RESUMO
The Ziwuling black goat is an indigenously in China, their offspring are frequently affected by congenital cryptorchidism. The extracellular matrix (ECM) contains cytokines and growth factors that regulate the development of the testis, and component changes often result in pathological changes. Cryptorchidism is closely related to structural changes in ECM. In this study, the histochemical staining, immunohistochemical, immunofluorescence and Western blot combined with semi-quantitative analysis was used to describe the distribution of the important ECM components Collagen type IV (Col IV), laminin (LN)and heparan sulfate proteoglycans (HSPG) in the normal and cryptorchid testes of Ziwuling black goats. Results showed that: The histochemical staining showed that the dysplasia of seminiferous tubules and decreased number of Sertoli cells in cryptorchidism, as well as sparse collagen fiber. Meanwhile, the distribution of reticular fibers is relatively rich. Furthermore, the PAS and AB staining in the interstitial vessels and lamina propria of seminiferous tubules is weak. The immunohistochemical and immunofluorescence revealed that Col IV, LN was strongly expressed in Leydig, Sertoli cells of normal testes and moderately positive in the spermatogonia and spermatids, but HSPG was not expressed in the spermatogonia. However, cryptorchidism, the expression of Col IV, LN and HPSG in Leydig, Sertoli cells significantly decreased, as well as the expression of Col IV and LN in capillary endothelial cells, but HSPG was moderately expressed in spermatogonia. Based on these data, the underdevelopment of spermatogenic epithelium, decreased synthesis function of collagen fibers and Leydig cells develop usually in the cryptorchidism were shown to be closely related to the abnormal metabolism of Col IV and LN. The positive expressed of HSPG in the spermatogonia of cryptorchid testes is related to the compensatory development of spermatogonia.(AU)

In this study, we aimed to evaluate the effect of concentrate supplementation containing crude glycerin (CG) before and during the breeding season on the physiological and blood biochemical variables as well as on the reproductive performance of Santa Inês ewes. A total of 24 ewes (12 pluriparous and 12 nulliparous ewes, 4 and 1.5 years old, weighing an average of 40 ± 5 and 27 ± 3 kg, respectively) were randomly assigned to three treatments consisting of different dietary concentrations of CG (0%, 5%, and 10 % of the total dry matter). The experiment lasted 63 days and was divided into three 21-day phases. In the first and second phases, the animals were subjected to flushing. In the second and third phases, we evaluated the animals' reproductive performance. Weather, physiological, and blood biochemical variables were also studied. The results indicated that the inclusion of CG did not influence significantly (P > 0.05) either the ewes' body weight or their body condition score. All ewes exhibited estrus during the breeding season. Their respiratory rate was significantly influenced by the time of day (P < 0.0001). A 10% CG supplementation did not drastically change the hematological and biochemical variables, which were within the reference ranges. However, the week of supplementation influenced sérum metabolites (P < 0.05). Thus, these results indicate that CG could be used in up to 10% in replacement of ground corn without significantly affecting the physiological, hematological, and biochemical parameters of the ewes or their reproductive performance before and during the breeding season.(AU)
Todas as ovelhas exibiram estro durante a estação reprodutiva.

Cryopreservation of epididymal sperm is a useful tool for preserving the genetic potential of valuable animal specimens. The domestic cat is used as a model to study and develop cryogenics for other felines. However, regulation of the entire cryopreservation process is essential for the success of this biotechnology. Thus, our aim was to evaluate the effects of glycerol equilibration time and freezethaw stages on the quality of epididymal sperm obtained from domestic cats. Epididymal sperm were recovered with TRIS and immediately evaluated for total motility (TM), vigor, viability, membrane functionality (HOST), and morphology. Then, TRIS-20% egg yolk was added to the samples, which were equally divided into two 1.5 mL tubes and refrigerated at 4 ºC for 1 hour. Subsequently, glycerol was added at a final concentration of 5%. The samples were incubated with glycerol (equilibration time) for either 5 or 10 minutes (groups G5 and G10, respectively) and then frozen. Thawing occurred at 37 ºC for 30 seconds. The samples were evaluated at all stages. A reduction in TM was observed only after thawing; however, it was higher in G5 (39.00 ± 4.07%) than in G10 (18.50 ± 4.54%). Vigor declined in both groups after thawing; however, they did not differ from each other. Sperm viability was maintained in G5 after glycerolization (53.60 ± 2.59%); in G10, sperm viability decreased in the glycerolized sample (48.80 ± 2.93%) when compared to that in the fresh sample (59.90 ± 1.74%). Post-thaw viability of G5 (33.80 ± 1.89%) was higher than that of G10 (18.80 ± 3.01%). In the HOST, a decrease in viability was only observed after thawing, with no difference between the groups (41.50 ± 2.84% for G5 and 40.20 ± 3.49% for G10). With regard to sperm morphology, normal sperm decreased while sperm with post-thaw secondary defects increased in both groups. In conclusion, a shorter equilibration time for glycerolization preserves epididymal sperm quality better and the freeze-thaw process is the most critical stage of thawing.(AU)

