The influence of pharmaceutical residues and heavy metals on living organisms has received global attention. The present study assessed the interactive effect of antibiotic residues and heavy metals in soil, as contaminated food with cadmium (Cd) and oxytetracycline (OTC) on the isopod Porcellio leavis. It was fed on fresh plant leaves contaminated with different concentrations of cadmium, Cd+OTC1000 ppm, Cd+OTC2000 ppm and Cd+OTC3000 ppm for 4 weeks. The changes in the feeding patterns, protein, lipid peroxidation (LPO), catalase activity (CAT), and total free amino acids (TFAA) were recorded. There were significant differences in the obtained results where Cd reduced the egestion ratio (ER) however, OTC enhanced this ratio. Biochemical analysis illustrated that combination between OTC and Cd inhibits the toxic effects of Cd at low concentration (1000 ppm), while at high concentration (3000 ppm) raise the toxicity. Detailed studies are required for further understanding of the interaction between OTC and heavy metals, and also its impact on soil animals and for improving soil risk evaluation.


A influência de resíduos farmacêuticos e metais pesados em organismos vivos tem recebido atenção global. O presente estudo avaliou o efeito de resíduos de antibióticos e metais pesados no solo, como alimentos contaminados com cádmio (Cd) e oxitetraciclina (OTC), sobre o isópode Porcellio laevis, o qual foi alimentado com folhas frescas de plantas contaminadas com diferentes concentrações de cádmio, Cd + OTC1000 ppm, Cd + OTC2000 ppm e Cd + OTC3000 ppm, por quatro semanas. As mudanças nos padrões de alimentação, proteína, peroxidação lipídica (LPO), atividade da catalase (CAT) e aminoácidos livres totais (TFAA) foram registradas. Houve diferenças significativas nos resultados obtidos, em que o Cd reduziu a taxa de excreção (ER), no entanto o OTC aumentou essa proporção. A análise bioquímica mostrou que a combinação entre OTC e Cd inibe os efeitos tóxicos do Cd em baixa concentração (1.000 ppm), enquanto, em alta concentração (3.000 ppm), aumenta a toxicidade. Estudos detalhados são necessários para uma maior compreensão da interação entre OTC e metais pesados, e seu impacto sobre os animais do solo, bem como para melhorar a avaliação de risco do solo.

Aegerolysin proteins ostreolysin A6 (OlyA6), pleurotolysin A2 (PlyA2) and erylysin A (EryA) produced by the mushroom genus Pleurotus bind strongly to an invertebrate-specific membrane sphingolipid, and together with a protein partner pleurotolysin B (PlyB), form transmembrane pore complexes. This pore formation is the basis for the selective insecticidal activity of aegerolysin/PlyB complexes against two economically important coleopteran pests: the Colorado potato beetle and the western corn rootworm. In this study, we evaluated the toxicities of these aegerolysin/PlyB complexes using feeding tests with two ecologically important non-target arthropod species: the woodlouse and the honey bee. The mammalian toxicity of the EryA/PlyB complex was also evaluated after intravenous administration to mice. None of the aegerolysin/PlyB complexes were toxic against woodlice, but OlyA6/PlyB and PlyA2/PlyB were toxic to honeybees, with 48 h mean lethal concentrations (LC50) of 0.22 and 0.39 mg/mL, respectively, in their food. EryA/PlyB was also tested intravenously in mice up to 3 mg/kg body mass, without showing toxicity. With no toxicity seen for EryA/PlyB for environmentally beneficial arthropods and mammals at the tested concentrations, these EryA/PlyB complexes are of particular interest for development of new bioinsecticides for control of selected coleopteran pests.

