We investigated the morphology, phylogeny of the 18S rDNA, and pH response of Oxytricha acidotolerans sp. nov. and Urosomoida sp. (Ciliophora, Hypotricha) isolated from two chemically similar acid mining lakes (pH~2.6) located at Langau, Austria, and in Lusatia, Germany. Oxytricha acidotolerans sp. nov. from Langau has 18 frontal-ventral-transverse cirri but a very indistinct kinety 3 fragmentation so that the assignment to Oxytricha is uncertain. The somewhat smaller species from Lusatia has a highly variable cirral pattern and the dorsal kineties arranged in the Urosomoida pattern and is, therefore, preliminary designated as Urosomoida sp. The pH response was measured as ciliate growth rates in laboratory experiments at pH ranging from 2.5 to 7.0. Our hypothesis was that the shape of the pH reaction norm would not differ between these closely related (3% difference in their SSU rDNA) species. Results revealed a broad pH niche for O. acidotolerans, with growth rates peaking at moderately acidic conditions (pH 5.2). Cyst formation was positively and linearly related to pH. Urosomoida sp. was more sensitive to pH and did not survive at circumneutral pH. Accordingly, we reject our hypothesis that similar habitats would harbour ciliate species with virtually identical pH reaction norm.

BACKGROUND: Cytosine methylation of DNA is conserved across eukaryotes and plays important functional roles regulating gene expression during differentiation and development in animals, plants and fungi. Hydroxymethylation was recently identified as another epigenetic modification marking genes important for pluripotency in embryonic stem cells. RESULTS: Here we describe de novo cytosine methylation and hydroxymethylation in the ciliate Oxytricha trifallax. These DNA modifications occur only during nuclear development and programmed genome rearrangement. We detect methylcytosine and hydroxymethylcytosine directly by high-resolution nano-flow UPLC mass spectrometry, and indirectly by immunofluorescence, methyl-DNA immunoprecipitation and bisulfite sequencing. We describe these modifications in three classes of eliminated DNA: germline-limited transposons and satellite repeats, aberrant DNA rearrangements, and DNA from the parental genome undergoing degradation. Methylation and hydroxymethylation generally occur on the same sequence elements, modifying cytosines in all sequence contexts. We show that the DNA methyltransferase-inhibiting drugs azacitidine and decitabine induce demethylation of both somatic and germline sequence elements during genome rearrangements, with consequent elevated levels of germline-limited repetitive elements in exconjugant cells. CONCLUSIONS: These data strongly support a functional link between cytosine DNA methylation/hydroxymethylation and DNA elimination. We identify a motif strongly enriched in methylated/hydroxymethylated regions, and we propose that this motif recruits DNA modification machinery to specific chromosomes in the parental macronucleus. No recognizable methyltransferase enzyme has yet been described in O. trifallax, raising the possibility that it might employ a novel cytosine methylation machinery to mark DNA sequences for elimination during genome rearrangements.

Experiments were conducted to evaluate the toxic effects ofmonocrotophos in ciliate models Paramecium caudatum and Oxytricha fallax. In acute toxicity studies higherconcentrations of monocrotophos caused marked increase in mobility of cells exhibiting rocking movements within two mins of exposure but were decreased after 30 mins. LC50 value by mortality curve for 3 hr acute toxicity test of Oxytricha fallax and Paramecium caudatum was found 307.744 +/- 33.27 mg l(-1) and 332.284 +/- 57.52 mg l(-1) respectively. Oxytricha fallax was found sensitive than Paramecium caudatum to monocrotophos. In acute exposure cells showed deformities such as swelling, oval shaped deformity and in higher concentrations shortening of longitudinal axis with blackening of cytoplasm occurred. The length of paramecia was reduced prominently. Similarly enlargement of contractile vacuole and stress egestion of food vacuoles was also observed. The morphological studies showed the changes in shape, size, colour and width of Paramecia and Oxytricha. Frequencies of macronuclear aberrations were significant showing deformities such as rod shaped, elongation, fragmentation, diffusion and total absence of nucleus and were concentration dependent. The data provided in the present study on interaction of pesticides with nuclear structure can be of immense value because most of these pesticides have been reported to have carcinogenic, mutagenic and teratogenic properties.