Artificial insemination with cooled semen is the most common practice in rabbit farms and any improvement on it helps to increase the efficiency and productivity of rabbit meat farms. Therefore, the aim of this study was to assess whether different cryoprotectant agents (CPA) as glycerol, N, N-Dimethylformamide (DMF) and N-Methyl-2-Pyrrolidone (NMP) can improve cooled rabbit sperm quality stored at 4ºC and 16ºC. Sperm samples were diluted with INRA 96® (Extender A), INRA 96® with 6% glycerol (Extender B) or 6% DMF (Extender C) or 6% NMP (Extender D) respectively and stored at 4ºC and 16ºC. Samples were then analysed at 4, 24, 48 and 72 hours after refrigeration by integrated sperm analysis system (ISAS®), eosin-nigrosin stain (vitality), hypo-osmotic swelling test (HOS test) and acrosome integrity test. Extender C showed higher percentage of motility, vitality and HOS test than extender B and D (p<0.05). Whereas sperm quality decreased over time (p<0.05), data showed that the addition of DMF kept the motility and sperm plasma membrane integrity after 24 hours of storage better than other diluents. These results suggest that the addition of DMF to INRA 96® exerts a protective effect on the membrane of spermatozoa improving seminal quality.(AU)

This experiment was conducted to evaluate the effect of crude glycerin at differing inclusion levels on performance and carcass characteristics of broilers and litter moisture. 960 broilers of 21-42 days of age were used in a completely randomized design with six crude glycerin inclusion levels (0, 3, 6, 9, 12, 15%), with eight replicates containing 20 broiler chickens per experimental unit. There was a quadratic effect (p0.05) on carcass yield, breast, or liver percentages. However, there was an increase (6.17%, p<0.05) in leg yield and a decrease in (p<0,05, 7.05%) the drumstick yield. For the wing yield and abdominal fat, a quadratic effect (p<0.05) was observed. The protein and fat deposition rates, as well as the dry matter content of the carcass, showed a quadratic effect (p<0.05) due to the inclusion levels of crude glycerin. Crude glycerin may be used in broiler diets from 21 to 42 days, at up to 6% without harming the performance of the broilers, the yield and carcass quality, litter moisture, and the viability of broilers. It is recommended that crude glycerin can be fed at 5.63% from 21 to 42 days, while for lower feed conversion it is recommended that the level of 3.72% is used.

Acute kidney injury is mostly reversible, and hepatocyte growth factor (HGF) has a relevant role in the tissue repair. MicroRNA (miR)-26a is an endogenous modulator of HGF. The role of miR-26a in the kidney repair process was evaluated in Wistar rats submitted to an acute kidney injury model of rhabdomyolysis induced by glycerol (6 mL/kg). Animals were evaluated 3, 12, 48, 96, and 120 hours after glycerol injection. Serum creatinine (SCr) and gene expression of HGF, c-met, signal transducer and activator of transcription 3 (STAT3), and miR-26a were estimated. Also, tubular NK52E cells were transfected with anti-miR26a and stimulated with Fe3+ for 24 hours to mimic the effects of myoglobin in vitro. SCr was highest after 48 hours. After 96 hours, SCr started to decrease, characterizing the recovery phase, with normalization after 120 hours. HGF expression increased during the onset phase (3 hours), with a low relationship with miR-26a. In contrast, in the recovery phase, the increase in miR-26a was coincident with HGF messenger RNA suppression, suggesting that in the recovery phase, miR-26a may have a role in HGF modulation. Fe3+ induced cellular death after 3 hours and proliferation after 24 hours. There was no correlation between miR-26a and STAT3 during the death phase; however, during the proliferation phase, an increase in STAT3 was paralleled with a decrease in miR-26a. miR-26a silencing induced increases in cell viability and the phosphorylated form of STAT3 protein expression in cells receiving Fe3+ . In conclusion, miR-26a may have a key role in modulating HGF levels after its proliferative effects have been triggered.