This study was designed to assess the impact of the mixture of cadmium (Cd) and zinc (Zn) on the bioaccumulation and the ultrastructural changes in the hepatopancreas of Porcellio laevis (Latreille, 1804) after 4 weeks of exposure to contaminated Quercus leaves under laboratory conditions. For each metal, four concentrations were used with four replicates for each concentration. Metal concentrations in the hepatopancreas and the rest of the body were determined using atomic absorption spectrometry. From the first week until the end of the experiment, a weight gain in P. laevis was observed particularly between the first and the end of exposure from 93.3 ± 18.22 mg fw to 105.22 ± 16.16 mg fw and from 106.4 ± 22.67 mg fw to 125.9 ± 23.9 mg fw for Mix1 and Mix4, respectively. Additionally, the determined metal trace elements (MTE) concentrations in the hepatopancreas were considerably higher compared to those in the rest of the body and seem to be dose-dependent. Using transmission electron microscopy (TEM), some alterations were highlighted in the hepatopancreas. The main observed alterations were (a) the destruction of the microvilli border in a considerable portion of cells, (b) the increase of the lipid droplets with different shapes and sizes, (c) the increase in the number of the mitochondria, and (d) the appearance of TE in the form of B-type granules. The obtained results confirmed the ability of P. laevis to deal with high amounts of MTE, suggesting its possible use in future soil's biomonitoring programs.

This study was conducted to compare metals bioaccumulation in the terrestrial isopod Armadillidium granulatum collected from Ghar El Melh lagoon. We focused on recognizing the effects of trace elements on hepatopancreas functional role. To this end, isopod specimens were exposed for 3 weeks to sediments contaminated with cadmium, copper, zinc, mercury, and nickel. Three concentrations were used in duplicate for each experimental condition. At the end of the experiment, metal body burdens were determined using flame atomic absorption spectrometry. Results of the bioaccumulation factor (BAF) showed that the species A. granulatum was classified as a Cu macroconcentrator (BAF > 2) and a Zn deconcentrator (BAF < 2). Dose dependent morphological and histological changes were observed in the hepatopancreas cells using transmission electron microscopy. The predominant features were: microvillus border disruption, condensation of the cytoplasm with increasing endoplasmic reticulum, mitochondria, lysosomes and granules that accumulated metals in B and S cells. The number of lipid droplets decreased especially after Cd, Zn, Hg, and Ni treatments. This study demonstrated that the terrestrial isopod A. gramulatum could be a good indicator of soil metal contamination.

Anthropogenic substances pollute freshwater systems worldwide, with serious, long-lasting effects to aquatic biota. Present methods of detecting elevated levels of trace metal pollutants are typically accurate but expensive, and therefore not suitable for applications requiring high spatial resolution. Additionally, these methods are not efficient solutions for the determination of long-term averages of pollution concentration. This is the rationale for the implementation of a biomonitoring programme as an alternative means of pollutant detection. This review summarises recent literature concerning the past and potential uses of the benthic isopod Asellus aquaticus as a biomonitor for pollution in freshwater systems. Recent studies indicate that A. aquaticus is well suited for this purpose. However, the mechanisms by which it bioaccumulates toxins have yet to be fully understood. In particular, the interactions between coexisting trace metal pollutants in the aquatic environment have only recently been considered, and it remains unclear how a biomonitoring programme should adapt to the effects of these interactions. It is evident that failing to account for these additional stressors will result in an ineffective biomonitoring programme; for this reason, a comprehensive understanding of the bioaccumulation mechanisms is required in order to reliably anticipate the effects of any interferences on the outcome.

Isopods of the genus Ligia are deemed as excellent pollution monitoring organisms for coastal zones. A species of Ligia cinerascens was chosen to be tested, which was collected from three coastal zones in Dalian, China. The concentrations of metals followed the order of Cu>As ≈ Cd>Pb. Ligia collected from a municipal sewage outlet in Heishijiao showed the highest level of metals. Ligia from a natural bathing beach in Changshan Island showed the lowest level of metals as expected. Ligia grown in the same site could accumulate different levels of metals due to their different body weight. Both males and females conformably accumulated heavy metals in bodies. Taken together, this work suggests that L. cinerascens can serve as a biomonitor species. Within the range of 0.3-0.9 g body weights, body weight rather than gender should be taken into account as a factor in the process of environmental biomonitoring.

