDT did not exceed the tolerable daily intake proposed by the WHO, but the EDI of p,p'-DDT exceeded the oral reference dose proposed by the US Environmental Protection Agency. Further research is required to clarify the reason for the high levels of DDT in beef, which seems to be the major intake route of DDT for women of reproductive age in Bangladesh.

Although the use of engineered nanomaterials continues to increase, how these materials interact with coexisting contaminants in the environment is largely unknown. The effect of silver (Ag) in bulk, ionic, and nanoparticle (NP; bare and polyvinyl pyrrolidone-coated) forms at 3 concentrations (0 mg/kg, 500 mg/kg, 1000 mg/kg, 2000 mg/kg; ion at 69 mg/kg, 138 mg/kg, 276 mg/kg) on the accumulation of field-weathered chlordane and dichlorodiphenyldichloroethylene + metabolites (DDX) by Eisenia fetida (earthworm) was investigated. Earthworm biomass and survival were unaffected by treatment. At the 500 mg/kg and 1000 mg/kg exposure levels, NP-exposed earthworms contained significantly greater Ag (194-245%) than did the bulk exposed organisms; NP size or coating had no impact on element content. Generally, exposure to Ag of any type or at any concentration significantly reduced pesticide accumulation, although reductions for DDX (35.1%; 8.9-47.0%) were more modest than those for chlordane (79.0%; 17.4-92.9%). For DDX, the reduction in pesticide accumulation was not significantly affected by Ag type or concentration. For chlordane, the 3 NP exposures suppressed chlordane accumulation significantly more than did bulk exposure; earthworms exposed to bulk Ag contained 1170 ng/g chlordane, but levels in the NP-exposed earthworms were 279 ng/g. At the 500 mg/kg exposure, the smallest coated NPs exerted the greatest suppression in chlordane accumulation; at the 2 higher concentrations, chlordane uptake was unaffected by NP size or coating. The findings show that in exposed earthworms Ag particle size does significantly impact accumulation of the element itself, as well as that of coexisting weathered pesticides. Human breast milk samples (n=87) collected between July 2004 and July 2005 from primipara and multipara mothers from Thessaloniki, Greece were analysed for six groups of persistent organic pollutants (POPs): polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethane and its metabolites (DDTs), chlordane compounds (CHLs), hexachlorocyclohexane isomers (HCHs) and hexachlorobenzene (HCB). DDTs [median: 410ng/g lipid weight (lw)], PCBs (median: 90ng/g lw) and HCHs (median: 40ng/g lw) were the predominantly identified compounds in all the breast milk samples. Levels of PBDEs (median: 1.5ng/g lw) in human breast milk samples from Thessaloniki, Greece were lower compared to other countries. Maternal age had a positive correlation with most compounds, but not with PBDEs. Women with a higher occupational exposure to PBDEs (i.e., working in office environments) had higher PBDE concentrations than all others and showed strong correlations, especially for BDE 47 and BDE 153. None of the analysed compounds showed any correlation with parity. Based on these levels, the daily intake of each group of POPs via human milk was calculated and compared with the tolerable daily intakes (TDI) or the reference doses (RfD). For the majority of samples (85 out of 87) a higher daily intake of PCBs than the TDI was calculated, while 11 out of 87 samples had a higher HCB intake than the TDI. The TDI and the RfD were not exceeded for DDTs and PBDEs, respectively. This is the first report of brominated flame retardants in human breast milk from Greece.

Chlordane bioremediation using actinobacteria mixed culture is an attractive clean-up technique. Their ability to produce bioemulsifiers could increase the bioavailability of this pesticide. In order to select a defined actinobacteria mixed culture for chlordane remediation, compatibility assays were performed among six Streptomyces strains. The strains did not show growth inhibition, and they were assayed for chlordane removal, either as pure or as mixed cultures. In pure cultures, all of the strains showed specific dechlorination activity (1.42-24.20 EU mg(-1)) and chlordane removal abilities (91.3-95.5%). The specific dechlorination activity was mainly improved with cultures of three or four microorganisms. The mixed culture consisting of Streptomyces sp. A2-A5-A13 was selected. Their ability to produce bioemulsifiers in the presence of glucose or chlordane was tested, but no significant differences were observed (p > 0.05). However, the stability of the emulsions formed was linked to the carbon source used. Only in chlordane presence the emulsions retained 100% of their initial height. Finally, the selected consortium showed a high degree of sporulation in the pesticide presence. This is the first study on the effects that chlordane exerts on microbe morphology and emulsifier production for a defined mixed culture of Streptomyces with ability to remediate the pesticide.

