[Antibody formation to o-aminoazotoluene via the administration of conjugated antigens synthesized by enzymatic and chemical methods]. / Obrazovanie antitel k o-aminoazotoluolu pri vvedenii kon''iugirovannykh antigenov, sintezirovannykh fermentativnymi i khimicheskimi metodami.

Two new methods are developed for synthesis of conjugated antigens of o-aminoazotoluene-bovine serum albumin (o-AAT--BSA): (1) from the microsomal fraction of the guinea pig liver which contains the cytochrome-P-450-dependent monooxygenase enzymic system and (2) from the m-chloroperbenzoic acid. A possible mechanism of the covalent binding of o-AAT with albumin under the effect of monooxigenases and of their chemical model is considered. Differences of antigenic determinants in conjugated antigens of o-AAT--BSA synthetized by the chemical and enzymic methods are detected.

Two methods are developed for synthesis of conjugated antigens to o-aminoazotoluene (o-AAT)-albumin: the enzymatic method--using horseradish peroxidase (HP) and the chemical one--diazomethod. A possible mechanism is suggested for the covalent binding of o-AAT to albumin which is catalyzed by HP. Antibodies to o-AAT are obtained by immunization of animals. An essential difference is established for antigenic o-AAT determinants in conjugates synthesized by the enzymatic and chemical methods.

[Dye and nickel allergy in the hairdressing salon]. / Farbstoff- und Nickelallergie im Friseurgewerbe.

In 73 hairdressers, out of a total number of 3,300 patients, patchtests were performed using substances of a standard series. Patients of various professions excluding hairdressers and patients belonging to an age group comparable with that of hairdressers, served as two control-groups. In the group of hairdressers the most frequently seen positive reactions were against p-phenylendiamin, nickelsulfat, pellidol, cobaltchlorid, p-aminodiphenylamin, and perubalsam. The incidence of positive reactions against p-phenylendiamin and pellidol in this group was significantly higher than in the control group, whereas the higher incidence of nickelsulfat allergy in hairdressers proved not to be significant.

Single injections of ortho-aminoazotoluene (OAT) or aminoazobenzene (AB) decrease the immune response of DD mice to the strain-nonspecific Krebs-2 tumor. The immunosuppressive action of AB is due to its toxicity for lymphoid cells, whereas the OAT effect is mediated by adrenal glands. After OAT injection,II-oxycorticosteroids level is elevated although their production by adrenal glands is not increased. The accumulation of glucocorticoids in blood after OAT injection is assumed to be the result of a decreased hormone utilization because of the blockade of receptor proteins or metabolizing enzymes in liver cells by a carcinogen.

The antigenic structure of the livers of mice and rats after a single hepatocarcinogenic treatment was investigated using the immunodiffuse and immunofluorescent methods. The next day after carcinogen application changes characteristic of hepatocellular tumors were observed: decrease in the synthesis of organospecific antigens and intensification of the synthesis of heteroorganic antigens.