The leaf-shredding crustacean Hyalella azteca, which is indigenous to Northern and Central America, is used to assess environmental risks associated with (metal-)contaminated sediments and to propose sediment quality standards also in Europe. Yet, it is unknown if H. azteca is protective for European crustacean shredders. We thus compared the sensitivity of H. azteca with that of the European species Asellus aquaticus and Gammarus fossarum towards copper- and cadmium-contaminated sediments (prepared according to OECD 218) under laboratory conditions employing mortality and leaf consumption as endpoints. H. azteca either reacted approximately fourfold more sensitive than the most tolerant tested species (as for cadmium) or its sensitivity was only 1.6 times lower than the highest sensitivity determined (as for copper), which should be covered by safety factors applied during risk assessments. Therefore, the results for the sediment type and the two heavy metals tested during the present study in combination with the existence of standardized testing protocols, their ease of culture, and short generation time, suggest H. azteca as suitable crustacean model shredder for assessing the toxicity of sediment-associated metals in Europe.

The present work aims to study the effects that acute exposure to low concentrations of silver nanoparticles (AgNPs) cause in digestive glands of terrestrial isopods (Porcellio scaber). The experiments were designed to integrate different analytical techniques, such as transmission electron microscopy, atomic absorption spectroscopy, proton induced X-ray emission, and Fourier transform IR imaging (FTIRI), in order to gain a comprehensive insight into the process from the AgNPs' synthesis to their interaction with biological tissues in vivo. To this aim, terrestrial isopods were fed with AgNPs having different shapes, sizes, and concentrations. For all the tested conditions, no toxicity at the whole organism level was observed after 14 days of exposure. However, FTIRI showed that AgNPs caused detectable local changes in proteins, lipids, nucleic acids and carbohydrates at the tissue level, to an extent dependent on the interplay of the AgNPs' properties: shape, size, concentration and dissolution of ions from them.

The bioaccumulation of and resistance to different heavy metals in soil was tested using Mongoloniscus sinensis, a terrestrial isopod endemic to china, and results show that: (1) the median lethal concentration (LC50) of Pb, Zn, Cd in the filter paper contact test after 48 h was 197.6, 503.7, 448.0 µg cm- 2, LC50 of Pb-Zn compounds was 173.8 and 440.8 µg cm- 2 and after 14 days of soil contamination LC50 was 2917.0, 2977.9, 5048.4 mg kg- 1, LC50 of Pb-Zn compounds was 1219 and 1463 mg kg- 1. Thus Zn turned out to be less toxic than Cd in the filter paper contact test, while their sequence of toxicity was reversed during the soil exposure test, which shows that M. sinensis can tolerate a dose of Zn and Cd. (2) analysis of body burdens showed that the sequence of internalized metal concentrations was Pb < Cd < Zn, which indicates that heavy metals in soil can be enriched and absorbed by M. sinensis, and that it is much more efficient at absorbing Zn and Cd than Pb.

Animals adjust behaviors to balance changes in predation risk against other vital needs. Animals must therefore collect sensory information and use a complex risk-assessment process that estimates risks and weighs costs and benefits entailed in different reactions. Studying this cognitive process is challenging, especially in nature, because it requires inferring sensory abilities and conscious decisions from behavioral reactions. Our goal was to address this empirical challenge by implementing psychophysical principles to field research that explores considerations used by desert isopods (Hemilepistus reaumuri) to assess the risk of scorpions that hunt exclusively from within their burrows. We introduced various combinations of chemical and physical cues to the vicinity of isopod burrows and recorded their detailed reactions on first encountering the cues. The isopods reacted defensively to scorpion odor but only when accompanied with excavated soil or other odors typically found near scorpion burrows. Isopods also reacted defensively to piles of excavated soil without scorpion olfactory cues, suggesting that isopods take precautions even against physical disturbances that do not necessarily reflect predator activity. Simultaneous presence of different cues provoked graded responses, possibly reflecting an additive increase in risk estimation. We conclude that wild isopods use defensive reactions toward environmental signals only when the integrated perceptual information implies an active scorpion burrow or when they lack data to refute this possibility.