Levels of organohalogen compounds (PCBs, chlordane, PBB 153, PBDEs, HCB) were determined in adipose tissue, liver, kidney and muscle of four polar bears which swam and/or drifted to Iceland in extremely malnourished condition. Since the colonization in the 9th century polar bears have been repeatedly observed in Iceland. However, in recent years three of the animals have clearly left their natural habitat in poor condition in May or June, i.e. at the end of the major feeding season. The fourth bear is believed to have drifted with melting ice to North-Eastern Iceland in mid-winter. The concentrations of the POPs were within the range or higher than the typical concentrations measured in polar bears from the East Greenland population. In addition to the targeted compounds, we tentatively detected Dechlorane 602 and its potential hydrodechlorinated Cl11-metabolite in all samples. Moreover, a polychlorinated compound which partly co-eluted with PCB 209 was detected in all liver samples but not in adipose tissue, kidney or muscle. The mass spectrum of the potential metabolite did not allow determining its structure. Polar bears are good swimmers and can reach Iceland from the ice edge of East Greenland within a few days. Potential reasons for the swims are briefly discussed.

Concentrations of polychlorinated biphenyls (PCBs), dichlorodiphenyl trichloroethane and its metabolites (DDTs), hexachlorocyclohexane isomers (HCHs), hexachlorobenzene (HCB) and chlordane compounds (CHLs) were determined in the blubber of males (20-25 years old) of Antarctic minke whales, Balaenoptera bonaerensis, from the International Whaling Commission (IWC) management Areas IV (70°-130°E) and V (130°E-170°W), south 60°S. The ranges of concentrations (ng g(-1) lipid wt.) for each compound were, PCBs: 7.7-89; DDTs: 29-340; HCHs: 0.20-4.3; HCB: 75-430; CHLs: 10-120, which were much lower than those in common minke whales, Balaenoptera acutorostrata, from the northern hemisphere. The levels of PCBs, HCHs, HCB and CHLs in Area IV were significantly higher than those in Area V, while the levels of DDTs in both areas were similar. For comparing the fate among four pesticides in the Antarctic Ocean avoiding the effect of variance due to food intake, the ratios of the pesticides to PCBs, which has an extremely high chemical stability and environmental persistence, were examined. The HCHs/PCBs ratio decreased by a factor of about 20 in a span of 16 years in both Areas IV and V, while temporal trends of DDTs/PCBs, HCB/PCBs and CHLs/PCBs ratios were not observed. These results indicate that PCBs, DDTs, HCB and CHLs levels did not vary or slightly decreased in Areas IV and V during the study period. However HCHs levels clearly decreased. Spatial differences seems to be related to differences in food intake among whales, and temporal differences seems to be related to the length stay of OCs in the Antarctic Ocean.

Temporal trends of persistent organic pollutants (POPs) in arctic foxes (Vulpes lagopus) from Svalbard, Norway, were investigated in relation to feeding habits and seasonal food availability. Arctic foxes from Svalbard forage in both marine and terrestrial ecosystems and the availability of their food items are impacted by climatic variability. Concentrations of polychlorinated biphenyls (PCBs), organochlorinated pesticides (OCPs) and brominated flame retardants (polybrominated diphenyl ethers [PBDEs] and hexabromocyclododecane [HBCDD]) were analyzed in the liver of 141 arctic foxes collected between 1997 and 2013. Stable carbon isotope values (δ13C) were used as a proxy for feeding on marine versus terrestrial prey. The annual number of recovered reindeer carcasses and sea ice cover were used as proxies for climate influenced food availability (reindeers, seals). Linear models revealed that concentrations of PCBs, chlordanes, p,p'-DDE, mirex and PBDEs decreased 4-11% per year, while no trends were observed for hexachlorobenzene (HCB) or ß-hexachlorocyclohexane (ß-HCH). Positive relationships between POP concentrations and δ13C indicate that concentrations of all compounds increase with increasing marine dietary input. Increasing reindeer mortality was related to lower HCB concentrations in the foxes based on the linear models. This suggests that concentrations of HCB in arctic foxes may be influenced by high mortality levels of Svalbard reindeer. Further, ß-HCH concentrations showed a positive association with sea ice cover. These results in addition to the strong effect of δ13C on all POP concentrations suggest that climate-related changes in arctic fox diet are likely to influence contaminant concentrations in arctic foxes from Svalbard.

