RESUMO

Helminths are a major challenge in dog breeding, particularly affecting young animals and posing a significant zoonotic risk. The widespread use of anthelmintics to treat gastrointestinal helminth infections in companion animals is common. However, these chemical products generate residues that can have adverse effects on animal, human and environmental health. In addition to the challenge of parasite resistance to treatment, there is an urgent need to explore and discuss complementary and sustainable methods of controlling helminthiases in these animals. In this context, nematophagous or helminthophagous fungi have emerged as a potential tool for the control of environmental forms of helminths. The purpose of this review is to emphasize the importance of these fungi in the control of free-living forms of helminth parasites in companion animals by highlighting the research that has been conducted for this purpose. In vitro experiments demonstrated the efficacy of fungi like Pochonia chlamydosporia, Arthrobotrys robusta, and Monacrosporium thaumasium in trapping and reducing helminth infective forms. These findings, along with soil contamination studies, suggest the feasibility of using helminthophagous fungi as a sustainable and effective strategy for environmental control. The current literature supports the potential of these fungi as an environmentally friendly solution for managing helminthiasis in dogs, benefiting both animal health and public welfare.

INTRODUCTION: The use of anthelmintic and antibiotic medicines is imperative to prevent the suffering of diseased stock in organic farming. However, their use must be minimised to comply with low input ideals and prevent the spread of resistance. Reducing such inputs first requires determining their current use, but information is lacking. The objective of this study was to benchmark the current use of anthelmintics and antibiotics in UK organic livestock farming. METHODS: Data were gathered by conducting a national survey of organic livestock farmers in the UK and by analysing records of requests for allopathic medicines. RESULTS: Key findings include (i) anthelmintics used in sheep constitute the greatest input of veterinary medicines in organic systems, (ii) farmers are incorporating alternative/support tools in helminth control to reduce anthelmintic requirements, (iii) the use of antibiotics is targeting individual animals, whereas the use of anthelmintics is targeting groups of animals. CONCLUSION: This study provides the first benchmark on the use of anthelmintics and antibiotics in UK organic livestock.

Infections with endoparasites are common in dogs. Some of these parasites are potentially zoonotic and therefore a public health concern. A survey was conducted in twenty-six small animal practices to evaluate the prevalence of endoparasites in Belgian and Dutch owned dogs older than 6 months as well as risk factors associated with infection. Out of 239 faecal samples screened (168 in Belgium and 71 in the Netherlands), 18 dogs were tested positive for at least one type of endoparasite with three dogs co-infected with two parasitic species. Toxocara sp. was the most frequently found endoparasite (4.6%). Three other dogs were positive for Angiostrongylus vasorum (1.4%) using the Baermann method and confirmed in one dog by the Angiodetect® test. Age and predation behaviour were identified as two risk factors associated with endoparasite infection. Although the majority (77%) of the owners in this study reported to administer at least one anthelmintic treatment per year, only a minority of them (24.3%) were aware of the risk to human health, indicating that owner awareness is sub-optimal. For dog owners, human toxocarosis and other potential zoonoses remain an underestimated health concern. The implementation of sustainable parasite control strategies should be promoted taking also into account the public health risk.

OBJECTIVE: To investigate the distribution of Oncomelania snails in Four-Lake Basin of Hubei Province, so as to provide scientific evidence for the development of snail control interventions. METHODS: The snail distribution was surveyed by means of the retrospective study and cross-sectional survey in the Four-Lake Basin in Spring of 2016. The data pertaining to the endemic situation of schistosomiasis were collected and analyzed in the Four-Lake Basin from 2008 to 2018, and the associations of snail habitat areas with the prevalence of Schistosoma japonicum in humans and bovines were examined. RESULTS: There were 16 003 snail habitats in the 9 schistosomiasis-endemic counties (districts) in the Four-Lake Basin in 2016, covering an area of 246 million m2. The mean density of living snails was 0.622 5 snails/0.1 m2, which was relatively high in weed-type settings (0.688 0 snails/0.1 m2) and ditches (0.706 4 snails/0.1 m2). Type I (current snail habitats, 53.03%) and Ⅱ- Ⅳ (historical snail habitats, 46.97%) settings were predominant snail habitats. A big change was seen in snail habitats in dry farmland, marshland and paddy farmland, and environmental modification resulted in satisfactory snail control effectiveness. In addition, the area of snail habitats positively correlated with the changes in the prevalence of S. japonicum infections in humans (r = 0.842, P < 0.01) and livestock (r = 0.695, P < 0.01). CONCLUSIONS: The integrate strategy with an emphasis on the control of the source of S. japonicum infections results in remarkable snail control effectiveness in the Four-Lake Basin; however, there is a risk of re-emerging snail habitats.