This study aimed to observe the type of asymmetry exhibited by Porcellio laevis sampled from 15 sites belonging to Tunisian industrialized areas. Physicochemical parameters such as pH, organic matter and CaCO3 contents were measured in soils. Moreover, Cd, Pb, Zn, and Cu concentrations were determined in both soils and woodlice. Additionally, 10 metrical traits were measured to evaluate the type of asymmetry on individuals: the basis, the second and the third articles of the antenna, the first article of the flagellum of the antenna and the merus, the carpus, and the propodus of the sixth and the seventh pereopods. Among the 531 measured individuals, 432 exhibited fluctuating asymmetry (FA) while the remaining individuals exhibited antisymmetry or directional asymmetry. The data obtained were analyzed using a multivariate statistical analysis. Contrary to our hypothesis, the results showed that individuals from contaminated sites have a low FA level, whereas those from uncontaminated sites have a high FA level, particularly females but with some exceptions. Variations in FA level in the traits and populations studied and its usefulness as a stress indicator were discussed.

Isopods are macrodecomposers in terrestrial ecosystems, contributing to soil organic matter breakdown and nutrient cycling. They have been considered sensitive in laboratory tests designed to evaluate contaminants effects and are considered likely candidates to have a standardized protocol for ecotoxicity tests. For this purpose, a reference substance should be proposed as positive control in laboratory tests. This work aimed to evaluate the avoidance behaviour of the isopod species Porcellio dilatatus to boric acid (H3BO3; BA). Interlaboratory comparison tests were carried out based on the ISO guideline for earthworms, using tropical artificial soil as substrate, in the concentrations of 0, 125, 250, 500, 750, 1000, 1300 and 1800 mg BA kg-1 soil. Avoidance behaviour was evaluated in group (six organisms per replicate) and in individual tests (one organism per replicate), in dual-section plastic boxes, which received control soil (not contaminated) in one side and tested soil (with BA) in another one. The percentage of organisms in each side was recorded after 48 h, and data were analysed using Fisher exact test (p < 0.05) and t-test. Results showed significant avoidance response in individual tests at 250 mg kg-1 and in group tests at 500 mg kg-1. Limited habitat function (< 20% of organisms) for both tests and laboratories were observed at the highest tested concentration of 1800 mg kg-1, indicating the low sensitivity of this species to BA in avoidance tests, if compared to other substances in literature. Results showed that BA can be used as reference substance until a better option will be proposed.

Biotic stressors have been demonstrated to change the toxicity of pollutants. While the combined effects of predator cues and pesticides are well documented, the interaction of predator stress with metals is a topic that has remained largely unexplored. In this laboratory experiment, the freshwater isopod Asellus aquaticus is exposed to predator cues and metal mixtures of Cd, Cu and Pb. We examined the effects on growth, respiration and, as behavioral parameters, feeding rate and activity. These were linked to the free ion activities (FIAs) in the water and the metal body concentrations. The findings reveal that Cu accumulation significantly influenced the growth rate, the feeding rate and the activity of isopods exposed to predator stress. Furthermore, we found a concentration-dependent interaction of the Cd + Pb mixtures on the feeding rate and a lower feeding rate for Cd and Pb predator exposed asellids. As several interactions were found between metals and predator stress, it demonstrates the importance of investigating how organisms and whole ecosystems respond to multiple stressors. A better understanding of these interactions will undoubtedly improve risk assessment and management.

Despite glyphosate-based herbicides are widely used in agriculture, forestry and gardens, little is known about its effects on non-target organisms. The present work evaluated the ecotoxicity of four formulated products (Roundup® Original, Trop®, Zapp® Qi 620 and Crucial®) on soil invertebrates. Screening ecotoxicity tests were carried out with soil and oat straw collected in a field experiment, besides laboratory-spiked soils. Screening tests included avoidance behaviour of earthworms (Eisenia andrei), collembolans (Folsomia candida) and isopods (Porcellio dilatatus) in single and multispecies tests; reproduction of collembolans (F. candida), and bait lamina in field. Non-avoidance behaviour was observed in standard tests (earthworms) in soil, neither in multispecies tests (earthworm + isopods) using oat straw, while for collembolans it occurred for the product Zapp® Qi 620 even at the recommended dose. Reproduction of F. candida was not impaired even at high doses in laboratory-spiked soils. Feeding activity on bait lamina test was impaired in treatment corresponding to the red label product, Crucial®. Results showed the relevance of bait lamina test on screening the impact of herbicides in the field. The findings highlight the importance of considering different formulations for the same active ingredient in risk assessment of pesticides.