Persistent organic pollutants (POPs) are thought to enhance the risk for later development of allergic disease like atopic dermatitis (AD). However, few studies have evaluated the effect of foetal exposure to various POPs on the development of AD in early infancy. Here, we describe the impact of foetal exposure to a number of POPs on the occurrence of AD in 7-month-old infants. The participants were 81 infants with or without AD who participated in a birth cohort study, where the concentrations of 15 polychlorinated biphenyl (PCBs) congeners, dichlorodiphenyltrichloroethane (p,p'-DDT), dichlorodiphenyldichloroethylene (p,p'-DDE), ß-hexachlorocyclohexane (ß-HCH), hexachlorobenzene (HCB), cis-nonachlor, trans-nonachlor, mirex, oxychlordane, and 27 polybrominated diphenyl ether (PBDEs) congeners were measured in the umbilical cord tissues collected immediately after birth. At 7 months, 27 of the 81 infants (33.8%) were diagnosed with AD. Of all POPs examined, total concentrations of 27 PBDE congeners were associated with a significantly decreased incidence of AD. Notably, the concentration of 27 PBDEs was significantly lower in AD infants than in non-AD infants (P<0.01), and the risk of AD development decreased with increasing PBDE levels. These results suggest that foetal exposure to PBDEs is a possible contributing factor to reducing AD in early infancy.

Peregrine falcon eggs were collected in South Greenland between 1986 and 2003 and analysed for 6 congeners of toxaphene and 5 chlordane-related pesticides (cis-chlordane, trans-chlordane, cis-nonachlor, trans-nonachlor and oxychlordane). Oxychlordane had the highest median concentration of 1448 ng/g lipid weight (lw) or 79 ng/g wet weight (ww) of all compounds. Of the toxaphene congeners, CHB-50 was the predominant congener, with a median concentration of 215 ng/g lw (15.5 ng/g ww). Chlordane-related concentrations were comparable with results from the USA, but lower than those from Canada and Norway. Toxaphene was considerably higher than in eggs of Norwegian peregrine falcon eggs, possibly reflecting different toxaphene usage in the areas of peregrine falcon migration. Toxaphene information in birds of prey is limited and comparisons with other species indicate differences in concentrations and organochlorine pattern. Temporal trends showed significant log-linear decreases for cis-chlordane and trans-chlordane, but no significant trends for other chlordanes or the toxaphene congeners, probably affected by the large variation between eggs, also of the same bird. Correlations between chlordanes and toxaphene in the Greenland peregrine falcon eggs might suggest similar sources of exposure.

The effect of multiwalled carbon nanotubes (MWCNT) or C60 fullerenes on the uptake of weathered chlordane or DDx (DDT + metabolites) by Cucurbita pepo (zucchini), Zea mays (corn), Solanum lycopersicum (tomato), and Glycine max (soybean) was investigated. The plants were grown in 50 g of soil with weathered chlordane (2150 ng/g) and DDx (118 ng/g) that was amended with 0, 500, 1000, or 5000 mg/kg MWCNT or C60. After 28 d, the root and shoot content of chlordane components and DDx was determined by GC-MS. Zucchini and tomato growth were unaffected by carbon nanomaterial coexposure, although C60 at 500 mg/kg reduced corn and soybean biomass by 36.5-45.0%. Total chlordane content ranged from 1490 (tomato) to 4780 (zucchini) ng; DDx amounts ranged from 77.8 (corn) to 395 ng (zucchini). MWCNT coexposure decreased chlordane and DDx accumulation 21-80% across all crops, depending on species and nanotube concentration. Conversely, C60 had species- and contaminant-specific effects on pesticide uptake, ranging from complete suppression of DDx uptake (corn/tomato) to 34.9% increases in chlordane accumulation (tomato/soybean). The data show that pesticide accumulation varies greatly with crop species and carbon nanomaterial type/concentration. These findings have implications for food safety and for the use of engineered nanomaterials in agriculture.