The present paper reviewed publications on the nematocidal activity of edible mushrooms (EM) and their potential use as sustainable tools for the control of parasitic nematodes affecting agriculture and livestock industry. Nematodes are organisms living in the soil and animals' guts where they may live as parasites severely affecting economically important crops and farm animals, thus causing economic losses to worldwide agriculture. Traditionally, parasitic nematodes have been controlled using commercial pesticides and anthelmintic (AH) drugs. Over the years, nematodes developed resistance to the AH drugs, reducing the usefulness of many commercial drugs. Also, the use of pesticides/anthelmintic drugs to control nematodes can have important negative impacts on the environment. Different EM have been not only used as food but also studied as alternative methods for controlling several diseases including parasitic nematodes. The present paper reviewed publications from the last decades about the nematocidal activity of EM and assessed their potential use as sustainable tools for the control of nematodes affecting agriculture and livestock industry. A reduced number of reports on the effect of EM against nematodes were found, and an even smaller number of reports regarding the potential AH activity of chemical compounds isolated from EM products were found. However, those studies have produced promising results that certainly deserve further investigation. It is concluded that EM, their fractions and extracts, and some compounds contained in them may have biotechnological application for the control of animal and plant parasitic nematodes.

Recent advances in systems biology have shifted vaccine development from a largely trial-and-error approach to an approach that promote rational design through the search for immune signatures and predictive correlates of protection. These advances will doubtlessly accelerate the development of a vaccine for schistosomiasis, a neglected tropical disease that currently affects over 250 million people. For over 15 years and with contributions of over 120 people, we have endeavored to test and optimize Sm-p80-based vaccines in the non-human primate model of schistosomiasis. Using RNA-sequencing on eight different Sm-p80-based vaccine strategies, we sought to elucidate immune signatures correlated with experimental protective efficacy. Furthermore, we aimed to explore the role of antibodies through in vivo passive transfer of IgG obtained from immunized baboons and in vitro killing of schistosomula using Sm-p80-specific antibodies. We report that passive transfer of IgG from Sm-p80-immunized baboons led to significant worm burden reduction, egg reduction in liver, and reduced egg hatching percentages from tissues in mice compared to controls. In addition, we observed that sera from Sm-p80-immunized baboons were able to kill a significant percent of schistosomula and that this effect was complement-dependent. While we did not find a universal signature of immunity, the large datasets generated by this study will serve as a substantial resource for further efforts to develop vaccine or therapeutics for schistosomiasis.

As genomic research continues to improve our understanding of the genetics of anthelmintic drug resistance, the revolution in DNA sequencing technologies will provide increasing opportunities for large-scale surveillance for the emergence of drug resistance. In most countries, parasite control in cattle and bison has mainly depended on pour-on macrocyclic lactone formulations resulting in widespread ivermectin resistance. Consequently, there is an increased interest in using benzimidazole drugs which have been used comparatively little in cattle and bison in recent years. This situation, together with our understanding of benzimidazole resistance genetics, provides a practical opportunity to use deep-amplicon sequencing to assess the risk of drug resistance emergence. In this paper, we use deep-amplicon sequencing to scan for those mutations in the isotype-1 ß-tubulin gene previously associated with benzimidazole resistance in many trichostrongylid nematode species. We found that several of these mutations occur at low frequency in many cattle and bison parasite populations in North America, suggesting increased use of benzimidazole drugs in cattle has the potential to result in widespread emergence of resistance in multiple parasite species. This work illustrates a post-genomic approach to large-scale surveillance of early emergence of anthelmintic resistance in the field.