Glycerin is a by-product from the biodiesel chain whose energy value can be beneficial to pig diets. In this study, eight barrows were distributed individually into metabolic cages in a completely randomized block design. The animals were fed diets containing 0, 5, 10 and 15% glycerin from soybean oil and subjected to the methods of total feces and urine collection and chromic oxide marker. We determined the digestibility coefficients (DC); the dietary levels of digestible dry matter (DM), organic matter (OM), crude protein (CP), mineral matter (MM), ether extract (EE), an neutral detergent fiber (NDF); digestible energy (DE); metabolizable energy (ME); and these last two variables corrected for the nitrogen content (DEn and MEn). Digestible OM values increased linearly with glycerin inclusion in the diets. The digestible CP of the diets showed an inversely proportional relationship with glycerin inclusion levels. Digestible MM decreased linearly when determined by the total collection method and quadratically when determined by the marker method. There was a linear increase in the digestible and metabolizable coefficients of energy, DE, DEn, ME and MEn of the diets as the glycerin inclusion level was increased. Inclusion of up to 15% glycerin obtained from soybean oil production increased the digestible fractions of organic matter and energy of the diets.(AU)

The objective of this study was to evaluate the effect of the dietary addition of different levels of glycerin on the performance, litter moisture, pododermatitis incidence, and carcass and parts yield of broilers. In total, 1,610 broilers were reared in 35 pens with 46 birds each. A completely randomized experimental design, with five treatments with seven replicates was applied. The experimental treatments were: T1: control diet; T2: dietary inclusion of 5% glycerin from 1-42 days of age; T3: dietary inclusion of 10% glycerin from 1-42 days of age; T4: dietary inclusion of 5% glycerin from 7-42 days of age; T5: dietary inclusion of 10% glycerin from 7-42 days of age. The diets containing glycerin fed since the pre-starter period improved broiler weight gain and feed conversion ratio, but did not influence feed intake or livability. At the end of the experiment, the production efficiency index of the broilers fed 10% glycerin during the entire rearing period was significantly reduced compared with the other treatments. Litter moisture in the pens of broilers fed 10% glycerin during the entire rearing period was higher compared to the other treatments since day 21.Diets containing 10% glycerin, both for the entire rearing period (1-42 days) or only after the pre-starter phase (7-42 days), influenced broiler performance and incidence of severe pododermatitis, reducing the production efficiency indexes at 42 days. Glycerin may be added up to 5% in broiler´s diets with no effect on performance, litter moisture and carcass yield, indicating that this co-product of the biodiesel industry can be used as an alternative feedstuff for broilers.

This study was realized to evaluate the production of gas and volatile fatty acids when glycerin was used to replace four forage species through the use of a semi-automated technique in vitro. The experimental design included 4 treatments and 4 replications. The treatments consisted of increasing levels of glycerin (0, 30, 50 and 70%), which was used to replace the forage plants Elephant grass, sugar cane silage, corn silage and Brachiaria grass. Replacement of the Brachiaria and Elephant grasses with glycerin linearly reduced the production of methane and carbon dioxide and ammonia nitrogen and had a positive effect on forage quality (p 0.05). With the addition of glycerin, the values for acetate concentration decreased (p 0.05); however, when added glycerin there was increased in propionic and butyric acid concentration (p 0.05). Substitution of forage species for glycerin by tests for semi-automatic technique in vitro reduce the production of greenhouse gases (CH4 and CO2) which reduction in energy loss, and increase volatile fatty acids propionic and butyric.