Cucurbita pepo ssp pepo (zucchini) accumulates significant levels of persistent organic pollutants in its roots, followed by unexpectedly high contaminant translocation to the stems. Most other plant species, including the closely related C. pepo ssp ovifera (squash), do not have this ability. To investigate the mechanism of contaminant accumulation, two cultivars each of parental zucchini and squash, as well as previously created first filial (F1) hybrids and F1 backcrosses (BC) of those parental cultivars, were grown under field conditions in a soil contaminated with weathered chlordane (2.29 microg/g) and DDX residues (0.30 microg/g; sum of DDT, DDE, DDD). The parental zucchini had stem-to-soil bioconcentration factors (BCF, contaminant ratio of stem to soil) for chlordane and DDX of 6.23 and 3.10; these values were 2.2 and 3.7 times greater than the squash, respectively. Chlordane and DDX translocation factors, the ratio of contaminant content in the stems to that in the root, were 2.1 and 3.2 times greater for zucchini than for squash. The parental zucchini and squash also differed significantly in chlordane component ratios (relative amounts of trans-nonachlor [TN], cis-chlordane [CC], trans-chlordane [TC]) and enantiomer fractions for the chiral CC and TC. Hybridization of the parental squash and zucchini resulted in significant differences in contaminant uptake. For both the three separate component ratios (CR) and two sets of enantiomer fraction (EF) values, subspecies specific differences in the parental generation became statistically equivalent in the F1 hybrid zucchini and squash. When backcrossed (BC) with the original parental plants, the zucchini and squash F1 BC cultivars reverted to the statistically distinct CR and EF patterns. This pattern of trait segregation upon hybridization suggests either single gene or single locus control for persistent organic pollutant (POP) uptake ability by C. pepo ssp pepo.

Chlordane, heptachlor, and their metabolites are chiral persistent organic pollutants that undergo enantiomeric enrichment in the environment. This study investigated the enantioselective metabolism of both chlordane isomers and heptachlor, major components of technical chlordane, by liver microsomes prepared from male rats treated with corn oil (CO) or inducers of CYP2B (PB; phenobarbital) and CYP3A enzymes (DX; dexamethasone), isoforms induced by chlordane treatment. The extent of the metabolism of all three parent compounds was dependent on the microsomal preparation used and followed the rank order PB > DX > CO. The mass balances ranged from 49 to 130% of the parent compound added to the microsomal incubations. Both cis- and trans-chlordane were enantioselectively metabolized to oxychlordane (EF = 0.45-0.89) and 1,2-dichlorochlordene (EF = 0.42-0.90). Heptachlor was metabolized enantioselectively, with heptachlor epoxide B (EF = 0.44-0.54) being the only metabolite. Interestingly, the direction on the enrichment for oxychlordane, 1,2-dichlorochlordene, and heptachlor epoxide differed depending on the microsomal preparation. These findings demonstrate that the direction and extent of the enantioselective metabolism of both chlordane isomers and heptachlor is P450 isoform-dependent and can be modulated by the induction of P450 enzymes.