Thirty-two Friesian cattle under a leaders/followers four-day rotation and passing eggs of trematodes and gastrointestinal nematodes (GIN) were studied in two trials for the integrated control of these helminths over two years. In the first trial, the effect of rotational pasturing was assessed on a group of leaders (milking cows, G-L1) and followers (dried-off cows and heifers, G-F1) supplemented daily with commercial nutritional pellets. In the second trial, leaders (G-L2) and followers (G-F2) were maintained under a rotational pasturing regime; the cows received daily commercial pelleted feed and heifers pellets manufactured with a blend of parasiticide fungi (3 × 105 chlamydospores of both Mucor circinelloides and Duddingtonia flagrans/kg pellet). Deworming via closantel and albendazole was performed in cows in each trial at the beginning of their drying periods, and fourteen days later, the fecal egg-count reductions (FECR) of Calicophoron daubneyi and GIN were from 94 to 100% (average 98 %), while the percentages of reduction of cattle shedding eggs (CPCR) were from 50 to 100% (average 77 % and 82 %, respectively). The heifers were dewormed one time only, at the beginning of each trial, and the values of FECR and CPCR were 100 % against C. daubneyi and 96 % and 83 %, respectively, against GIN. Over a period of 24 months, significantly higher numbers of helminth egg-output were observed in G-L1, with the lowest numbers in G-F2. C. daubneyi egg output was reduced by 5 % (G-L1) and 42 % (G-F1) at the end of trial 1 and by 83 % (G-L2) and 100 % (G-F2) at the end of trial 2; the numbers of GIN egg-output decreased by 13 % (G-L1) and 18 % (G-F1) at the end of trial 1, and by 72 % (G-L2) and 85 % (G-F2) at the end of trial 2. No adverse effects were detected in cattle taking pellets enriched with fungal spores (G-F2). It is concluded that long-term ingestion of spores of M. circinelloides and D. flagrans provides a valuable tool to improve the effect of rotational grazing and to lessen the risk of infection by C. daubneyi and GIN in dairy cattle, and accordingly, the performance of integrated control programs.

Interval treatment control programmes used widely in equine helminth control have favoured the development of anthelmintic resistance worldwide. Best practice guidelines have been designed to address resistance and include the requirement for improved pasture hygiene to break helminth transmission cycles, along with anthelmintic application informed by the results of diagnostic tests to reduce selection pressure for resistance. Using an online questionnaire, this study examined uptake of measures recommended in these guidelines by UK horse owners. The survey comprised 58 questions spanning grazing management, anthelmintic use and use of faecal egg count (FEC) testing to inform treatment decisions. Analysis was carried out using a combination of Chi-square and Mann-Whitney tests. In total, 705 owners responded and, following specific exclusion criteria, the responses of 652 individuals were analysed. The majority of the respondents owned <20 horses on private premises or livery yards in England. The main outputs of the survey were as follows. Overall, 60.9% of respondents used FEC tests to inform the requirement to administer anthelmintics, with macrocyclic lactones the most frequently-used anthelmintics. Of the respondents, 38% obtained advice on anthelmintic choice from their veterinarians; however, many respondents (43.8%) purchased anthelmintics via the internet. Encouragingly, 74.4% of respondents stated that they practiced good pasture hygiene by removing dung from pasture. Generally, there were differences between the responses of participants who based anthelmintic treatments on FEC testing (targeted treatments; TT) and those who practiced calendar-based anthelmintic treatments (interval treatments; IT). Briefly, the "key" findings from the Chi-square analysis included higher levels of satisfaction with the level of knowledge about equine parasites/parasitic diseases and higher levels of concern about anthelmintic resistance from TT-respondents compared to IT-participants. Confusion on the interpretation of quarantine recommendations was identified in this study group and there was poor uptake of testing for anthelmintic effectiveness. Overall, compared to previous reports, this study indicated improved engagement of UK horse owners with some helminth control practices recommended to reduce the spread of anthelmintic resistance. However, a proportion of respondents did not utilise these practices and there were still important gaps in the use of appropriate quarantine and efficacy testing. These identified gaps must be taken into consideration in knowledge dissemination activities in the future.

