RESUMO

Silicone intraocular lenses are undergoing clinical investigation for use in the United States. We compared the ability of silicone intraocular lenses to activate the complement system in human sera with that of polymethylmethacrylate intraocular lenses with polypropylene loops using a radioimmunoassay that measures levels of activated complement fragments. Sera incubated with polymethylmethacrylate lenses with polypropylene loops had higher levels of C3a and C5a, but not C4a, than control sera incubated without intraocular lenses. On the other hand, there were no differences in levels of C3a, C4a, and C5a between sera incubated with silicone lenses and control sera. These results suggest that polymethylmethacrylate lenses with polypropylene loops activate the alternative pathway of complement, while silicone lenses do not.

RESUMO

Surgical stress animal models were established by performing laparotomies on mice. It was found that this type of stress could suppress the natural killer (NK) cytotoxicity and the proliferation of spleen cells induced by conA or lipopolysaccharide (LPS). Dynamic changes showed that the stress-mediated immunosuppression was reversible, as the responses to conA and LPS would be restored with time. The sensitivity to the stress-mediated suppression was different according to variations in immunological parameters. Furthermore, the macrophages in spleen were tested by isolation by the plastic-adherent procedure. The results showed clearly that these adherent cells (Plastic Adherent Cells, PAC) possessed an immunosuppressive effect on mitogen-induced lymphocyte proliferation in post-operative mice, but not in normal mice. Treatment of mice with indomethacin blocked the PAC-mediated immunosuppression. Surgical stress appeared to increase the level of prostaglandins, which in turn induced the production of suppressor PAC.

RESUMO

Metronidazole administered orally in doses of 20 and 200 mg/kg daily suppressed granuloma formation around Schistosoma mansoni eggs which were injected intravenously and lodged in the pulmonary microvasculature of mice. The same doses did not suppress granuloma formation in animals which had previously been sensitized to the eggs. Nonspecific granulomatous inflammation around divinyl benzene copolymer beads was unaffected by the drug. In a daily dose of 20 mg/kg, metronidazole inhibited delayed footpad reactions to soluble schistosome egg antigen, but 200 mg/kg on alternate days failed to suppress skin allograft rejection. The drug appears to suppress selectively some aspects of cell-mediated immunity.

RESUMO

Due to the increase in the frequency of allergic disorders it is necessary also to consider allergic reactions in the case of unspecific lesions in the oral region. Intolerance of drugs, resins, and metals can be demonstrated by elimination and exposure tests, but also by epicutaneous tests. Diagnostic measures and therapy vary with the type of allergy in each case.