Concentrations of persistent organochlorine compounds (OCs) including polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), and polychlorinated biphenyls (PCBs) in the liver and adipose tissue of Japanese cadavers were measured, and their toxicokinetics were examined in association with hepatic cytochrome P450 (CYP) 1A protein expression levels. Total 2,3,7,8-tetrachlorodibenzo-p-dioxin toxic equivalents (TEQs) were 66±74 and 65±57 pg/g lipid weight (mean±S.D.) in the liver and adipose tissue, respectively. Total PCBs (sum of 62 congeners targeted), p,p'-dichlorodiphenyl-dichloroethylene (p,p'-DDE) and ß-hexachlorocyclohexane (ß-HCH) were detected at concentrations over 1 µg/g lipid in both tissues of some specimens. For most of the dioxin-like congeners, total PCBs, p,p'-DDE, oxychlordane, α- and ß-HCH, hexachlorobenzene (HCB), and tris(4-chlorophenyl)methane (TCPMe), age-dependent increases in concentrations were found in the adipose tissue of males. No such age-dependent trend was observed in the liver, suggesting that there are different mechanisms underlying the hepatic concentrations of OCs. Immunoblot analyses indicated detectable expression of hepatic CYP1A2 protein, whereas no CYP1A1 protein was detected. The CYP1A2 expression levels were positively correlated with concentrations (on wet weight basis) of 2,3,4,7,8-P5CDF, the dominant TEQ-contributed congeners in the liver, indicating the induction of this CYP. Hepatic CYP1A2 protein levels were strongly correlated with the liver to adipose concentration (L/A) ratios of PCDD/F congeners with more than 5 chlorine atoms. Together with higher concentrations of the congeners in the liver than in the adipose tissue, the observation on L/A ratios of highly chlorinated PCDD/Fs suggests that induced hepatic CYP1A2 protein is involved in their sequestration in this human population, as observed in model animals (rodents). Nonetheless, the magnitude of hepatic sequestration (L/A ratio) of PCDD/Fs in this human population was lower than in other mammals and birds, reported previously. This study emphasizes the fact that toxicokinetics of some OCs can be affected at least partly by CYP1A2 protein levels in humans. For the extrapolation of their toxicokinetics from model animals to humans, knowledge on the induction and sequestration potencies of CYP1A is necessary.

This paper analyzes the influence of mussel physiological conditions on bioaccumulation of organochlorine pollutants. A direct relationship between reproductive stage, lipid content and accumulation of PCBs, DDTs and HCHs was observed. The organochlorine compounds accumulated by mussels cultured in the Ría of Vigo show levels below the limits set by the European legislation for human consumption, ranging between 12.50-58, 2.21-10.80 and 0.50-2.69 µg kg(-1) dry weight for PCBs, DDTs and HCHs, respectively. Other organochlorine pesticides normally present in estuarine areas, such as hexachlorobenzene (HCB), aldrin, chlordane and nonachlor were not detected. The proportion of PCBs, between 2 and 7 times higher than OCPs, indicates a pollution of predominantly industrial origin in the sampling area. Several molecular indices were used to identify the source of accumulated OCPs by Mytilus, showing recent inputs of technical DDT, mainly in summer-autumn, and continued discharges of lindane. On the other hand, other pesticides containing DDT impurities, such as dicofol, can contribute to DDT pollution in the Ría of Vigo. Multivariate analysis suggests the usefulness of including physiological parameters such as the gonad condition index and lipid content of sentinel organisms in environmental biomonitoring of lipophilic pollutants, as well as in the determination of their origin.

Legacy organochlorine contaminants were determined in adipose tissues from 294 polar bears (Ursus maritimus) sampled in East Greenland in 23 of the 28years between 1983 and 2010. Of 19 major legacy contaminants and congeners (ΣPCB, 4 PCB congeners (CB153, 180, 170/190), ΣDDT, p,p'-DDE, p,p' -DDD and p,p'-DDT, α- and ß-hexachlorocyclohexane (HCH), HCB, octachlorostyrene, dieldrin, oxychlordane, cis- and trans-chlordane, cis- and trans-nonachlor, heptachlor epoxide and BB-153), 18 showed statistically significant average yearly declines of -4.4% (range: -2.0 to -10.8%/year) among subadult polar bears (i.e. females<5years, males<6years). For example, the ∑PCB concentrations declined 2.7 fold from 22730ng/g lw (95% C.I.: 12470-32990ng/g lw) in 1983-1986 to 8473ng/g lw (95% C.I.: 6369-9776ng/g lw) in 2006-2010. Similar but fewer statistically significant trends were found for adult females and adult males likely due to smaller sample size and years. Despite declines as a result of international regulations, relatively high levels of these historic pollutants persist in East Greenland polar bear tissues.