Two experiments were performed to determine whether oral administration of copper oxide capsules controlled helminthic infections in Lacaune sheep without acute collateral effects on animal health. In experiment 1, 48 multiparous lactating sheep (60.1 ±â€¯8.5 kg) were stratified according to initial number of eggs (Haemonchus contortus) per gram of feces (EPG) and were assigned randomly to 1 of two treatments (24 sheep/treatment): no oral administration (control) or oral administration of two copper capsules (treated; approximately 58 mg copper/kg body weight). Blood and fecal samples were collected on days 0, 15 and 45. Animals treated with copper capsules showed lower of EPG, eosinophils, acetylcholinesterase (AChE) in whole blood, and lower butyrylcholinesterase (BChE) activity in serum. Treated sheep had higher erythrocyte numbers, hemoglobin concentrations, hematocrit, and lymphocyte numbers. In experiment 2, 12 male lambs negative for helminths and coccidia were assigned randomly to one of two treatments (six lambs/treatment): control or treated (one copper capsule; approximately 58 mg copper/kg body weight); the experiment was designed to determine whether the results of experiment 1 were due to treatment or parasitism. Blood samples were collected on days 0, 5, 10 and 15 and fecal samples were collected on days 0, 7 and 15. Treated animals showed greater concentrations of lymphocytes; however, treatment had no effect on other hemogram variables, AChE and BChE activities, or levels of alanine aminotransferase, aspartate aminotransferase, gamma-glutamyl transferase, creatinine, urea, albumin, total protein, and reactive oxygen species. These data suggest that copper capsules in dairy sheep efficiently controlled H. contortus infections. Treatment was not harmful to lambs during the first 15 days, i.e. there were no signs of acute toxicity.

BACKGROUND: Endoparasites are considered a major health problem of South American camelids as shown in a recent survey among German and Austrian camelid owners. Although prophylactic and therapeutic measures such as application of anthelmintics are commonly used, treatment efficacy is usually not assessed. Owners have expressed significant concerns regarding the effect of antiparasitic therapy, so this study aimed to evaluate the outcome of anthelmintic treatment in German alpaca herds with different drugs. RESULTS: Overall, 617 samples from 538 clinically healthy alpacas > 1 year-old from 27 farms (n = 11-157 animals/herd) were examined. The most common parasites detected by flotation were Eimeria spp. (75.1%) followed by strongylids (55.0%), Nematodirus spp. (19.3%), cestodes (3.1%) and Trichuris (2.7%). After initial coproscopical examination by flotation and strongylid egg quantification by the McMaster technique, positive animals excreting at least 150 eggs per gram of faeces were included in a faecal egg count reduction test (FECRT) using fenbendazole (n = 71 samples), moxidectin (n = 71) or monepantel (n = 66). Pre-treatment larval cultures (n = 23 positive pooled farm samples) revealed Haemonchus (87% of the farms), Cooperia (43.5%), Trichostrongylus (21.7%), Ostertagia (13.0%), Nematodirus and Oesophagostomum (4.3% each). Fenbendazole treatment reduced egg excretion by 45%, moxidectin by 91% and monepantel by 96%. On the farm level, 13/18 farms that used fenbendazole, 6/6 farms that used moxidectin and 2/5 farms that used monepantel had individual FECR values < 90% (fenbendazole) or < 95% (moxidectin, monepantel). Haemonchus and Cooperia were overrepresented on the farms with reduced treatment efficacy. CONCLUSIONS: Gastrointestinal strongylids are common in German alpacas and fenbendazole in particular was not sufficiently effective to reduce strongylid egg excretion. Although the FECRT could not unambiguously determine anthelmintic resistance in the present study, the finding that small ruminant strongylids, especially Haemonchus, are common in alpacas indicates that determination of effective anthelmintic doses, monitoring of efficacy and adapted (selective) treatment regimens must be implemented as part of sustainable deworming practices in this species in accordance with recommendations for ruminants.

Knowledge regarding helminth control strategies and nematode infection of horses in Ireland is limited and only one study has been published recently. This present study was designed to investigate the current helminth control strategies followed by horse owners in Ireland. A questionnaire was formulated to collect data on general grazing, pasture management and deworming strategies including the use of fecal egg counts. Questionnaires were emailed to 700 members of the Irish Thoroughbred Breeders Association and Horse Sport Ireland. Only 78 questionnaires were returned. Respondents indicated that horses are grazed for 16-24 h day-1 during the summer and autumn (89% and 65%, respectively). Removing feces from the pasture was implemented by 37.6% of respondents. Few (22.2%) owners kept horses off pasture after worming. Overall, ivermectin and moxidectin were the most commonly administered anthelmintics in 2014 by 75% and 62% of respondents, respectively. Benzimidazole and pyrantel drugs were used by 53% and 35% of respondents, respectively. The majority of farms (81.4%) treated horses 4-5 times per year and 74.2% only estimated the weight of the horses visually. The findings of this study illustrates that many stud managers/owners do not follow best practice with regard to helminth control and more education is needed.