As the production and usage of nanomaterials are increasing so are the concerns related to the release of the material into nature. Tungsten carbide (WC) is widely used for its hard metal properties, although its use, in for instance tyre studs, may result in nano-sized particles ending up in nature. Here, we evaluate the potential long-term exposure effects of WC nanoparticles on a pelagic (Daphnia magna) and a benthic (Asellus aquaticus) organism. No long-term effects were observed in the benthic system with respect to population dynamics or ecosystem services. However, long-term exposure of D. magna resulted in increased time to first reproduction and, if the particles were resuspended, strong effects on survival and reproductive output. Hence, the considerable differences in acute vs. long-term exposure studies revealed here emphasize the need for more long-term studies if we are to understand the effects of nanoparticles in natural systems.

RESUMO

The exposure to pesticides by nontarget soil biota has long been regarded as a serious downside of modern agricultural regimes and the subject of heated debate. Of utmost relevance is the exposure to pesticide mixtures because their effects have been shown to not necessarily reflect the individual toxicity of their components, and even the simple addition of effects may lead to consequences not clearly anticipated. In the present study, a multiple biomarker approach was employed to identify the mechanistic and time effects underlying several single and mixture treatments of chlorpyrifos (CPF) and mancozeb (MCZ) in juveniles and adults of the terrestrial isopod Porcellionides pruinosus. The effects of the individual pesticides and the mixtures at recommended doses were mostly transitory under these controlled conditions and one-pulse exposure. Whereas imbalances were identified on detoxification and oxidative stress-related enzymes, the isopods generally showed the ability to recover through the end of the experiment. However, juveniles displayed greater vulnerability than adults. Most of the differences between life stages occurred in energy-related parameters where distinct performances and stress-handling behaviors were observed, suggesting higher metabolic costs to juveniles. Our results stress that understanding the time dependence of the underlying mechanisms governing the joint effects of the pesticides can help in assessing and anticipating the effects of the pesticide mixtures. Moreover, we emphasize the importance of taking life stage-related differences into consideration when evaluating the environmental risks of pesticides and pesticide mixtures. Silver nanoparticles (AgNPs) are efficiently converted during the wastewater-treatment process into sparingly soluble Ag sulfides (Ag2 S). In several countries, sewage sludge is used as a fertilizer in agriculture. The bioavailability of sulfidized Ag to the terrestrial isopod Porcellio scaber was investigated. Sewage sludge containing transformed AgNPs was obtained from a laboratory-scale sewage-treatment plant operated according to Organisation for Economic Co-operation and Development (OECD) guideline 303a. The results of transmission electron microscopy with energy dispersive X-ray of sludge samples suggest that AgNPs were completely transformed to Ag2 S. Adult isopods were exposed to OECD 207 soil substrate amended with the AgNP spiked sludge for 14 d (uptake phase) followed by an elimination phase in unspiked soil of equal duration. Most of the Ag measured in P. scaber at the end of the uptake phase was found in the hindgut (71%), indicating that only a minor part of the estimated Ag content was actually assimilated by the isopods with 16.3 and 12.7% found in the carcass and hepatopancreas, respectively. As a result of this, the Ag content of the animals dropped following transition to unspiked sludge within 2 d to one-third of the previously measured Ag concentration and remained stable at this level until the end of the elimination period. The present study shows that Ag2 S in sewage sludge is bioavailable to the terrestrial isopod P. scaber. Environ Toxicol Chem 2018;37:1606-1613. © 2018 The Authors. Agricultural land-use frequently results in short pulse exposures of insecticides such as pyrethroids in river systems, adversely affecting local invertebrate communities. In order to assess insecticide-induced effects, stream mesocosms are used within higher tier aquatic risk assessment. Regulatory acceptable concentrations (RACs) derived from those studies are often higher compared with tier 1 RACs. Hence, the present mesocosm study evaluates this aspect using a pulse exposure scenario typical for streams and the pyrethroid insecticide etofenprox. A 6-h pulse exposure with measured concentrations of 0.04, 0.3 and 5.3µgL-1 etofenprox was used. We considered abundance, drift and emergence of invertebrates as structural endpoints and the in situ-measured feeding rates of the isopod Asellus aquaticus as functional endpoint. Most prominent effects were visible at 5.3µgL-1 etofenprox which caused adverse effects of up to 100% at the individual and population level, as well as community structure alterations. Transient effects were observed for invertebrate drift (effect duration ≤24h) and for the invertebrate community (9 days after exposure) at 0.3µgL-1 etofenprox. Furthermore, 0.04µgL-1 etofenprox affected the abundance of the mayfly Cloeon simile (decrease by 66%) and the feeding rate of A. aquaticus (decrease by 44%). Thus, implications for the functional endpoint leaf litter breakdown in heterotrophic ecosystems may be expected. A hypothetical RAC derived from the present mesocosm study (0.004µgL-1) is in line with the official tier 1 RAC (0.0044µgL-1) and thus shows that the present mesocosm study did not result in a higher RAC.

