RESUMO

A selective and sensitive analytical method was developed for enantiomeric separation and determination of N-methyl-DL-aspartic acid (NMA). The method involved the conversion of each enantiomer into N-ethoxycarbonylated (S)-(+)-2-octyl ester derivative for the direct separation by gas chromatography-mass spectrometry (GC-MS). The diastereomeric derivatives showed characteristic mass spectral properties for analysis by selected ion monitoring mode (SIM) and enabling enantioseparation on an achiral capillary column. Two enantiomers were baseline separated, and the detection limits for N-methyl-L-aspartic acid (NMLA) and N-methyl-D-aspartic acid (NMDA) were 0.07 and 0.03 ng/g, respectively. When applied to rat brain tissues for absolute configuration of NMA, only NMDA was determined, while NMLA was monitored as lower than the limit of detection.

Drinking water utilities in Europe are faced with a growing presence of organic micropollutants in their water sources. The aim of this research was to assess the robustness of a drinking water treatment plant equipped with reverse osmosis and subsequent activated carbon filtration for the removal of these pollutants. The total removal efficiency of 47 organic micropollutants was investigated. Results indicated that removal of most organic micropollutants was high for all membranes tested. Some selected micropollutants were less efficiently removed (e.g. the small and polar NDMA and glyphosate, and the more hydrophobic ethylbenzene and napthalene). Very high removal efficiencies for almost all organic micropollutants by the subsequent activated carbon, fed with the permeate stream of the RO element were observed except for the very small and polar NDMA and 1,4-dioxane. RO and subsequent activated carbon filtration are complementary and their combined application results in the removal of a large part of these emerging organic micropollutants. Based on these experiments it can be concluded that the robustness of a proposed treatment scheme for the drinking water treatment plant Engelse Werk is sufficiently guaranteed.

RESUMO

Gamma-aminobutyrate acid, L-glutamate, and N-methyl-D-aspartate were separated by capillary electrophoresis and detected by the use of whole-cell and outside-out patch-clamp techniques on freshly dissociated rat olfactory interneurons. These neuroactive compounds could be identified from their electrophoretic migration times, unitary channel conductances, and power spectra that yielded corner frequencies and mean single-channel conductances characteristic for each of the different agonist-receptor interactions. This technique has the sensitivity to observe the opening of a single ion channel for agonists separated by capillary electrophoresis.