The anthelmintic activity of an injectable eprinomectin formulation (Eprecis® 20 mg/mL) was evaluated in 150 naturally infected dairy sheep raised in 3 semi-intensive flocks. All ewes were at the same stage of lactation and grazed on natural pastures. Ewes did not receive any anthelmintic treatment for at least 4 months prior to the experiment. In each flock, 50 ewes were selected and randomly allocated to control (C) or treatment (T) groups (n = 25 per group). Groups were balanced according to the ewes' bodyweight (BW) and fecal egg count (FEC) measured seven days before eprinomectin administration (day-7). On study day 0, ewes in group T, received 0.2 mg/kg BW of eprinomectin subcutaneously (Eprecis® 20 mg/mL, Ceva). Ewes in group C were left untreated. Fecal samples were collected on day 0, 7, 14, 21 and 28 post-treatment to assess FEC and for coprocultures. Ewes were weighed on day 0 and 28. Overall and within-flock efficacy of eprinomectin was calculated throughout the experimental period. No local or general adverse reaction after injection was observed. The most prevalent parasite genera were Teladorsagia, Haemonchus and Trichostrongylus. Following treatment, the overall mean FEC of C and T groups differed significantly (P < 0.001). Overall and within-flock efficacy of eprinomectin was 99.8%-100.0% and 99.7%-100.0%, respectively. Contrary to C group, ewes treated with injectable eprinomectin increased their BW during the study (-0.5 kg vs. + 1.5 kg, P < 0.001). In this field study, a single subcutaneous injection of eprinomectin to dairy sheep, at 0.2 mg/kg BW, resulted in excellent curative anthelmintic activity; egg counts remain low for at least 28 days after treatment.

This study aimed to assess current worm control practices used by Australian alpaca farmers with an online questionnaire survey. The questionnaire contained questions about farm demography and general husbandry practices, farmers' knowledge about gastrointestinal nematodes (GINs) and their importance, the use of worm control strategies and anthelmintics, and grazing management. A link for the questionnaire survey was sent to all (n = 954) registered members of the Australian Alpaca Association in July 2015. The response rate for the questionnaire was 25% (239/954). The majority of respondents were from small (≤ 50 alpacas; 64%, 153/239) followed by medium (50-100 alpacas; 24%, 57/239) and large (>100 alpacas; 12%, 29/239) farms. Findings revealed that the majority of respondents kept Huacaya alpacas to produce high-quality fibre and alpacas were usually kept with other domestic ruminants (e.g. cattle and sheep). Although half of alpaca farmers (114/220) perceived that GINs were an important health problem of alpacas, with Haemonchus spp. being the most common nematode, the majority of them (174/220) used anthelmintics for nematode control. Macrocyclic lactones, a commercial combination of four anthelmintics (abamectin, albendazole, closantel and levamisole) and monepantel were the three most commonly used dewormers by Australian alpaca farmers. Although a significant proportion (166/213) of respondents used a quarantine drench for alpacas, very few respondents were aware of strategic deworming and the issue of anthelmintic resistance. Alpaca farmers mostly used anthelmintics at the dose rate recommended for sheep (47%, 79/167) and cattle (9%, 15/167), though some used 1.5 (31%, 51/167) and 2 (13%, 22/167) times the dose rate recommended for sheep. The majority of small herds used anthelmintics at the dose rate recommended for sheep and cattle while medium and large herds used anthelmintics at 1.5 to 2 times the dose rate recommended for sheep. This study provides invaluable insights into the demography of alpaca farms in Australia, husbandry practices used by alpaca farmers and their knowledge about worms and their control, thereby paving the way for developing guidelines for the control of GINs of alpacas.