Substitution of forage species for glycerin by tests for semi-automatic technique in vitro reduce the production of greenhouse gases (CH4 and CO2) which reduction in energy loss, and increase volatile fatty acids propionic and butyric.(AU)

This study evaluated the metabolic response of broilers fed diets containing increasing crude glycerine levels in two bioassays. Birds were house in metabolic cages, and were distributed according to a completely randomized experimental design with five treatments of 4 replicates each (1st assay: 5 birds/ cage; 2nd assay: 1-20 days = 8 birds/ cage, and 21-42 days = 4 birds/cage). Treatments consisted of a control diet based on corn and soybean meal, and four other diets containing 2.5%, 5.0%, 7.5% and 10.0% glycerine derived from biodiesel. In experiment I, there was no effect (p>0.05) of glycerol level on liver weight or blood parameters. Serum blood glycerol levels of the birds fed 10% crude glycerine increased during the first nine days of diet intake (p 0.05). In experiment II, water intake increased (p 0.05) in the birds fed 7.5 and 10.0% crude glycerine at 4 and 8 days of age. Feed intake increased (p 0.05) on days 8 and 12 in birds fed 2.5 and 7.5% glycerine. Fecal moisture increased (p 0.05) in birds fed diets with 5.0, 7.5, and 10.0% crude glycerine on days 16 and 20. Ileal content moisture was not different (p>0.05) among treatments when birds were 42 days old. High dietary glycerine levels may induce metabolic change in broilers, such as increased blood glycerol level, water intake and fecal moisture.(AU)

Pregnancy rates using stallions's frozen semen are variable; moreover, some breeds present low freezability. One hundred and four (104) mares were inseminated and separeted into two experiments to evaluate fertility of frozen semens of Criollo stallions (n=5), with 5% of dimethylformamide (DMF) or 5% of glycerol (GLY) as cryoprotectants. In Experiment I, the inseminations were made during pre-ovulation with fresh and frozen semen with DMF. In Experiment II, the mares were inseminated during post-ovulation with fresh semen, DMF and GL. The inseminations with frozen semen were performed deep in the uterine horn ipsilateralis to the dominant follicle. Control mares were inseminated with fresh semen in the uterus. Only one estrus period per mare was used to evaluate the pregnancy rates of the groups. Pregnancy diagnosis through ultrasonography was performed on the 15th day post-ovulation. The mean post-thawing motility was 40% and 20%, respectively, for frozen semen with DMF and GLY (P<0.05). All stallions had higher post-thawing motility when using DMF. In Experiment I, pregnancy rates were 12% (5/42) and 62% (20/32), respectively for DMF and fresh semen (P<0.0001). In Experiment II, pregnancy rates were 40% (4/10), 10% (1/10) and 70% (7/10), respectively for DMF, GLY (P<0.05) and fresh semen (P<0.0001). Insemination with frozen semen performed with more frequent follicular control showed the best results. The low sperm motility after thawing was attributed to the GLY used in experiment II. The DMF can be used alternatively to Criollos Stallions semen feezing.

The objective was to evaluate the effect of crude glycerin levels in supplement on carcass characteristics and live weight components of suckling lambs maintained on ryegrass pasture. Thirty two suckling lambs were used and were distributed among the treatments: 0, 10, 20 and 30% crude glycerin, in replacement of corn, in the isoproteic supplement (18% CP) provided daily in amounts equivalent to 2% of body weight. The levels of crude glycerin had negative linear effects on hot and cold carcass weight, hot and cold carcass yield but an increasing effect was observed on gastrointestinal content and fasting losses. The shoulder and rib weights decreased, while the leg proportion increased with the elevation of crude glycerin use. In the tissue composition of the shoulder, it was observed that the subcutaneous fat, intermuscular fat and total fat decreased linearly according to the levels of crude glycerin. Regarding body weight components, differences were observed in the proportions of skin, rumen, omasum, large intestine, which increased, while the kidney fat decreases linearly. The use of crude glycerin in up to 30% in the supplement provided in creep feeding for suckling lambs kept in ryegrass pasture influences carcass characteristics and corporal components.
