RESUMO

Crinum x powellii 'Album' belongs to the Amaryllidaceae medicinal plant family that produces a range of structurally diverse alkaloids with potential therapeutic properties. The optimal conditions for in vitro tissue growth, morphogenesis, and alkaloid biosynthesis remain unclear. Auxin and light play critical roles in regulating plant growth, development, and alkaloid biosynthesis in several Amaryllidaceae plants. Here, we have succeeded in showing, for the first time, that the combination of auxin and light significantly influence C. x powellii "Album" in vitro tissue growth, survival, and morphogenesis compared to individual treatments. Furthermore, this combination also upregulates the expression of alkaloid biosynthetic genes and led to an increase in the content of certain alkaloids, suggesting a positive impact on the defense and therapeutic potential of the calli. Our findings provide insights into the regulation of genes involved in alkaloid biosynthesis in C. x powellii "Album" callus and underline the potential of auxin and light as tools for enhancing their production in plants. This study provides a foundation for further exploration of C. x powellii "Album" calli as a sustainable source of bioactive alkaloids for pharmaceutical and agricultural applications. Furthermore, this study paves the way to the discovery of the biosynthetic pathway of specialized metabolites from C. x powellii "Album", such as cherylline and lycorine.

Three new flavanols, (2R,3S)-7-methoxy-flavan-3-ol (1: ), (2R,3S)-7-hydroxy-flavan-3-ol (2: ), and (2R,3S)-2'-hydroxy-7-methoxy-flavan-3-ol (3: ), together with two known flavans (4: and 5: ), were isolated from the chloroform extract of Crinum asiaticum. Their structures were elucidated by various spectroscopic methods, including 1D and 2D NMR, HR-ESI-MS, and CD data. The isolated compounds 1: and 3: -5: showed inhibitory activity toward LPS-induced nitric oxide (NO) production. Further investigation of the NF-κB pathway mechanisms indicated that 1: and 3: -5: inhibited the LPS-induced IL-6 production and p65 subunit phosphorylation of NF-κB in RAW264.7 cells, with an effective dose of 10 µM.

Toxicity of lead in soil is well documented and established. Phytoremediation has gained attention as a cheap, easily applicable, and eco-friendly clean-up technology. Chemical methods are used to assess exact levels and type of pollutants but heavy metal content in soil can also be evaluated indirectly by estimation of phytotoxicity levels using bioassays. Plant bioassays through fast germinating cereals can indicate not only the level of pollution and its effects on growth and survival but also the progress of phytoremediation process. The performance of barley Hordeum vulgare L. seedlings as bioassay for assessment of changes in the levels of lead (Pb) at three concentrations, i.e., 300 (T(1)), 600 (T(2)), and 1,200 ppm (T(3)) in the soil was evaluated while testing the efficiency of Crinum asiaticum L. as a phytoremedial tool. At the first assessment, i.e., 30 DAT (days after treatment) shoot and root lengths of seedlings decreased with increasing concentrations of Pb. As the study progressed, a decrease in levels of Pb was accompanied by better germinability and growth of barley. At 120 DAT seedling growth in all the treatments were comparable to control. In T(1), T(2), and T(3) soils, 74.5%, 83.7%, and 91.2% reduction in lead content was observed at 120 DAT. Highly significant correlations between decreasing pollutant (Pb) content in the soil, seed germination, and seedling growth of barley H. vulgare were found. The differences in root and shoot length as well as overall growth pattern are indicative of the suitability of barley as a bio-monitoring tool.

The alkaloids cherylline (1), crinamidine (2), crinine (3), epibuphanisine (4), lycorine (5), powelline (6), undulatine (7), 1-epideacetylbowdensine (8), and 3-O-acetylhamayne (9) were identified in the in vitro propagated bulblets of Crinum moorei. In addition, crinine, powelline, and undulatine were detected in the solidified Murashige and Skoog (MS) medium. The identity of the alkaloids was confirmed by comparing retention times and mass spectra with known samples. Light, as well as benzyladenine (BA) and charcoal supplementation of the tissue culture medium, influenced the levels of specific alkaloids in both the bulblets and media.

RESUMO

The distribution and seasonal variation of alkaloids from Crinum macowanii were investigated. The alkaloids lycorine, 1-O-acetyllycorine, crinine, powelline, crinamine, crinamidine, 3-O-acetylhamayne, 1-epideacetylbowdensine and cherylline were isolated from this plant using gas chromatography. Significant organ-to-organ variations were detected for the alkaloids crinine, lycorine, powelline, crinamidine, 3-O-acetylhamayne and crinamine. Crinine, crinamidine and 1-epideacetylbowdensine showed significant seasonal variation.