AIMS: To investigate the production responses and cost-benefit of administering a controlled-release anthelmintic capsule (CRC) to pregnant yearling ewes prior to lambing. METHODS: Yearling ewes from two commercial sheep flocks (A, n=489; B, n=248) in the North Island of New Zealand were enrolled in the study. Prior to lambing, CRC containing albendazole and abamectin were administered to half the ewes while the other half remained untreated. Ewe liveweights and body condition scores were measured prior to lambing, at weaning and, for Flock B, prior to subsequent mating. Lambs were matched to dams shortly after birth and the weight and number of lamb weaned per ewe were determined. A cost-benefit analysis was undertaken for Flock B considering the increased weight of lamb weaned per ewe, and the weight of ewes at the next mating and the benefit in terms of lambs born. RESULTS: The mean weight at weaning of treated ewes was greater for treated than untreated ewes by 2.76 (95% CI 0.64-4.88) kg in Flock A (p<0.001) and 2.35 (95% CI -0.41-5.12) kg in Flock B (p=0.003); the weight of lamb weaned per ewe was greater for treated than untreated ewes by 1.43 (95% CI -0.71 to -3.49) kg in Flock A (p=0.041) and 3.97 (95% CI 1.59-6.37) kg in Flock B (p<0.001), and ewe liveweight prior to subsequent mating was greater for treated than untreated ewes in Flock B by 4.60 (95% CI 3.6-5.6) kg (p<0.001). There was no difference in the percentage of lambs reared to weaning between treated and untreated ewes in either flock (p>0.8). The overall cost-benefit of treatment for Flock B was NZ$9.44 per treated ewe. CONCLUSIONS AND CLINICAL RELEVANCE: Pre-lambing CRC administration to yearling ewes resulted in increased ewe weaning weights and weight of lamb weaned in both the flocks studied. There was an economic benefit in the one flock where this was assessed.

The usefulness of a strategy involving the biological control of soil-transmitted helminths (STHs) with zoonotic potential is tested. A blend of spores of two filamentous fungi, Mucor circinelloides and Duddingtonia flagrans, was provided to one group (G-2) of puppies (five Griffon bleu de Gascogne and five Gascon Saintongeois) shedding eggs of Toxocara canis, Toxascaris leonina, Trichuris vulpis, and Ancylostoma caninum. Other identical group (G-1) remained without spores. Fecal samples were collected daily and divided into two portions, one sample was immediately analyzed by the flotation test and the other was examined 28 days later. Viability of eggs of T. canis, T. leonina, and T. vulpis in fecal pats was ascertained by microscopic examination; the numbers of larvae of A. caninum were assessed in each group. In the feces of G-2, spores of both fungi were observed 1 day after beginning the assay. Viability of eggs of T. canis was reduced by 57%, T. leonina by 61% and T. vulpis by 53% in the fecal pats of G-2, and by 87%, 92%, and 85%, respectively, in G-1. The numbers of third-stage larvae of A. caninum were 59% lower in G-2 than that in G-1. By considering the results obtained and the absence of adverse effects in the puppies of G-2, it is concluded that the presence of the filamentous fungi in the feces has a high potential to avoid the transmission of these parasites. Further studies are in progress to determine the appropriate way of administration to pets.

Most animal research is less evidence-based for females, with the majority of studies conducted on males. Since immune responses vary between males and females, sexual dimorphism in immunity contributes, among other things, to sex-based differences post-vaccination. However, the issue of sex effects in animal vaccine research is rarely considered in vaccine study design. Previously, we have evaluated the efficacy of cathepsin L3 (FhCL3-1 and FhCL3-2) and B3 proteases (FhCB3) from juvenile Fasciola hepatica as vaccines against fasciolosis in male rats. Their administration resulted in reductions in liver fluke recovery in the range of 47-63% when compared with an infection control group. Here, we investigated if the protective effect of vaccination with these proteins can also be observed for female rats. The data indicates females were not protected from F. hepatica infection when vaccinated with juvenile cathepsins. Only in the FhCL3-2 vaccinated group was a low, non-significant, reduction in worm burden observed (21%). Although liver fluke mean body lengths and wet weights were reduced in vaccinated animals when compared with the infection controls, these effects were adjuvant- not vaccine-induced, while for males changes in these parameters were related primarily to vaccination. Specific humoral responses throughout the study were evident; however, trends in antibody responses in females replicated trends observed previously for male humoral responses. Formerly, elevated levels of FhCL3-1 and FhCL3-2 specific IgG1 and IgG2a were suggested to be correlated with protection. Here, despite increased and clear responses of these antibodies, protection was not observed. Hence, in the present study the roles of IgG1 and IgG2 in liver fluke reduction are questionable. Results demonstrated in our study show that observations obtained in one sex are not always applicable to the other sex. Hopefully, the findings of the study will stimulate discussion of the issue of sex impacts on post-vaccination outcomes and will encourage researchers to consider sex in their future vaccine studies.