We investigated how different carbonized materials, or chars (olive mill pomace biochar (BC) and urban greens pruning residues hydrochar (HC)) affect an important member of soil fauna, the terrestrial isopod Porcellio scaber. The selection behavior of isopods towards chars after a 48 h exposure was studied in (i) soil-char amended tests with single and multiple choices, and (ii) tests with chars offered as pure material. Finally, we exposed the isopods to char-amended soils for a period of 14 days to follow the effect on food consumption, body mass and activities of enzymes that are commonly altered upon stressor exposure (acetylcholinesterase, AChE, and glutathione S-transferase, GST). We showed that isopods are able to select between char amended and un-amended soil and different forms of char amendments: a clear preference for BC, and avoidance of HC were evidenced. The preferences remained the same when the chars were sterilized leading to the conclusion that initial microorganism composition was not the reason for selection, but selection was governed by other chars' physico-chemical properties. It remains to be elucidated which of these properties were the dominant reason for the selection. We also showed that isopods intentionally use BC as food at a similar rate to alder leaves. Medium-term exposure to HC resulted in adverse effects on isopods because it led to reduced feeding and growth, in addition to increasing GST activity, although no alterations in AChE activity were found. We suggest that behavioral tests with P. scaber could be used as a fast, reliable and economically feasible screening method for determining the safety of chars for the soil environment. Results represent significant contribution in the field of char toxicity testing, highlighting the importance of tests with isopods as important members of soil meso fauna, with the aim of influencing environmental policies and quality standards.

Metal pollution is a serious threat to environmental health. While the aquatic isopod Asellus aquaticus L. (Isopoda) is an important decomposer of freshwater ecosystems, very little research has reported its long-term or incipient lethal concentrations for metals. Moreover, the lethal concentrations at a certain percentage (LCxs) that can be found in the literature are often based on unmeasured concentrations, which could lead to a severe underestimation of the actual toxicity. In the present exploratory study, the 1-, 4-, 7-, and 14-d LC10, LC20, and LC50 values of copper, cadmium, and lead for adult A. aquaticus were determined. The LCx values were calculated using the nominal concentrations, the effective concentrations, and the free ion activities. Incipient lethal values were determined as well. In general, surprisingly lower lethal concentrations were found than in other studies. Furthermore, the present study shows that lethal concentrations based on free ion activities were generally much lower than nominal and effective concentrations. Although almost all LC values were higher than the environmental quality standards (EQS), the Cu 14-d LC10 and several (incipient) Pb LC10 and LC20 values, all calculated with free ion activities, were lower than the EQS. We conclude that lethal or effective concentrations based on free ion activities provide a more realistic view, which is vital for environmental policy making.
