OBJECTIVES: To investigate adaptive strategies of Tupaia belangeri to environmental factors in different populations, 12 locations were selected, including higher and lower altitude areas. RESULTS: Total of 96 and 90 metabolites were annotated in serum and liver respectively, which were mainly concentrated in primary metabolites. Principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) showed that the locations were divided into two groups in serum metabolites, but each group had a few samples overlap. The samples of each group overlap to some degree in the liver metabolites. The tricarboxylic acid (TCA) cycle occupies a central position in metabolism. The concentrations of TCA intermediates, lipid metabolites and amino acid metabolites were higher in higher altitude areas, and the concentrations of carbohydrate and glycolysis intermediates were higher in lower altitude areas. CONCLUSIONS: Different areas adapted to the changes of environmental and altitude by regulating the concentration of metabolites in serum and liver, and revealed the adaptive mechanism of T. belangeri in different living environments.

Systematic classification and determination of various cells in normal peripheral blood of artificially feeding Tupaia belangeri chinensis of different ages and genders and evaluation of the effectiveness of an automatic blood cell classification counter for measuring tree shrew blood cells. Child, young and adult tree shrews (forty for each group) were randomly selected, half male and half female. After the animals were stable, the peripheral blood of each group was collected through the femoral vein, and the morphology of various blood cells of the tree shrew was observed and classified by the manual microscopic counting method and by an automatic blood cell classification counter. The Reference intervals of the normal peripheral blood cell absolute count, cell diameter and white blood cell percentage in tree shrews of different ages and genders has been calculated. White blood cell count and neutrophil relative count increased with age, while lymphocyte relative count decreased. The white blood cell count, neutrophil relative count, and lymphocyte relative count in the child group, as well as lymphocyte relative count in the young group, significantly differed according to gender (P<0.05), and the differences in other indicators were not significant. The Bland-Altman plot and the Passing-Bablok scattergram showed that the change trend of each indicator was consistent but exhibited large systematic differences between methods. Differences in peripheral blood cells exist among different age groups and different genders. An automatic blood cell classification counter is not suitable for the absolute count of blood cells in the tree shrew.

RESUMO

A novel beta-defensin 1-like antimicrobial peptide (ß-defensin 1TB) containing 36 amino acid residues was purified and characterized from the serum of the tree shrew, Tupaia belangeri. Its amino acid sequence was determined as DHYLCVKNEGICLYSSCPSYTKIEGTCYGGKAKCCK, by Edman degradation, mass spectrometry analysis, and cDNA cloning. Evolution analysis indicated that ß-defensin 1TB showed maximal similarity to the ß-defensin 1 identified from cotton-top tamarin, Saguinus oedipus. ß-defensin 1TB exerted potential antimicrobial activities against wide spectrum of microorganisms including Gram-negative and -positive bacteria and fungi. It showed little hemolitic activity to human or rabbit red cells. To the best of our knowledge, this is the first report of antimicrobial peptide from Tupaiidae.

The developmental life-history of tree-shrews conforms with the general primate pattern. Consequently, elucidation of the tree-shrew's neuroendocrine reproductive axis could shed light on the mechanisms that underlie human pubertal development. In the present study, we examined plasma gonadotropin concentrations in male tree-shrews from birth to sexual maturity, and related them to changes in the androgenic and gametogenic status of the testis. A hypogonadotropic infantile phase, during which a stable population of primordial cells is established, extended from birth to approximately Day 30. Following a short juvenile phase (Days 30-40), a pubertal phase of accelerated reproductive development was initiated between Days 40-55. At this time, FSH and LH levels rose and testosterone concentrations reached peak levels coincident with the descent of the testes, accelerated growth in the reproductive tract and the onset of spermatogenesis. To test whether this developmental peak in testosterone secretion is an important determinant in the normal onset of puberty, we exposed male tree-shrews prematurely to high circulating androgen levels for various periods and then examined the impact on key components of the developing reproductive axis. The testosterone implants failed to initiate spermatogenesis and the testes remained in an infantile state for the duration of the treatment, whereas implant removal led to the development of full spermatogenic activity. In both normal and experimental situations, low levels of FSH were associated with a lack of spermatogenic activity while the progression of germ cell development was precisely correlated with rising FSH levels. Taken together, these data establish a comprehensive picture of reproductive development in the male tree-shrew, and also provide support for the hypothesis that FSH plays a primary role in the initiation of spermatogenesis.