Helminths are common pathogens of ruminants. These are controlled by anthelmintics; however, resistance is a major issue. More sustainable practices need to be deployed to protect anthelmintic efficacy. There is no published information on how farmer/prescriber interactions at anthelmintic purchase shape application of best practice on-farm. To determine experiences of UK ruminant farmers as they relate to anthelmintic purchasing/provision of best practice advice at the point-of-sale, an online survey was distributed. This explored farmer experiences in purchasing anthelmintics from the three UK animal medicines' prescribers (veterinarians, Suitably Qualified Persons [SQPs], veterinary pharmacists) and investigated farmer attitudes to anthelmintic use and resistance. First, farmers were grouped according to whom they purchased anthelmintics from, including those that purchased from > 1 prescriber type. The results indicated there was no significant difference between the type of prescriber from whom farmers bought anthelmintics and route (face-to-face, telephone, online) through which they purchased them. Farmers who bought from veterinarians were more likely to view prescriber knowledge as more important. When asked who influenced purchase, there was a significant difference; farmers tended to be most influenced by the prescriber they purchased from. Those who bought from various types of prescriber were most influenced by veterinarians. Farmers who purchased from veterinarians were significantly more likely to state their seller discussed/recommended faecal egg count (FEC) testing and management strategies to reduce anthelmintic use than those who purchased from SQPs or from > 1 prescribing channel. When grouped according to purchase route, farmers that bought through face-to-face interactions were significantly more likely to state they valued prescriber knowledge of parasites/anthelmintics than farmers that purchased via the telephone or internet. Farmers that purchased online were significantly less likely to consider prescriber advice. Analysis was then applied based on the type of livestock farmed. Generally, sheep farmers undertook FEC testing more than cattle farmers. Relatively few farmers stated they conducted anthelmintic sensitivity testing, with the majority of cattle farmers having not tested at all. This was despite a high level of concern for anthelmintic resistance in all types of farmers. 'Cattle only' farmers viewed convenience factors as more important than those that managed sheep or sheep/cattle. The results suggest that UK farmers that bought anthelmintics from veterinarians were more likely to be exposed to diagnostic-led control advice. Gaps in advice provision, particularly in relation to efficacy testing, were identified in all farmer groups.

Anthelmintic treatments to ewes around lambing time, often with long acting products, have become common practice on UK sheep farms, but these treatments have also been shown to be highly selective for anthelmintic resistance in New Zealand and Australia, with field data supported by modelling results. The purpose of this study was to determine (1) the effect of treating or withholding anthelmintic treatments and (2) the effect of treatment of ewes with a persistent or non-persistent anthelmintic, on early infection in lambs in the UK. Faecal egg count data for 10-16 weeks old lambs collected over a three year period (2012-2014) was analysed. Samples were grouped according to whether the ewes on the farm had or had not been treated with an anthelmintic at lambing. For both analyses, data for early infection were analysed by generalised linear mixed model. There was no effect of withholding or treating ewes on subsequent early infection in lambs. In addition, there was no effect of region, farm type or management type on the faecal egg counts. There was, however an effect of year, with lambs having lower counts in 2014 than in 2012 and an interaction between year and ewe treatment, with data suggesting lower infection levels over time for those farms withholding anthelmintic treatments altogether. There was no effect of drug type on early infection in lambs nor region, farm type or treatment on the lamb faecal egg counts. However, there was an effect of year and an interaction between year and drug type with lower egg count over time with the short acting drugs. Our study supports data generated by other researchers suggesting that the practice of treating ewes at lambing to reduce contamination on pasture and minimise subsequent disease may not in fact always result in lower levels of infection in lambs. The study also demonstrated no significant benefit in early infection in lambs when ewes were treated with long acting compared to short acting anthelmintics. This provides further evidence to support the potential benefits of a more targeted approach to anthelmintic treatment on sheep farms.
